Process for the preparation of 1- (3 ', 4'-diogyphenyl) -2-aminopropanol-1- The present invention is a process for the preparation of 1- (3', 4'-dioxyphenyl) -2-aminopropanol-1 , which is characterized in that a derivative of 1- (3 ', 4'-diogyphenyl) -propanone-1, in which the hydrogen atoms of the hydroxyl groups are substituted, is produced by the action of nitrous acid, the corresponding isonite roso compound subject to the reduction and cleaves off the groups protecting the hydroxyl groups.
The process product, known per se, whose hydrochloride melts at 178 ° C., has an adrenaline-like effect and is used accordingly in therapy. <I> Example: </I> 64 g of 3,4-dibenzyloxypropiophenone in <B> 500 </B> cm3 'benzene are slowly mixed with 18.7 g of butyl nitrite while introducing hydrochloric acid gas. After the liquid has stood for several hours at room temperature, the excess benzene is distilled off in vacuo.
The crystalline residue is recrystallized from dilute alcohol after washing with hexahydrobenzene. The 3,4-dibenzyloxyisoriitro- sopropiophenone melts at <B> 117 '</B> C.
35 g of the isonitro compound obtained are dissolved in 250 cm 'of alcohol and catalytically hydrogenated with nickel and hydrogen. The catalyst is filtered off with suction and the filtrate is evaporated in vacuo. The residue that remains is taken up in dry ether and the 3,4-dibenzy loxyphenylpropanolainine chlorohydrate is precipitated therefrom by introducing hydrochloric acid gas. The hydrochloric acid salt obtained is then dissolved without further purification in 6-7 times the amount of absolute alcohol and reduced to the end with palladium and hydrogen.
As soon as the calculated amount of hydrogen is absorbed, the catalyst is filtered off and the alcoholic solution is completely evaporated to dryness in a vacuum. The 3,4-dioxyphenylpropanolamine chlorohydrate is obtained from the residue obtained by recrystallization from methanol and with the addition of acetone. The anhydrous compound melts at 178 C.
