EMI0001.0042
in which R, Ari and Arz have the meanings given above, condensed, the substituent R hydrolyzed to an oxoalkyl group and the latter being converted into an oxyalkyl radical by reduction.
The compounds that can be prepared according to the invention have therapeutically interesting properties, in particular antipyretic, analgesic and antiphlogistic effectiveness.
An embodiment of the process according to the invention is characterized in that a substituted 1-valonic acid ester of the formula II, in which X and x2 are lower alkoxy radicals and R has the meaning given above, is treated with a hydrazobenzene or
a hydrazobenzene derivative of the formula
EMI0001.0070
where Z1 and Z are hydrogen or easily cleavable acid radicals, in particular Z1 is an acetyl radical and Ari and Arz have the meaning given above, ring-closed in the presence of a basic, preferably an alkaline condensation agent to give a compound of the formula IV.
One can also proceed in such a way that one. a substituted malonic acid dilialide of the formula II, in which XI and X are chlorine or bromine, in the presence of a base, suitable for acid binding agent based on hydrazobenzene or a substituted hydrazobenzene of the formula
EMI0002.0017
can act.
Instead of substituted malonic acid diesters or dihalides, substituted malonic acid ester halides of the formula II, in which X1 is a lower alkoxy group and X # is chlorine or bromine, can also be used as starting materials,
and in a first stage corresponding to the second embodiment mentioned in the presence of an acid-binding agent to give substituted 3Ialonsänreester-hy draziden of the formula
EMI0002.0031
where X is a hydrocarbon radical, in particular a lower alkyl radical, is condensed and, in the second stage, corresponding to the first-mentioned embodiment, if necessary, heated in the presence of an alkaline condensing agent to close the ring. will.
The conversion of the substituent R in the -1 position of the pyrazolidine ring into an oxyalkyl group for conversion to compounds of formula I of the condensation products obtained takes place by hydrolysis and reduction.
The choice of the starting materials of the formula II and thus the hydrolysis and reduction processes necessary for converting the intermediate products of the formula IV into the end substances depend in particular on the distance between the hydroxyl groups present in the end substance and the pyrazolidine ring and on whether these are primary, should be of a secondary or tertiary nature.
Groups which can be converted into the hydroxyl group by hydrolysis and reduction are the carbalkoxy groups and aeetalized oxo groups.
The carbalkoxy group to be converted into the hydroxyl group (the group must be located in the "" position or further away from the two geminal carbalkoxy groups, since an aetalised oxo group can also be closer to the geminal carbalkoxy groups. groups are located.
The oxo group can as well as with two molecules of a monohydric alcohol. a molecule of a glycol, most advantageously with. ltliyleii - lyko1, zii be connected to an acetal group.
The new 1,2-diaryl-4-oxyalkyl-3,: 5-dioxopyrazolidines are mostly colorless crystalline substances, which see both common organic solvents and, due to the presence of an acidic hydrogen atom, as enolation in dilute aqueous solutions Dissolve alkalis easily, and also with other inorganic ingredients,
as well as easily form monobasic salts with organic bases. In this context, a peculiarity of compounds according to the invention with a hydroxyl group on the pyrazolidine ring;) should be pointed out (see example under c).
When a solution of an alkali salt of the reduction product of 1.2-1) i-plienyl-4 - ('O'oxo-butyl) -3.5-dioxo-pyrazolidine is acidified, a low-cutting compound (Snip. F9-91) can be isolated which is similar Lich rage how analogous compounds with the 11y droxylgruppe in another position of the 4-alkyl radical can be dissolved in sodium hydroxide solution again with salt formation.
However, if the precipitated substance is allowed to crystallize in an organic solvent, or if it is left to stand in an acidic medium for a longer period of time, then one obtains substances with a higher melting point, which dissolve very slowly in aqueous alkalis.
B. from acetone at 16:; - 16-1. Spectrometric investigations as well as the great differences in the melting points as well as in the rate of dissolution in alkalis make it possible that we are not just looking at a very careful demarcation between enolforni and ketoforni,
the latter optionally as a cyclobalba acetal of the formula
EMI0003.0021
acts, but that there is a deeper difference.
While the low-grade form is probably the keto form according to general formula I or, if applicable, the tautomeric enol form:
#n acts, it is to be assumed that the higher, incising forms do not contain a yi-azolidine ring, but the ss-lactone ring, i.e. This means that it looks at the ß-caprolactone-a-carboxylic acid-N.N'-diplnenylhydrazide of the formula
EMI0003.0039
acts.
Successful by brief heating in an alkaline medium. In any case, it is necessary to loosen the higher melting forms again and to keep the lower melting form from them by acidification and rapid separation.
It can therefore be assumed that the salts of 1,2-1) iplienyl-4- (y-oxy-a.lkyl) -3,5-dioxo-pyrazolidines have the same structure, at least in aqueous solution, as the salts of z. B. 1,2-diphenyl-4-alkyl-3,5-dioxo-pyrazolidines, regardless of which form of the free connec tion the salts were obtained by dissolving in alkalis.
In the following examples, parts mean parts by weight; these are related to parts of volume like g to Gina. The temperatures are given in centigrades.
Examples a) 1,2-1) iphenyl-4- (3 ',' -äthylenedioxy-butyl) -3,5-dioxo-pyrazolidine. 274 parts (3,3-ethylene-dioxy-butyl) -malonic acid-d.iäthy-lester are abs in 100 parts by volume. Dissolved benzene and mixed with 57 parts of sodium metal derivative and 184 parts of hydrazobenzene. @There is warming. The reaction mass is 15 hours. Long cooked on the Rüek river.
After cooling, it is poured into water, separated off and the aqueous portion is washed twice with benzene. The benzene solutions are washed 3 times with 'normal sodium carbonate solution and the combined aqueous solutions are acidified with normal hydrochloric acid. The precipitated 1,2-diphenyl-4- (3 ', 3' -äthylenedioxy-butyl) -3,5-dioxo-pyrazolidine can be recrystallized from alcohol. M.p .: 165-167.
b) 1., 2-1) iphenyl-4- (3'-oxobutyl) -3,5-dioxo-p3-razolidine. 36.6 parts of 1,2-diphenyl-4- (3 ', 3'-äthylendioxy-buty 1) -3,5-dioxo-pyr azolidine who in 750 parts by volume of acetone with 0.35 parts of p-toluenesulfonic acid for 7.8 hours heated at the back. Then the solution is filtered, mixed with 1500 parts of water and left to stand at 5 for 24 hours.
The precipitated 1,2-diphenyl-4- (3'-oxobutyl) -3,5-dioxopyrazolidine is filtered off with suction and washed with 50% acetone: mp 115.5-116.5 from alcohol-water ( Occasionally a crystal form melting at 127.5-128.50 is obtained.
c) 1,2-Diphenyl.-4- (3'-oxyblityl) -3,5-dioxopyrazolidine. By reduction with sodium borohydride: 50 parts of 1,2-diphenyl-4- (3'-oxobutyl) -3,5-dio, xo-pyrazolidine become methanol and 171 parts by volume in 1300 parts by volume Dissolved in sodium hydroxide solution and mixed with 250 parts by volume of 10% methanolic sodium borohydride solution.
This solution is left to stand for 24 hours, with water cooling to ensure that the temperature does not exceed 40. Then it is filtered and with. 2500 parts by volume of 2N hydrochloric acid. After 48 hours of standing at 5, the mixture is filtered off with suction and the 1,2-diphenyl-4- (3'-oxy-butyl) -3,5-dioxo-pyrazolidine is washed with 50% strength methanol. From acetone: 163 to 164.
From the aqueous-inethanolisehen mother liquor, additional amounts of the end product can be obtained by pouring it out with chloroform.
The crystallized compound from Sinp. i 163--164 is rather difficult to dissolve in alcohol, somewhat more easily in acetone, and even more easily in chloroform. In the cold, aqueous or alcoholic aqueous alkalis do not dissolve until after hours of shaking. The dissolution process can be accelerated considerably by gentle heating.
A compound is obtained from the alkaline-aqueous solution by rapid precipitation with acid which melts at 89-91 and dissolves very easily in alcohol and acetone, as well as in aqueous alkalis. By crystallizing from alcohol and acetone, the compound of melting point 163-164 is obtained again.
If, in the above example, one works in aqueous instead of aqueous-methanolic solution, then upon precipitation of finite 2N hydrochloric acid the readily alkali-soluble compound which melts at 89-91 is obtained. B. acetone can be converted into the preparation melting at 163 to 164 ".
By catalvtisehe reduction 5 parts of 1,2-diphenyl-4- (3'-ohobutyl) -3,5-dioao-pyrazolidine are dissolved in 100 parts of alcohol and 50 parts of 1N NaOH and added to 5 parts. Nickel catalyst (Railey) shaken at normal temperature and normal pressure in an H .. atmosphere. After absorption of the calculated amount H, comes. the hydrogenation practically comes to a standstill.
The catalyst is filtered off and 50 parts of 2N HCl are added to the filtrate. After standing for several days, at 5, the precipitated 1, 2-diplienyl-4- (j '-oty btityl) -3, 5-dioxo-pyra-zolidine is sucked off and finite 50% alcohol is washed out. The product is with the by reduction with. NiBH4 manufactured identically.
