Description
Verfahren zur Herstellung einer 31olekülverbindung des 2-phenylchinolin-4-ca.rbon- sauren Kalziums mit 1-Phenyl-2,3-dimethyl-4-dimethylamino-5-pyrazolon. Gegenstand vorliegender Erfindung ist ein Verfahren zur Herstellung einer Molekül verbindung des 2 -phenylchinolin - 4- carbori- sauren Kalziums mit 1-Phenyl-2,3-dimetbyl- 4-dimethylarnino-5-pyrazolon.
Diese Molekülverbindung wird hergestellt, indem die vorerwähnten Stoffe in einer dem Molekülverhältnis entsprechenden Menge bis zur Erreichung einer vollständigen, klaren Schmelze in Gegenwart von Wasser erhitzt werden.
Bei dieser Arbeitsweise tritt mitunter keine vollkommene Erstarrung des Reaktions produktes ein. Dieser Übelstand kann beho ben werden, wenn das warme Reaktionspro dukt vorerst einer allmählichen Abkühlung innerhalb eines 5 bis 50 unterhalb des Er starrungsproduktes der Molekülverbindung liegenden Temperaturbereiches unterworfen wird.
<I>Beispiel:</I> 25 gr 2 - phenylchinolin - 4 - carbonsaures Kalzium werden in der Reibschale mit 21,56 gr 1-Phenyl-2,3-dimethyl-4-dimethyl- amino-5-pyr@azolori unter Zusatz von 10 cm' Wasser verrieben. Das so erhaltene Gemenge wird auf eine Temperatur von<B>1100</B> erhitzt und nach Ablauf von je 10 bis 15 Minuten durchgerührt.
Der Reaktionsprozess ist been det, wenn eine vollkommen klare Schmelze erfolgt ist, in der weder unveränderte Teil chen der Reaktionskomponenten sichtbar sind, noch das Aufsteigen von Wasserdampfbläs- cben wahrnebinbar ist. Dieser Zustand wird bei Anwendung der erwähnten Menge in 2 bis 21/z Stunden erreicht. Man lässt die Schmelze erkalten und pulverisiert sie in üb licher Weise.
Die nach dem beschriebenen Verfahren hergestellte Molekülverbindung zeigt gegen über den Ausgangsstoffen ein völlig verän dertes Verhalten.
Während nämlich das Kalziumsalz der 2-Phenylchinolin - 4 - carbonsäure in Wasser nahezu unlöslich ist, erweist sich die be schriebene Molekülverbindung als gut löslich in Wasser; sie gibt eine Lösung mit einem Gehalte von 1,5%.
Besonders bemerkenswert ist das Verhal ten der vorbeschriebenen Molekülverbindung gegenüber verdünnter Salzsäure. Durch Salz säure werden die Salze der 2-Phenylchinolin- 4-carbonsäure leicht zersetzt und es wird die freie, in -Wasser nahezu unlösliche Säure ausgeschieden. Die vorerwähnte Molekülver bindung zeigt sich dagegen gegenüber Salz säure ziemlich widerstandsfähig, indem sie sich bei dieser Behandlung ganz langsam, erst nach längerem Stehen, zersetzt. Grosse Haltbarkeit zeigt die nach dem vorliegenden Verfahren hergestellte Molekülverbindung beim Auflösen in genuinem Magensaft.
Die beschriebenen Eigenschaften bleiben auch er halten, wenn eine der Reaktionskomponenten in einem gewissen, ein bestimmtes Mass nicht überschreitenden Überschuss, zur Anwendung gelangt.
Die vorbeschriebene Molekülverbindung wird als Heilmittel verwendet und hat sich besonders bei echter Gicht mit entzündlichen Gelenkaffektionen, bezw. chronischem Arthri- tismus bei drei- bis viermaligen Gaben pro os von 0,5 bis 0,6 gr pro Tag bewährt. Die Molekülverbindung kann ferner auch als Salbe oder in Form von Suppositorien zur Anwendung gelangen.

The present invention relates to a process for the preparation of a molecular compound of 2-phenylquinoline-4-carboric acid calcium with 1-phenyl-2,3-dimetbyl-4-dimethylamino-5-pyrazolone.
This molecular compound is produced by heating the aforementioned substances in an amount corresponding to the molecular ratio until a complete, clear melt is achieved in the presence of water.
With this procedure, there is sometimes no complete solidification of the reaction product. This inconvenience can be remedied if the warm reaction product is initially subjected to gradual cooling within a temperature range from 5 to 50 below the temperature range lying below the solidification product of the molecular compound.
<I> Example: </I> 25 grams of 2 - phenylquinoline - 4 - carboxylic acid calcium are placed in the mortar with 21.56 grams of 1-phenyl-2,3-dimethyl-4-dimethyl-amino-5-pyr @ azolori under Addition of 10 cm 'of water rubbed in. The mixture obtained in this way is heated to a temperature of <B> 1100 </B> and stirred after 10 to 15 minutes each time.
The reaction process is over when a completely clear melt has taken place, in which neither unchanged particles of the reaction components are visible nor the rising of water vapor bubbles can be perceived. This condition is reached in 2 to 21/2 hours when using the amount mentioned. The melt is allowed to cool and pulverized in the usual way.
The molecular compound produced by the process described shows a completely different behavior compared to the starting materials.
While the calcium salt of 2-phenylquinoline - 4 - carboxylic acid is almost insoluble in water, the molecular compound described proves to be readily soluble in water; it gives a solution with a content of 1.5%.
The behavior of the above-described molecular compound towards dilute hydrochloric acid is particularly remarkable. The salts of 2-phenylquinoline-4-carboxylic acid are easily decomposed by hydrochloric acid and the free acid, which is almost insoluble in water, is excreted. The above-mentioned molecular compound, on the other hand, is quite resistant to hydrochloric acid, as it decomposes very slowly during this treatment, only after standing for a long time. The molecular compound produced according to the present process shows great durability when dissolved in genuine gastric juice.
The properties described are retained even if one of the reaction components is used in a certain excess that does not exceed a certain level.
The above-described molecular compound is used as a remedy and has particularly proven itself in real gout with inflammatory joint affections, respectively. chronic arthritis with three to four times doses per os of 0.5 to 0.6 g per day. The molecular compound can also be used as an ointment or in the form of suppositories.
