Verfahren zur Herstellung neuer Phthalazine Gegenstand der Erfindung ist ein Verfahren zur Herstellung von 1-Hydrazino - 4 - (pyridyl-methyl)- phthalazinen, ihrer Salze und quaternären Ammo- niumverbindungen, vorzugsweise von 1-Hydrazino-4- [pyridyl-(2')- oder -pyrid'yl-(4')-methyl]-phthalazinen.
Die neuen Phthalazine besitzen wertvolle pharma kologische Eigenschaften. Sie zeigen eine ausge prägte hypotensive Wirkung und können dementspre chend als blutdrucksenkende Mittel therapeutische Verwendung finden. Besonders hervorzuheben ist in dieser Hinsicht das 1-Hydrazino-4-[pyridyl-(4')-me- thyl]-phthalazin der Formel

Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, d'ass man ein gegebenenfalls im aroma tischen Kern substituiertes 1-X-4-(Pyridyl-methyl)- phthalazin oder eine quaternäre Ammoniumverbin- dung davon, worin X einen austauschfähigen Rest, wie eine freie oder verätherte Mercaptogruppe, z. B. eine Alkylmercaptogruppe, eine reaktionsfähig ver- esterte oder verätherte Oxygruppe, z.
B. ein Halogen atom, vorzugsweise Chlor, oder eine Phenoxygruppe, oder eine Aminogruppe, z. B. eine unsubstituierte oder mono- oder disubstituierte Aminogruppe, wie eine Alkyl- oder Alkylenaminogruppe bedeutet, mit Hydrazin umsetzt.
Bei dem erfindungsgemässen Verfahren können die Ausgangsstoffe auch unter den Reaktionsbedin- gungen entstehen. So kann man reaktionsfähige Stick stoffderivate von o-(Pyridyl-acetyl)-benzoesäure, wie z.
B. die entsprechenden Thioamide, Amidine oder Iminoäther, vorzugsweise jedoch das Nitril, gegebenen falls in Form ihrer Derivate mit funktionell abgewan delter Ketogruppe, wie Acetale, Thioacetale, Enol- äther oder Acylate, oder auch 1-Amino-3-(pyridyl- methyl)-isoindolenine mit Hydrazin umsetzen, wobei intermediär 1-Amino-4-(pyridyl-methyl)-phthalazine entstehen.
Die erfindungsgemässe Umsetzung kann in An- oder Abwesenheit von Verdünnungsmitteln, allen falls auch in Gegenwart von Kondensationsmitteln, durchgeführt werden, wobei man ausserdem in Ge genwart von Katalysatoren, wie Kupferpulver, arbei ten kann. Das als Ausgangsstoff verwendete Pyridyl- methyl-phthalazin kann auch in Form seiner quater- nären Ammoniumverbindungen, insbesondere der Niederalkylammoniumverbindüngen, vorliegen.
Je nach der Arbeitsweise erhält man die neuen Hydrazinverbindungen in Form der freien Basen oder ihrer Salze. Aus diesen können in üblicher Weise die freien Basen gewonnen werden. Von letz teren lassen sich durch Umsetzung mit Säuren, die zur Bildung therapeutisch verwendbarer Salze geeig net sind, Salze gewinnen, wie z.
B. der Halogen wasserstoffsäuren, Schwefelsäure, Salpetersäure, Phosphorsäure, Rhodanwasserstoffsäure, Essigsäure, Propionsäure, Oxalsäure, Malonsäure, Bernstein säure, Zitronensäure, Weinsäure, Äpfelsäure, Me- thansulfonsäure, Äthansulfonsäure, Oxyäthansulfon- säure, Benzol- oder Toluolsulfonsäure oder Amino- säuren. Tertiäre Basen lassen sich in üblicher Weise,
vorzugsweise durch Behandlung mit reaktionsfähigen Estern niederer Alkanole, quaternisieren.
Die neuen Hydrazinophthalazine, ihre Salze und quarternären Ammoniumverbindungen können in Form pharmazeutischer Präparate Verwendung fin den, welche sie in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen Trägermaterial enthalten.
In den folgenden Beispielen sind die Tempera turen in Celsiusgraden angegeben.
<I>Beispiel 1</I> Aus 14,2 g 1-Oxo-4-[pyridyl-(4')-methyl]-1,2- d'ihydro-phthalazin gemäss den untenstehenden An gaben erhältliches, rohes 1-Chlor-4-[pyridyl-(4')-me- thyl]-phthalazin wird in 60 cm33 Methanol und 54 cm3 Hydrazinhydrat gelöst und 1 Stunde auf dem Wasserbad unter Rückfluss gekocht.
Die Lösung wird unter vermindertem Druck ver dampft und der Rückstand durch Erwärmen bei 60 - und 12 mm weitgehend vom Hydrazinhydrat befreit und in 250 cm3 absolutem Äthanol gelöst. Die Lö sung wird filtriert und wieder im Vakuum zur Trockne gebracht. Der Rückstand wird nochmals in 250 cm3 absolutem Äthanol gelöst, filtriert und mit 60 cm3 2,75n äthanolischer Salzsäure versetzt.
Es entstehen gelbbraune Kristalle, die isoliert und aus 400 cm3 95o/oigem Äthanol unter Zusatz von etwa 20 cm3 Wasser umkristallisiert werden. Man erhält so das in Wasser spielend lösliche 1-Hydrazino-4- [pyridyl-(4')-methyl]-phthalazin-dihydrochlorid der Formel

als Monohydrat vom F. 279-281 . Daraus lässt sich mit der berechneten Menge Natronlauge die freie Base gewinnen; F. 287-289 (nach Verflüssigung bei 88-133 und nachfolgendem Erstarren bei 183 ).
Das als Ausgangsstoff verwendete 1-Chlor-4- [pyridyl-(4')-methyl]-phthalazin kann wie folgt dar gestellt werden: 44,6 g der durch Kondensation von Phthalsäure- anhydrid und 4-Methyl-pyridin erhältlichen Verbin dung (y-Pyrophthalon) werden mit 200 cm3 Hydra zinhydrat 3 Stunden im Ölbad von 135 unter Rück fluss erwärmt. Die beim Kühlen sich abscheidenden Kristalle werden abgesaugt und aus Äthanol um kristallisiert.
Man erhält so das 1-Oxo-4-[pyridyl- (4')-methyl]-1,2-dihyd'ro-phthalazin als gelblicheKri- stalle vom F. 208-210 . Die Verbindung bildet ein Hydrochlorid vom F. 280-282 .
14,2 g 1-Oxo-4-[pyridyl-(4')-methyl]-1,2-dihydro- phthalazin werden mit 60 cm3 Phosphoroxychlorid 21/2 Stunden auf dem Wasserbad erwärmt. Die ge bildeten violetten Brocken werden zusammen mit dem Phosphoroxychlorid langsam in Eis eingetra gen und die entstandene violette Lösung in dünnem Strahl in ein Gemisch aus 450 cm3 Wasser und 270 cm3 konz. Ammoniak unter starkem Rühren und Kühlung einlaufen gelassen. Die Innentemperatur soll dabei nicht über 10 ansteigen.
Man saugt den rötlichen Niederschlag ab, wäscht ihn gründlich mit Wasser und saugt ihn trocken. Das so erhaltene 1-Chlor-4-[pyridyl-4')-methyl]-phthalazin wird direkt, wie oben geschildert, weiter verarbeitet. Durch Um lösen aus Äthanol kann es aber auch gereinigt wer den. Es schmilzt dann bei 170 .
<I>Beispiel 2</I> Das durch 21/2stündiges Erhitzen unter Rückfluss von 14,2 g 1-Oxo-4-[pyridyl-(2')-methyl]-1,2-dihy- dro-phthalazin mit 60 cm3 Phosphoroxychlorid in einem Ölbad von 125 und weitere Aufarbeitung gemäss Beispiel 1 erhältliche rohe 1-Chlor-4-[pyridyl- (2')-methyl]-phthalazin wird mit 120 cm33 Methanol und 108 cm3 Hydrazinhydrat unter Rückfluss eine Stunde auf dem Wasserbad gekocht.
Das Methanol und das Hydrazinhydrat werden im Vakuum (12 mm) völlig verdampft und der Rückstand zweimal je in 250 cm3 absolutem Äthanol gelöst und zur Trockne gebracht und wieder in 250 cm-' absolutem Äthanol gelöst. Man versetzt diese Lösung mit 70 cm3 2,3n äthanolischer Salzsäure und saugt nach einigen Stun den von etwas rötlichem Niederschlag ab. Das Filtrat wird völlig zur Trockne verdampft, der Rückstand mit 25 cm3 absolutem Äthanol übergossen und kristalli sieren gelassen.
Die Kristalle werden isoliert und: aus 95 o/oigem Äthanol umkristallisiert. Das so er haltene, in Wasser spielend lösliche 1-Hydrazino-4- [pyridyl-(2')-methyl]-phthalazin-dihydrochlorid der Formel _

bildet ein Monohydrat. Es schmilzt bei 230-232 unter Aufschäumen; die Schmelze wird dann wieder fest und schmilzt erneut bei etwa 290 .
Das als Ausgangsstoff verwendete 1-Oxo-4-[pyri- dyl-(2')-methyl]-1,2-d-ihydro-phthalazin wird aus dem durch Umsetzung von Phthalsäureanhydrid mit 2-Me- thyl-pyridin erhältlichen Kondensationsprodukt (a- Pyrophthalon) gemäss den. Angaben von Beispiel 1 für die entsprechende Pyridyl-(4)-methyl-Verbindung gewonnen. Es schmilzt bei 179-182 , sein Hydro- chlorid bei 261-264 .
<I>Beispiel 3</I> <B>16,5</B> g 1- Oxo - 4 - [pyridyl - (4') - methyl]-7-acet- amino-1,2-dihyd'ro-phthalazin-hydrochlorid werden mit<B>100</B> cm3 Phosphoroxychlorid 3 Stunden auf dem Wasserbad erhitzt. Es tritt keine sichtbare Reaktion ein.
Die Suspension wird auf 600 g Eis ausgetragen, die entstandene klare Lösung in dünnem Strahl in Wasser, das durch Zugabe von 310 cm s konz. Ammoniak dauernd alkalisch gehalten wird, einlau fen gelassen, der entstehende Niederschlag des 1-Chlor -4-[pyridyl-(4')-methyl]-7-acetamino-phthal- azins abgesaugt, gründlich gewaschen und sofort in einer Lösung von 81 cm3 Hydrazinhydrat in 90 cm3 Methanol 2 Stunden unter Rückfluss gekocht. Die leicht flüchtigen Anteile werden unter vermindertem Druck verdampft.
Dann wird zur Entfernung von Hydrazinhydrat noch zweimal mit 200 cm3 Äthanol verdampft. Der kristalline Rückstand wird in wenig Methanol gelöst, von einem darin unlöslichen, hoch schmelzenden Produkt getrennt und eingeengt. Man erhält gelbliche Kristalle des 1-Hydrazino-4-[pyri- dyl-(4')-methyl]-7-acetamino-phthalazins der Formel

Es schmilzt bei 143-145 unter Aufschäumen, wird bei etwa 180 wieder fest und schmilzt nicht mehr bis 315 .
Das in Äthanol mit 2,5n alkoholischer Salz säure hergestellte Dihydrochlorid schmilzt bei 264 bis 267 . Es ist hygroskopisch; das kristallwasser haltige Produkt schmilzt tiefer.
Das in diesem Beispiel verwendete Ausgangs material wird wie folgt gewonnen: Das durch Erhitzen von 4-Nitro-phthalsäure- anhydrid mit y-Picolin in o-Dichlorbenzol bei 180 nach den bekannten Verfahren gewonnene 5-Nitro-y- pyrophthalon wird mit der 4fachen Menge Hydrazin- hydrat 3 Stunden unter Rückfluss gekocht.
Die Nitro- gruppe wird dabei reduziert, und man erhält gelb braune Kristalle des 1-Oxo-4-[pyridyl-(4')-methyl]- 7-amino-1,2-dihydro-phthalazins vom F. 298 (Hy- drochlorid nach Umkristallisieren aus Äthanol F. über 300 ).
Das gewonnene Phthalazon wird in fein zerrie benem Zustand mit der 6fachen Menge Acetanhydrid 3 Stunden gekocht, das nach dem Erkalten abge trennte Produkt in Eisessig gelöst und mit 2,4n alkoholischer Salzsäure versetzt. Das sich sofort ab scheidende Hydrochlorid des 1-Oxo-4-[pyridyl-(4')- methyl]-7-acetamino-1,2-dihyd'ro-phthalazins ist ana lysenrein und kann direkt verwendet werden.
<I>Beispiel 4</I> 27 g 1-Methylmercapto-4-[pyridyl-(4')-methyl]- phthalazin werden mit 200 cm3 Äthanol und 150 cm3 Hydrazinhydrat 4 Stunden auf dem Wasserbad unter Rückfluss gekocht. Das Äthanol wird unter vermin dertem Druck entfernt und weiter wie in Beispiel 1 aufgearbeitet. Das mit alkoholischer Salzsäure schliesslich gewonnene Dihydrochlorid ist mit dem nach Beispiel 1 gewonnenen Produkt identisch.
Das verwendete 1-Methylmercaptoderivat wird aus dem in Beispiel 1 beschriebenen 1-CWor-4- [pyridyl-(4')-methyl]-phthalazin mit überschüssiger Natrium-methylmercaptan-Lösung in Methanol im geschlossenen Rohr bei 135-140 gewonnen und direkt weiter verarbeitet.
<I>Beispiel S</I> 24 g 1-Amino-4-[pyridyl-(4')-methyl]-phthalazin werden in 200 cm3 Äthanol und 200 cm3 Hydrazin- hydrat bis zur Beendigung der Ammoniakentwick- lung, das sind etwa 12 Stunden, unter Rückfluss ge kocht. Man filtriert die heisse Lösung und arbeitet sie, wie in Beispiel 1 beschrieben, auf das Dihydro- chlorid des 1 Hydrazino-4-[pyridyl-(4')-methyl]- phthalazins auf.
Das als Ausgangsstoff verwendete 1-Amino-4- [pyridyl-(4') methyl]-phthalazin wird aus dem in Bei spiel 1 beschriebenen 1-Chlor-4-[pyridyl-(4')-methyl]- phthalazin gewonnen, indem man 35 g davon mit 400 cm3 Äthanol, in welchem 34 g Ammoniak gelöst sind,
im geschlossenen Gefäss 5 Stunden auf 175 erhitzt und nach Abtrennen der Hauptmenge Ammo- niumchlorid sofort zur Trockne verdampft und den Rückstand gegebenenfalls aus Alkohol-Essigester kristallisiert. Er kann auch roh zur oben beschrie benen Umsetzung verwendet werden.

<I> Example 1 </I> From 14.2 g of 1-oxo-4- [pyridyl- (4 ') - methyl] -1,2-d'ihydro-phthalazine according to the information below, raw 1- Chlor-4- [pyridyl- (4 ') methyl] -phthalazine is dissolved in 60 cm33 methanol and 54 cm3 hydrazine hydrate and refluxed on a water bath for 1 hour.

The 1-oxo-4- [pyridyl- (4 ') - methyl] -1,2-dihydrophthalazine is thus obtained as yellowish crystals with a melting point of 208-210. The compound forms a hydrochloride of F. 280-282.
14.2 g of 1-oxo-4- [pyridyl- (4 ') - methyl] -1,2-dihydrophthalazine are heated with 60 cm3 of phosphorus oxychloride on a water bath for 21/2 hours. The violet chunks formed are slowly introduced into ice together with the phosphorus oxychloride and the violet solution formed is poured into a mixture of 450 cm3 of water and 270 cm3 of conc. Let ammonia run in with vigorous stirring and cooling. The internal temperature should not rise above 10.
The reddish precipitate is filtered off with suction, washed thoroughly with water and sucked dry. The 1-chloro-4- [pyridyl-4 ') - methyl] phthalazine obtained in this way is processed further directly as described above. But it can also be cleaned by dissolving it from ethanol. The methanol and the hydrazine hydrate are completely evaporated in vacuo (12 mm) and the residue is dissolved twice in 250 cm3 of absolute ethanol and brought to dryness and redissolved in 250 cm3 of absolute ethanol. This solution is mixed with 70 cm3 of 2.3N ethanolic hydrochloric acid and, after a few hours, the somewhat reddish precipitate is filtered off. The filtrate is completely evaporated to dryness, the residue is poured with 25 cm3 of absolute ethanol and allowed to crystallize.
The crystals are isolated and: recrystallized from 95% ethanol. The 1-hydrazino-4- [pyridyl- (2 ') -methyl] phthalazine dihydrochloride of the formula, which is easily soluble in water, is obtained in this way.

The dihydrochloride produced in ethanol with 2.5N alcoholic hydrochloric acid melts at 264 to 267. It is hygroscopic; the product containing water of crystallization melts deeper.

The 1-methyl mercapto derivative used is obtained from the 1-CWor-4- [pyridyl- (4 ') - methyl] phthalazine described in Example 1 with excess sodium methyl mercaptan solution in methanol in a closed tube at 135-140 and directly further processed.
<I> Example S </I> 24 g of 1-amino-4- [pyridyl- (4 ') - methyl] phthalazine are dissolved in 200 cm3 of ethanol and 200 cm3 of hydrazine hydrate until the evolution of ammonia has ended, that is about 12 hours, refluxed ge. The hot solution is filtered off and worked up, as described in Example 1, on the dihydrochloride of 1 hydrazino-4- [pyridyl- (4 ') - methyl] phthalazine.
The 1-amino-4- [pyridyl (4 ') methyl] phthalazine used as starting material is obtained from the 1-chloro-4- [pyridyl (4') methyl] phthalazine described in Example 1 by 35 g of it with 400 cm3 of ethanol, in which 34 g of ammonia are dissolved,
Heated in a closed vessel to 175 for 5 hours and, after separating off the main amount of ammonium chloride, immediately evaporated to dryness and the residue crystallized from alcohol / ethyl acetate, if necessary. It can also be used raw for the implementation described above.
