Exemple 2: On ajoute graduellement 9 g d'anhydride a - (n - butyl) -,a - éthylsuccinique à 10 cm3 d'hydroxyde d'ammonium concentré et 10 cm3 d'éthanol, à température ordinaire.
Process for -preparation of succinimide. The invention relates to a process for the preparation of a succinimide having chemotherapeutic activity, a- (n-butyl) -a-ethylsuccinimide, of formula
EMI0001.0008
According to the invention, this siiccinimide is prepared by reacting a- (n-butyl) -a-ethylsuccinic acid or a- (n-butyl) anhydride
-a-ethylsuccinic with at least one chemical equivalent of ammonia, usually in the form of ammonium hydroxide, and heating the intermediate product thus formed to a temperature between 100 and 350 C.
For carrying out the first part of the process, it is advantageous, although not essential, to use an inert solvent such as water, a lower aliphatic alcohol or a mixture of such alcohols: ether, benzene , toluene or the like, to facilitate the reaction of suecinic acid or anhydride with ammonia.
It is recommended to use several equivalents of ammonia per equivalent of a- (n-butyl) -a-ethyl succinic acid or anhydride. The intermediate products are different depending on which of the two above-mentioned raw materials was used and also on the amount of ammonia used.
When a- (n-butyl) -a-ethylsuccinic anhydride is employed with one equivalent of ammonia, the intermediate product is the half-amide, while if two or more equivalents of ammonia are employed, the intermediate product is the ammonium salt of α- (n-butyl) -α-ethylsuccinic acid half-amide.
In the case where a- (n-butyl) -a-ethylsuccinic acid is used as the raw material, the intermediate product is the mono- or di-ammonium salt of a- (n-butyl) acid. -a-ethyl-succinic. These intermediates can be converted to the desired succinimide by heating them to a temperature between 100 and 350 C.
The preferred temperature for this conversion is between 190 and 210 C, at which temperature the reaction is complete after about one hour.
The product obtained by the process according to the invention is a new substance having a melting point of 59-61 C and has valuable therapeutic properties and is particularly useful for the treatment of the mild illness type of epilepsy.
The invention is illustrated by the following examples Example 1: 10 g of a- (n-butyl) ra-ethylsuccinic acid are gradually added to 10 cms of concentrated ammonium hydroxide and then removed by distillation of water and excess ammonia. The temperature of the residue is raised and maintained at 190 ° C. until no more ammonia is given off.
The cooled residue is dissolved in ether, heated with animal charcoal and filtered, and the filtrate is diluted with petroleum ether. On cooling, it precipitates a white product, which is the desired a- (n-butyl) - a-ethylsuccinimide; P. F. 59-61 C. Yield: 33 % .
Example 2: 9 g of a - (n - butyl) -, a - ethyl succinic anhydride are gradually added to 10 cm3 of concentrated ammonium hydroxide and 10 cm3 of ethanol, at room temperature . Ethanol and water are removed from the reaction mixture by distillation, and the residue is heated at 200 C for about an hour.
The cooled residue is then treated as described in Example 1, and the desired α- (n-butyl) = α-ethylsuccinimide is obtained; Mp. 59-61 C. Yield: 30%.
