Verfahren zur Darstellung von Androsteron. Es ist bereits bekannt, dass man Stoffe nach Art der männlichen Keimdrüsenhormone durch Oxydation von Sterinen darstellen kann.
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Darstellung von Andro- steron, welches dadurch gekennzeichnet ist, dass man 3-Epiacylätioallocholansäure in das entsprechende Säurechlorid überführt, das Säurechlorid durch Behandlung mit Natrium azid in das entsprechende Azid und dieses durch Erhitzen in das Isocyanat überführt, das Isocyanat zum Amin verseift und das Amin durch Behandlung mit unterchloriger Säure in die Chlorylverbindung überführt,
aus dieser mit Hilfe von Alkali Salzsäure abspaltet und das so entstandene Imin mit Hilfe einer Säure zum Androsteron verseift.
Das neue Verfahren schafft die Möglich keit, das Androsteron mit besseren Ausbeu ten als nach den bekannten Verfahren her zustellen. Androsteron ist eine an sich bekannte Substanz, die dem männlichen Sexualhormon entspricht.
Beispiel: 2 g 3-epi-Acetosyätioallocholansäure wer den in 20 cm' trockenem Benzol und 6 g Thionylchlorid 11/a Stunden erhitzt. Darnach wird die Reaktionslösung im Vakuum ein gedampft und der- kristallisierte Rückstand in 20 cm' Aceton aufgenommen. Die Aceton lösung wird nun auf 0 gekühlt und mit 0,5 g Natriumazid in 5 cm' Wasser ver setzt. Die trübe Lösung, aus der sich bald ein Niederschlag abscheidet, wird 20 Stun den 'bei 0 aufbewahrt; darnach wird mit Wasser versetzt und mit Toluol ausgeschüt telt.
Nach Trocknen über Natriumsulfat wird die Toluollösung 15 Minuten zum Sieden erhitzt und darnach im Vakuum eingeengt. Der Rückstand wird nun in 15 cm3 Äther und 15 eins Benzol gelöst, mit 100 g 60- prozentiger Schwefelsäure unterschichtet und 30 Minuten intensiv gerührt. Dabei scheidet sich das 3-Acetoxyaridrostan-17-aininsulfat ab.
Es wird mit alkoholischer Lauge in Frei heit gesetzt und kann aus seiner ätheriselien Lösung als Sulfat oder Hydrochlorid abge schieden werden. Zersetzungspunkt des Sul fates um 350 . Ausbeute 0,5 g.
0,2 g des 3-Epiacetoxyaridrostan-17-amiris werden in Äther gelöst und zur Lösung bei 0 die berechnete Menge einer ätherischen Lösung von HOCL einfliessen gelassen. Die Reaktionslösung wird im Vakuum einge dampft und der Rückstand finit einer Na- trium-Methylatlösung (bereitet ans 0,2 g Natrium und 1.5 cm' Alethanol abs.) 45 Mi nuten unter Rückfluh erhitzt.
Darnach wird in schwefelsäurehaltiges Wasser gegossen und die Mischung nach 24 Stunden ausgeäthert. Der Rückstand der ätherischen Lösung wird firn Hochvakuum destilliert und das Destillat mit wässerigem Methanol unigelöst. Ausbeute an Androsteron 0.1 g. Schmelzpunkt<B>1820.</B>

The reaction solution is evaporated in vacuo and the residue is refluxed for 45 minutes with a sodium methylate solution (prepares 0.2 g of sodium and 1.5 cm of absolute alcohol).
It is then poured into water containing sulfuric acid and the mixture is extracted with ether after 24 hours. The residue of the ethereal solution is distilled in a high vacuum and the distillate is undissolved with aqueous methanol. Yield of androsterone 0.1 g. Melting point <B> 1820. </B>
