Abstract

L'invention concerne des procédés de préparation de matrices d'ADN pour le séquençage de produits de transcription unicellulaires d'ARN provenant d'une population de cellules. Les procédés comprennent la répartition des cellules provenant de ladite population dans des volumes de réaction séparés de sorte qu'une pluralité de volumes de réaction séparés contiennent chacun une seule cellule isolée, lesdites cellules ayant été traitées avec un agent fixateur avant la répartition. Les cellules isolées sont ensuite perméabilisées ou dissociées, et l'ADNc est préparé par une transcription inverse, suivie d'une amplification. L'invention concerne également une nouvelle chimie pour une production efficace de matrices d'ADN à partir de récepteurs de lymphocytes T ou d'immunoglobulines dans des monocellules.
