Verfahren zur Herstellung von wässerigen Lösungen, welche eine Additionsverbindung von 4-Dimethylamino-antipyrin enthalten Die Erfindung betrifft ein Verfahren zur Herstellung von wässerigen Lösungen, welche eine Additionsverbindung aus 4-Dimethy-1- amino-antipyriri (1-Phenyl-2,3-dimethyl-4-di- methylamino-pyrazolon) und einem Lösungs vermittler enthalten, welches ermöglicht, sta bile, verdünnbare sowie mischbare Lösungen hoher Konzentration mit 4-Dimethylamino- antipyrin-Wirksamkeit herzustellen, bei denen der verwendete,
chemisch an das 4-Dimethyl- amino-antipyrin gebundene Lösungsvermitt ler die physiologische Wirkung des 4 - Di- methylamino-antipyrins unterstützt.
Das 4-Dimethylaniino-antipyrin besitzt be kanntlich eine geringe Wasserlöslichkeit; seine gesättigte wässerige Lösung enthält höchstens 51/o des Stoffes. Für therapeutische Zwecke - insbesondere für parenterale Anwendung - ist jedoch die Herstellung höher konzen trierter wässeriger Lösungen des 4-Dimethyl- amino-antipyrins von grosser Bedeutung, wes halb bereits verschiedentlich versucht wurde, die Wasserlösliehkeit dieses Stoffes durch Zu gabe eines Lösungsvermittlers zu steigern.
Die bisher hierzu verwendeten Stoffe wichen jedoch in ihrer chemischen Struktur von 4- Dimetlivlamino-antipyrin meist sehr erheblich ab, so dass sich entweder die therapeutische Indikation des Ausgangsstoffes änderte oder unerwünschte Nebenerscheinungen eintraten. Trotz der erzielten höheren Konzentration der Lösung waren also die Ergebnisse keines wegs befriedigend.
Nach dem Verfahren gemäss der Erfin dung lassen sich nun diese Nachteile beseiti gen. Das erfindungsgemässe Verfahren ist da durch gekennzeichnet, dass 4-Dimethylamino- antipyrin in Gegenwart von 4-Amino-anti- pyrin in Wasser gelöst wird, und zwar in zusammen mindestens lüprozentiger Konzen tration, wobei sich eine Additionsverbindung zwischen den genannten Komponenten bildet.
Die Erfindung beruht auf der Feststel lung, dass bei Einbringen eines Gemisches oder einer Schmelze von 4-Dimethylamino- antipyrin und 4-Amino-antipyrin in Wasser sich eine Additionsverbindung aus je einem Mol dieser Komponenten bildet, welche besser wasserlöslich ist als 4-Dimethylamino-anti- pyrin, so dass eine beständige wässerige Lö sung hoher Konzentration entsteht. Die Ver- wendVng einer mit dem Ausgangsstoff che misch sehr ähnlichen Verbindung als Lösungs vermittler bringt weitere Vorteile mit sich.
plan verwendet in der Regel beide Kom ponenten in praktisch äquimolekularen Men gen, da die Additionsverbindung aus j e einem Mol der Komponenten sich bildet. Da die Molekulargewichte der beiden Komponenten relativ wenig verschieden sind und sich wie 1 : 0;88 verhalten, verwendet man in der Regel praktisch gleiche Gewichtsmengen der beiden Komponenten. Dass eine Additionsverbindung vorliegt, geht aus dem Schmelzpunktdia- gramm sowie aus dem Absorptionsspektrum hervor.
Die so entstehende, wässerige Lösung der chemischen Additionsverbindung besitzt in pharmakologischer Hinsicht alle Eigenschaf ten des 4-Dimethylamino-antipyrins.
Falls man nicht äquimolekulare Mengen der Ausgangsstoffe verwendet, so erhält man eine Lösung, die neben der Additionsverbin dung noch die überschüssige Komponente ge löst enthält.
Nach dem erfindungsgemässen Verfahren können also stabile wässerige Lösungen der Additionsverbindung hergestellt werden, die mehr als je 5 %, vorzugsweise mehr als je 1.5 %- jeder der beiden Komponenten enthal- ten, also Lösungen,
deren Gesamtkonzentra- tion an beiden Komponenten mindestens 10 %, vorzugsweise über 30 % beträgt.
Weiter erweist es sich bei der nach der Erfindung hergestellten Lösung als besonders günstig, dass bei Aufnahme von 4-Dimethyl- amino-antipyrin durch den Organismus 4-- Amino-antipyrin als Abbauprodukt entsteht und dieses bei wesentlich geringerer Toxizi- tät praktisch die gleiche physiologische Wir kung hat wie 4-Dimetbylamino-antipyrin. Die wässerige Lösung einer Additionsverbindung beider Substanzen muss daher die gleichen physiologischen Eigenschaften aufweisen wie eine 4-Dimethylamino-antipyrin-Lösung glei cher Gesamtkonzentration,
das heisst also des doppelten Gehaltes an 4-Dimethyla.mino-anti- pyrin allein.
So wurde zum Beispiel eine stabile Lö sung von je etwa 25 0/0 4-Dimethylamino- antipyrin und 4-Amino-antipyrin erhalten, die also nach dem oben Gesagten in ihrer thera peutischen Wirkung einer etwa 50prozentigen Lösung von 4-Dimethylamino-a.ntipyrin ent spricht. Dieses überraschende Ergebnis be deutet jedenfalls einen wesentlichen Fort- schritt gegenüber den bisher bekannten Resul taten.
Die nach der Erfindung hergestellten Lö sungen zeigen nicht nur eine auffallend gute Stabilität gegen Verdünnung, sondern auch gegen Zusatz von Säuren und Laugen sowie insbesondere auch bei Mischung mit. Penicillin, Sulfonamiden oder andern Injektionspräpa raten, wie vor allem auch mit Vitamin Bi, Vitamin-B-Komplex sowie Coffeinsalzen. Diese vorteilhaften Eigenschaften des Präparates sind darauf zurückzuführen, dass die Lö sung beider Komponenten nicht - wie viel fach bei andern Präparaten - durch Bildung eines Alkalisalzes, also durch Ionisierung, sondern durch Bildung einer Additionsverbin dung vor sieh geht,
wodurch eine besondere Beständigkeit gegen physikalische und che mische Einflüsse gewährleistet wird.
Auf Grund umfangreicher Versuchsreihen haben sich verschiedene Varianten des erfin dungsgemässen Verfahrens bewährt..
So kann entweder eine Mischung der bei den Komponenten oder auch eine aus beiden Substanzen hergestellte Schmelze in Wasser gelöst werden, welche Massnahme sieh insbe sondere zur Erreichung extrem hoher Kon zentrationen als vorteilhaft erweist.
Auch das getrennte Einbringen beider Stoffe nacheinander in Wasser ist selbstver ständlich ohne weiteres möglich.
Die Temperatur ist für den Ablauf des Verfahrens von untergeordneter Bedeutung, so dass die Lösung sowohl bei Zimmertem peratur als auch unter Kühlung oder Er wärmung erfolgen kann.
Bei gleichfalls schwerlöslichen Gemischen von 4-Dimethylamino-antipyrin mit andern Stoffen - wie solche vielfach therapeutische Verwendung finden - wird eine analoge Ver besserung der Löslichkeit in Wasser erzielt, wenn 4-Amino-antipyrin dazugegeben bzw. ein Teil des 4-Dimethylamino-antipyrins durch 4-Amino-antipyrin ersetzt wird. Als Zusatz stoffe - die selbstverständlich ebenso gut auch der fertigen Lösung zugesetzt werden können -kommen vor allem Salicylate, Bar- bitursäure- oder Purinderivate in Betracht.
<I>Beispiel 1</I> Je 10 Teile 4-Dimethylamino-antipyrin und 4-Amino-antipyrin werden gemischt und da nach in Wasser zu 100 Teilen gelöst, so dass eine auf 4-Dimethylamino-antipyrin bezogen 10prozentige, insgesamt - also in der thera peutischen Wirkung - jedoch 20prozentige Lösung entsteht.
<I>Beispiel 2</I> Eine Schmelze von je 2.5 Teilen 4-Dime- thylamino-antipyrin und 4-Amino-antipyrin wird in Wasser zu 100 Teilen gelöst, so dass eine insgesamt 50prozentige Lösung entsteht. <I>Beispiel 3</I> In eine wässerige Suspension von 4-Di- methylamino-antipyrin wird die gleiche Menge pulverisiertes 4-Amino-antipyrin in kleinen Portionen eingerührt oder umgekehrt.
<I>Beispiel 4</I> Im gewünschten Mengenverhältnis wird eine eventuell unter Erwärmung bereitete Lö sung von 4-Amino-antipyrin mit 4-Dimethyl- amino-antipyrin versetzt oder umgekehrt.
<I>Beispiel 5</I> Ein Gemisch von 15 Teilen 4-Dimethyl- amino-antipyrin, 15 Teilen 4-Amino-antipyrin und 6 Teilen Coffei-- am-natrium-benzoicum wird in Wasser zu 100 Teilen gelöst. In glei- eher Weise kann man auch bei andern Zu sätzen als C offein vorgehen.

4-Dimethylaniino-antipyrine is known to have a low solubility in water; its saturated aqueous solution contains not more than 51% of the substance. For therapeutic purposes - especially for parenteral use - the production of higher concentrated aqueous solutions of 4-dimethylamino-antipyrine is of great importance, which is why various attempts have already been made to increase the solubility of this substance in water by adding a solubilizer.
The substances used to date for this purpose, however, differed in their chemical structure from 4-dimetlivlamino-antipyrine mostly very considerably, so that either the therapeutic indication of the starting material changed or undesired side effects occurred. Despite the higher concentration of the solution achieved, the results were by no means satisfactory.
According to the method according to the invention these disadvantages can now be eliminated. The method according to the invention is characterized in that 4-dimethylamino-antipyrine is dissolved in water in the presence of 4-amino-antipyrine, in total at least 1% Concentration, an addition compound being formed between the components mentioned.
The invention is based on the determination that when a mixture or a melt of 4-dimethylamino-antipyrine and 4-amino-antipyrine is introduced into water, an addition compound is formed from each mole of these components, which is more water-soluble than 4-dimethylamino- antipyrine, so that a constant aqueous solution of high concentration is created. The use of a compound that is chemically very similar to the starting material as a solubilizer has further advantages.

The solutions prepared according to the invention show not only a strikingly good stability against dilution, but also against the addition of acids and alkalis and especially when mixed with. The separate introduction of the two substances one after the other in water is of course easily possible.
The temperature is of minor importance for the course of the process, so that the solution can take place both at room temperature and with cooling or heating.
In the case of similarly sparingly soluble mixtures of 4-dimethylamino-antipyrine with other substances - as such are often used therapeutically - an analogous improvement in solubility in water is achieved if 4-amino-antipyrine is added or part of the 4-dimethylamino-antipyrine is carried out 4-amino-antipyrine is replaced. Salicylates, barbituric acid or purine derivatives are particularly suitable as additives - which can of course also be added to the finished solution.
