Description
Verfahren zur Gewinnung eines sedativen Wirkstoffes Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Gewinnung eines nachfolgend als Re- serpin bezeichneten sedativen Wirkstoffes aus Pflan zen der Rauwolfia-Arten in reiner Form insbeson dere aus dem Wurzelmaterial von Rauwolfia serpen- tina Benth.
Es wurde gefunden, dass man zu Reserpin in reiner Form gelangen kann, wenn man einen sedativ wirksamen, mit Hilfe eines Alkohols aus Pflanzen material von Rauwolfia-Arten erhaltenen, nicht mit Wasser behandelten Rückstand mit einem wässrigen, schwach sauren Mittel behandelt, wobei man eine wässrige Lösung erhält, welche das entsprechende Reserpinsalz enthält,
dass man das Reserpinsalz mit einem mit Wasser höchstens teilweise mischbaren organischen Lösungsmittel behandelt und dass man alsdann das Reserpin unter Verwendung eines Ad sorptionsmittels isoliert. Dabei kann man z. B. die wässrige saure Reserpinsalzlösung direkt mit einem mit Wasser höchstens teilweise mischbaren organi schen Lösungsmittel behandeln, oder man kann auch ein aus der wässrigen sauren Lösung gewonnenes festes Salz des Reserpins mit dem organischen Lö sungsmittel behandeln.
Reserpin bildet, aus Aceton in der Hitze um kristallisiert, schwach gelbliche Kristalle vom F. 262 bis 263 (unter Zersetzung); [a.]D - -117 (Chloro form). Es lässt sich auch aus einer Mischung von Chloroform und Äther umkristallisieren. Reserpin ist eine schwache Base und bildet ein schwerlösliches Hydrochlorid vom F. 22111 (unter Zersetzung) und ein Pikrat vom F. 183-186 . Das leichtlösliche Acetat ist nur in überschüssiger verdünnter Essig säure beständig.
Ausserdem ist das Reserpin unlös lich in Wasser und Petroläther, mässig löslich in Ben zol, Äther, kaltem Alkohol und kaltem Aceton und leicht löslich z. B. in Chloroform, 10%iger Essig- säure oder verdünnter Phosphorsäure.
Als Ausgangsmaterial bedient man sich z. B. eines rohen, zur Trockne eingedampften Alkohol- wie Methanolextraktes des fein zerriebenen Pflanzen materials von Rauwolfia-Arten, insbesondere des Wurzelmaterials von Rauwolfia serpentina Benth.
Man kann z. B. wie folgt vorgehen: Der oben genannte Extrakt wird mit dem wässrigen sauren Mittel, insbesondere der wässrigen Lösung einer nie deren Fettsäure, wie z. B. von Ameisensäure, Essig säure, Propionsäure, oder einer Phosphorsäure oder eines sauren Salzes einer mehrbasischen Säure, mehr mals extrahiert. Die so erhaltene sedativ wirksame saure Lösung kann zur Weiterverarbeitung entweder auf ein kleineres Volumen eingeengt oder mit Wasser verdünnt oder unverändert belassen werden.
Dieser Auszug wird dann mit einem mit Wasser höchstens teilweise mischbaren organischen Lösungsmittel, vor zugsweise mit einem halogenierten Kohlenwasser stoff, wie Chloroform, Äthylenchlorid, Methylen- chlorid oder Trichloräthylen, oder auch mit Benzol oder Essigester, mehrmals extrahiert. Die organische Lösung wird dann neutral gewaschen, wodurch die Base wieder freigesetzt wird, und zur Trockne ein gedampft.
Anschliessend wird der Eindampfrückstand an einem Adsorptionsmittel, insbesondere einem mit telstarken Adsorptionsmittel, wie mässig aktivem Alu miniumoxyd, Kieselsäure, Hyflo (Markenprodukt), oder einem andern Silikat, chromatographiert. Das Reserpin lässt sich durch Petroläther-Benzol-Ge- mische eluieren. Es kristallisiert in Gegenwart von wenig Aceton spontan. Die Reinigung kann z. B.
durch Umkristallisieren aus heissem Aceton, Alkohol oder aus einer Chloroform-Äther-Mischung vorge- nommen werden. Das Reserpin soll therapeutische Verwendung finden.
Im nachfolgenden Beispiel besteht zwischen Ge wichtsteil und Volumteil die gleiche Beziehung wie zwischen Gramm und Kubikzentimeter. Die Tempe raturen sind in Celsiusgraden angegeben.
<I>Beispiel</I> 1000 Gewichtsteile gemahlene Wurzeln von Rau- wolfia serpentina Benth. werden mit 1000 Volum- teilen Methanol angefeuchtet und drei- bis viermal mit je 2000 Volumteilen Methanol unter 10- bis 16- stündigem Rühren und jeweiligem Abnutschen extra hiert. Mit Vorteil wird von der zweiten Extraktion an auf etwa 65 erwärmt.
100 Gewichtsteile des eingedampften Methanol extraktes werden mit 70 Gewichtsteilen Hyflo Super Cel vermischt und mit 100 Volumteilen 15 01o iger Essigsäure angefeuchtet.
Hierauf wird das Gemisch drei- bis viermal mit je 150 Volumteilen 15o/oiger Essigsäure extrahiert, wobei es jeweils zer rieben, abgenutscht und der Rückstand mit 100 Volumteilen 15 % iger Essigsäure gewaschen wird. Die vereinigten Essigsäureauszüge werden mit total 750 Volumteilen Chloroform extrahiert, die Extrakt lösung mit Wasser gewaschen,
mit Natriumsulfat ge trocknet und im Vakuum zur Trockne eingedampft, wobei man 20 Gewichtsteile Rückstand erhält. Der Rückstand wird in 200 Volumteilen Benzol bei 30 bis 40 gelöst, vom ungelösten Rückstand abfiltriert und das Filtrat eingedampft. 34 Gewichtsteile des so erhaltenen Rückstandes werden in 400 Volum- teilen Benzol gelöst, mit 100 Volumteilen Petrol- äther versetzt, wobei ein Teil der Substanz ausflockt, und auf eine Säule von 800 Gewichtsteilen Alumi niumoxyd gegeben.
Dann wird mit einem Gemisch von Petroläther und Benzol im VolumverhäItnis 1:4 nachgewaschen, gegebenenfalls unter Druck, wobei zuerst ein inaktives hellgelbes Öl, hernach etwa 0,6 Gewichtsteile einer inaktiven Kristallfraktion vom F. 238-239 und anschliessend die oft von einem roten Farbstoff begleitete sedativ wirksame Substanz eluiert wird.
Nun wird das erste Lösungsmittelge- misch nacheinander durch Benzol und ein Gemisch von Benzol und Aceton im Volumverhältnis 2:1 ersetzt, wodurch der letzte Rest des aktiven Mate rials aus der Säule entfernt wird. Die Eluatfraktionen werden nach dem Eindampfen mit wenig Aceton ver setzt; sie kristallisieren dann meist innert kurzer Zeit. Durch Abnutschen und Waschen mit einem Gemisch aus gleichen Volumteilen Aceton und Äther und mit Aceton kann die kristallisierte Substanz leicht isoliert werden.
Man erhält 4,5-5 Gewichts teile eines Rohkristallisates vom F. etwa 250 . Die weitere Reinigung geschieht durch Umlösen aus hei ssem Aceton oder aus einem Gemisch von Chloro form und Äther, wobei das fast farblose Reserpin vom F. 262-263 (unter Zersetzung), [u] D =-118 (Chloroform), erhalten wird.

that the reserpine salt is treated with an organic solvent which is at most partially miscible with water and that the reserpine is then isolated using an adsorbent. You can z. B. treat the aqueous acidic reserpine salt solution directly with a water-at most partially miscible organic solvent, or you can also treat a solid salt of reserpine obtained from the aqueous acidic solution with the organic solvent.
Reserpine, crystallized from acetone in the heat, forms pale yellowish crystals with a melting point of 262 to 263 (with decomposition); [a.] D - -117 (chloro form). It can also be recrystallized from a mixture of chloroform and ether. Reserpine is a weak base and forms a sparingly soluble hydrochloride of F. 22111 (with decomposition) and a picrate of F. 183-186. The easily soluble acetate is only stable in excess dilute acetic acid.
In addition, reserpine is insoluble in water and petroleum ether, moderately soluble in benzene, ether, cold alcohol and cold acetone and easily soluble in z. B. in chloroform, 10% acetic acid or dilute phosphoric acid.
As a starting material one uses z. B. a raw, evaporated to dryness alcohol such as methanol extract of the finely ground plant material of Rauwolfia species, especially the root material of Rauwolfia serpentina Benth.
You can z. B. proceed as follows: The above extract is mixed with the aqueous acidic agent, in particular the aqueous solution of a fatty acid never used, such as. B. of formic acid, acetic acid, propionic acid, or a phosphoric acid or an acidic salt of a polybasic acid, extracted several times. The sedative acidic solution obtained in this way can either be concentrated to a smaller volume for further processing or diluted with water or left unchanged.
This extract is then extracted several times with an organic solvent that is at most partially miscible with water, preferably with a halogenated hydrocarbon such as chloroform, ethylene chloride, methylene chloride or trichlorethylene, or with benzene or ethyl acetate. The organic solution is then washed neutral, whereby the base is released again, and evaporated to dryness.
The evaporation residue is then chromatographed on an adsorbent, in particular a medium-strong adsorbent such as moderately active aluminum oxide, silica, Hyflo (branded product), or another silicate. The reserpine can be eluted with petroleum ether-benzene mixtures. It crystallizes spontaneously in the presence of a little acetone. The cleaning can e.g. B.
by recrystallization from hot acetone, alcohol or a chloroform-ether mixture. The reserpine is said to be used therapeutically.
In the example below, the relationship between part by weight and part by volume is the same as that between grams and cubic centimeters. The temperatures are given in degrees Celsius.
<I> Example </I> 1000 parts by weight of ground roots of Rauwolfia serpentina Benth. are moistened with 1000 parts by volume of methanol and extracted three to four times with 2000 parts by volume of methanol each with 10 to 16 hours of stirring and suction filtration. It is advantageous to heat to around 65 from the second extraction.
100 parts by weight of the evaporated methanol extract are mixed with 70 parts by weight of Hyflo Super Cel and moistened with 100 parts by volume of 150% acetic acid.
The mixture is then extracted three to four times with 150 parts by volume of 15% acetic acid, each time being rubbed and suction filtered and the residue is washed with 100 parts by volume of 15% acetic acid. The combined acetic acid extracts are extracted with a total of 750 parts by volume of chloroform, the extract solution is washed with water,
dried with sodium sulfate and evaporated to dryness in vacuo, giving 20 parts by weight of residue. The residue is dissolved in 200 parts by volume of benzene at 30 to 40, the undissolved residue is filtered off and the filtrate is evaporated. 34 parts by weight of the residue obtained in this way are dissolved in 400 parts by volume of benzene, 100 parts by volume of petroleum ether are added, some of the substance flocculating, and the solution is poured onto a column of 800 parts by weight of aluminum oxide.
Then it is washed with a mixture of petroleum ether and benzene in a volume ratio of 1: 4, if necessary under pressure, whereby first an inactive light yellow oil, then about 0.6 part by weight of an inactive crystal fraction of F. 238-239 and then often of a red dye accompanying sedative substance is eluted.
Now the first solvent mixture is successively replaced by benzene and a mixture of benzene and acetone in a volume ratio of 2: 1, whereby the last remainder of the active material is removed from the column. After evaporation, the eluate fractions are added with a little acetone; they then usually crystallize within a short time. The crystallized substance can easily be isolated by suction filtration and washing with a mixture of equal parts by volume of acetone and ether and with acetone.
4.5-5 parts by weight of crude crystals from F. about 250 are obtained. The further purification is done by dissolving from hot acetone or from a mixture of chloroform and ether, whereby the almost colorless reserpine of F. 262-263 (with decomposition), [u] D = -118 (chloroform), is obtained.
