Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von a-Dialkylamino-aeetaniliden der Formel :

In den obigen Formeln haben RI, R2. R3, R4 und X die gleiche Bedeutung wie weiter oben beschrieben, während Hal ein Halogenatom bedeutet.
Vorzugsweise verwendet man als Alkyl magnesiumhalogenid das Äthylmagnesium- bromid, und als Dialkylaminoessigsäureester einen Dialkylaminoessigsäureäthylester.
Bei der Herstellung von Acetaniliden aus den Magnesiumhalogenidverbindungen von Anilin oder dessen im Benzolkern alkylsubsti- tuierten Derivaten und einem Fettsäureester wurde die letzte Verfahrensstufe, das heisst die Hydrolyse der erhaltenen Komplexverbin- dung, üblicherweise mittels Säuren durchge- führt. Die Darstellung von a-Dialkylamino acetaniliden unter Anwendung von Dialkvl- aminofettsäureestern nach der Grignard-Me- thode war indessen bisher nicht gelungen, indem sieh nach der sauren Hydrolyse eine Abtrennung des Reaktionsproduktes als unmöglich erwies.
Es war daher völlig überrasehend, dal3 naeh dieser methode α-Dialkylamino-acet- anilide bequem und in guter Ausbeute zugänglich sind, wenn man den Hydrolyseschritt entweder in neutralem Medium, wie Wasser, oder besonders zweekmässig in ganz sehwach sauren Medien, wie sie zum Beispiel die Hydrolyseacidität eines Ammoniumsalzes liefert, ausführt.
Die Erfindung wird dureh die fogenden Beispiele näher erläutert :
Beispiel 1
Diäthylamino-acet-2,6-dimethyl-anilid (Lidocain)
Zu einer Lösung von Äthylmagnesium- bromid, hergestellt dureh Umsetzung von 38, 2 g Magnesiumspäne und 191 g Äthylbromid in 1100 em3 abs. Äther, werden bei 0 unter guter Aussenkühlung, 153 g vie.-meta Xylidin zugetropft. Unter lebhafter Gasentwicklung (Äthan) ist die Reaktion in kurzer Zeit beendet.
Man lässt das erhaltene Reak tionsprodukt (vic.-meta-Xylidin-magnesinm- bromid) 1/2 Stunde bei Zimmertemperatur stehen, kühlt wieder auf 0 ab, und lässt, unter Schiitteln, eine Lösung von 160g Diäthylaminoessigsäureäthylester in 400 cm3 abs. Äther rasch eintropfen. Schliesslich erhitzt man 1/2 Stunde am Rückflusskühler, um die Reaktion zu beendigen. Man kühlt wieder auf 0 ab und zersetzt das gebildete Komplexsalz unter gutem Schütteln und Zusatz von 500 cm3 Eiswasser, welches 2, 5 g Ammoniumehlorid enthält. Die Athersehicht wird abgetrennt und über Natriumsulfat t getrocknet.
Ahan dampft dann den Äther weg. Der Rück- stand, eine Mischung von Lidoeain und vic. meta-Xylidin, wird dann im Vakuum fraktioniert. Man erhält bei 5 mm Druck 2 Fraktionen : eine bei 90-95 (vie.-meta-Xylidin) und die zweite bei 182-183 (Lidocain).
Schmelzpunkt F = 68, 5-69 .
Ausbeute : bis 90% der Theorie.
Beispiel 2 Dimethylamino-acet-anilid
Diese Verbindung wird analog wie im Bei spiel 1 aus 3, 8: g Magnesium, 19, 1 g Äthbyl- bromid und 11, 7 g Anilin in 115 cm3 abs.
Äther (Herstellung von Anilin-magnesiumbromid) und 13, 1 g Dimethylaminoessigsäure- äthylester und naehfolgende Hydrolyse des erhaltenen Komplexsalzes, hergestellt. Aus becte : 83% ; F = 38 .
Beispiel 3
Dimethylamino-acet-2,6-dimethyl-anilid
Die Herstellung erfolgt analog wie im Beispiel 1 aus Xylidin-magnesium-bromid und Dimethylaminoessigsäureäthylester und nachfolgender Hydrolyse.
Ausbeute : 87% ; F = 75, 5 .

In the above formulas, RI, R2. R3, R4 and X have the same meaning as described above, while Hal denotes a halogen atom.
Ethylmagnesium bromide is preferably used as the alkyl magnesium halide and an ethyl dialkylaminoacetate as the dialkylaminoacetic acid ester.
In the production of acetanilides from the magnesium halide compounds of aniline or its derivatives alkyl-substituted in the benzene nucleus and a fatty acid ester, the last process stage, that is the hydrolysis of the complex compound obtained, was usually carried out by means of acids. The preparation of α-dialkylamino acetanilides using dialkylamino fatty acid esters according to the Grignard method, however, had hitherto not been successful, since separation of the reaction product after acidic hydrolysis proved impossible.
It was therefore completely surprising that, with this method, α-dialkylamino-acet-anilides can be obtained conveniently and in good yield if the hydrolysis step is carried out either in a neutral medium, such as water, or, especially, in very weakly acidic media, such as those used in Example provides the hydrolysis acidity of an ammonium salt.
The invention is illustrated in more detail by the following examples:
example 1
Diethylamino-acet-2,6-dimethyl-anilide (lidocaine)
To a solution of ethylmagnesium bromide, prepared by reacting 38.2 g of magnesium shavings and 191 g of ethyl bromide in 1100 em3 abs. Ether, 153 g of vie.-meta xylidine are added dropwise at 0 with good external cooling. The reaction is over in a short time with vigorous evolution of gas (ethane).
The reaction product obtained (vic.-meta-xylidine-magnesine-m-bromide) is left to stand for 1/2 hour at room temperature, cooled again to 0, and a solution of 160 g of ethyl diethylaminoacetate in 400 cm3 of abs is left under shaking. Drip in ether quickly. Finally, the mixture is heated on the reflux condenser for 1/2 hour in order to terminate the reaction. It is cooled again to 0 and the complex salt formed is decomposed with thorough shaking and the addition of 500 cm3 of ice water which contains 2.5 g of ammonium chloride. The ether layer is separated off and dried over sodium sulfate.
Ahan then steams the ether away. The residue, a mixture of Lidoeain and vic. meta-xylidine, is then fractionated in vacuo. Two fractions are obtained at 5 mm pressure: one at 90-95 (vie.-meta-xylidine) and the second at 182-183 (lidocaine).
Melting point F = 68.5-69.
Yield: up to 90% of theory.
Example 2 Dimethylamino-acet-anilide
This compound is made analogously to Example 1 from 3, 8: g magnesium, 19.1 g ethyl bromide and 11.7 g aniline in 115 cm3 abs.
Ether (production of aniline magnesium bromide) and 13.1 g of ethyl dimethylaminoacetate and subsequent hydrolysis of the complex salt obtained. From becte: 83%; F = 38.
Example 3
Dimethylamino-acet-2,6-dimethyl-anilide
The preparation takes place analogously to Example 1 from xylidine magnesium bromide and dimethylaminoacetic acid ethyl ester and subsequent hydrolysis.
Yield: 87%; F = 75.5.
