Verfahren zur Konservierung von tierischen Organpräparaten In neuer Zeit werden. Organpräparate als solche oder zur Herstellung von Wirkstoffen therapeutisch verwendet, die unter grösstmöglicher Schonung des Zel.lgefüges mit Hilfe der Gefriertrocknung konser viert worden sind. Zur Arbeitsweise der Gefrier trocknung gehört es bekanntlich, die lebendfrisch ent nommenen Organteile sofort möglichst rasch tiefzu kühlen und dann bei Temperaturen, die zumindest unterhalb des Gefrierpunktes liegen, das Wasser abzu- sublimieren.
Vorliegende Erfindung betrifft ein Verfahren zur Konservierung von tierischen Organpräparaten durch Gefriertrocknung. Dieses Verfahren ist dadurch ge kennzeichnet, dass man die frischen Organe oder Organteile vor der Abkühlung zur Gefriertrocknung kurze Zeit auf Temperaturen zwischen 25 und 85 erhitzt.
Alls den so konservierten Organpräparaten lassen sich, z. B. durch Extraktion, therapeutische Wirk stoffpräparate, wie Hormonpräparate usw., herstellen.
Durch das erfindungsgemässe Verfahren lässt sich die Ausbeute, das heisst die therapeutische Wirksam keit der konservierten Zellpräparate ganz bedeutend steigern. Man kann daher bei jeder Applikation die Dosierung entsprechend niedriger wählen. Diese Ausbeutesteigerung erlaubt es, aus einer gegebenen Anzahl von Spendentieren einen vermehrten Betrag an therapeutischen Wirkstoffen zu produzieren. Das ist für die Herstellung von therapeutisch verwend baren Organpräparaten wichtig, denn der wesentliche Teil der Arbeit erstreckt sich auf die Auswahl und ärtzliche Untersuchung der Spendentiere sowie auf die operative Gewinnung der einzelnen Organe.
Die erfindungsgemäss konservierten Präparate können für Injektionszwecke in Ringerlösung aufge schwemmt werden. Man kann sie aber auch in Salben oder Pillen einarbeiten. Durch die nachfolgend angeführten Versuche wird diese Ausbeutesteigerung bei Verwendung von nach dem erfindungsgemässen Verfahren konser vierten Organpräparaten veranschaulicht. <I>I.
Testung der Wirkungserhöhung</I> Testobjekt für diese Untersuchungen waren Mäuse, denen operativ beide Eierstöcke entfernt wurden, und die sich somit in einem Hormon (Oestrogen)Mangeizustand befanden.. Dieser Oestro- genmangel ist an den Zellformen des Vaginalepithels im mikroskopischen Ausstrichpräparat abzulesen. Das Auftreten sog. Schollen ist dabei ein Mass für den Grad der Wiederherstellung des normalen Zustandes.
Zur Testung der Wirksamkeit vorbehandelter Präpa rate wurde eine Gruppe ovariectomierter Mäuse mit unbehandelten Zellen,, eine andere Gruppe mit vor behandelten Zellen gespritzt und die durchschnitt liche Schollenzahl beider Gruppen über einen Monat hinweg verglichen. Die Wirkungssteigerung oder der Wirkungsabfall ist nachfolgend in 1/o der mittleren Schollenzahl der Kontrollgruppe ausgedrückt.
II. <I>Durchführung der</I> Vorbehandlung <I>der Zellen</I> Die Organzellen wurden unter sterilen Kautelen aus dem frisch getöteten Versuchstier entnommen, mit Scheren fein zerkleinert und dann (für Versuchs zwecke) in abgewogene Portionen aufgeteilt. Jeweils eine Portion wurde ohne eine besondere Vorbehand- lung gefriergetrocknet und Kontrolltieren injiziert.
Die anderen Portionen wurden in sterile und dicht verschliessbare Kölbchen oder Gläschen gefüllt und in diesen 15 Minuten auf + 37 bzw. + 60 bzw. + 80 erhitzt, indem das Kölbchen in ein Wasserbad dieser Temperatur eingetaucht wurde. Anschliessend wurden die Kölbchen in einem Bad fliessenden Lei- tungswassers auf etwa + 10 C abgekühlt, wozu 2 Mi nuten benötigt wurden.
Unmittelbar danach erfolgte das Einfrieren des Präparates durch Eintauchen in ein Kohlensäure-Aceton-Bad. Die Trocknung wurde bei -25 C durchgeführt. Suspendierung der Zellen wie üblich in Ringerlösung. Injektion intramuskulär.

The present invention relates to a method for preserving animal organ preparations by freeze-drying. This process is characterized in that the fresh organs or parts of organs are briefly heated to temperatures between 25 and 85 for freeze-drying before cooling.
All of the organ preparations preserved in this way can, for. B. by extraction, therapeutic active substance preparations, such as hormone preparations, etc., produce.
The method according to the invention allows the yield, that is to say the therapeutic effectiveness of the preserved cell preparations, to be increased quite significantly. You can therefore choose the dosage correspondingly lower for each application. This increase in yield makes it possible to produce an increased amount of therapeutic agents from a given number of donated animals. This is important for the production of therapeutically usable organ preparations, because the essential part of the work extends to the selection and medical examination of the donated animals as well as the operative extraction of the individual organs.
The preparations preserved according to the invention can be suspended in Ringer's solution for injection purposes. But you can also work them into ointments or pills. Testing the increased effectiveness </I> The test object for these investigations were mice whose ovaries had been surgically removed and were therefore in a hormone (estrogen) deficiency state. This estrogen deficiency can be read from the cell shapes of the vaginal epithelium in a microscopic smear. The appearance of so-called clods is a measure of the degree of restoration of the normal state.
To test the effectiveness of pretreated preparations, a group of ovariectomized mice was injected with untreated cells, another group with pre-treated cells and the average number of clots in both groups was compared over a month. II. <I> Carrying out the </I> pretreatment <I> of the cells </I> The organ cells were removed from the freshly sacrificed test animal under sterile conditions, finely comminuted with scissors and then (for test purposes) divided into weighed portions. One portion each was freeze-dried without any special pretreatment and injected into control animals.
The other portions were filled into sterile and tightly sealable flasks or jars and heated to +37 or +60 or +80 in these 15 minutes by immersing the flask in a water bath at this temperature. The small flasks were then cooled in a bath of running tap water to about + 10 ° C, which took 2 minutes.
