Description

  

  Procédé de résolution du     thréo-1-p-nitrophényl-2-amino-propanediol-1,3        racémique     en \ses isomères optiques.    La présente invention a pour objet -un  nouveau procédé de résolution du     thréo-1-p-          nitrophényl-2-amino-propanediol-1,3    racémi  que en ses isomères optiques.  



  Le dédoublement de ce     produit    en ses iso  mères optiques par l'intermédiaire de sels  d'acides     optiquement-    actifs a déjà été décrit  dans la littérature (cf. par exemple Am. Soc.  71, p. 2467 [1949] et le brevet des États-Unis  d'Ain. N  2483884).  



  On a trouvé que ce dédoublement pouvait  être réalisé d'une façon plus économique et  avec un rendement comparable en produit       optiquement    actif de pureté égale, sans avoir  recours à l'emploi d'un acide     optiquement     actif.  



  Selon le procédé de l'invention, la résolu  tion est obtenue en transformant cette base  racémique en un sel et en soumettant ce sel,  en solution aqueuse et en présence d'un sel  d'un de ses     isomères    optiques, à une précipi  tation fractionnée au moyen d'une     solution     alcaline.  



  Selon le mode de réalisation préféré du  procédé de l'invention, on dissout     soins    forme  de sels (par exemple chlorhydrates ou ni  trates) un mélange de base racémique et de  base dextrogyre et on opère     ensuite    une pré  cipitation fractionnée des bases par addition  d'une solution alcaline. Il précipite d'abord  une base dextrogyre presque     pitre,    puis de la  base     lévogyre    mélangée à un peu de base racé-         inique    et ensuite la base racémique renfer  mant encore     une    faible quantité de base dex  trogyre.

   La base lévogyre obtenue dans la  deuxième fraction est ensuite purifiée par  précipitation fractionnée de la solution  aqueuse de son     chlorhydrate.        Ces    précipita  tions sont     effeetuées    à température ordinaire.  



  La quantité de base dextrogyre à ajouter       peut        être        comprise        entre        25        et        100        %        en        poids          de        la        base        racémique,        mais        c'est        avec        50%     qu'on a obtenu les rendements optima.  



  Les bases dextrogyre     (1''e    fraction) et racé  mique     (3-e    fraction) sont réutilisées dans  l'opération suivante.  



  Chacun des deux isomères optiques que  l'on obtient d'après le procédé de résolution  peut servir de produit intermédiaire pour la  préparation de composés à activité antibioti  que. .  



  Outre que le procédé selon l'invention pré  sente l'avantage de mettre en     oeuvre    des acides  courants tels que l'acide chlorhydrique, qui  sont bien meilleur marché que les acides     opti-          quement    actifs, il permet de travailler en so  lution aqueuse concentrée et à température  ordinaire.  



  Lorsqu'on opère en présence d'un sel de la  base lévogyre au lieu d'un sel de la base  dextrogyre, la séparation se fait moins facile  ment.  



  L'exemple suivant montre     comment    le pro  cédé de l'invention peut être réalisé en pra-      tique. Les pouvoirs rotatoires ont été mesurés  sur une solution aqueuse du     chlorhydrate    à       une        concentration        de        2,4%        en        base.     



       Exemple:     Dans 600     cm3    d'eau et 57,5     cin3    d'une  solution d'acide chlorhydrique à 443. g par  litre, on dissout en chauffant vers 70  100 g  de     thréo-1-p-nitrophényl-2-amino-propanediol-          1,3    racémique et 50 g de la base dextrogyre  correspondante. On traite la solution au noir  animal et on filtre. Lorsque la température  est redescendue vers 50 , on coule en une  demi-heure 28     cm3    d'une solution de  soude à -397 g par litre. On agite pendant  une nuit en maintenant la température  à 20 . Le précipité qui se forme est filtré, lavé  avec 50     cms    d'eau et séché.

   On obtient ainsi  62 g de base dextrogyre presque pure  [a] =     -27 .     



  D On réunit les eaux mères et les eaux de  lavage et précipite une deuxième fraction en  ajoutant 10     en-i3    de la même solution de soude.  On agite pendant une nuit à 20 , filtre le pré  cipité formé et lave avec 50     cm-3    d'eau. On  obtient ainsi 31,5     g    de produit [a] D = -16  6       renfermant        environ        58        %        de        base        lévogyre.     



  Les eaux-mères sont traitées avec 32     cm3     de soude, donnant une troisième fraction  [a]<B>"</B> =     -I-    1  66 constituée en majeure partie  de base racémique.  



  La base     lévogyre        constituant    la deuxième  fraction est purifiée de la façon suivante:  74 g de base     lévogyre    impure provenant  de plusieurs opérations sont     dissous,    vers 60  à 70 ,     dans    236 cm- d'eau et 60     cm3    de solu  tion d'acide chlorhydrique à 209,8 g par litre.    On précipite une première fraction par coulée  de 38,5     cms    de soude diluée à 200,8 g par  litre. On agite pendant une nuit. à 20 , filtre  et lave avec 25     cm3    d'eau.

   Après séchage dans  une étuve à vide à 60 , on obtient 35     g    de  base lévogyre [a]     21    = - 26  2. Cette base est.  à un état de pureté convenable pour la pré  paration du     chloramphénicol.     



  Des eaux-mères on récupère encore 32,2 g  de base [a] D =     -10     qui est purifiée avec la  base impure provenant. d'une opération ulté  rieure.

  The splitting of this product into its optical isomers by means of salts of optically active acids has already been described in the literature (cf. for example Am. Soc. 71, p. 2467 [1949] and the patent of United States of Ain. N 2483884).



  It has been found that this resolution can be achieved in a more economical manner and with a comparable yield of optically active product of equal purity, without having to resort to the use of an optically active acid.



  According to the process of the invention, the resolution is obtained by transforming this racemic base into a salt and by subjecting this salt, in aqueous solution and in the presence of a salt of one of its optical isomers, to fractional precipitation. using an alkaline solution.



  According to the preferred embodiment of the process of the invention, a mixture of racemic base and dextrorotatory base is dissolved in the form of salts (for example hydrochlorides or trates) and then a fractional precipitation of the bases is carried out by addition of an alkaline solution. It precipitates first an almost clogged dextrorotatory base, then of the levorotatory base mixed with a little racemic base and then the racemic base still containing a small amount of dex trorotatory base.

   The levorotatory base obtained in the second fraction is then purified by fractional precipitation of the aqueous solution of its hydrochloride. These precipitations are carried out at ordinary temperature.



  The amount of dextrorotatory base to be added may be between 25 and 100% by weight of the racemic base, but it is with 50% that the optimum yields have been obtained.



  The dextrorotatory (1''e fraction) and racemique (3-rd fraction) bases are reused in the following operation.



  Each of the two optical isomers obtained from the resolution method can serve as an intermediate for the preparation of compounds with antibiotic activity. .



  In addition to the fact that the process according to the invention has the advantage of using common acids such as hydrochloric acid, which are much cheaper than optically active acids, it makes it possible to work in a concentrated aqueous solution and at room temperature.



  When operating in the presence of a salt of the levorotatory base instead of a salt of the dextrorotatory base, the separation takes place less easily.



  The following example shows how the process of the invention can be carried out in practice. The rotatory powers were measured on an aqueous solution of the hydrochloride at a concentration of 2.4% by base.



       Example: In 600 cm3 of water and 57.5 cin3 of a hydrochloric acid solution at 443. g per liter, it is dissolved by heating at around 70 100 g of threo-1-p-nitrophenyl-2-amino-propanediol - 1.3 racemic and 50 g of the corresponding dextrorotatory base. The solution is treated with animal charcoal and filtered. When the temperature has fallen to 50, 28 cm3 of a sodium hydroxide solution at -397 g per liter are poured in in half an hour. Stirred overnight while maintaining the temperature at 20. The precipitate which forms is filtered, washed with 50 cms of water and dried.

   62 g of almost pure dextrorotatory base [a] = -27 are thus obtained.



  D The mother liquors and the washing liquors are combined and a second fraction is precipitated by adding 10 en-i3 of the same sodium hydroxide solution. Stirred overnight at 20, filter the precipitate formed and washed with 50 cm-3 of water. In this way 31.5 g of product [a] D = −16 6 is obtained, containing approximately 58% of levorotatory base.



  The mother liquors are treated with 32 cm3 of soda, giving a third fraction [a] <B> "</B> = -I- 1 66 consisting mainly of a racemic base.



  The levorotatory base constituting the second fraction is purified as follows: 74 g of impure levorotatory base originating from several operations are dissolved, at around 60 to 70, in 236 cm 3 of water and 60 cm 3 of hydrochloric acid solution at 209.8 g per liter. A first fraction is precipitated by pouring 38.5 cm 3 of sodium hydroxide diluted to 200.8 g per liter. Stir overnight. at 20, filter and wash with 25 cm3 of water.

   After drying in a vacuum oven at 60, 35 g of levorotatory base [a] 21 = - 26 2 are obtained. This base is. in a state of purity suitable for the preparation of chloramphenicol.



  A further 32.2 g of base [a] D = -10 is recovered from the mother liquors, which is purified with the impure base obtained. of a subsequent operation.
