EMI0001.0005
Process for production of a new indoline. The present patent relates to a process for the preparation of an indoline, which is characterized in that the compound of the formula
EMI0001.0005
treated with methyl bromide.
The 1,1-dimethyl-6- (N-methyl-carbamyloxy) indolinium bromide of the formula obtained in this way
EMI0001.0009
that melts between 158 and 170 is new. It has a pharmacological effect similar to physostigmine and is said to be used as a medicinal product.
Example: 1.75 parts by weight of 1-methyl-6-oxy-indoline are mixed with 0.1 parts by weight of triethylamine and then with 4 parts by volume of methyl isocyanate in portions with cooling, with gradually all the substance in Solution works. The mixture is then left to stand at 20 for 24 hours, some of the 1-methyl-6- (N-methyl-carbamyloxy) indoline of the formula
EMI0001.0023
precipitates as coarse crystals.
Recrystallized from ether and sublimed in a high vacuum at 0.1 mm Hg / 80, it melts at 98.
To recover the remaining parts, the crystals are suctioned off, the mother liquor dissolved in 100 parts by volume of ether and extracted twice with 20 parts by volume of 2N sodium hydroxide solution with the addition of ice in an atmosphere of nitrogen.
The ether solution is washed three times with water and saturated sodium chloride solution, dried with sodium sulfate and evaporated in a water jet vacuum, the residue crystallizing. The above-mentioned compound is likewise obtained from the raw crystals by sublimation, crystallization and repeated sublimation.
1 part by weight of 1-methyl-6- (N-methyl-carbamyloxy) indoline is heated to 80 in a sealed tube with 5 parts by weight of methyl bromide in 5 parts by weight of benzene during 12-15 hours. This gives 1,1-dimethyl-6- (N-methyl-carbamyloxy) indolinium bromide of the formula
EMI0002.0001
which after recrystallization from a mixture of methanol and ether melts between 158 and 170.
The 1-methyl-6-oxyindoline used as starting material can be obtained as follows: 60 parts by weight of 1-methyl-6-oxy-oxindole [represented analogously to the 5-oxy compound according to Julian, Pikl and Wantz, Am.
Soc. 57, 2029 (1935)] are dissolved warm in 2000 parts by volume of tetrahydrofuran, cooled, slowly added to 60 parts by weight of lithium aluminum hydride in 300 parts by volume of ether in an atmosphere of carbon dioxide-free stick materials and the mixture is refluxed for 4 hours.
After cooling ver, one sets carefully with 600 parts by weight of ice, then with further cooling with 120 parts by volume of concentrated hydrochloric acid, the tetrahydrofuran is distilled on the water bath, diluted with 600 parts by volume of water and a further 120 parts by volume of concentrated hydrochloric acid are added. The Congo-acidic solution is extracted three times with 500 parts by volume of ether each time, with the exclusion of air, brought to a pH of about 9 with concentrated aqueous ammonia and extracted with ether.
The latter ether extract is washed with water and saturated sodium chloride solution, dried with sodium sulfate and evaporated. The oily residue is distilled at 12 mm Hg J 171-175, a colorless, slowly turning yellowish oil passing over, which solidifies after some time. Recrystallized from ether and sublimed twice at 0.1 mm pressure and 100, the melts 1- Met.h@#1-6-oxy-indoline at 94-97. It is extreme against air and light. sensitive.
