Verfahren zur Darstellung eines Acridinderivates. In dem Patent Nr. 94950 wurde ein Ver fahren zur Darstellung von<B>3.9</B> . 7-Diamino- äthoxy-acridin beschrieben, welches darauf beruht, dass man im 3 . 9 .7-Amino-halogen- äthoxy-acridin das Halogen gegen die Amino- gruppe umtauscht, indem man entweder di rekt mit Ammoniak erhitzt, oder zunächst das Halogen gegen eine Alkyloxy- oder Aryl- oxygruppe und diese dann weiter mittelst Ammoniak gegen die Aminogruppe tauscht.
Es wurde nun gefunden, dass als geeig netes Mittel zur Überführung der in 9-Stel- lung befindlichen negativen Substituenten in die Aminogrup.pe anstatt des Ammoniaks auch Derivate des Ammoniaks, wie A.nnmon- salze, Hydroxylamin und Acylderivate des Ammoniaks, wobei erforderlichenfalls der Säurerest nachfolgend abzuspalten ist, ver wendet werden können.
<I>Beispiel 1:</I> 34 Teile salzsaures 3 . 7. 9-Amino- < ithoxy- chloracridin werden in 170 Teilen Phenol gelöst und bei zirka<B>80'</B> 12,5 Teile Ammon- karbonat kristallisiert eingetragen. Unter Rühren wird bis 140' erhitzt, 10 Minuten bei dieser Temperatur gehalten, mit Aceton verdünnt und vom Reaktionsprodukt abge saugt.
Die wässerige Lösung desselben wird zwecks Abscheidung von Verunreinigungen ammoniakalisch gemacht und aus dem Fil trat das salzsaure 3.9.7-Diamino-äthoxy- acridin durch Ansäuren mit Salzsäure abge schieden.
Natronlauge fällt aus der wässe rigen Lösung des Salzes, das bei 124' unter Aufschäumen schmelzende gelbe 3.9.7.Di- amino-äthoxy -acridin. <I>Beispiel 2</I> 27 Teile 3.7.9-Amino-äthoxy-chloracri- din werden in 135 Teilen Phenol mit 12 Tei len Ammonchlorid oder Ammonsulfat 6 Stun den auf 130 bis 140 erhitzt und das salz saure 3.9.7-Diamino-äthoxy-acridin durch Zusatz von Aceton abgeschieden. Das Salz kristallisiert in gelben Nadeln.
<I>Beispiel 3</I> 28 Teile 3.7.9-Amino-diäthoxy-aeridin, erhalten aus 3. 7..9-Amino-äthoxy-chloracri.- den und Natriumäthylat, werden mit 7 Tei len Harnstoff und 150 Teilen Phenol oder Kresol langsam bis zum Sieden erwärmt und einige Zeit auf dieser Temperatur gehalten. Das Phenol wird im Vakuum abdestilliert. Der Rückstand wird in warmer verdünnter Salzsäure aufgenommen, filtriert und aus der Lösung durch Zusatz von Kochsalz das salzsaure 3.9.7-Diamino-äthoxy-aeridin ab geschieden.
Beispiel 1 Teil 3. 7.9-Amino-äthoxy-chloracridin wird mit 5 Teilen Harnstoff rasch auf 180 bis 200' erhitzt. Die Reaktionsmasse wird mit verdünnter Natronlauge ausgekocht und der Rückstand aus verdünntem Alkohol um kristallisiert. Man erhält das 3.9.7-Di- amino-äthoxy-aeridin in gelben Nädelchen vom S .
P .124 . ' <I>Beispiel 5</I> 2,7 Teile 3. 7. 9-Amino-äthoxy-chloracri- din werden mit 0,7 Teilen Acetamid in 15 Teilen Phenol langsam zum Sieden erhitzt und kurze Zeit im Kochen erhalten. Beien Verdünnen der Schmelze mit Aceton fällt das Hydrochlorid des 3.7. 9-Amino-äthoxy- aeetamino-aeridins aus; beim Erwärmen mit 50 loiger Schwefelsäure entsteht aus diesem <B>3.9</B> . 7-Diamino-äthoxy-acridin.
<I>Beispiel ö:</I> <B>2,7</B> \feile 3 . 7 . 9-Amino-äthoxy-chloracri- din werden mit 2,3 Teilen Hydroxylamin- chlorhydrat und 2,7 Teilen wasserfreiem Natriumacetat mit 15 Teilen Phenol langsam zum Sieden erhitzt. Es tritt lebhafte Gas entwicklung ein. Die Reaktionsmasse wird mit verdünnter Natronlauge erwärmt. Das ausgeschiedene 3.9.7-Diamino-äthoxy-acri- din wird über sein salzsaures Salz gereinigt.
<I>Beispiel 7:</I> 1 Teil 3-Amino-7-äthoxy-9-phenoxy-acri- din, erhältlich aus 3.7.9-Aminoäthoxy-chlor- acridin und Natriumphenolat, wird mit 0,5 Teilen Ammonehlorid in 5 Teilen p-Kresol 3,5 Stunden zum Sieden erhitzt.
Mittelst Ace- ton und Salzsäure wird dann ein gelbes Salz abgeschieden, dieses in Wasser gelöst und nach der Beseitigung von Verunreinigungen durch Ammoniakzugabe, durch Ansäuern mit Salz säure das salzsaure 3.9.7-Diamino-ä.tlioxy- acridin zur Abscheidung gebracht.
<I>Beispiel 8</I> 2,7 Teile 3. 7.9-Amino-äthoxy-chloracri- din werden mit 1,3 Teilen Ammoncarbonat in 10 Teilen Amyl- oder Äthylalkohol im Rohr 2 Stunden auf 180 erhitzt. Das ausgeschie dene Reaktionsprodukt wird abgesaugt, mit Aceton gewaschen, dann in Wasser gelöst, ammoniakalisch gemacht und aus dem Fil trat mittelst Natronlauge das 3.9.7-Di.- amino-äthoxy-aeridin abgeschieden.
Beispiel <I>9:</I> 2,7 Teile 3. 7 . 9-Amino-äthoxy-chloracri- din werden in 13,5 Teilen Phenol gelöst, 2,7 Teile Dicyandiamid zugegeben, zum Sie den erhitzt und so eine Stunde gehalten. Die Schmelze wird dann mit Aceton verdünnt, von der dabei erhaltenen Abseheidung ab gesaugt, diese in Wasser gelöst, ammoniaka- lisch gemacht und aus dem Filtrat mittelst Salzsäure 3.9.7 -Diamino-äthoxy-acridin- hy drochlorid abgeschieden.
Process for the preparation of an acridine derivative. In the patent no. 94950 a method for the representation of <B> 3.9 </B>. 7-diamino-ethoxy-acridine described, which is based on the fact that in the 3rd 9 .7-Amino-haloethoxy-acridine exchanges the halogen for the amino group by heating either directly with ammonia, or first the halogen for an alkyloxy or aryloxy group and this then by means of ammonia for the amino group exchanges.
It has now been found that, instead of ammonia, derivatives of ammonia such as ammonia salts, hydroxylamine and acyl derivatives of ammonia can also be used as a suitable means for converting the negative substituents in the 9-position into the amino group, where necessary the acid residue is subsequently to be split off can be used.

Sodium hydroxide solution falls from the aqueous solution of the salt, the yellow 3.9.7.Diamino-ethoxy-acridine which melts at 124 'with foaming. <I> Example 2 </I> 27 parts of 3.7.9-amino-ethoxy-chloroacridine are heated in 135 parts of phenol with 12 parts of ammonium chloride or ammonium sulfate for 6 hours to 130 to 140 and the salt acid 3.9.7- Diamino-ethoxy-acridine deposited by adding acetone. The salt crystallizes in yellow needles.
<I> Example 3 </I> 28 parts of 3.7.9-amino-diethoxy-aeridine, obtained from 3. 7-9-amino-ethoxy-chloroacrides and sodium ethylate, are mixed with 7 parts of urea and 150 parts Phenol or cresol slowly heated to boiling and kept at this temperature for some time. The phenol is distilled off in vacuo. The residue is taken up in warm dilute hydrochloric acid, filtered and the hydrochloric acid 3.9.7-diamino-ethoxy-aeridin separated from the solution by adding sodium chloride.
Example 1 Part 3. 7.9-Amino-ethoxy-chloroacridine is heated rapidly to 180 to 200 'with 5 parts of urea. The reaction mass is boiled with dilute sodium hydroxide solution and the residue is recrystallized from dilute alcohol. The 3.9.7-di-amino-ethoxy-aeridin is obtained in yellow needles from the S.
P .124. Example 5 2.7 parts of 3. 7. 9-Amino-ethoxy-chloroacridine are slowly heated to boiling with 0.7 parts of acetamide in 15 parts of phenol and kept in the boil for a short time. When the melt is diluted with acetone, the hydrochloride of 3.7. 9-amino-ethoxy-aeetamino-aeridins from; when heated with 50% sulfuric acid, <B> 3.9 </B> is produced from this. 7-diamino-ethoxy-acridine.
<I> Example ö: </I> <B> 2,7 </B> \ file 3. 7th 9-Amino-ethoxy-chloroacridine are slowly heated to boiling with 2.3 parts of hydroxylamine chlorohydrate and 2.7 parts of anhydrous sodium acetate with 15 parts of phenol. Vigorous evolution of gas occurs. The reaction mass is heated with dilute sodium hydroxide solution. The precipitated 3.9.7-diamino-ethoxy-acridine is purified using its hydrochloric acid salt.
<I> Example 7: </I> 1 part of 3-amino-7-ethoxy-9-phenoxy-acridine, obtainable from 3.7.9-aminoethoxy-chloro-acridine and sodium phenolate, is mixed with 0.5 part of ammonium chloride in 5 parts of p-cresol heated to boiling for 3.5 hours.
A yellow salt is then deposited using acetone and hydrochloric acid, which is then dissolved in water and, after the impurities have been removed by adding ammonia and acidifying with hydrochloric acid, the hydrochloric acid 3.9.7-diamino-a.tlioxy-acridine is deposited.
Example 8 2.7 parts of 3. 7,9-amino-ethoxy-chloroacridine are heated to 180 in a tube with 1.3 parts of ammonium carbonate in 10 parts of amyl or ethyl alcohol. The excreted reaction product is filtered off with suction, washed with acetone, then dissolved in water, made ammoniacal, and the 3.9.7-di.-amino-ethoxy-aeridine was deposited from the fil by means of sodium hydroxide solution.
Example <I> 9: </I> 2.7 parts 3. 7. 9-Amino-ethoxy-chloroacridine are dissolved in 13.5 parts of phenol, 2.7 parts of dicyandiamide are added to the heated and held for one hour. The melt is then diluted with acetone, sucked off from the resulting deposit, dissolved in water, made ammoniacal and separated from the filtrate by means of hydrochloric acid 3.9.7-diamino-ethoxy-acridine hydrochloride.
