Verfahren zur Darstellung von kristallisierten Estern der weiblichen Keimdrüsenhormone. Bisher ist die Darstellung kristallisierter Ester der weiblichen Keimdrüsenhormone immer so ausgeführt worden, dass man aus gehend von den kristallisierten weiblichen Keimdrüsenhormonen (Follikelhormon der For mel C1sH2208, Follikelhormonhydrat der For mel CisEs40s) durch Anwendung der üblichen Acylierungsmethoden zu den kristallisierten Estern dieser Hormone gelangt. (Butenandt, Untersuchungen d.weibl. Sexualhormons,<B>1931,</B> S. 61 und S.
62.) Da dieses Verfahren in al kalischem Medium durchgeführt wird, bildet sich vorgängig der Acylierung die Alkaliver- bindung des Hormons.
Aus den rohen, die Keimdrüsenhormone enthaltenden (Ölen, wie sie im Gange der Isolierung dieser Stoffe aus den natürlichen Vorkommen erhalten werden, sind kristalli- aerte Ester der Hormone bisher nicht darge- stellt worden.
Man hat aber beispielsweise durch Acetylieren des rohen (Öls aus Follikel- flüssigkeit (Doisy, Ralls und Jordan, Endo- orinology 10, 273 [1926]) ein öliges Acetyl- derivat erhalten und Butenandt (Untersuchun gen über das weibliche Sexualhormon, 1931, S.
59) beschreibt ein öliges Benzoat, das durch Benzoylierung eines aus Schwangerenharn stammenden Rohhormonöls mit einer Wirk samkeit von 1 Million Mäuse-Einheiten pro Gramm gewonnen wurde.
Wir haben nun gefunden, dass man auch aus Rohhormonölen, sogar solchen mit einer Wirksamkeit weit unter der von Butenandt verwendeten von 1 Million Mäuse-Einheiten pro Gramm, überraschenderweise kristalli sierte Ester der Keimdrüsenhormone erhalten kann, wenn man das Rohhormonöl mit Al- kalien behandelt, die Alkalisalze der Keim drüsenhormone, nach Abtrennung der neu tralen Substanzen, in wässeriger Lösung acy- liert und das Acylprodukt des Hormons zur Kristallisation bringt.
Das so erzielbare Re- sultat ist um so überraschender, als man durch Behandlung der Rohhot-monöle mit Al- kalien bekanntlich kein kristallisiertes Hormon abscheiden kann.
Man kann beispielsweise so vorgehen, dass man das Rohhormonöl mit verdünntem wäs serigen Alkali auskocht und die Neutralteile durch Filtrieren von der wässerigen alkali schen Hormonlösung abtrennt, oder dass man das rohe Hormonöl in einem mit Wasser nicht mischbaren Lösungsmittel, beispielsweise Äther etc., löst und die Lösung nun mit kon zentrierter Alkalilauge ausschüttelt, wobei die Neutralteile in der ätherischen Lösung ver bleiben und das gesamte Hormon an das Al kali gebunden wird.
Nach Abtrennen der ätherischen Hormonlösung von dem Alkali salz des Hormons wird dieses mit Wasser zu einer Lösung verdünnt und hierauf der Ein wirkung von Acylierungsmitteln unterworfen.
Es sei bemerkt, dass das Rohhormonöl verschiedene Follikelhormone, darunter auch Föllikelhormonhydrat bezw. deren Derivate, enthält, und dass unter dem Ausdruck "Fol- likelhormon" hier diese Vielheit der Hormone verstanden wird. Beispiel <I>1:</I> 10 gr eines Hormonöls von ca._ <B>300000</B> Mäuse - Einheiten pro Gramm werden, mit 50 ;oiger wässeriger Kalilauge erwärmt.
Nach dem Erkalten wird die alkalische Lösung durch Filtrieren vom ungelösten Öl befreit und in der üblichen Weise nach Schotten- Baumann benzoyliert. Das ausgeschiedene Reaktionsprodukt wird in Äther aufgenom men. Nach dem Verdampfen des Äthers ver bleibt ein hellgelbes Öl, das beim Verreiben mit Alkohol kristallisiert. Ausbeute ca. 0,7 bis 0,8 gr. Durch Umkristallisieren des Roh- kristallisates aus Äthanol wird das Benzoat des Follikelhormons rein erhalten.
Beispiel <I>2:</I> 10 gr eines Hormonöls von ca. 250 000 Mäuse-Einheiten pro Gramm werden :in<B>600</B> emi ilther gelöst und mit 10 gr einer Xatronlauge von 40 % gründlich durchgeschüttelt. Nach Abtrennen der ätherischen Lösung von dem schmierig bis pulverig ausfallenden Alkalisalz kann die ätherische Lösung zur Gewinnung der neutralen Substanzen verdampft werden.
Die Alkaliverbindungen werden in ca. 100 Teilen Wasser gelöst und nun mit Benzoyl- chlorid unter gutem Umschütteln versetzt. Nach Vollendung der Reaktion hat sich das Benzoat des Follikelhormons zum Teil als feines Pulver abgesetzt und kann nun durch Filtrieren von der alkalischen Mutterlauge abgetrennt werden. Es wird nach gutem Waschen mit Wasser aus Alkohol umkristal lisiert und in einer Ausbeute von 0,4 bis 0,6 gr mit einem Schmelzpunkt von 204 o nach vor herigem Sintern bei ca. 200 o erhalten.
Beispiel <I>3:</I> 10 gr eines Hormonöls von ca.<B>300000</B> Mäuse - Einheiten pro Gramm werden mit 5 %iger wässeriger Kalilauge erwärmt. Nach dem Erkalten wird die alkalische Lösung durch Filtrieren vom ungelösten Öl befreit und in der üblichen Weise nach Schotten- Baumann mit Acetylchlorid acetyliert. - Das ausgeschiedene Reaktionsprodukt wird in 4ther aufgenommen. Nach dem Verdampfen des Äthers verbleibt ein, hellgelbes Öl, das leim Verreiben mit Alkohol --kristallisiert.
Durch Umkristallisieren des Rohkristallisates aus verdünntem Äthanol wird das Acetat des Follikelhormons rein erhalten.

We have now found that, even from crude hormone oils, even those with an activity far below the 1 million mouse units per gram used by Butenandt, surprisingly crystallized esters of the gonadal hormones can be obtained if the crude hormone oil is treated with alkali, the alkali salts of the gland hormones, after separation of the neutral substances, acylated in aqueous solution and brings the acyl product of the hormone to crystallization.
The result that can be achieved in this way is all the more surprising since, as is known, no crystallized hormone can be separated off by treating the raw hot mono-oils with alkalis.
One can, for example, proceed in such a way that the crude hormone oil is boiled out with dilute aqueous alkali and the neutral parts are separated from the aqueous alkaline hormone solution by filtering, or the crude hormone oil is dissolved in a water-immiscible solvent, such as ether, etc. shake the solution with concentrated alkali, whereby the neutral parts remain in the ethereal solution and the entire hormone is bound to the alkali.
After separating the ethereal hormone solution from the alkali salt of the hormone, this is diluted with water to form a solution and then subjected to the action of acylating agents.
It should be noted that the crude hormone oil contains various follicle hormones, including follicle hormone hydrate, respectively. their derivatives, and that the term "follicle hormone" is understood here to mean this multiplicity of hormones. The alkali compounds are dissolved in approx. 100 parts of water and benzoyl chloride is then added with thorough shaking. After completion of the reaction, the benzoate of the follicular hormone has partially settled as a fine powder and can now be separated from the alkaline mother liquor by filtration. It is recrystallized from alcohol after thorough washing with water and obtained in a yield of 0.4 to 0.6 gr with a melting point of 204 o after prior sintering at about 200 o.
