Le 1-p-nitrophényl-2-dichloracétamido-pro- panediol-1,3 utilisé à titre de matière pre mière et les esters représentés par la formule précédente peuvent exister sous forme d'iso mères de structure (thréo et érythro) ainsi que sous forme d'isomères optiques.
La pré sente invention vise la préparation d'esters de la forme diastéréoisomérique thréo , dis tincte de la forme érythro . Les groupes fixés sur les deux atomes de carbone asymé triques des isomères thréo desdits esters pos sèdent la même configuration ou le même arrangement spatial relatif que les groupes fixés sur les deux atomes de carbone asymé- triques de la pseudo-éphédrine -et de la thréose. En raison de la difficulté de représenter ces différences de structure sous forme gra phique,
on utilisera les formules développées courantes, avec une indication placée au-des sous ou à côté de la formule, désignant la con figuration particulière du composé. Quand figure l'indication forme thréo , comme ci dessus, ceci indique que l'on doit interpréter la. formule dans son sens générique, c'est-à-dire comme représentant les isomères thréo dextro gyre et lévogyre, sous forme séparée ainsi que sous forme de leur mélange racémique. Ce dernier est ïme nouvelle substance cristallisée; la. forme racémique (DL) fond à.
90-91 C, tandis que sa forme optiquement active D fond à 90 C et possède un pouvoir rotatoire [a] D = + 5 l dans l'acétate d'éthyle.
Toutes les formes optiques (D, L et DL) de ce nouveau composé cristallisé présentent des propriétés thérapeutiques importantes et sont utilisées éomme antibiotiques dans le même but que le chlorainphénicol. Les iso mères DL et D sont particulièrement appro priés à l'emploi dans des buts thérapeutiques. Le produit obtenu par le procédé selon l'in vention ne présente pas du tout le goût très fort et désagréable, semblable à celui de la quinine, du chloramphénicol et de ses esters 3-0-aeylés contenant 7 ou moins de 7 atomes de carbone dans le groupe acyle.
En outre, le composé obtenu selon l'invention est chimique ment stable et peut être administré sous foi-me de suspensions, d'élixirs, etc., à des enfants et à d'autres malades qui ne sont pas capables d'avaler des capsules et des pilules enrobées. La dose usuelle est de 25 à 250 oing par kilo et par jour, suivant la nature et l'intensité de la maladie traitée.
Lorsqu'on utilise dans le procédé selon l'invention comme dérivé de l'acide palmitique un halogénure de palmitoyle ou l'anhydride de l'acide palmitique, la réaction est de pré férence effectuée dans des conditions anhy dres. On obtient les meilleurs résultats lors qu'on utilise ium catalyseur basique tel que la pyridine, la triéthylamine, la N,N#-diméthyï- aniline, la N=éthylpipéridine ou une amine tertiaire analogue.
On a également constaté qu'un halogénure de palmitoyle réagit plus lentement et donne des rendements plats éle vés en le produit désiré que l'anhydride pal mitique. La température utilisée pour le pro cédé peut varier dans d'assez larges limites, mais en général, il n'est pas recommandable d'utiliser une température excédant 150 C. Lorsqu'on emploie un catalyseur basique, il est rarement nécessaire de chauffer le mé lange réactionnel à une température supé rieure à 60 C puisque la réaction procède assez rapidement à des températures compri ses entre environ 20 et. 35 C.
On obtient les meâlleurs résultats lorsqu'on utilise deq quantités approximativement. équivalentes de l'agent d'acylation et du propanediol de dé part. Il va de soi qu'on peut utiliser un excès jusqu'à environ 25 % de l'in ou de l'autre des produits mis en réaction, mais on doit prendre soin d'éviter des excès trop grands de l'agent d'acylation.
Des trop grands excès de ce dernier diminuent sensiblement le r en dernent en le produit désiré par le fait de la tendance à former des produits diacylés dans ces conditions. Pour effectuer l'acylation, on pourrait aussi utiliser l'acide palmitique en présence d'acide chlorhydrique sec ou en pré sence de P205, en formant ainsi le chlorure ou l'anhydride de l'acide palmitique in situ.
Le procédé selon l'invention est illustré plus en détail par les exemples suivants: <I>Exemple 1:</I> On ajoute 1674 g de chlorure de palmi- toyle à 1870 g de D-(-)-thréo-1-p-nitrophé- r_yl - 2 - dichloracétamido - propane -1,3 - diol (ehloramphénicol) dans 2700 cm3 de pyridine et on agite la solution pendant ime heure. On verse le mélange dans 16 litres d'eau et on recueille le solide.
La recristallisation du pro duit brut au sein du benzène donne le D-(+ )- thréo-1-p-nitrophényl - 2 - dichloracétamido- 3 palmitoyloxy-propane-1-ol à l'état pur; point de fusion 90 C: [a] = 5 I. dans l'acé- D tate d'éthyle.
Exemple <I>2:</I> On ajoute 9 g de chlorure de palinitoyle à une solution de 10 g de DL-thréo-1-p-nitro- phényl - 2 - dichloracétamido-propane-1,3 -diol (racémique du chloramphénicol) dans 40 cm3 de pyridine et on abandonne au repos le mé lange ainsi obtenu à la température ambiante pendant vingt minutes.
On verse le mélange réactionnel dans 400 cm3 d'eau; le DL-thréo- I - p - nitrophényl ..2 -dichloracétamido- 3 - pal mitoyloxy-propane-1-ol se sépare et est pari- fié par recristallisation au sein de benzène.
<I>Exemple 3:</I> On chauffe au reflux (environ 82 C) pen dant vingt heures un mélange formé de 3,2 g de D - (-) - thréo -1-p - nitrophényl -2-dichlor- acétamido-propane-1,3-diol (chloramphénicol), 2,9 g de chlorure de palmitoy le et 200 cens de benzène. On concentre la solution limpide à un volume de 1.00 cms et on étend au moyen d'éther de pétrole.
Le D-(+)-thréo-1-p-nitro phényl-2-dichloracétamido-3-palmitoyloxy-pro- pane-1-ol qui se sépare est recueilli et purifié par recristallisation au sein de benzène; point de fusion 90 C; [a] D --- 5 1 dans l'acétate d'éthyle.
Exemple On ajoute 30 g de chlorure de palmitoyle à un mélange composé de 32 g de D-(-) thr éo-1-p-nitrophény1 a<B>(</B> 2-diehloracétamido-pro- pane-1,3-diol, 50 cm3 de benzène et 9 cm-3 de pyridine. On agite le mélange pendant un court instant à la température ambiante, puis on l'étend au moyen d'un volume égal d'éther. On lave le mélange à l'eau, on le sèche et on le concentre.
