Procédé de préparation d'un polypeptide
La présente invention a pour objet un procédé de préparation d'un dérivé tétrapeptidique, terme intermédiaire de la synthèse de l'ocytocine, hormone nonapeptidique dont la formule de constitution suivante :

a été démontrée par Du Vigneaud et collaborateurs, J. Am. Chem. Soc., 1954, 76, 3115.
Le dérivé tétrapeptidique préparé par le procédé faisant l'objet de la présente invention est un ester de la S-trityl-L-cystéinyl-L-propyl-L-leucyl-glycine composé de formule L dont la préparation figure au
schéma annexé et sera décrite plus loin. Comme on le voit, ce composé est très proche du tronçon tétrapeptidique terminal de l'ocytocine et s'apparente très étroitement à l'ester de la S-benzyl-L-cystéinyl-
L-propyl-L-leucyl-glycine de formule B [Ch. Ressler et V. Du Vigneaud (J. Am. Chem. Soc., 1954, 76, 3107)] qui, après transformation en amide, a servi à Du Vigneaud et collaborateurs (l. c) à la synthèse de l'ocytocine.

Pour arriver à ce tétrapeptide de formule B,
Ch. Ressler et V. Du Vigneaud ont employé une voie assez laborieuse. Utilisant comme agent de blocage de la fonction amine, au cours de leurs synthèses peptidiques, le chloroformiate de benzyle, composé coûteux, peu stable, lacrymogène et dont la préparation comporte la manipulation du phosgène et effectuant la condensation peptidique par la méthode aux anhydrides mixtes, ils ont d'abord préparé le dérivé N-benzyl-oxycarbonylé de la L-leucine, lequel condensé avec le glycinate d'éthyle, leur a fourni le N-benzyloxy-carbonyl-L-leucyl-glycinate d'éthyle. Une hydrogénolyse catalytique fournit le Lleucyl-glycinate d'éthyle. Après ces deux étapes,
on condense le produit obtenu avec la N-benzyloxycarbonyl-L-proline par le procédé aux anhydrides mixtes avec utilisation du chlorure d'isovaléryle, ce qui fournit le N-benzyloxy-carbonyl-L-prolyl-L-leu- cyl-glycinate d'éthyle. Une nouvelle hydrogénolyse pour éliminer le groupement protecteur conduit au L-prolyl-L-leucyl-glycinate d'éthyle. Ce composé est condensé avec le dichlorure de bis- (N-benzyloxycarbonyl)-L-cystinyle obtenu en une étape à partir de la benzyloxy-carbonyl-L-cystine, ce qui conduit au bis- (N-benzyloxy-carbonyl)-L-cystinyl-bis- (Le prolyl-L-leucyl-glycinate d'éthyle) après un total de 6 étapes.
Pour passer de ce composé au peptide de formule B, il faut encore 4 étapes. On saponifie la fonction ester en acide correspondant, soumet ce produit à la réduction par le sodium dans l'ammo- niaque liquide pour transformer le pont cystinique en fonction cystéine, traite au chlorure de benzyle pour obtenir le dérivé S-benzylé du tétrapeptide qu'on estérifie à nouveau.
Pour l'industrie, cette synthèse comporte l'inconvénient d'astreindre à une élimination répétée d'un agent de blocage par hydrogénolyse, d'obliger à bloquer à nouveau et à effectuer peu avant la fin de la synthèse une réduction au sodium dans l'ammoniaque liquide, opération difficile à réaliser sur une échelle industrielle et de soumettre un ester à la saponification préliminaire avant cette réduction, ce qui entraîne une nouvelle estérifi- cation finale.
Vis-à-vis de ce procédé en 10 stades, le procède selon l'invention permet de parvenir au tétrapeptide ester de formule L en 6 stades, facilement réalisables n'utilisant qu'un blocage unique, le dérivé final présentant en outre l'avantage de se laisser scinder plus facilement que les composés S-benzylés pour lesquels la régénération de la fonction mercaptan libre nécessite une nouvelle intervention du sodium dans l'ammoniaque liquide.
Selon le présent procédé, on fait réagir la S, N ditrityl-L-cystéine en présence d'un carbodiimide
N, N'-disubstitué avec un ester de L-proline, isole l'ester du dipeptide formé, le saponifie en S, N ditrityl-L-cystéinyl-L-proline que l'on condense en présence d'un carbodiimide N, N'-disubstitué avec un ester de L-leucine, isole l'ester du tripeptide formé, le saponifie en S, N-ditrityl-L-cystéinyl-L-propyl-L- leucine que l'on condense en présence d'un carbodiimide N, N-disubstitué avec un ester de glycine, isole l'ester du tétrapeptide formé et soumet cet ester, d'une part, à une détritylation sélective sur l'azote et, d'autre part,
à une saponification pour obtenir le tétrapeptide libre correspondant.
La préparation de la S, N-ditrityl-cystéine a déjà été décrite par la titulaire dans le brevet suisse
N 357729, sa préparation ne figurant donc dans l'exemple qu'à titre de rappel.
Le mode opératoire décrit dans l'exemple qui suit peut évidemment être employé au départ des aminoacides racémiques ou des énantiomorphes. On peut notamment faire varier les températures, changer de solvant, employer d'autres esters d'aminoacides que ceux mentionnés ou les préparer par d'autres voies que celles indiquées, les saponifier par toute autre méthode connue ou employer comme agent de condensation un autre carbodiimide disubstitué que le dicyclohexylcarbodiimide ou utiliser toute autre méthode de condensation peptidique connue.
Les points de fusion sont des points de fusion instantanée déterminés sur bloc de Maquenne. Les formules des composés figurent sur le schéma annexé.
Exemple
Stade 1 : Préparation du SJN-ditrityl-L-cystéinyl-L-
prolinate de méthyle de formule E
a) Condensation de la S, N-ditrityl-L-cystéine de
formule C avec le prolinate de méthyle de for
mule D (R = CHg,)
27, 5 g de S, N-ditrityl-cystéinate de diéthylamine cristallise sont dissous dans 150 ce de chloroforme.
On lave cette solution avec 41 cm : 3 d'acide chlorhy
drique normal, puis à l'eau, sèche sur sulfate de soude et évapore à sec. D'autre part, on dissout
6, 66 g de chlorhydrate de prolinate de méthyle dans 25 cil de chlorure de méthylène ; on glace à ¯ 10 {} C et ajoute, sans dépasser 0t) C, 4, 8 cm3 de diéthylamine, puis 200 cm3 d'éther. Le chlorhydrate de diéthylamine, insoluble dans l'éther précipite. On le filtre et évapore la solution du prolinate de méthyle à sec sous vide.
La S, N-ditritylcystéine de formule C préparée comme on vient de l'indiquer est dissoute dans 50 ce de chlorure de méthylène. On refroidit à 00 C et ajoute le prolinate de méthyle de formule D également dissous dans 50 cm3 de chlorure de méthylène et refroidi à 0ç) C. Dans ce mélange réactionnel, on introduit 9 g de dicyclohexylcarbodiimide et abandonne une nuit à 20 C. Le lendemain, on ajoute
1 cm3 d'acide acétique pour transformer l'excès de dicyclohexylcarbodiimide en dicyclohexylurée. Après une demi-heure on filtre la dicyclohexylurée qu'on lave au chlorure de méthylène.
Le filtrat, réuni au chlorure de méthylène de lavage, est lavé à l'acide chlorhydrique dilué, à l'eau, séché et évaporé à sec.
L'ester du dispeptide de formule E brut (R = CH) ainsi obtenu peut servir directement à la deuxième étape de la synthèse, c'est-à-dire être saponifié en dipeptide de formule F.
Pour la préparation de la S, N-ditrityl-L-cystéine de formule C, qui est donnée ici à titre de rappel, on procède comme suit : dans une solution, refroidie à 00 de 25 g de chlorhydrate de L-cystéine [a] 20 = + 5, 50 (c= 1 /o, acide chlorhydrique N) dans 480 cm3 d'eau, on introduit, sous courant d'azote et agitation mécanique, 480 cm3 d'éther et 80 cm de diéthylamine et refroidit à-5fM C. Sans interrompre l'agitation, on introduit 120 g de chlorure de trityle et poursuit l'agitation pendant trois heures. Le mélange réactionnel devient une masse pâteuse assez difficile à agiter.
On ajoute alors 150 à 200 ce de chloroforme, décante la phase aqueuse qui ne renferme que du chlorhydrate de diéthylamine et le chlorhydrate de cystéine qui n'a pas réagi. La couche organique est lavée à nouveau avec 300 ce d'eau, séchée sur sulfate de soude et concentrée. On ajoute alors 100cm3 d'alcool contenant 1 ce de diéthylamine et distille le solvant jusqu'à ce qu'il ait entraîné tout le chloroforme. Le sel de diéthylamine de la N, S-ditrityl-L-cystéine cristallise.
On complète la cristallisation par addition progressive de 100 cm3 d'éther. On obtient ainsi 55 g de sel se présentant
sous forme d'aiguilles [a] 2D0= +71 + 1 (c=2 /o,
chloroforme) solubles en chloroforme, très peu solu
bles en alcool, insolubles dans l'eau et l'éther. Les
réactions à la ninhydrine sont négatives. A partir des
eaux mères de cristallisation on obtient un deuxième
jet de produit portant le rendement global à 70 g
environ. En reprenant ce sel par l'eau acidulée, on le
décompose avec mise en liberté de la N, S-ditrityl-L
cystéine de formule C qui peut être facilement
extraite par l'éther dans lequel elle est assez soluble.
L'évaporation de l'éther fournit le produit brut à l'état amorphe.
Le chlorhydrate de L-prolinate de méthyle de formule D (R = CH3) est préparé selon J. Schumann et R. A. Boissonnas (Helv. Chim. Acta, 1952, 35, 2240), par action d'un courant d'acide chlorhydrique sec sur une suspension de L-proline dans le méthanol. On introduit 6 g de L-proline dans 120 cm de méthanol saturé à 20 C d'acide chlorhydrique sec ; on abandonne 24 heures à température ambiante, évapore à sec sous vide.
On obtient finalement 8 g de chlorhydrate de L-prolinate de méthyle sous forme d'une huile jaune pâle, prête à l'emploi pour la condensation, soit un rendement de 94 /0. b) Condensation de la S, N-ditritylcystéine avec le
chlorhydrate de prolinate de benzyle de formule
D (R=CGH5-CH2-)
On fait réagir pendant une nuit 6, 8 g de sel de
S, N-ditritylcystéine et 2, 5 g de chlorhydrate de prolinate de benzyle dissous dans 50 cm3 de chlorure de méthylène-en présence de 2, 2 g de dicyclohexylcarbodiimide. Après un traitement identique à celui décrit sous a) on isole du dipeptide de formule E (R=-CH.,-CH) sous forme d'huile.
Pour la préparation du chlorhydrate de L-prolinate de benzyle D (R=CgHs-CHo-) on procède comme suit :
15 g de L-proline sont mis en suspension fine dans 150 cm3 d'alcool benzylique pur. On refroidit vers +10 C et sature à cette température d'acide chlorhydrique gazeux sec. Après avoir abandonné quelque temps, on chauffe sous vide à 90"C pour éliminer l'eau formée et l'acide chlorhydrique. On laisse refroidir et ajoute un grand excès d'éther. Le chlorhydrate de prolinate de benzyle cristallise en petits prismes incolores. On glace, filtre, lave à l'éther et sèche sous vide. On recueille 31 g de produit blanc, [ =-42" 1 (c==2"/o alcool absolu) identique au composé décrit par R. E. Neumann et
E. L. Smith (J. Biol.
Chem., 1951, 193, 97).
Stade 2 : Préparation de la S, N-ditrityl-L-cystéinyl
L-proline de formule F
a) L'ester méthylique du dipeptide de formule E brut huileux obtenu comme on vient de l'indiquer dans le stade 1 a) est traité au reflux par 100 cm3 d'alcool, 30 cm3 de potasse méthanolique et 30 cm3 d'eau. Après refroidissement, on verse dans un mélange d'eau et de glace et ajoute de l'acide chlor hydrique normal jusqu'à pH 1 (environ 90 cm3 d'acide chlorhydrique N). Le précipité formé est extrait au chloroforme. La solution chloroformique est lavée à l'eau jusqu'à neutralité, séchée et évaporée à sec. On reprend le résidu par 150 cm3 d'éther anhydre et porte au reflux en ajoutant 5 cm3 de diéthylamine. On laisse refroidir et ajoute 2 crd, d'eau.
Le sel de diéthylamine du dipeptide de formule F cristallise en aiguilles. On ajoute 150 cm3 d'éther de pétrole léger et abandonne une nuit à la température ambiante. On obtient ainsi 22 g de sel de diéthylamine de formule F cristallisant avec 1/2 molécule d'eau et suffisamment pur pour la condensation avec le leucinate de méthyle. Rendement : 70%, F. = vers 150¯ C (dÚc.), [α]20D = + 92¯ ¯ 2 (c = 2 /0, éthanol absolu).

b) L'ester benzylique du dipeptide de formule E dont la préparation a été décrite dans le stade 1 b) est saponifié au reflux avec 6 cm3 de potasse métha- nolique à 20 /0 et 6 cm3 d'eau. On ajoute 50 g d'eau glacée, acidifie à l'acide chlorhydrique à pH 2, 3 et effectue une extraction au chloroforme. L'extrait chloroformique est lavé à l'acide chlorhydrique N/10, à l'eau, séché sur sulfate de magnésium et évaporé à sec.
Le résidu est repris par 50 ce d'éther et 2 ce de diéthylamine. On refroidit et ajoute 1 cm3 d'eau. On recueille 5, 8 g de sel de diéthylamine de la S, N-ditrityl-L-cystéinyl-L-proline renfermant Va molécule d'eau, identique à tous points de vue au produit décrit à la fin de a). Rendement 74 /0.
Stade 3 : Préparation du S, N-ditrityl-L-cystéinyl-L-
prolyl-L-leucinate d'éthyle (H) (R = C2Hs)
Les matières premières utilisées pour cette préparation sont la SN-ditrityl-Lcystéinyl-L-proline de formule F, préparée selon le stade 2 et le chlorhydrate du L-leucinate d'éthyle (G) (R = CgHg) de préparation classique sur lequel on opère directement.
15, 7 g de sel de diéthylamine de la S, N-ditrityl- L-cystéinyl-L-proline précédemment décrit sont dissous dans 100 cm3 de chlorure de méthylène. On sèche la solution sur sulfate de magnésium et ajoute 4, 1 g de chlorhydrate de L-leucinate d'éthyle, puis 4, 5 g de dicyclohexylcarbodiimide. On abandonne quatre heures à une température de 25 à 300 C, ajoute 1 ce d'acide acétique et filtre la dicyclo hexylurée au bout d'une demi-heure. Le précipité de dicyclohexylurée est lavé au chlorure de méthylène, les liqueurs de lavage sont jointes au filtrat et le mélange est lavé à l'acide chlorhydrique, à l'eau, séché sur sulfate de magnésium et évaporé à sec.
Le résidu fournit l'ester du tripeptide de formule H à l'état suffisamment pur pour la saponification ultérieure.
Stade 4: PrÚparation de la S,N-ditrityl-L-cystÚinyl L-prolyl-L-lecscine de forrnule I
La totalité de l'ester du tripeptide de formule H obtenu selon le stade 3 est saponifiée à l'ébullition par 50 cm3 d'alcool Ó 95% et 40 cm : 3 de soude normale. On refroidit, verse dans l'eau glacée et extrait le précipité formé au chloroforme. L'extrait chloroformique est lavé à l'acide chlorhydrique N/5 et à l'eau, séché sur sulfate de magnésium et évaporé à sec. On reprend le résidu par 20 cm3 d'éther auxquels on ajoute 200 cm3 d'éther de pétrole léger.
Le tripeptide de formule 1 se sépare sous forme d'un produit pulvérulent, soluble dans tous les solvants usuels sauf l'éther de pétrole, suffisamment pur pour la transformation en tétrapeptide. Rendement quantitatif, F. = 120 C (déc.), [a] = + 80" 2 (c = 2 0/o, chloroforme).

L-prolyl-L-leucyl-glycinaíe de méthyle de for
mule K (R= CH,)
8, 25 g de S, N-ditrityl-L-cystéinyl-L-prolyl-L-leu- cine de formule I, obtenus selon le stade 4, sont introduits dans une solution refroidie à 0 C de 1, 5 g de chlorhydrate de glycinate de méthyle de formule
J dans 50 cm3 de chlorure de méthylène et 1, 3 cm3 de diéthylamine. On ajoute à la solution 2, 3 g de dicyclohexylcarbodiimide et abandonne trois heures à 25-30 C. On refroidit à la température ambiante, ajoute 0, 5 cm d'acide acétique et filtre après une demi-heure.
Le filtrat est lavé à l'acide chlorhydrique et à l'eau, séché sur sulfate de magnésium et évaporé à sec sous vide. On reprend par 50 cm : 3 d'éther.
L'ester méthylique du tétrapeptide de formule K cristallise en feuillets incolores (6 g). Rendement 68"/o. Recristallisé en alcool, il se présente sous forme de grands prismes incolores, F. environ 130 C (déc.) [a] 20 = + 1120 2 (c = 2 0/o, chloroforme) solubles en chloroforme, éthanol chaud, peu solubles en acétone, insolubles dans l'éther et l'eau.

2 g d'ester méthylique du tétrapeptide de formule
K sont saponifiés au reflux par 20 cm3 de méthanol et 10 cm3 de soude N/2 ; on refroidit, ajoute de l'eau glacée, acidifie à plll à 2 par addition d'acide chlorhydrique N et extrait au chloroforme.
L'extrait chlo-
roformique est lavé à l'eau jusqu'à neutralité, séché
sur sulfate de magnésium et évaporé à sec. Par
reprise au benzène, le produit de formule K cristal
lise en fines aiguilles incolores : 1, 65 g (rendement
80 /0). F. 135-1400 C puis 190o C, [a] 20 115o
2 (c 2 0/o, chloroforme). Le produit qui renferme Va mol. de benzène de solvatation est soluble dans l'acétone, l'alcool et le chloroforme, très peu soluble dans le benzène, insoluble dans l'eau et l'éther.

Stade 6 : Détritylation sélective à l'azote du S, N ditrityl-L-cystéinyl-L-prolyl-L-leucyl-glycinate de
méthyle de formule K (R = CH3) par l'acétone
chlorhydrique et prÚparation du S-trityl-L
cystéinyl-L-prolyl-L-leucyl-glycinate de méthyle de formule LR = CH3)
4, 4 g d'ester méthylique de formule K sont mis en suspension dans 20 cm3 d'acétone, on ajoute 5 cmS d'acide chlorhydrique 5 N et abandonne vingt minutes à 35O C. On ajoute 60 cm3 d'eau, extrait au chloroforme. Les extraits chloroformiques réunis sont lavés à l'acide chlorhydrique, séchés sur sulfate de magnésium et évaporés à sec sous vide.
Le résidu huileux, repris par l'éther, se prend en masse et fournit 3, 2 g de chlorhydrate du tétrapeptide S-tritylé de formule L (R = CH3) (rendement : 94 /o).
F. environ 150¯C, [α]20D = -17¯¯ l (c=2%, chloroforme), soluble en chloroforme, benzène, acétone, eau insoluble dans l'éther et les acides dilués aqueux.

En opérant comme il a été indiqué dans l'exem- ple 6 et en faisant précéder l'extraction chloroformique du peptide détritylé à l'azote par une neutralisation à l'acétate de soude, on obtient au départ de 1, 5g du produit de formule K (R = H), après traitement à l'éther, 1 g de peptide détritylé, soit un rendement de 95%.
Apr¯s recristallisation en éthanol-éther, le produit se présente en aiguilles [a] D
-28 2 (c = 2"/o, éthanol absolu), renferment Va molécule d'eau de cristallisation. Il est soluble
dans les acides et alcalis dilués aqueux, dans l'alcool chaud, peu soluble en chloroforme, insoluble dans l'eau et l'éther.
Process for preparing a polypeptide
The subject of the present invention is a process for preparing a tetrapeptide derivative, an intermediate term in the synthesis of oxytocin, a nonapeptide hormone, of which the following constitutional formula:

The tetrapeptide derivative prepared by the process forming the subject of the present invention is an ester of S-trityl-L-cysteinyl-L-propyl-L-leucyl-glycine compound of formula L, the preparation of which appears in
attached diagram and will be described later. As can be seen, this compound is very close to the terminal tetrapeptide stretch of oxytocin and closely resembles the S-benzyl-L-cysteinyl ester.
L-propyl-L-leucyl-glycine of formula B [Ch. Ressler and V. Du Vigneaud (J. Am. Chem. Soc., 1954, 76, 3107)] which, after transformation into an amide, was used by Du Vigneaud et al. (L. C) for the synthesis of oxytocin.

To arrive at this tetrapeptide of formula B,
Ch. Ressler and V. Du Vigneaud took a rather laborious approach. Using as an agent for blocking the amine function, during their peptide syntheses, benzyl chloroformate, an expensive compound, not very stable, tear gas and whose preparation involves the manipulation of phosgene and carrying out the peptide condensation by the mixed anhydrides method, they first prepared the N-benzyl-oxycarbonyl derivative of L-leucine, which condensed with ethyl glycinate, provided them with ethyl N-benzyloxy-carbonyl-L-leucyl-glycinate. Catalytic hydrogenolysis affords ethyl eucyl glycinate. After these two steps,
the product obtained is condensed with N-benzyloxycarbonyl-L-proline by the mixed anhydride process using isovaleryl chloride, which provides N-benzyloxy-carbonyl-L-prolyl-L-leu-cyl-glycinate d 'ethyl. Further hydrogenolysis to remove the protective group results in ethyl L-prolyl-L-leucyl-glycinate. This compound is condensed with bis- (N-benzyloxycarbonyl) -L-cystinyl dichloride obtained in one step from benzyloxy-carbonyl-L-cystine, which leads to bis- (N-benzyloxy-carbonyl) -L -cystinyl-bis- (Ethyl prolyl-L-leucyl-glycinate) after a total of 6 steps.
To go from this compound to the peptide of formula B, 4 more steps are needed. The ester function is saponified to the corresponding acid, this product is subjected to reduction with sodium in liquid ammonia in order to convert the cystine bridge into a cysteine function, treated with benzyl chloride to obtain the S-benzyl derivative of the tetrapeptide which we esterify again.
For industry, this synthesis has the drawback of requiring repeated elimination of a blocking agent by hydrogenolysis, of forcing blocking again and carrying out, shortly before the end of the synthesis, a reduction with sodium in the Liquid ammonia, an operation difficult to carry out on an industrial scale, and to subject an ester to preliminary saponification before this reduction, which leads to a new final esterification.
With respect to this 10-step process, the process according to the invention makes it possible to obtain the tetrapeptide ester of formula L in 6 stages, easily achievable using only a single blocking, the final derivative also exhibiting the advantage of being split more easily than the S-benzyl compounds for which the regeneration of the free mercaptan function requires a new intervention of sodium in liquid ammonia.
According to the present process, S, N ditrityl-L-cysteine is reacted in the presence of a carbodiimide.
N, N'-disubstituted with an L-proline ester, isolates the ester of the dipeptide formed, saponifies it into S, N ditrityl-L-cysteinyl-L-proline which is condensed in the presence of an N-carbodiimide, N'-disubstituted with an L-leucine ester, isolates the ester from the tripeptide formed, saponifies it to S, N-ditrityl-L-cysteinyl-L-propyl-L-leucine which is condensed in the presence of a N, N-disubstituted carbodiimide with a glycine ester, isolates the ester of the tetrapeptide formed and subjects this ester, on the one hand, to selective nitrogen detritylation and, on the other hand,
to saponification to obtain the corresponding free tetrapeptide.
The preparation of S, N-ditrityl-cysteine has already been described by the proprietor in the Swiss patent
N 357729, its preparation therefore appearing in the example only as a reminder.
The procedure described in the example which follows can obviously be employed starting from the racemic amino acids or from the enantiomorphs. It is in particular possible to vary the temperatures, change the solvent, use other amino acid esters than those mentioned or prepare them by other routes than those indicated, saponify them by any other known method or use another condensing agent. disubstituted carbodiimide than dicyclohexylcarbodiimide or use any other known peptide condensation method.
The melting points are instantaneous melting points determined on a Maquenne block. The formulas of the compounds are shown in the attached diagram.
Example
Stage 1: Preparation of SJN-ditrityl-L-cysteinyl-L-
Methyl prolinate of formula E
a) Condensation of S, N-ditrityl-L-cysteine from
formula C with methyl prolinate from for
mule D (R = CHg,)
27.5 g of crystallized S, N-ditrityl-cysteinate diethylamine are dissolved in 150 cc of chloroform.
This solution is washed with 41 cm: 3 of chlorhy acid
drique normal, then with water, dried over sodium sulphate and evaporated to dryness. On the other hand, we dissolve
6.66 g of methyl prolinate hydrochloride in 25 cil of methylene chloride; ice is carried out at ¯ 10 {} C and added, without exceeding 0t) C, 4.8 cm3 of diethylamine, then 200 cm3 of ether. Diethylamine hydrochloride, insoluble in ether precipitates. It is filtered and the methyl prolinate solution evaporated to dryness under vacuum.
The S, N-ditritylcysteine of formula C prepared as just indicated is dissolved in 50 cc of methylene chloride. Cooled to 00 C and added the methyl prolinate of formula D also dissolved in 50 cm3 of methylene chloride and cooled to 0 ° C. In this reaction mixture, 9 g of dicyclohexylcarbodiimide are introduced and left overnight at 20 C. The next day, we add
1 cm3 of acetic acid to convert excess dicyclohexylcarbodiimide into dicyclohexylurea. After half an hour, the dicyclohexylurea is filtered off and washed with methylene chloride.
The filtrate, combined with washing methylene chloride, is washed with dilute hydrochloric acid, with water, dried and evaporated to dryness.
The ester of the crude dispeptide of formula E (R = CH) thus obtained can be used directly for the second stage of the synthesis, that is to say be saponified into the dipeptide of formula F.
For the preparation of S, N-ditrityl-L-cysteine of formula C, which is given here as a reminder, the procedure is as follows: in a solution, cooled to 00, of 25 g of L-cysteine hydrochloride [a ] 20 = + 5.50 (c = 1 / o, N hydrochloric acid) in 480 cm3 of water, are introduced, under a stream of nitrogen and mechanical stirring, 480 cm3 of ether and 80 cm3 of diethylamine and cooled to -5fM C. Without interrupting the stirring, 120 g of trityl chloride are introduced and stirring continued for three hours. The reaction mixture becomes a pasty mass which is quite difficult to stir.
150 to 200 cc of chloroform are then added, decanting the aqueous phase which contains only diethylamine hydrochloride and unreacted cysteine hydrochloride. The organic layer is washed again with 300 cc of water, dried over sodium sulfate and concentrated. 100cm3 of alcohol containing 1 cc of diethylamine are then added and the solvent is distilled off until it has entrained all the chloroform. The diethylamine salt of N, S-ditrityl-L-cysteine crystallizes.
The crystallization is completed by the gradual addition of 100 cm3 of ether. 55 g of salt are thus obtained, presenting
in the form of needles [a] 2D0 = +71 + 1 (c = 2 / o,
chloroform) soluble in chloroform, very slightly soluble
wheat in alcohol, insoluble in water and ether. The
Reactions to ninhydrin are negative. From
mother liquors of crystallization, a second
product stream bringing the overall yield to 70 g
about. By taking up this salt in acidulated water, it is
decomposes with release of the N, S-ditrityl-L
cysteine of formula C which can be easily
extracted by ether in which it is quite soluble.
Evaporation of the ether provides the crude product in an amorphous state.
Methyl L-prolinate hydrochloride of formula D (R = CH3) is prepared according to J. Schumann and RA Boissonnas (Helv. Chim. Acta, 1952, 35, 2240), by the action of a stream of dry hydrochloric acid on a suspension of L-proline in methanol. 6 g of L-proline are introduced into 120 cm 3 of methanol saturated at 20 C with dry hydrochloric acid; left for 24 hours at room temperature, evaporated to dryness under vacuum.
Finally, 8 g of methyl L-prolinate hydrochloride are obtained in the form of a pale yellow oil, ready for use for condensation, ie a yield of 94/0. b) Condensation of S, N-ditritylcysteine with
Benzyl prolinate hydrochloride of the formula
D (R = CGH5-CH2-)
6.8 g of salt is reacted overnight.
S, N-ditritylcysteine and 2.5 g of benzyl prolinate hydrochloride dissolved in 50 cm3 of methylene chloride in the presence of 2.2 g of dicyclohexylcarbodiimide. After a treatment identical to that described under a), dipeptide of formula E (R = -CH., - CH) is isolated in the form of an oil.
For the preparation of benzyl L-prolinate hydrochloride D (R = CgHs-CHo-), the procedure is as follows:
15 g of L-proline are placed in a fine suspension in 150 cm3 of pure benzyl alcohol. Cooled to +10 C and saturated at this temperature with dry gaseous hydrochloric acid. After standing for some time, the mixture is heated under vacuum at 90 ° C. to remove the water formed and the hydrochloric acid. Allowed to cool and a large excess of ether added. Benzyl prolinate hydrochloride crystallizes in small colorless prisms. Ice, filtered, washed with ether and dried under vacuum. 31 g of white product are collected, [= -42 "1 (c == 2" / o absolute alcohol) identical to the compound described by RE Neumann and
E. L. Smith (J. Biol.
Chem., 1951, 193, 97).
Stage 2: Preparation of S, N-ditrityl-L-cysteinyl
L-proline of formula F
a) The methyl ester of the crude oily dipeptide of formula E obtained as just indicated in stage 1 a) is refluxed with 100 cm3 of alcohol, 30 cm3 of methanolic potassium hydroxide and 30 cm3 of water. After cooling, it is poured into a mixture of water and ice and normal hydrochloric acid is added to pH 1 (approximately 90 cm3 of N hydrochloric acid). The precipitate formed is extracted with chloroform. The chloroform solution is washed with water until neutral, dried and evaporated to dryness. The residue is taken up in 150 cm3 of anhydrous ether and brought to reflux while adding 5 cm3 of diethylamine. Leave to cool and add 2 crd of water.
The diethylamine salt of the dipeptide of formula F crystallizes in needles. 150 cm3 of light petroleum ether are added and leave overnight at room temperature. 22 g of diethylamine salt of formula F are thus obtained crystallizing with 1/2 molecule of water and sufficiently pure for condensation with methyl leucinate. Yield: 70%, F. = around 150¯ C (dÚc.), [Α] 20D = + 92¯ ¯ 2 (c = 2/0, absolute ethanol).

b) The benzyl ester of the dipeptide of formula E, the preparation of which has been described in stage 1 b) is saponified under reflux with 6 cm3 of methanol potassium hydroxide at 20/0 and 6 cm3 of water. 50 g of ice-cold water are added, acidified with hydrochloric acid to pH 2.3 and extraction is carried out with chloroform. The chloroform extract is washed with N / 10 hydrochloric acid, with water, dried over magnesium sulfate and evaporated to dryness.
The residue is taken up in 50 cc of ether and 2 cc of diethylamine. Cooled and added 1 cm3 of water. 5.8 g of diethylamine salt of S, N-ditrityl-L-cysteinyl-L-proline are collected, containing the water molecule, identical in all respects to the product described at the end of a). Yield 74/0.
Stage 3: Preparation of S, N-ditrityl-L-cysteinyl-L-
ethyl prolyl-L-leucinate (H) (R = C2Hs)
The raw materials used for this preparation are SN-ditrityl-Lcysteinyl-L-proline of formula F, prepared according to stage 2 and the hydrochloride of ethyl L-leucinate (G) (R = CgHg) of conventional preparation on which we operate directly.
15.7 g of the diethylamine salt of S, N-ditrityl-L-cysteinyl-L-proline described above are dissolved in 100 cm 3 of methylene chloride. The solution is dried over magnesium sulfate and 4.1 g of ethyl L-leucinate hydrochloride, then 4.5 g of dicyclohexylcarbodiimide. It is left for four hours at a temperature of 25 to 300 C, 1 cc of acetic acid is added and the dicyclohexylurea filtered off after half an hour. The dicyclohexylurea precipitate is washed with methylene chloride, the washing liquors are added to the filtrate and the mixture is washed with hydrochloric acid, with water, dried over magnesium sulphate and evaporated to dryness.
The residue provides the ester of the tripeptide of formula H in a sufficiently pure state for subsequent saponification.
Stage 4: Preparation of S, N-ditrityl-L-cystÚinyl L-prolyl-L-lecscine from Formula I
All of the ester of the tripeptide of formula H obtained according to step 3 is saponified at the boiling point with 50 cm3 of 95% alcohol and 40 cm: 3 of normal sodium hydroxide. Cooled, poured into ice water and the precipitate formed is extracted with chloroform. The chloroform extract is washed with N / 5 hydrochloric acid and with water, dried over magnesium sulfate and evaporated to dryness. The residue is taken up in 20 cm3 of ether to which 200 cm3 of light petroleum ether are added.
The tripeptide of formula 1 separates out in the form of a pulverulent product, soluble in all the usual solvents except petroleum ether, sufficiently pure for conversion into the tetrapeptide. Quantitative yield, F. = 120 C (dec.), [A] = + 80 "2 (c = 20 / o, chloroform).

8, 25 g of S, N-ditrityl-L-cysteinyl-L-prolyl-L-leucine of formula I, obtained according to stage 4, are introduced into a solution cooled to 0 C of 1, 5 g of hydrochloride. of methyl glycinate of formula
J in 50 cm3 of methylene chloride and 1.3 cm3 of diethylamine. 2.3 g of dicyclohexylcarbodiimide are added to the solution and left for three hours at 25-30 C. The mixture is cooled to room temperature, 0.5 cm of acetic acid is added and filtered after half an hour.
The filtrate is washed with hydrochloric acid and with water, dried over magnesium sulfate and evaporated to dryness under vacuum. The residue is taken up in 50 cm: 3 of ether.
The methyl ester of the tetrapeptide of formula K crystallizes in colorless sheets (6 g). Yield 68 "/ o. Recrystallized in alcohol, it is in the form of large colorless prisms, F. about 130 C (dec.) [A] 20 = + 1120 2 (c = 20 / o, chloroform) soluble in chloroform , hot ethanol, poorly soluble in acetone, insoluble in ether and water.

The combined chloroform extracts are washed with hydrochloric acid, dried over magnesium sulphate and evaporated to dryness under vacuum.
The oily residue, taken up in ether, solidifies and gives 3.2 g of hydrochloride of the S-trityl tetrapeptide of formula L (R = CH3) (yield: 94 / o).
