Es wurde gefunden, dass man Verbindungen der Formel

erhalten kann, wenn man tertiäre Aralkylaminonitrile der Formel

mit Organomagnesiumhalogeniden der Formel Y-Mg-Halogen umsetzt und die erhaltenen Reaktionsprodukte hydrolytisch zersetzt.
In diesen Formeln bedeuten R, und R2 gleiche oder verschiedene Alkyl-, Oxyalkyl-, Cycloalkyl-, Aryl-oder Aralkylgruppen oder auch zusammen mit dem N einen gegebenenfalls ein weiteres Heteroatom enthaltenden heterocyclischen Ring, z. B. den Pyrrolidin-, Piperidinoder Morpholinring, Y einen gegebenenfalls substituierten Alkyl-, Cycloalkyl-, Aryl-oder Aralkylrest, R Wasserstoff, eine Alkyl-oder Alkoxygruppe und Z einen geraden oder verzweigten Alkylenrest mit 2 oder 3 Kohlenstoffatomen.
Die Umsetzung kann in der gleichen Weise und unter den gleichen Bedingungen wie in der Hauptpatentschrift angegeben durchgeführt werden.
Die tertiären Aralkylaminonitrile lassen sich nach bekannten Methoden (vgl. Houben-Weyl, Methoden der organ. Chemie, 4. Aufl., 8. Bd., S. 279 ff.) herstellen.
Umsetzungen von tertiären Aralkylaminonitrilen der obigen Formel sind aus der Literatur noch nicht bekannt.
Das erfindungsgemässe Verfahren bietet besondere Vorteile, wenn man von Nitrilen ausgeht, die aus technisch zugänglichen Aldehyden erhältlich sind und wenn die Grignard-Verbindungen ebenfalls leicht zu gänglich sind.
So ist das 1- (p-Isopropyl-phenyl)-2-methyl-3-dimethylamino-3-phenyl-propan der Formel

leicht aus Cyclamenaldehyd über das entsprechende 3- (p-Isopropylphenyl)-2-methyl-1-dimethylaminobutyronitril durch Umsetzung mit Phenylmagnesiumhalogenid zu gewinnen, während die Herstellung derselben Verbindung aus Phenyldimethyl-aminoacetonitril und 2- (p-Isopropyl-phenyl)-l-methyl-äthylma- gnesiumhalogenid der Formel

infolge der schlechten Zugänglichkeit der letzteren Substanz ausserordentlich erschwert wäre.
Die nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen wirken stark spasmolytisch, insbesondere auch am Uterus, coronar-und vasodilatorisch ; sie beeinflussen ausserdem den Blutdruck.
Beispiel 1 1, 5-Diphenyl-l-methyl-2-dimethylaminopentan

a) 2-Phenyl-2-methyl-1-dimethylamino-propionitril
Zu 73 g (0,5 Mol) Hydratropaaldehyd gibt man unter Schütteln 52 g (0,5 Mol) Natriumbisulfit als 40% ige wässrige Lösung, wobei sich eine feste Substanz abscheidet, die mit 74 g (0,5 Mol) Dimethylamin (40% ig) und 36 g Kaliumcyanid (0,55 Mol), in wenig Wasser gelöst, etwa 20 Std. geschüttelt wird.
Das entstandene feste Nitril wird abgesaugt und aus Petroläther umkristallisiert. F. = 130 . Ausbeute 87% d. Th. b) 1, 5-Diphenyl-l-methyl-2-dimethylaminopentan
Zu der in üblicher Weise aus 2,3 g Magnesiumspänen und 19,9 g y-Phenyl-propylbromid in 50 cm3 abs. Ather bereiteten Grignardverbindung gibt man portionsweise unter Rühren 9,4 g 2-Phenyl-2-methyl 1-dimethylamino-propionitril in 70 cm3 abs. Äther zu und erwärmt nach beendeter Zugabe noch ll/2 Std. unter Rückfluss. Nach dem Erkalten wird das Reaktionsgemisch durch verdünnte Salzsäure in Eis zersetzt. Man trennt die ätherische Schicht, die verworfen wird, ab und macht die wässrig-saure Lösung durch Zugabe von Ammoniak alkalisch.
Das abgeschiedene 61 wird mit Ather extrahiert und nach dem Verjagen des Lösungsmittels im Vakuum destilliert. Man erhält ein fast farbloses 01 vom Kpo, = 119 in einer Ausbeute von 64% d. Th.
Das durch Lösen des Amins in Ather, Versetzen mit ätherischer Salzsäure und Kristallisieren aus Äthanol/Äther erhältliche farblose Hydrochlorid schmilzt bei 130-131 .
Beispiel 2 l-Phenyl-3-piperidino-3- (4methoxy-phenyl)-propan

a) 3-Phenyl-l-piperidino-butyronitril
Zu 42,5 g (0,5 Mol) Piperidin, das mit Salzsäure neutralisiert wird, gibt man 73 g (0,5 Mol) Hydrozimtaldehyd und 36 g (0,55 Mol) Kaliumcyanid in konzentrierter wässriger Lösung und schüttelt das Gemisch über Nacht. Das entstandene 01 wird mit Äther extrahiert und nach dem Verjagen des Lösungsmittels im Vakuum destilliert. Das Nitril wird als gelbliches 01 vom Kpo. g = 134-135 in einer Ausbeute von 65% d. Th. erhalten.
4) 1-Phenyl-3-piperidino-3- (4-methoxy-phenyl)-propan
Zu einer Grignardverbindung aus 2,3 g Magnesiumspänen und 18,7 g p-Brom-anisol in 50 cm3 abs.
Benzol fügt man unter Rühren langsam 11,4 g 3 Phenyl-1-piperidino-butyronitril in 100 cm3 abs. Benzol und erwärmt noch 11/2 Std. auf 50-70 . Nach dieser Zeit zersetzt man durch Zugabe von Eis und verdünnter Salzsäure, trennt von der Benzolschicht ab, die verworfen wird, und fügt Ammoniak bis zur alkalischen Lösung zu. Das abgeschiedene 01 nimmt man in Benzol auf und destilliert es nach dem Abdampfen des Lösungsmittels im Vakuum. Man erhält das Amin als schwach gelbliches 01 vom Kpo, os = 146 in einer Ausbeute von 84% d. Th. Durch Behandlung mit ätherischer Salzsäure entsteht das Hydrochlorid als kristalline, farblose Substanz vom F. = 203-204 .
Beispiel 3 1- (4-Isopropyl-phenyt)-2-methyl-3-dimethylamino- butan

a) 3- (4-Isopropyl-phenyl)-2-methyl-l-dimethylamino- butyronitril
Die aus 95 g (0,5 Mol) Cyclamenaldehyd und 52 : (0,5 Mol) Natriumbisulfit als 40% ige wässrige Lösung erhaltene Anlagerungsverbindung wird mit 56 g (0,5 Mol) Dimethylamin als 40% ige wässrige Lösung und einer Lösung von 35 g (0,55 Mol) Kaliumcyanid in wenig Wasser versetzt und einen Tag gerührt. : entstandene Reaktionsprodukt wird in Äther aufgenommen und nach der Entfernung des Lösungsmittels i. V. destilliert. Das Nitril ist eine fast farblose Flüssigkeit vom Kplo = 165 . Die Ausbeute beträgt 80% d.
Th. b) 1- (4-Isopropyl-phenyl)-2-methyl-3-dimethylamino- butan.
Die aus 2,3 g Magnesium und 14,2 g Methyljodid erhaltene Grignardverbindung wird unter Rühren tropfenweise mit 12,2 g 3- (4-Isopropyl-phenyl)-2-me thyl-1-dimethylamino-butyronitril in Äther versetzt und noch eine Stunde unter Rückfluss erhitzt. Nach dieser Zeit wird wie unter Beispiel 2b beschrieben aufgearbeitet. Das Amin ist eine farblose Flüssigkeit vom Kpols = 96 . Ausbeute 8,0 g. Das farblose Hydrochlorid schmilzt bei 107 .
Beispiel 4
I-Phenyl-3-piperidino-dodecan

Zu der in üblicher Weise aus 2,3 g Magnesium und 20,7 g n-Nonylbromid hergestellten Grignardverbindung fügt man unter Rühren portionsweise 11,4g 3-Phenyl-1-piperidino-butyronitril in 70 cm3 abs. Äther zu und arbeitet wie in Beispiel lb beschrieben auf.
Das in einer Ausbeute von 69% d. Th. erhaltene gelbliche Amin hat einen Kpo, 03 = 151-152 . Das Hydrochlorid ist kristallin und farblos und schmilzt bei 159-160 .
Beispiel S 1, 4-Diphenyl-l-methyl-2-morpholino-butan

a) 2-Phenyl-2-methyl-1-morpholino-propionitril
Die Herstellung des Nitrils erfolgt wie in Beispiel la angegeben aus 36,5 g Hydratropaaldehyd, 26 g Natriumbisulfit, 21,8 g Morpholin und 18 g Kaliumcyanid. Das abgeschiedene feste Nitril wird abgesaugt, mit Ather gewaschen und mit Benzin ausgekocht. Man erhält farblose Kristalle vom F. = 125 in einer Ausbeute von 79% d. Th. b) 1, 4-Diphenvl-1-methyl-2-morpholino-butan
Zu der aus 2,3 g Magnesium und 18,5 g ss-Phenyl- äthylbromid in Xther bereiteten Grignardverbindung werden nach dem Verjagen des Äthers 50 cm3 abs.
Benzol und dann unter Rühren 11, 5g des obigen Nitrils in 70 cm3 abs. Benzol gegeben. Man erhitzt dann noch 2 Std. unter Rückfluss und arbeitet wie in Beispiel 2b beschrieben auf. Das Amin hat einen Kpo, 07 = 155-156 und bildet ein farbloses Hydrochlorid vom F. = 157 . Die Ausbeute beträgt 47% d. Th.
Beispiel 6 1- (4-Isopropyl-phenyl)-2-methyl-3-pyrrolidino-4-phenyl-butan

a) 3- (4-Isopropyl-phenylJ-2-methyl-1-pyrrolidino- butyronitril
Das Nitril wird, wie in Beispiel la angegeben, aus 47,5 g Cyclamenaldehyd, 26 g Natriumbisulfit, 17,8 g Pyrrolidin und 18 g Kaliumcyanid als feste Substanz erhalten, welche, aus Äthanol umkristallisiert, bei 99 schmilzt. Die Ausbeute beträgt 66% d.
Th. b) 1- (4-Isopropyl-phenyl)-2-methyl-3-pyrrolidino-4 phenyl-butan
13,5 g des Nitrils in 70 cm abs. Äther lässt man in die Grignardverbindung aus 2,3 g Magnesium und 12,6 g Benzylchlorid unter Rühren eintropfen und arbeitet wie in Beispiel lb beschrieben weiter auf Das Amin ist ein farbloses 01 vom Kpo, o4 = 153 . Die Ausbeute beträgt 73% d. Th. Das farblose Hydrochlorid schmilzt bei 172 . Ferner kann man als Ausgangsstoffe u. a. die in der folgenden Tabelle aufgeführten Aralkylaminonitrile der Formel

Process for the preparation of tertiary amines
It has been found that compounds of the formula

can be obtained if one tertiary aralkylaminonitriles of the formula

with organomagnesium halides of the formula Y-Mg-halogen and the reaction products obtained are hydrolytically decomposed.
In these formulas, R 1 and R 2 denote identical or different alkyl, oxyalkyl, cycloalkyl, aryl or aralkyl groups or, together with the N, a heterocyclic ring optionally containing a further hetero atom, e.g. B. the pyrrolidine, piperidine or morpholine ring, Y an optionally substituted alkyl, cycloalkyl, aryl or aralkyl radical, R hydrogen, an alkyl or alkoxy group and Z a straight or branched alkylene radical with 2 or 3 carbon atoms.
The reaction can be carried out in the same way and under the same conditions as indicated in the main patent specification.
The tertiary aralkylaminonitriles can be prepared by known methods (cf. Houben-Weyl, Methods of Organ. Chemistry, 4th edition, 8th volume, p. 279 ff.).
Conversions of tertiary aralkylaminonitriles of the above formula are not yet known from the literature.
The process according to the invention offers particular advantages when starting from nitriles which are obtainable from industrially available aldehydes and when the Grignard compounds are also easily accessible.

Ether-prepared Grignard compound is added in portions with stirring, 9.4 g of 2-phenyl-2-methyl-1-dimethylamino-propionitrile in 70 cm3 of abs. Add ether and, after the addition is complete, reflux for a further 11/2 hours. After cooling, the reaction mixture is decomposed by dilute hydrochloric acid in ice. The ethereal layer, which is discarded, is separated off and the aqueous-acidic solution is made alkaline by adding ammonia.
