Verfahren zur Herstellung eines basischen Esters. Nach Untersuchungen von Fromherz und andern Autoren (Arch. exp. Path. u. Pharm. 173 124 [1933]) besitzen basische Ester von aliphatischen Carbonsäuren keine nennens werte krampflösende Wirkung.
Auch die in der Patentliteratur bekannt gewordenen ali- phatischen Carbonsäureester, wie der Isovale- riansäure- bezw. a. - Bromisovaleriansäure- bezw. Isopropylallylessigsäure- bezw. Di- äthylessigsäureester des 3-Diäthylamino-2,2- dimethyl-l-proponals, haben ein sehr schwa ches, neurotrop-atropinartiges Verhalten.
Nicht viel besser verhalten sich die von @al- pern (Arcb. internat. de Pharmacodyn. 59 149 [1988]) erwähnten Ester, wie der Äthyl- butylessigsäure- bezw. Dibutylessigsäureester des Diäthylaminoäthanols, der Dibutylessig- säureester des Diäthylamino-(1)-propanols- (3), der Essigsäure-,
Propionsäure-, n-Butter- säure-, Diäthylessigsäure-, Äthylbutylessig- säure- bezw. Dibutylessigsäureester des Di- äthylamino-(1)-pentanols-(4).
Demgegenüber wurde überraschenderweise gefunden, dass Piperidin-alkanolester der Di- n - butylessigsäure hervorragende krampf lösende Eigenschaften besitzen. Ein solches= Verhalten war nicht zu erwarten. Zur Darstellung der Ester setzt man z. B.
reaktionsfähige.Abkömmlinge der Di-n-butyl- essigsäure, d. h. ihre Halogenide, Ester oder ihre Anhydride in An- oder Abwesenheit von Kondensationsmitteln mit Piperidin-alkano- len um, oder man lässt reaktionsfähige Ester der Piperidin-alkanole, gegebenenfalls in Ge genwart säurebindender Mittel, auf die oben genannte Säure bezw. ihre Salze einwirken.
Unter reaktionsfähigen Estern von Pipe- ridin-alkanolen sind insbesondere Ester mit Halogenwasserstoffsäuren, mit Ary1sulfon- säuren und dergleichen zu verstehen.
Gegenstand vorliegenden Patentes ist ein Verfahren zur Herstellung eines basischen Esters. Das Verfahren ist dadurch gekenn zeichnet, dass man eine Verbindung der Formel

worin X und Y zwei, mit Ausnahme eines in ihnen enthaltenen Sauerstoffatoms, sich bei der Reaktion abspaltende Reste bedeuten, einwirken lässt.
Die neue Verbindung, der Di-n-butylessig- säure-ss-piperidino-äthylester, ist eine Base vom Siedepunkt 167 bis 169 unter 11 mm Druck. Sie lässt sich mit anorganischen oder organischen Säuren in wasserlösliche Salze überführen. Sie soll therapeutische Verwen dung finden.
<I>Beispiel:</I> 19 Teile Di-n-butylessigsäurechlorid wer den unter Rühren zu 13 Teilen ss-Piperidino- äthylalkohol gegeben und das Gemisch unter Rühren kurze Zeit auf 160 erhitzt, wobei unter Wärmeentwicklung ein klares, braunes 01 entsteht, das vorteilhaft noch warm mit Wasser versetzt wird. Die wässrige Lösung wird einige Male ausgeäthert und hierauf die Base mit konzentriertem Ammoniak frei gemacht.
Diese wird in Äther aufgenommen und nach einmaligem Waschen mit Wasser und Trocknen der ätherischen Lösung wird das Lösungsmittel abdestilliert. Der erhaltene Di-n-butylessigsäure-ss-piperidino-äthylester siedet unter 11 mm Druck bei 167 bis 169 .
Zum gleichen Endprodukt gelangt man auch durch Verestern von Di-n-butylessig- säure mit ss-Piperidino-äthylalkohol, oder auch durch Umestern von z. B. Di-n-butyl- essigsäure-methyl- oder -phenylester mit ss- Piperidino-äthylalkohol. Ferner kann man auch ein Salz der Di-n-butylessigsäure mit einem reaktionsfähigen Ester des ss-Piperi- dino-äthylalkohols z.
B. mit ss-Piperidino- äthylchlp,rid oder mit Toluolsulfonsäure-ss-pi- peridino-äthylester umsetzen und gelangt so ebenfalls zum. Di-n-butylessigsäure-ss-piperi- dino-äthylester.
