Description
Verfahren zur Darstellung von 17-lllethyl-dihydrofollikelhormon. Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Darstellung von 17- Methyl-dihydrofollikelhormon, welches da durch gekennzeichnet ist, dass man eine Ver bindung, die sich vom Follikelhormon der Formel Cl$H220.> dadurch ableitet, dass sie an Stelle der Hydroxylgruppe einen Rest be sitzt, der bei Hydrolyse durch die Hydroxyl- gruppe ersetzt wird, z.
B. eine Ester- oder Äthergruppe oder Halogen, mit einer für die Umwandlung von Ketonen in Methylkarbi- nole geeigneten metallorganischen Verbin dung.
beispielsweise nach der Methode von Grignard mit Methylmagnesiumjodid, um setzt, das dabei gebildete metallorganische Umsetzungsprodukt mit Hilfe hydrolysie- rend wirkender Mittel aufspaltet und an schliessend die an Stelle der Hydroxylgruppe des Follikelhormons befindliche Gruppe durch Hydrolyse in die Hydroxylgruppe überführt.
Bei der Aufarbeitung des Reak tionsgemisches erhält man dann 17-Methyl- dihYdrofollikelhormon der Formel C11112602 in kristalliner Form.
Das erfindungsgemässe Verfahren sei im folgenden anhand beispielsweise gegebener Formelbilder veranschaulicht, in denen mit R eine durch Hydrolyse in die Hy droxylgruppe überführbare Gruppe, zum Beispiel eine Ester- oder Äthergruppe oder Halogen, be-

<I>Beispiel:</I> Zu einer ätherischen Lösung von Methyl- uragnesiumjodid, hergestellt aus 4 g trockenen Magnesiumspänen, 12,5 cm' Methyljodid und 160 cm' trockenem Äther in Gegenwart einer Spur Jod wird unter gutem Rühren eine Lö sung von 5 g Follikelhormonbenzoat vom Schmelzpunkt 216 bis 22.0 allmählich in 150 cm'<B>'</B> trockenem Benzol hinzulaufen ge- la:
ssen. Die Reaktionsflüssigkeit wird nach erfolgter Zugabe der Benzoatlösung noch zirka 111 Stunden zum Sieden erhitzt, ,dann nach dem Erkalten mit Wasser und ver dünnter Salzsäure (1 : 5) unter Eiskühlung zersetzt ünd in Äther aufgenommen. Die ätherische Lösung wird nacheinander mit Nasser, wässriger Natriumthiosulfatlösung und wieder mit Wasser gewaschen, getrock net und .abgedampft.
Der erhaltene Rückstand wird durch 3- stündiges Erhitzen mit 5 % iger methylalko- holischer Kalilauge auf dem Dampfbad ver seift.
Nach dem Erkalten wird die Ver- seifungslösung mit Wasser verdünnt, mit ver dünnter Salzsäure (1 :5) angesäuert, mit Äther ausgezogen und die ätherische Lösung nacheinander mit verdünnter Sodalösung (zur Entfernung der Benzoesäure) und dann gründlichst mit 5 %iger wässriger Kalilauge extrahiert.
Nach dem Ansäuernder Kalium hydroxydauszüge scheidet sich -das Methyl- @d:.hydrofollikelhormon in kristalliner Form ab und kann durch Filtration oder Auf nehmen in Äther isoliert werden. Ausbeute nahezu quantitativ.
Das verfahrensgemäss erhältliche Reak- tionsprodukt ist neu; es besteht entsprechend der Theorie aus einem Gemisch zweier stereoisomerer Methylkarbinole und zeigt in konzentrierter Schwefelsäure im Lichte der Analysenquarzlampe eine blaugrüne Farbe und Fluoreszenz.
Der physiologische Wir kungswert des Präparates im Allen-Doisy- Test beträgt bei subkutaner Verabfolgung in wässriger Lösung und sechsmaliger Unter teilung 3 bis 4 Millionen Ratteneinheiten pro Gramm, während das Follikelhormon' der Formel C1gH2202 nur eine @@Tirk@@mh@it voll etwa 1,2 Millionen Ratterreinlreitull I110 Gramm aufweist.
