worin R einen Alkylrest darstellt, mit einem Acryl säureester oder einem ss-Halogenpropionsäureester umsetzt, den erhaltenen Diester cyclisiert,
den er haltenen Ketoester mit einem ss-ungesättigten alipha- tischen oder cycloaliphatischen Kohlenwasserstoff halogenid umsetzt und anschliessend in beliebiger Reihenfolge einerseits hydriert und anderseits verseift und decarboxyliert.
Die als Ausgangsmaterial benötigten 1-Carb- allcoxymethyl -1,2,3,4 - tetrahydroisochinoline, deren isocyclischer Ring durch Alkoxygruppen, deren zwei zusammen eine Alkylendioxygruppe bilden können, substituiert sein kann, können nach an sich bekann ten Methoden (vgl. z. B.
J. Chem. Soc. London 1951, 3472 und 1953, 2465) durch Umsetzung der in ss-Stellung entsprechend substituierten Äthylamine mit Malonsäureester, Cyclisierung der gebildeten Halbesteramide und nachfolgende katalytische Hy drierung gewonnen werden.
In der ersten Stufe des erfindungsgemässen Ver fahrens wird das Carbalkoxymethyl-tetrahydroisochi- nolin mit Acrylsäureester oder mit ss-Halogen- propion.säureester kondensiert. Die Kondensation er folgt zweckmässig bei einer Temperatur zwischen 50 und 100 C. Falls als.
Kondensationskomponente Acrylsäureester gewählt wird, ist ein weiteres Lö- sungsmittel überflüssig. Es ist jedoch zweckmässig, einen überschuss an Acrylsäureester, z. B. die zehn fache theoretische Menge, zu verwenden.
Dagegen erfolgt die Kondensation mit einem ss-Halogen-propionsäureester vorteilhaft in einem organischen Lösungsmittel, wie z. B. Aceton oder Benzol, unter Zugabe eines säurebindenden Mittels, wie z. B. Pottasche.
Die erhältlichen ss-[1-Carbalkoxymethyl-tetra- hydroisochinolyl-(2)]-propionsäureester sind Verbin dungen basischen Charakters; sie sind löslich in Säuren, dagegen unlöslich in Wasser oder Alkalien.
In der zweiten Stufe des erfindungsgemässen Verfahrens werden die Diester durch Ringschluss in die entsprechend substituierten 2-Oxo-3-carbalkoxy- 1,2,3,4,6,7-hexahydro-11bH-benzo[a]chinolizine über- geführt. Die Cyolisierung erfolgt zweckmässig in einem inerten Lösungsmittel, wie z. B. Benzol, Äther, Toluol, in Gegenwart eines gebräuchlichen Konden sationsmittels, wie z.
B. einem Alkalialkoholat, Bor- trifluorid, Natriumhydrid. In einer bevorzugten Aus führungsform wird der Diester in trockenem Benzol in Gegenwart von Natriumäthylat unter fortwähren der azeotroper Entfernung des gebildeten Alkohols erhitzt. Die gebildeten Ketoester sind basische Ver bindungen, die sich leicht in Säuren und in den ge bräuchlichen organischen Lösungsmitteln, wie z. B. Athanol, lösen lassen.
Sie zeigen die typische Enol- reaktion mit Eisenchlorid. Sie stellen wertvolle Aus gangsstoffe für die Gewinnung anders substituier ter Benzo[a]chinolizine dar.
In einer weiteren Stufe des erfindungsgemässen Verfahrens werden die Ketoester durch Umsetzung mit einem ,B - ungesättigten Kohlenwasserstoff halogenid in 3-Stellung substituiert. Dabei kommen. insbesondere Allylbromid, Methallylchlorid, y,y D methyl-allylbromid, Propargylbromid und Cyclo- hexen-(2) yl-bromid in Frage.
Die Umsetzung erfolgt zweckmässig in wässeriger Lauge in Gegenwart einer katalytischen Menge Kupfer. Nach einer be vorzugten Ausführungsform des Verfahrens wird der Ketoester mit dem Halogenid in Gegenwart von wenig Kupferpulver gerührt und die berechnete Menge wässerige Lauge zugegeben. Die gebildeten.
3-substituierten 2 - Oxo - 3 -carbalkoxy -1,2,3,4,6,7- hexahydro-11bH-benzo[a]chinolizine sind basische Verbindungen, welche mit Eisenchlorid keine Enol- reaktion zeigen.
Durch Verseifung und Decarboxylierung erhält man die entsprechenden 3-substituierten 2-Oxo-Ver- bindungen. Dabei ist es zweckmässig, die Verseifung und Decarboxylierung durch Erwärmen in alkali schem Medium durchzuführen. Nach einer zweck mässigen Ausführungsform wird der Ketoester in alkoholischer Natronlauge gekocht.
Die gewonnenen im isocyclischen Ring gegebenenfalls substituierten 2 - Oxo - 3 - alkenyl - 1,2,3,4,6,7 -hexahydro -11bH- benzo[a]chinolizine sind Basen, die mit den ge bräuchlichen organischen und anorganischen Säuren wasserlösliche Salze bilden.
Die Hydrierung zu den entsprechenden 2-Oxo- chinolizinen gelingt z. B. mit katalytisch angeregtem Wasserstoff. Als Katalysatoren werden zweckmässig Raney-Nickel oder Palladium-Kohle verwendet. Es ist vorteilhaft, die Reduktion in einem inerten orga nischen Lösungsmittel, wie z. B. Methanol oder Äthanol, durchzuführen..
Nach einer weiteren Ausführungsform des erfin dungsgemässen Verfahrens kann man die 3-substi- tuierten 2-Oxo-3-carbalkoxy-benzo-chinolizine zu den entsprechenden 3-substituierten 2-Oxo-3-carb- alkoxy-Verbindungen reduzieren.. Die Hydrierung kann z. B. mit katalytisch angeregtem Wasser stoff durchgeführt werden. Die Hydrierung wird zweckmässig in einem Lösungsmittel, z. B. in einem Alkohol, durchgeführt. Als Katalysatoren können z.
B. Raney-Nickel oder Palladium-Kohle verwen det werden. Die erhaltenen in 3-Stellung durch Carbalkoxy und Alkyl substituierten Verbindungen zeigen keine Enolreaktion mit Eisenchorid.
Die Verscifung und Decarboxylierung dieser Ketoester führt zu den 3-substituierten 2-Oxo- 1,2,3,4, 6,7 - hexahyd@ro -11 bH - benzo[a]chinolizinen;. Die Verseifung und Decarboxylierung gelingt sowohl durch Anwendung saurer wie alkalischer Mittel. In einer bevorzugten Ausführungsform wird verdünnte Mineralsäure, z. B. 3n Salzsäure verwendet.
Die in 3-Stellung durch einen gesättigten Kohlenwasserstoffrest substituierten Benzo[a]chino- lizine sowie deren Salze besitzen wertvolle pharma zeutische Eigenschaften; insbesondere eine Narkose potenzierende Wirkung. Sie sollen als Heilmittel oder als Zwischenprodukte zur Herstellung von Heilmitteln Verwendung finden.
<I>Beispiel</I> 54 g 1-Carbäthoxymethyl-6,7-d'imethoxy-1,2,3,4- tetrahydro-isochinolin und 100 cms Acrylsäureäthyl- ester werden während 24 Stunden unter Rückfluss, gekocht. Nach dem Erkalten wird mit dem gleichen Volumen Äther verdünnt und die basischen Teile mit 3n Salzsäure extrahiert. Der Salzsäureextrakt wird unter Eiskühlung mit Ammoniaklösung bis zur alkalischen Reaktion auf Phenolphthalein versetzt und die ausgeschiedenen Basen in Äther aufgenom men.
Nach dem Trocknen der Ätherlösung über Natriumsulfat wird filtriert, eingeengt und der Rück stand im Hochvakuum destilliert. Man erhält 65 g ss-(1-@Carbäthoxymnethyl-6,7-dimethoxy-1,2,3,4 tetra- hydro-isochinolinyl-(2)]-propionsäure-äthylester, der bei 0,02 mm zwischen 197-201 C destilliert. nD = 1,5235. Das Destillat erstarrt nach einigem Stehen. Schmelzpunkt nach dem Umlösen aus tief siedendem Petroläther 37-39 C.
Die gleiche Verbindung kann auch wie folgt hergestellt werden: 9,0 g 1-Carbäthoxymethyl'-6,7-dimethoxy-1,2,3,4- tetrahydro-isochinolin werden in 30 cm?, Aceton gelöst, 15 g trockenes Kaliumcarbonat und 6 g ss-Brom-propionsäure-äthysester zugegeben und unter Rühren während 24 Stunden am Rückfluss gekocht. Nach dem Filtrieren und Waschen des Rückstandes mit Aceton wird eingeengt.
Der Rückstand wird in Äther gelöst und die basischen Teile mit 3n Salz säure extrahiert. Nach dem Versetzen des Salzsäure extraktes mit Ammoniak bis zur Reaktion auf Phenolphthalein wird mit Äther extrahiert, der Extrakt mit Natriumsulfat getrocknet, filtriert, ein geengt und der Rückstand im Hochvakuum destil liert.
Man erhält 6,5 g,ss-[1-Carbäthoxymethyl-6,7-d'i- methoxy -1,2,3,4 - tetrahydro - isochinolinyl- (2)]-pro- pionsäure-äthylester, der im Hochvakuum unter 0,02 mm bei 190-200 C destilliert. Das Destillat erstarrt nach dem Abkühlen und eine aus Petrol- äther umgelöste Probe schmilzt bei 37-39 . Der Mischschmelzpunkt mit dem oben erhaltenen Pro dukt zeigt keine Depression.
2,49g Natrium werden in 90 cm3 absolutem Äthylalkohol gelöst und das Lösungsmittel im Wasserstrahlvakuum abdestilliert. Zum trockenen Natriumäthylat werden nun 34,2 g i3-[1-Carbäthoxy methyl-6,7-dimethoxy-1,2,3,4-tetrahydro-isochinolinyl- (2)]-propionsäure-äthylester, welcher in 600 cm?, trockenem Benzol gelöst ist, zugegeben, unter Rühren erhitzt und der bei der Kondensation gebildete Alkohol azeotrop abdestillie.rt. Die Destillation wird unterbrochen, wenn die Temperatur von 78 C er reicht ist.
Nun wird noch während 1 Stunde am Rückfluss gekocht, gekühlt und mit 3n Natron lauge und Wasser extrahiert, bis die Benzollösung mit Eisenchlorid keine Enol@reaktion mehr gibst. Die wässerigen alkalischen Extrakte werden ver einigt, mit Eis gekühlt und Kohlensäure eingeleitet. Man erhält auf diese Weise 18g 2-Oxo-3-carbäthoxy- 9,10 - dimethoxy - 1,2,3,4,6,7 - hexahydro - 11bH- benzo[a]chinolizin, welches nach dem Umlösen aus Alkohol bei 114-115 C schmilzt.
Das vermittels alkoholisch-ätherischer Salzsäure bereitete Hydro chlorid schmilzt nach dem Umlösen aus Alkohol- Äther bei 193-194 C.
Die Cyclisierung kann auch wie folgt durchge führt werden: 36,5 g j3-[1-Carbätiioxymethyl-6,7-dimethoxy- 1,2,3,4 - tetrahyd'ro - isochinolinyl - (2)]-propion.säure- äthylester werden in 100 cm2- absolutem Benzol ge löst, mit 2,9g Natriumhydrid versetzt und 1 Stunde am Rückfluss gekocht. Anschliessend wird nach der oben angegebenen Weise aufgearbeitet.
Man erhält 15 g 2-Oxo-3-carbäthoxy-9,10-dimethoxy-1,2,3,4,6,7- hexahydro-11bH-benzo[a]chinolizin vom Schmelz punkt 114-115 C.
<B>19,8</B> g 2 - Oxo - 3 -carbäthoxy- 9,10 - d'imethoxy- 1,2,3,4,6,7-hexahydro-11bH-benzo[a]chinolizin wer den mit 60 cm3 Wasser und 5,6 cm?, frisch destillier- tem Allylbromid versetzt, 200 mg Kupferpulver zu gegeben und stark gerührt. Dazu tropft man 61 cm3 In Natonlauge. Nach der Zugabe wird noch während 30 Minuten gerührt, das ausgeschiedene Öl in Äther aufgenommen, die Ätherlösung nacheinander mit 3n Natronlauge und Wasser gewaschen und dann mit 3n Salzsäure extrahiert.
Das Hydrochlorid des 2 - Oxo - 3 - allyl - 3 - carbäthoxy - 9,10 - d'imethoxy- 1,2,3,4,6,7-hexahydro-11bH-benzo[a]chinolizin kri stallisiert nach dem Stehen über Nacht. 10,8g Hy- drochlorid vom Schmelzpunkt 156 C. Die aus dem Hydrochlorid mit Alkali freigesetzte Ketoester-Base kristallisiert aus Alkohol in Form von Prismen vom Schmelzpunkt 118 C. Die Ketoester-Base gibt keine Farbreaktion beim Versetzen mit alkoholischer Eisenchloridlösung.
10 g 2-Oxo-3-allyl-3-carbäthoxy-9,10-dimethoxy- 1,2,3,4,6,7 hexahydro-11bH-benzo[a]chinolizin wer den in 300 cm3 50/0 alkoholischer Natronlauge wäh rend 3 Stunden am Rückfluss gekocht. Nach dem Konzentrieren im Vakuum wird mit Wasser ver- setzt, filtriert, getrocknet und aus Essigester-Petrol- äther umgelöst.
Man erhält 3,5g 2-Oxo-3-allyl-9,10=@ dimethoxy-1,2,3,4,6,7-hexahydro-11bH-benzo[a]chi- nolizin vom Schmelzpunkt 114-116 C. Hydro- bromid Schmelzpunkt 205 C. Oxim Schmelzpunkt 164 C.
Nach der gleichen Arbeitsweise können aus 2-lOxo - 3 - carbäthoxy- 9,10-I d'imethoxy-1,2,3,4,6,7- hexahydro-11bH-benzo[a]chinolizin durch Alkenylie- rung mit Methallylchlorid und y,y-Dimethylallyl- bromid nach Verseifung und Decarboxylierung mit alkoholischer Natronlauge folgende Chinolizinketone gewonnen werden:
2-Oxo - 3 - methallyl-9,10-d'irnethoxy-1,2,3,4,6,7- hexahydro-11bH-benzo[a]chinolizin vom Schmelz punkt 138 C; 2 - Oxo - 3 - y,y - d'imethylallyl - 9,10 - dimethoxy- 1,2,3,4,6,7-hexahydro-11bH-benzo[a]chinolizin vom Schmelzpunkt <B>1310C.</B> Hydrobromid Schmelzpunkt 190 C.
10 g des oben erhaltenen 2-Oxo-3-allyl-9,10-di- methoxy-1,2,3,4,6,7 -hexahyd'ro-11bH-benzo[a]chi- nolizin werden in 300 cm3 Methanol gelöst und bei Raumtemperatur über 3 g Raney Nickel hydriert. Nach Aufnahme der theoretischen Menge Wasser stoff wird unterbrochen, der Katalysator filtriert und, eingeengt. Aus Alkohol-Wasser kristallisieren 8,0 g 2-Oxo-3 -.n-propyll-9,10-d'iinethoxy-.1,2,3,4,6,7-hexa- hydro-11bH-benzo[a]chinolizin vom Schmelzpunkt 102 .
In analoger Weise führt die katalytische Hydrie rung der oben erhaltenen Ketone in Feinsprit über Palladium-Kohle-Katalysator zu den folgenden Chinolizinen: 2 - Oxo - 3 - isobutyl-9,10-d!imethoxy-1,2,3,4,6,7- hexahydro-11bH-benzo[a]chinolizin vom Schmelz punkt 123 C. Hydrobromid 202 C.
2 - Oxo - 3 -isoamyl-9,10-dimethoxy-1,2,3,4,6,7- hexahyd'ro-11bH-benzo[a]chinolizin vom Schmelz punkt 106 C. Hydrobromid Schmelzpunkt 196 C.
2 - Oxo - 3 -n-propyl-9,10-dimethoxy-1,2,3,4,6,7- hexahydro-11bH-benzo[a]chinolizin kann aus 2-Oxo- 3 - allyl - 3 - carbäthoxy - 9,10 - dimethoxy-1,2,3,4,6,7- hexahydro-11bH-benzo[a]chinolizin auch auf fol gende Weise erhalten werden:
9,4 g 2-Oxo-3-allyl-3-carbäthoxy-9,10-dixnethoXy 1,2,3,4,6,7-hexahydro-11bH-benzo[a]chinolizin-hy- drochlorid werden in 300 cm3 Feinsprit gelöst und über 3 g Palladium-Kohle-Katalysator (5%) hydriert. Nach der Aufnahme der theoretischen Menge Wasserstoff wird filtriert, zur Trockne eingeengt, in 300 cm-' 3n Salzsäure gelöst und während 6 Stunden am Rückfluss gekocht. Nach dem Waschen mit Äther wird mit Natronlauge alkalisch gestellt und die ba sischen Teile mit Äther ausgeschüttelt.
Nach dem Lösen des Rückstandes in Aceton und Versetzen mit alkoholischer Salzsäure kristallisieren 4,5 g 2-Oxo - 3 -n-propyl-9,10-dimethoxy-1,2,3,4,6,7-hexa- hydro -11 bH - benzo[a]chinolizin - hydrochlorid vom Schmelzpunkt 200-202 C.
Die aus dem Hydro- chlorid auf übliche Weise gewonnene Base schmilzt nach dem Umlösen aus Methanol-Wasser bei 102 C. Das Präparat ist mit der weiter oben dargestellten Base vom Schmelzpunkt 102 C identisch. Schmelz- punkt des Oximes bei 158 C.
