On peut utiliser comme produit de départ soit la forme racémique,
soit une des formes optiquement actives de ladite allo-oxazoline. On préfère utiliser le racémique ou la forme optiquement active lévogyre de cette oxazo- line. De préférence, on effectue la réaction à une température voisine de la température ordinaire ou à une température élevée, par exemple sur un bain-marie. De préférence, on effectue la réaction dans Lui. solvant organique anhydre, tel que le dioxane.
Le produit obtenu par le procédé selon l'invention est une nouvelle substance cristal lisée. La forme racémique (DL-allo) fond à l33-134 C; l'isomère optique D fond à 101 C et possède -un pouvoir rotatoire [a] D --. -I- 12 5 à une concentration de 4% dans le méthanol; l'isomère optique L fond à 1.14--116 C et possède un pouvoir rotatoire [a] = D -12 36 à une concentration de 40/0 dans le méthanol.
Toutes les formes optiques (DL, L et D) de cette nouvelle substance sont des produits intermédiaires importants pour la préparation dé produits ayant des propriétés antibiotiques.
Le produit de départ du procédé selon l'invention peut être préparé de la faon sui vante: On dissout 1,93 g de chlorhydrate de DL- allo -1- p-nitr ophényl - 2-amino-propanediol-1,3 (point de fusion = 206-208 C) dans 13 cm3 de pyridine anhydre. On ajoute à cette solu tion 1,8 g de chlorhydrate de dichloracétimido- éthyl-éthei du p. f. = 80 C.
(Ce dernier a été préparé en faisant réagir du dichloracéto- nitrile avec. de l'éthanol.) Le mélange est laissé au repos pendant deux heures à la tempéra ture ordinaire. On remarque alors une préci pitation de chlorhydrate de pyridine et on verse le mélange dans 100 cm3 d'eau. On ob tient alors, par cristallisation et filtration, de la DL-allo-2-dichlorométhyl-4-p-nitrophényl- hy droxyméthyl-oxazoline-2, laquelle, après la vage à l'eau et séchage à 100 C, fond à 167 à. 168 C et constitue la forme racémique du produit de départ.
On peut préparer l'isomère optique L-allo du produit de départ en opé rant de façon analogue, mais en partant du chlorhydrate de L - allo -1- p - nitrophényl - 2 - a.mino-propanediol-1,3.
Les exemples suivants montrent comment le procédé selon l'invention peut être mis en ceuvre. Exemple <I>1:</I> On chauffe pendant 20 minutes sur un bain-marie 6 g de DL-allo-2-diclilorométhyl-4- p-nitrophénylhydroxyméthyl-oxazoline-2 (p. f. = 167-168 C) avec 45 em3 de dioxane an- hydre contenant 15 % d'acide chlorhydrique sec.
On verse ce mélange sur 200 em3 d'eau, on essore le produit qui cristallise, lave à l'eau et sèche dans le vide sur de l'acide sul furique. On obtient 5 g de DL-allo-1-p-nitro- phényl-2-dichlor acétamido-3-chloro-propanol-7 du point de fusion<B>133</B> à 134 C.
<I>Exemple. 2:</I> 0n dissout 1,86 g de L-allo-2-dichloro- méthyl-4-p- riitrophénylhydroxt-méthy 1- oxa- zoline-2 (p. f. =142 C, [a] D =-3 7 [e = 4% clans l'acétone])
à la température ordinaire dans 19 cm?, de dioxane contenant 13% d'acide chlorhydrique sec. On laisse reposer pendant trois heures, on chasse le solvant et l'excès d'acide chlorhydrique par évaporation sous pression réduite à température ordinaire. On reprend le résidu par 2 cm3 de méthanol et on ajoute 25 cm-' d'eau. Il cristallise un pro duit qu'on filtre, lave à l'eau et. sèche dans le vide sur de l'acide sulfurique.
On obtient ainsi 1,36 g de L-allo-1-p-nit.rophényl-2-diehloracét- amido-3-chloro-propaziol-1 du point de fusion 114 à 116 C; [a] ri = - 12 36 (e = 4 % clans le méthanol).
Exerrzple <I>3:</I> En opérant. dans les mêmes conditions que dans l'exemple 2, mais en utilisant 0,62 g clé D - allo - 2 - diehlor ométhyl - 4 - p -nitrophényl- hydroxyméthyl-oxazoline-2, on obtient, 0,27 -çr de D-allo-1-p-nitrophényl-2-dichloraeétamido- 3-cliloro-propanol-1;

<I> Example. 2: </I> 0n dissolve 1.86 g of L-allo-2-dichloromethyl-4-p-riitrophenylhydroxt-methyl-oxazoline-2 (mp = 142 C, [a] D = -3 7 [e = 4% in acetone])
at room temperature in 19 cm ?, of dioxane containing 13% dry hydrochloric acid. Allowed to stand for three hours, the solvent and the excess hydrochloric acid are removed by evaporation under reduced pressure at room temperature.
