herstellen.

) Alu miniumchlorid in 300 g trockenem Nitrobenzol setzt man 74,3 g (0,5 mol.)
Analyse:

EMI0001.0005
Due to its bronehospasmolytic properties, it is used in therapy to cure asthma. As is known, it can be done in a simple manner by reducing the corresponding ketone

produce. The reduction is preferably carried out by catalytic hydrogenation, e.g.
B. with nickel, palladium or platinum as a catalyst. After Corrigan c. s. (D. Am. Chem. Soe., 71, 530, 1949) the ketone yields moderate yield by reacting a large excess of the tertiary butylamine with 1- (3'4'-dioxyphenyl) -2-chlorine -acetophenone obtained.
The latter compound can by means of the difficult reaction of pyrocatechin with. Chloracetyl chloride are formed.
In the process of the present invention, better yields can be achieved than according to the precaution given by Corrigan, while the goal is achieved with a significantly smaller amount of the relatively expensive tertiary butylamine.
The process of the present invention for the preparation of 1- (3'4'-dioxyphenyl) -2-tertiary butylamino-ethanol is characterized in that a compound of the formula
EMI0001.0035
where R1 and R2 are alkyl groups, is reacted with the compound of the formula N = C - CHz 'NH - C - (CH3) 3 in anhydrous solution in the presence of aluminum chloride,
in the compound of the formula thus obtained
EMI0001.0045
the imino group is converted into the keto group by hydrolysis and the latter is converted into the secondary alcohol group by reduction, the groups R10- and R20- being converted into OH groups by hydrolysis before the keto group is converted into the secondary alcohol group.
You can also use a salt of aminonitrile with a strong inorganic acid, preferably hydrochloric acid, when carrying out the process. The inventive condensation is preferably carried out in a solvent with pronounced polarity that does not itself participate in the reaction, such as. B. nitrobenzene, ortho-nitrotoluene and ortho-nitroanisole. Nitrobenzene is preferably used as the solvent.
The reaction time when carrying out the process according to the invention is between a few hours and a few days; the reaction temperature is preferably kept between 0 and 70.degree. When using easily split ethers, it can happen that one or both ether groups are saponified during the condensation.
Embodiment: A solution of 150 g (1.13 mol.) Of aluminum chloride in 300 g of dry nitrobenzene is added 74.3 g (0.5 mol.) Of tertiary biitylaminoacetonitrile hydrochloride and 69 g (0.5 mol.) Of vegetable oil . Then, over a period of 6 hours, with stirring and cooling (temperature below 30 ° C.), dry hydrogen chloride gas is introduced into the reaction liquid.
When the introduction of hydrogen chloride gas has ended, the reaction mixture is left to stand overnight. The viscous reaction liquid is then poured into 750 cm3 of water. After cooling, the crystals formed are filtered off with suction and washed with a mixture of 7 parts by volume of acetone and 3 parts by volume of alcohol.
50 g of 1- (3'4'-dimethoxyphenyl) -2-tertiarybutylamino-ethanone hydrochloride are obtained, i.e. H. 35% of the theoretical yield. The substance melts at 228.5-229.5 C with decomposition. It can be further purified by crystallization from ethanol. will. Analysis:

46 g of this substance are demethylated with 140 cm3 of a 480% hydrogen bromide solution. The resulting hydrobromide is dissolved in 200 cm3 and the solution is made ammoniacal. The base is filtered off with suction, suspended in 75 cm3 and 2-N sulfuric acid is added until the solid substance is completely dissolved.
The solution was then evaporated to dryness and acetone was added to the residue. 24 g of 1- (3'4'-dioxy-phenyl) -2-tert-butylamino-ethanone sulfate are obtained. The substance melts at 253-254 C with decomposition.
15g of 1- (3'4'-dioxy-phenyl) -2-tertiary-bi-itylamino-ethanone sulfate are dissolved in as little water as possible and the solution with 1.5 g of active charcoal and a solution of 250 mg of palladium chloride in water (with 3 drops of concentrated hydrochloric acid). The mixture is shaken with hydrogen at normal pressure until the calculated amount of hydrogen is absorbed. After the hydrogen uptake has ended, the catalyst is suctioned off and the pH of the filtrate is adjusted to a value of 6-7 using aqueous ammonia.
The solution is then evaporated to dryness in vacuo under nitrogen and the residue is crystallized by crystallization from absolute methanol. 11 g of 1- (3'4'-dioxyphenyl) - 2 - tert-butylamino - ethanol sulfate are obtained, i.e. H. 73% of the theoretical yield. The substance melts at 157-157.5 C with decomposition. The free base can be produced from the sulfate using aqueous ammonia.
