  Verfahren zur Herstellung neuer Aldehyd-Derivate
Gegenstand des schweiz. Patentes   Nr.    306510 ist ein Verfahren zur Herstellung eines neuen Aldehydderivates, das erhalten wird durch Umsetzung von Methionin mit Formaldehyd und eine bedeutende bakterizide und bakteriostatische Wirkung aufweist.



   Es wurde nun gefunden, dass die Umsetzungsprodukte, wie sie erhalten werden durch Umsetzung von schwefelfreien   Monoamino-mono- oder -dicarbon-    säuren oder Diamino-monocarbonsäuren mit Aldehyden, die 1-10 C-Atome enthalten, in alkalischer bis neutraler Lösung in freier Form oder als Salze ebenfalls eine bakterizide und bakteriostatische Wirkung gegen eine grosse Anzahl von gram-negativen und gram-positiven Bakterien besitzen, die weit stärker ist als die der einzelnen Reaktionskomponenten, selbst wenn diese gemeinsam appliziert werden,
Als aliphatische, schwefelfreie Monoaminomonokarbonsäuren kommen z. B.   oder      bB-Alanin,    Glykokoll usw., als Monoamino-dikarbonsäuren z. B.



  Glutaminsäure, Asparaginsäure usw. und als Diamino-monokarbonsäuren z. B. Arginin oder Lysin in Betracht.



   Als Aldehyde mit   1--10    Kohlenstoffatomen kann man sowohl aliphatische als auch karbozyklische oder heterozyklische Aldehyde verwenden. Bevorzugt werden Formaldehyd, Acetaldehyd, Benzaldehyd, Anisaldehyd, Furfural, Nitrofurfural usw.



   Zur Durchführung der Umsetzung ist die Anwesenheit äquivalenter Mengen an Basen nicht er  forderlich;    es genügt, wenn das an sich saure Reaktionsgemisch so weit alkalisiert wird, dass sein   pH-Wert    mindestens 7 beträgt.



   Als Basen können z. B. Metallhydroxyde verwendet werden, wobei diejenigen der Alkalimetalle mit dem entstandenen Aldehyd-Aminosäure-Umsetzungsprodukt in Wasser leicht lösliche Salze bilden, während die mit Erdalkalien entstehenden Salze schwerer löslich sind. Die Erdalkalisalze können auch in einfacher Weise durch doppelte Umsetzung der Alkalisalze mit Erdalkalisalzlösungen erhalten werden.



   Beispiel 1
10 Teile a-Alanin, suspendiert in 50 Teilen Wasser, werden durch Zugabe von 15 Teilen   1SB/oiger    Natronlauge unter Erwärmen auf 50 bis 600 C in Lösung gebracht. Dann gibt man bei dieser Temperatur unter Rühren 8,5 Teile 40   Vol.o/cr    Formaldehydlösung zu und lässt unter Rühren auf Zimmertemperatur abkühlen.



   Die erhaltene klare Lösung filtriert man anschliessend zur Entfernung der Na-ionen durch eine mit einem Kationen-Austauschharz (wie  Amberlite  oder  Dowex  [Markenprodukte]) beschickte Säule, engt das Filtrat im Vakuum weitgehend ein und isoliert das N-Oxymethyl-cc-alanin durch Ausfällung mit Aceton. Es bildet nach Umkristallisieren aus wässrigem Aceton farblose Kristalle, die in Wasser leicht löslich sind.



   Beispiel 2
Verwendet man bei einem Ansatz gemäss Beispiel 1 zur Alkalisierung 22,5 Teile   200/obiger    Natronlauge (= 1 Äquivalent), so erhält man ein Re  aktionsgemisch,    aus welchem durch Eindampfen im Vakuum direkt das Na-Salz des N-Oxymethyl-aalanins gewonnen werden kann. Dieses stellt ein farbloses, bis   3000 C    nicht schmelzendes Pulver dar, das in Wasser mit alkalischer Reaktion sehr leicht löslich ist.



   Beispiel 3
In eine Suspension von 10 Teilen a-Alanin in 50 Teilen Wasser trägt man unter Rühren 4,5 Teile festes Calciumhydroxyd ein. Dann lässt man unter Rühren 8,5 Teile 40   Vol.O/o    Formaldehydlösung zutropfen, engt im Vakuum weitgehend ein und bringt  das entstandene N-Oxymethyl-a-alanin durch Behandlung mit abs. Alkohol zur Kristallisation. Die Verbindung stellt ein farbloses, bis   300  C    nicht schmelzendes Pulver dar, das in Wasser mässig löslich ist.



   Verwendet man unter sonst gleichbleibenden Bedingungen in den vorstehenden Beispielen statt a-Alanin das   fi-Alanin,    so erhält man N-Oxymethyl  fl-alanin,    das ähnliche Eigenschaften aufweist wie das isomere a-Derivat.



   Beispiel 4
Zu einer Lösung von 10 Teilen Glykokoll in 75 Teilen   4 0/obiger    Natronlauge gibt man 10 Teile 40   Vol.O/o    Formaldehydlösung und lässt anschliessend mehrere Stunden stehen.



   Die erhaltene klare Lösung filtriert man anschliessend zur Entfernung der Na-ionen durch eine mit einem Kationen-Austauschharz (wie  Amberlite  oder  Dowex ) beschickte Säule, engt das Filtrat im Vakuum weitgehend ein und isoliert das N-Oxymethyl-glykokoll durch Ausfällung mit Aceton. Es bildet nach Umkristallisieren aus wässrigem Aceton farblose Kristalle, die in Wasser leicht löslich sind.



   Beispiel 5
Verwendet man bei einem Ansatz gemäss Beispiel 4 zur Auflösung 90 Teile   6 0/obiger    Natronlauge (= 1 Äquivalent), so erhält man ein Reaktionsgemisch, aus welchem durch Eindampfen im   Va-      kuum    direkt das Na-Salz des N-Oxymethyl-glykokolls gewonnen werden kann. Dieses stellt ein farbloses, bis   300o    nicht schmelzendes Pulver dar, das in Wasser mit alkalischer Reaktion sehr leicht löslich ist.



   Beispiel 6
In eine Lösung von 10 Teilen Glykokoll in 50 Teilen Wasser trägt man unter Rühren 5,5 Teile festes Calciumhydroxyd ein. Dann lässt man unter Rühren 10 Teile 40   Vol.O/o    Formaldehydlösung zutropfen, engt im Vakuum weitgehend ein und bringt das entstandene N-Oxymethyl-glykokoll durch Behandlung mit abs. 10 Teile Glutaminsäure, suspendiert in 40 Teilen Wasser, werden durch Zugabe von 20- Teilen   15 0/obiger    Natronlauge in Lösung gebracht. Dann gibt man unter Rühren 6 Teile 40   Vol.O/o    Formaldehydlösung zu, lässt mehrere Stunden stehen und filtriert die erhaltene klare Lösung anschliessend zur Entfernung der Na-ionen durch eine mit einem Kationenaustauschharz (wie    Amberlite      oder  Dowex ) beschickte Säule. Das Filtrat wird im Vakuum eingeengt, dann trennt man eventuell ausgeschiedene Glutaminsäure ab und isoliert das Reaktionsprodukt durch Ausfällung mit abs. Alkohol. Man erhält so die N-Oxymethyl-glutaminsäure als farblose, in Wasser leicht lösliche Kristalle.



   Beispiel 8
Verwendet man bei einem Ansatz gemäss Beispiel    7 zur Alkalisierung 36 Teile 15 15 /oiger Natronlauge    (= 2 Äquivalente), so erhält man ein Reaktionsgemisch, aus welchem durch Eindampfen im Vakuum direkt das Dinatriumsalz der N-Oxymethylglutaminsäure gewonnen werden kann. Dieses stellt ein farbloses, bis   300o C    nicht schmelzendes Pulver dar, das in Wasser mit alkalischer Reaktion sehr leicht löslich ist.



   Beispiel 9
10 Teile Glutaminsäure, suspendiert in 50 Teilen Wasser, werden durch Zugabe von 50 Teilen 150/oiger Kalilauge in Lösung gebracht, dann gibt man unter Rühren 6 Teile 40   Vol.  /o    Formaldehydlösung und später 20 Teile 400/oige Calciumchloridlösung zu. Nach mehrstündigem Stehen wird das Reaktionsprodukt durch Zugabe von 30 Teilen Alkohol ausgefällt, abgenutscht und wiederholt mit verdünntem Alkohol gewaschen. Das so erhaltene Dicalciumsalz der N-Oxymethylglutaminsäure ist ein farbloses, bis   300o C    nicht schmelzendes Pulver, das in Wasser mässig löslich ist.



   Beispiel 10
10 Teile Asparaginsäure, suspendiert in 40 Teilen Wasser, werden durch Zugabe von 25 Teilen   15  /oiger    Natronlauge in Lösung gebracht. Dann gibt man unter Rühren 5,6 Teile 40   Vol.O/o    Formaldehydlösung zu, - lässt mehrere Stunden stehen und filtriert die erhaltene klare Lösung anschliessend zur Entfernung der Na-ionen durch eine mit einem Kationenaustauschharz   (wie  Amberlite       oder   Dowex  )    beschickte Säule. Das Filtrat wird im Vakuum eingeengt, dann isoliert man das Reaktionsprodukt durch Ausfällung mit abs. Alkohol und erhält so die N-Oxymethyl-asparaginsäure als farblose, in Wasser leicht lösliche Kristalle.



   Beispiel 11
Verwendet man bei einem Ansatz gemäss Bei   spiel 10 zur Alkalisierung 40 Teile 15 15 /oige Natron-    lauge (= 2 Äquivalente), so erhält man ein Reaktionsgemisch, aus welchem durch Eindampfen im Vakuum direkt das Dinatriumsalz der N-Oxymethylasparaginsäure gewonnen werden kann. Dieses stellt ein farbloses, bis   300o C    nicht schmelzendes Pulver dar, das in Wasser mit alkalischer Reaktion sehr leicht löslich ist.



   Beispiel 12
In eine Suspension von 10 Teilen Asparaginsäure in 70 Teilen Wasser trägt man unter Rühren 6 Teile festes Calciumhydroxyd ein, eventuell vorhandene Verunreinigungen werden aus der entstandenen Lösung durch Filtration entfernt. Man lässt nun 6 Teile 40   Vol.O/o    Formaldehydlösung zufliessen und fällt nach einigem Stehen das Dicalciumsalz der N-Oxymethyl-asparaginsäure durch Zugabe von Alkohol aus. Dieses stellt ein farbloses, bis   300  C     nicht schmelzendes Pulver dar, das in Wasser mässig löslich ist.



   Beispiel 13
10 Teile Arginin-monochlorhydrat, gelöst in 60 Teilen Wasser, werden unter Rühren mit 3,5 Teilen festem Calciumhydroxyd versetzt, dann gibt man zu der entstandenen Lösung 7,2 Teile 40   Vol.8/o    Formaldehydlösung zu. Man lässt einige Stunden stehen, engt anschliessend im Vakuum weitgehend ein, fällt das Reaktionsprodukt durch Zugabe von Alkohol aus, nutscht ab und wäscht gründlich mit Alkohol.



  Das so erhaltene Formaldehydderivat des Arginin-Ca stellt ein farbloses, bis   300o C    nicht schmelzendes Pulver dar, das in Wasser mässig löslich ist.
