Description
Verfahren zur Darstellung von a, Y-Diogy-f, ss-dimethyl-buttersäure- (3',4'-diogy-butyl)-amid. In den schweizerischen Patentschriften Nr. 227706, 233770 und 239703 werden Ver fahren zur Gewinnung therapeutisch wert voller Oxyalkylamide der a,.y-Dioxy-ss"f-di- methyl-buttersäure durch Umsetzung von d(-)-a-Oxy-ss,ss-dimethyl-y-butyrolacton mit Oxyalkylaminen beschrieben.
Es wurde nun gefunden, dass man zu sehr reinem 3,4-Dioxy-butylamid der<I>a,1-</I> Dioxy-ss,ss-dimethyl-buttersäure in besonders bequemer Weise dadurch gelangen kann, dass man 3,4-(Dimethyl-methylen-dioxy)-butyl- amin mit a-Oxy-ss,ss-dimethyl-y-butyrolacton umsetzt und das entstehende 3,4-(Dimethyl- methylen-dioxy)-butylamid der a,y-Dioxy- ss,
ss - dimethyl - buttersäure einer milden, sauren Hydrolyse unterwirft. Die Umsetzung verläuft nach folgendem Schema:

Bei dieser Umsetzung ist überraschend, dass es gelingt, aus dem Kondensationsprodukt den Acetonrest abzuspalten ohne Lösung der Amidbindung, denn sowohl Pantothensäure als auch die Oxyalkylamide der a,y-Dioxy- ss,ss-dimethyl-buttersäure sind im Vergleich zu andern Säureamiden ausserordentlich leicht verseifbar. Bei der Hydrolyse wird die optische Aktivität nicht beeinflusst.
Das als Ausgangsmaterial benötigte 3,4- (Dimethyl-methylen-dioxy)-butylamin (II) kann wie folgt bereitet werden: 8-Oxy-y-va- lerolacton (Leuchs und Mitarbeiter, Berichte der Deutschen Chemischen Gesellschaft, Bd. 40 [1907] S.801; Bd. 42 [1909] S.1228;
Winterfeld, Arch. Pharm> Bd. 277 [1939] S. 227) wird durch zweitägiges Stehen mit der 6fachen Menge bei 0 mittels mit trocke nem NH3 gesättigtem Methanol in das 4,5-Di- oxy-valeriansäureamid übergeführt, das als farbloses, dickes<B>01</B> erhalten wird. Dieses wird hierauf in etwa 100 Teilen trockenem Aceton gelöst; hierauf wird etwas trockenes Salzsäuregas eingeleitet, bis die Lösung gegen Kongo eben sauer reagiert.
Nach ein tägigem Stehen wird mit etwas fester Pott asche geschüttelt, filtriert und eingedampft. Das als Rückstand verbleibende 3,4-(Di- methyl - methylen - dioxy) - valeriansäureamid destilliert im Vakuum bei 0,1 mm unter 125 und kristallisiert aus Aceton-Äther in farb losen Plättchen vom Schmelzpunkt 92 .
Der Hofmannsche Abbau dieses Amids liefert die gesuchte Base (II), die im Vakuum von 11 mm bei 74-75 als leichtbewegliche, farb lose Flüssigkeit siedet und aus der Luft leicht C02 sowie Wasser anzieht. Sie liefert ein bei 157-158 schmelzendes Pikrat.
Zur Kondensation wird das 3,4-Dimethyl- methylen-dioxy)-butylamin zweckmässig mit der äquimolaren Menge a-Oxy-ss,ss-dimethyl- y-butyrolacton zur Reaktion gebracht, was vorteilhaft in einem indifferenten Lösungs-- mittel geschieht, in dem beide Komponenten löslich sind. Die Umsetzung vollzieht sieh bereits bei Zimmertemperatur nach etwa 24 Stunden und kann durch Erwärmen beschleu nigt werden.
Das so erhaltene 3,4-(Dimethyl- methylen-dio@xy)-butylamid der a,y-Dioxy- ss,ss-dimethyl-buttersäure bildet ein farbloses 0l, das sich im Hochvakuum uniersetzt destillieren lässt. Unter einem Druck von 0,1 mm destilliert die Verbindung bei einer Badtemperatur von etwa 130 .
Das a,y-Dioxy-ss,ss-dimethyl-buttersäure- 3',4'- (dimethyl -methy len) - dio.xy) -butylamid (III) liefert bei milder, saurer Hydrolyse das 3,4-Dioxy-butylamid der d-a,y-Dioxy-ss,ss- dimethyl-buttersäurer (IV) als farbloses, hy groskopisches., sehr zähflüssiges<B>01,</B> dessen Analysenwerte auf die Formel CloH"0;;N passen und das sich auch im Vakuum nicht uniersetzt destillieren lässt.
Es ist identisch mit der in der schweizerischen Patentschrift Nr. 239703 beschriebenen Verbindung. Das Verfahrenserzeugnis besitzt bei der Prüfung an Ratten alle physiologischen Eigenschaften der Pantothensäure. Es ist auch wertvoll als Zwischenprodukt.
Beispiel: 2 Gewichtsteile d,l-Dimethyl-methylen- 3,4-dioxy-butylamin und 1,7 Gewichtsteile <B>d(-)</B> - a - Oxy - ss.,ss - dimethyl - y - butyrolacton werden in 12 Gewichtsteilen wasserfreiem Benzol gelöst und die Lösung eine Stunde unter Ausschluss von Feuchtigkeit am Rück fluss gekocht. Anschliessend bringt man die Lösung durch Eindampfen im Vakuum zur Trockne.
Zur Weiterverarbeitung ist eine weitere Reinigung nicht erforderlich, obgleich es möglich ist, die Verbindung bei 0,1 mm Druck zu destillieren. Das. Destillationspro- dukt gibt bei der Analyse mit der Theorie übereinstimmende Werte für Kohlenstoff, Wasserstoff und Stickstoff.
7 Gewichtsteile des rohen Kondensations produktes werden in 50 Raumteilen n/10 Schwefelsäure gelöst und bei 18 stehen gelassen. An Hand der Bestimmung des ab gespaltenen Acetons lässt sich nachweisen, dass die Verseifung unter diesen Bedingun gen nach 30 Minuten beendet ist.
Die Lösung wird durch Einrühren von überschüssigem frisch gefälltem Bariumcarbonat neutralisiert und durch eine mit Bariumcarbonat und wenig ausgekochter Kohle überschichtete Nutsche filtriert. Das Filtrat enthält das ge suchte d(+)-2,4-Dioxy-3,3-dimethyl-butter- säure-(3',4'-dioxybutyl)-amid, das beim Ein engen der Lösung im Vakuum hinterbleibt. Es ist identisch mit der in dem schweizeri schen Patente Nr. 239703 beschriebenen Ver bindung.
Process for the preparation of a, Y-diogy-f, ss-dimethyl-butyric acid- (3 ', 4'-diogy-butyl) -amide. In Swiss patents Nos. 227706, 233770 and 239703, methods are used to obtain therapeutically valuable oxyalkylamides of a, .y-dioxy-ss "f-dimethyl butyric acid by reacting d (-) - a-oxy-ss , ss-dimethyl-y-butyrolactone described with oxyalkylamines.
It has now been found that very pure 3,4-dioxy-butylamide of the <I> a, 1- </I> dioxy-ss, ss-dimethyl-butyric acid can be obtained in a particularly convenient manner by adding 3, 4- (Dimethyl-methylen-dioxy) -butyl-amine with a-oxy-ss, ss-dimethyl-y-butyrolactone and the resulting 3,4- (dimethyl-methylen-dioxy) -butylamide of a, y-dioxy - ss,
ss - dimethyl - butyric acid is subjected to a mild, acidic hydrolysis. The implementation proceeds according to the following scheme:

In this reaction it is surprising that it is possible to split off the acetone residue from the condensation product without dissolving the amide bond, because both pantothenic acid and the oxyalkylamides of α, γ-dioxy- ss, ß-dimethylbutyric acid are extremely light compared to other acid amides saponifiable. The optical activity is not influenced by the hydrolysis.
The 3,4- (dimethyl-methylen-dioxy) -butylamine (II) required as starting material can be prepared as follows: 8-oxy-y-valerolactone (Leuchs and coworkers, reports of the German Chemical Society, Vol. 40 [ 1907] p. 801; vol. 42 [1909] p. 1228;
Winterfeld, Arch. Pharm> Vol. 277 [1939] p. 227) is converted into 4,5-dioxy-valeric acid amide by standing for two days in 6 times the amount at 0 using methanol saturated with dry NH3, which is a colorless, thick <B> 01 </B> is obtained. For condensation, the 3,4-dimethylmethylene-dioxy) -butylamine is expediently reacted with the equimolar amount of a-oxy-ss, ss-dimethyl-y-butyrolactone, which is advantageously done in an inert solvent in both components are soluble. The reaction takes place after about 24 hours at room temperature and can be accelerated by heating.
The 3,4- (dimethylmethylene-dio @ xy) -butylamide of α, γ-dioxy- ss, ß-dimethyl-butyric acid thus obtained forms a colorless oil which can be distilled in a high vacuum without stopping. The compound distills under a pressure of 0.1 mm at a bath temperature of about 130.
The a, y-dioxy-ss, ss-dimethyl-butyric acid- 3 ', 4'- (dimethyl-methylene) - dio.xy) -butylamide (III) yields the 3,4-dioxy with mild, acidic hydrolysis butylamide of da, y-dioxy-ss, ss-dimethyl-butyric acid (IV) as a colorless, hygroscopic., very viscous <B> 01, </B> whose analysis values match the formula CloH "0 ;; N and that Cannot be distilled in full, even in a vacuum.
It is identical to the compound described in Swiss Patent No. 239703. When tested on rats, the process product has all the physiological properties of pantothenic acid. It is also valuable as an intermediate.
Example: 2 parts by weight of d, l-dimethyl-methylene-3,4-dioxy-butylamine and 1.7 parts by weight of <B> d (-) </B> - a - oxy - ss., Ss - dimethyl - y - butyrolactone are dissolved in 12 parts by weight of anhydrous benzene and the solution refluxed for one hour with exclusion of moisture. The solution is then brought to dryness by evaporation in vacuo.
No further purification is required for further processing, although it is possible to distill the compound at 0.1 mm pressure. The. When analyzed, the distillation product gives values for carbon, hydrogen and nitrogen that are consistent with theory.
7 parts by weight of the crude condensation product are dissolved in 50 parts by volume n / 10 sulfuric acid and left to stand at 18. Based on the determination of the acetone split off, it can be demonstrated that the saponification is complete under these conditions after 30 minutes.
The solution is neutralized by stirring in excess freshly precipitated barium carbonate and filtered through a suction filter covered with barium carbonate and a little boiled charcoal.
