Verfahren zur Herstellung von Tachysterol oder andern Verbindungen mit Tachysterolstruktur Die Erfindung bezieht sich auf ein Verfahren zur Herstellung von Tachysterol oder andern Trans- Verbindungen der Formel:

worin R ein gesättigter oder ein eine oder mehrere Doppelbindungen aufweisender aliphatischer Kohlen wasserstoffrest ist, oder von Estern dieser Verbin dungen.
Unter einem Tachysterol wird eine Verbindung verstanden, die durch Bestrahlung mit Ultraviolett licht einer Lösung eines 3-Hydroxy-delta-5,7-sterols entsteht, welches im Absorptionsspektrum ein Maximum bei 2810 A aufweist.
Die oben angegebene Struktur ist in einer Publi kation von Inhoffen [Chemische Berichte 87, 1407 bis 1425 (l954)] angegeben.
Es wurde nun gefunden, dass man, Tachysterol oder Transverbindungen der Formel I oder Ester dieser Verbindungen dadurch herstellen kann, dass einer neutralen oder schwachalkalischen Lösung einer Cis-Verbindung der Formel:

die bei 2625 10 A ein Maximum im Absorptions- spektrum hat, oder eines Esters einer solchen Ver bindung, ein Katalysator zugesetzt wird, der Cis- Isomere in Trans-Isomere umwandelt.
Präcalciferol, eine Verbindung der Formel II, entsteht z. B. bei Bestrahlung von 3-Hydroxy- delta-5,7-sterol mit Ultraviolettlicht und hat bei 2625 10 A ein Maximum im Absorptionsspek trum.
Es ist bekannt, d'ass es verschiedene Steroide gibt, die an der Stelle 3 des Sterolskelettes eine Hy- droxylgruppe gebunden haben und bei denen zwi schen den Kohlenstoffatomen 5, 6 und 7, 8 je eine Doppelbindung auftritt.
Entsprechend der vorstehend angegebenen Defi nition gibt es verschiedene Verbindungen, die alle als Tachysterol bezeichnet werden; können, z. B. Tachystero12, eine Verbindung, welche man beispiels weise durch Bestrahlung von Ergosterol bzw. durch Reduktion des Bestrahlungsproduktes erhalten kann.
Entsprechend der hier angewandten Bezeichnungen des Bestrahlungsproduktes von Ergosterol nennt man das Bestrahlungsprodukt von 7-Dehydrocholesterol, Tachysterol3. Das aus Tachysterol, durch Reduktion erhaltene Produkt kann als Dihydrotachysterol, be zeichnet werden.
Es ist bekannt, dass Dihydrotachy- sterol, eine Verbindung ist, die einen stark erhöhen den Einfluss auf den, Calciumspiegel des Blutes hat. Die Anmelderin hat gefunden, dass dies auch zutrifft bei Dihydrotachysterol, enthaltenden Gemischen.
Windaus hat in Annalen der Chemie Bd. 499, Seiten 190 und ff. von 1932 ein Verfahren, beschrie ben, um durch Ultraviolettbestrahlung von Ergosterol Tachystero4 herzustellen. Der Verfasser meldet, dass Tachysterol2 bei Ultraviolettbestrahlung aus Ergo- sterol erzeugt wird, und dass die Umwandlung bei Anwendung von Ultraviolettlicht mit einer Wellen- läng e von weniger als 2840 A besonders ausgeprägt ist.
Indem das rohe Bestrahlungsgemisch, nach vor hergehender Entfernung von nicht umgewandeltem Ergosterol, mit 3,5 Dinitro-4methyl-benzoylchlorid in Pyridin gelöst wird, entsteht der entsprechende Ester von Tachystero12. Diese Verbindung lässt sich aus Aceton umkristallisieren und schmilzt bei 154 bis 155 .
Diese Herstellung von Tachystero12 hat verschie dene Nachteile.
In der Praxis gibt es eine geringe maximale Aus beute an Tachystero4 pro Zeiteinheit und Lichtquant bei der Bestrahlung.von Ergosterol mit kurzwelligem Ultraviolettlicht mit einer Wellenlänge, bei der die Ausbeute an Tachysterol pro Lichtquantum maximal ist (bei einer Wellenlänge von weniger als 2840 A). Man verwendet für die Bestrahlung mit Licht des angegebenen Wellenlängenbereiches Niederdruck Quecksilber-Dampfröhren.
Diese Röhren haben eine verhältnismässig geringe Leistung. Infolgedessen müssen viele Röhren verwendet werden oder die Reaktion muss längere Zeit fortgesetzt werden, um eine zufriedenstellende Ausbeute zu erhalten.
Ein anderer Nachteil ist der, dass bei der Be strahlungsreaktion eine grosse Anzahl von Neben produkten erzeugt wird, und dass die Isolierung und Reinigung von Tachysterol, verhältnismässig um- ständlich ist.
Wenn man nun, gemäss der Erfindung, das Tachysterol oder dessen Ester aus Präcalciferol oder dessen Estern herstellt, werden die obgenannten Nachteile vermieden.
Ein Verfahren zur Herstellung des gemäss der Erfindung erforderlichen Ausgangsmaterials der For mel II, z. B. Präcalcifero4 aus Ergosterol oder eines andern Präcalciferols aus 7-Dehydrocholesterol (so genanntes Präcalcifero13), ist durch Velluz in Bulle tin Soc. Chin. 1949, Seite 501 beschrieben.
Katalysatoren, die die Umwandlung von Cis- in Trans-Verbindungen zustande bringen können, sind z. B. Jod oder Eosin unter der Wirkung von Licht, das während der Reaktion absorbiert wird.
Die Erfindung ist besonders von Bedeutung, um aus Präcalciferol, oder aus Präcalciferol, bzw. aus den Estern derselben, Tachysterol, oder Tachystero13 bzw. die Ester derselben herzustellen.
Es ist sehr wichtig, dass während der Reaktion das Lösungsmittel keine saure Reaktion aufweist. Es wurde gefunden, dass sogar die Bildung von Säure spuren einen beeinträchtigenden Einfluss auf die Aus beute an Tachysterol hat. Aus diesem Grunde ist es empfehlenswert, dem verwendeten: Lösungsmittel eine Verbindung zuzusetzen, die dazu fähig ist, gege benenfalls erzeugte Säuren zu binden.
Solche für diesen Zweck geeignete Verbindungen sind z. B. tertiäre Amine, die den Katalysator nicht inaktivieren, wie Pyridin, Kollidin und Chinolin.
Da auch die verwendeten Lösungsmittel unter der Wirkung des Katalysators eine Säure bilden könnten, ist es empfehlenswert, das Lösungsmittel mit einiger Sorgfalt auszuwählen. Dazu besonders geeignet sind Kohlenwasserstoffe einzeln, oder im Gemisch, z. B. Petroleumäther, Ligroin, Benzin. Weiter wurden sehr gute Resultate mittels Benzol, Toluen und Zyklohexan erzielt.
Es können auch Äther, z. B. Diäthyläther, und weiter Schwcfelkohlenstoff verwendet werden. Wenn die Umwandlung von Präcalciferol in Tachysterol in einer Flüssigkeit stattfindet, die im vorangehenden Abschnitt erwähnt wurde und kein säurebindendes Mittel beigefügt wird, so ist es empfehlenswert, die Konzentration von Präcalciferol niedrig zu halten, das heisst niedriger als 1 mg Präcalciferol pro 100 cm3 Lösungsmittel, da sonst das sogenannte Isotachysterol entsteht.

Es hat sich erwiesen, dass die erfindungsgemässe Umwandlung von Präcalciferol in Tachysterol wenig von dem Vorhandensein von Produkten beeinträch tigt wird, die bei der Bildung von Präcalciferol ent stehen, z. B. die Nebenprodukte, der Bestrahlung eines 3-Hydroxy-delta-5,7-sterols.
Zur Herstellung eines Tachysterols oder eines Esters desselben kann man daher sowohl von dem rohes Präcalciferol enthaltenden. Produkt als auch von Präcalciferolen oder den Estern derselben aus gehen, die einer Reinigung unterworfen werden.
Um ein möglichst reines Tachysterol oder ein Ester desselben herzustellen, ist als Ausgangsmaterial vorzugsweise ein möglichst reines Präcalciferol oder ein, Ester desselben zu wählen. Man kann Präcalci- ferol am einfachsten in dem reinen Zustand gemäss dem Verfahren nach Velluz herstellen.
Eine möglichst grosse Ausbeute an Tachysterol, berechnet nach 3 Hydroxy-delta-5,7-sterol, wird hin gegen erzielt, wenn das rohe Bestrahlungsprodukt eines 3-Hydroxy-5,7-sterols ohne Zwischenreinigung dem Einfluss des Katalysators unterworfen wird.
Bei der Bestrahlungsreaktion wird nämlich sowohl ein Präcalciferol als auch ein Tachysterol erzeugt. In folge der Umwandlung von Präcalciferol in Tachy- sterol mittels eines Katalysators, wird die Maximal- erzeugung von Tachysterol, berechnet, nach 3 Hy- droxy-delta-5,7-sterol oder einem Ester desselben,
als Ausgangsprodukt erzielt.
Wenn das rohe Bestrahlungsprodukt von einem 3-Hydroxy-delta-5,7-sterol als Ausgangsprodukt zur Herstellung eines Tachysterols verwendet wird, ist es möglich, die katalytische Umwandlung des erzeug ten Präcalciferols in ein Tachysterol in demselben Lösungsmittel durchzuführen, das bei der Bestrah lung verwendet wurde; wenigstens, wenn die Lösung sich dazu eignet, die katalytische Umwandlung in ihr stattfinden zu lassen. Solche Lösungsmittel sind z. B. Benzen, Zyklohexan oder Diethyläther.
Die Reinigung des rohen Tachysterols braucht dem Fachmann keine besonderen Schwierigkeiten, zu bereiten. Wenn das Tachysterol dadurch erhalten, ist, dass das rohe Bestrahlungsprodukt mit dem Kataly sator behandelt wurde, so dass das rohe Tachysterol noch 3-Hydroxy-delta-5,7-sterol enthalten wird, kann letztere Verbindung durch Umkristallisierung oder durch Ablagerung mit Digitonin leicht entfernt werden.
Aus dem Rest kann das Tachysterol da durch isoliert werden, dass die Verbindung in ein leicht kristallisierbares Ester umgewandelt wird, z. B. ein Ester von 3,5-Dinitro-4-methyl-benzoesäure.
Die Umwandlung von Präcalciferol in Tachy- sterol lässt sich nicht mit Gewissheit erklären. Auf Grund der gemäss der Erfindung erzielten Resultate kann angenommen werden, dass ein Präcalciferol, anders als bisher vorausgesetzt wurde, nachstehende Cis-Struktur besitzt:

Aus dieser Formel ist er sichtlich, dass ein Präcalciferol als ein Cis-Isomer eines Tachysterols betrachtet werden kann, welches Trans-Struktur besitzt.
<I>Beispiele</I> 1. Durch alkalische Hydrolyse von 3,5-Dinitro- benzoesäure-ester von Prävitamin D2 (Ve11uz u. a. Bulletin Soc. Chim. 2949, Seite 501 ff.) wurde Prä- vitamin D2 als farbloser, harzartiger Stoff mit einem Ultraviolettabsorptionsmaximum bei 2630 A erhal ten. Davon wurde 199 mg in 100 cm3 Petroleumäther (Siedestrecke 40 bis 60 ) gelöst.
Dieser Lösung wurde eine Lösung von 0,25 mg Jod in 0,1 em3 Petroleumäther zugesetzt. Das Gemisch wurde in einer Stickstoffatmosphäre mit einer Glühlampe von 200 W belichtet. Nach 10 Minuten ergab sich bei spektro-photometrischer Untersuchung, dass das Ultraviolettabsorptionsspektrum der Lösung, die für Tachysterol, kennzeichnende Form aufwies, das heisst mit einem Absorptionsmaximum bei 2810 A.
2. Eine 0,2 %-Lösung von Ergosterot in Äthanol wurde mit Ultraviolettlicht längerer Wellenlängen als 2650 A belichtet.
Die Bestrahlung wurde fortgesetzt, bis 35% des Ergosterols umgewandelt war. Aus der auf diese Weise erhaltenen Lösung, die nicht nur nicht umgewandeltes Ergosterol, sondern auch eine grosse Menge Prävitamin D2 und eine kleine Menge Lumistero12 und Tachystero12 enthielt, wurde der grösste Teil des nicht umgewandelten Ergosterols da durch entfernt,
dass die Lösung schnell bei Zimmer temperatur auf ein kleines Volumen eingedampft wurde, worauf der Rest bei -5 C kristallisiert und das niedergeschlagene Ergosterol abfiltriert wurde. Das auf diese Weise erhaltene Filtrat wurde im Vakuum trockengedampft. Von dem Rest wurde 1,5 mg in<B>100</B> cm3 Zyklohexan gelöst. Diese Lösung wies ein Ultraviolettabsorptionsmaximum bei 2630 A auf; E i % des gelösten Stoffes ist 193.
Der Zyklohexanlösung wurde 0,02 mg Jod in 0,15 cm3 Zyklohexan zugesetzt. Nach einer Belich tung von 10 Minuten in einer Stickstoffatmosphäre mit einer Lampe von 200 W wurde das Ultraviolett absorptionsspektrum gemessen. Es ergab sich, dass es ein Maximum bei 2800 A (Ei% des gelösten Stoffes = 302) hatte, was auf die Bildung von Tachy- sterol2 aus Prävitamin D2 deutete.
3. 500 mg Prävitamin D2 wurde in 30 cm-' Di- äthyläther gelöst, dem 10 mg Pyridin zugesetzt wor den war. In der Lösung wurde 10 mg Jod gelöst, worauf das Gemisch in einem Glaskolben, in dem die Luft durch Stickstoff ersetzt war, während 40 Minuten in diffusem Tageslicht gehalten wurde. Nach dieser Zeitspanne wurde das Gemisch in einem Scheidetrichter unter Stickstoff mit einer verdünnten Thiosulfatlösung geschüttelt, um das Jod zu entfer nen.
Die ätherische Lösung wurde nach Trocknen abdestilliert, der Rest wurde in einigem cm3 Benzen gelöst, das im Vakuum abdestilhert wurde, um Feuchtigkeitsspuren zu entfernen. Der Rest wurde in 5 cm3 wasserfreiem Benzen gelöst, worauf dieser Lösung 3 cm3 wasserfreies Pyridin und darauf eine Lösung von 0,5 g 3,5-Dinitro-4-methyl-benzoyl- chlorid in Benzol zugesetzt wurde,
unter Abschluss gegen Feuchtigkeit und in einer Stickstoffatmosphäre. Nachdem das Gemisch bei Zimmertemperatur wäh rend 18 Stunden aufbewahrt worden war, wurde Tachystero12 Dimtro-methyl-benzesäureester dadurch isoliert, dass dem Reaktionsgemisch Diäthyläther und Wasser zugesetzt wurde und der ätherische Extrakt mit verdünnter Säure, wässeriger Bicarbonatlösung, einer Salzlösung und Wasser gewaschen wurde, worauf der Extrakt getrocknet und einer Vakuum destillation unterworfen wurde.
Der Ester wurde aus Aceton umkristallisiert und ergab sich entsprechend dem Mischschmelzpunkt und der Röntgenaufnahme pulverisierter Präparate- identisch mit auf andere Weise hergestellten Tachysterol2 Estern.
Das daraus durch alkalische Hydrolyse zu erhaltende Tachystero12 hat in einer Ätherlösung ein Ulltraviolettabsorptions- spektrum, das dem eines auf andere Weise herge stellten Tachystero12 Präparats identisch ist mit einem Ultraviolettabsorptionsmaximum von 2810 A (E1% = 700). Auch das Infrarotabsorptionsspektrum ist identisch mit dem eines bekannten Tachystero12 Präparats.
4. 2 mg Prävitamin D2 wurde in 100 cm3 Zyklo- hexan gelöst. Der Lösung wurde etwa 0,25 cm3 einer gesättigten Lösung von Eosin in Zyklohexan zugesetzt. Die Lösung wurde in einem Glaskolben in dem direkten Sonnenlicht belichtet.
Nach 30 Minuten wurde das Ultraviolettabsorptionsspektrum be- stimmt. Das Maximum bei 2630 A des Prävitamins D2 war nach 2700-2800 A verschoben, woraus die Bildung von Tachysterol2 ersichtlich war.

A tachysterol is understood to mean a compound which is formed by irradiation with ultraviolet light of a solution of a 3-hydroxy-delta-5,7-sterol which has a maximum at 2810 A in the absorption spectrum.
The structure given above is given in a publication by Inhoffen [Chemical Reports 87, 1407 to 1425 (1954)].
It has now been found that tachysterol or trans compounds of the formula I or esters of these compounds can be prepared by adding a neutral or slightly alkaline solution of a cis compound of the formula:

Precalciferol, a compound of the formula II, is formed e.g. B. upon irradiation of 3-hydroxy delta-5,7-sterol with ultraviolet light and has a maximum in the absorption spectrum at 2625 10 A.
It is known that there are various steroids which have a hydroxyl group attached to position 3 of the sterol skeleton and in which a double bond occurs between carbon atoms 5, 6 and 7, 8.
According to the definition given above, there are various compounds, all of which are referred to as Tachysterol; can e.g. B. Tachystero12, a compound which can be obtained, for example, by irradiating ergosterol or by reducing the irradiation product.
According to the names used here for the radiation product of ergosterol, the radiation product of 7-dehydrocholesterol is called tachysterol3. The product obtained from tachysterol by reduction can be referred to as dihydrotachysterol.
It is known that dihydrotachosterol is a compound that has a strong influence on the calcium level of the blood. The applicant has found that this also applies to mixtures containing dihydrotachysterol.
Windaus has in Annalen der Chemie Vol. 499, pages 190 and ff. Of 1932, a process described ben to produce Ergosterol Tachystero4 by ultraviolet irradiation. The author reports that Tachysterol2 is generated from ergot with ultraviolet radiation and that the conversion is particularly pronounced when ultraviolet light with a wavelength e of less than 2840 A is used.
By dissolving the crude irradiation mixture in pyridine with 3,5 dinitro-4methyl-benzoylchloride, after prior removal of unconverted ergosterol, the corresponding ester of tachystero12 is formed. This compound can be recrystallized from acetone and melts at 154 to 155.
This production of tachystero12 has various disadvantages.
In practice, there is a low maximum yield of Tachystero4 per unit of time and light quantum during irradiation of ergosterol with short-wave ultraviolet light with a wavelength at which the yield of tachysterol per light quantum is maximum (at a wavelength of less than 2840 A). Low-pressure mercury vapor tubes are used for irradiation with light of the specified wavelength range.
These tubes have a relatively low power. As a result, many tubes must be used or the reaction must be continued for a long time in order to obtain a satisfactory yield.
Another disadvantage is that a large number of by-products are generated during the irradiation reaction and that the isolation and purification of Tachysterol is relatively laborious.
If, according to the invention, the tachysterol or its ester is produced from precalciferol or its esters, the above-mentioned disadvantages are avoided.
A process for the preparation of the required according to the invention starting material of the For mel II, for. B. Precalciferol from ergosterol or another precalciferol from 7-dehydrocholesterol (so-called precalciferol), is by Velluz in Bulle tin Soc. Chin. 1949, page 501.
Catalysts that can bring about the conversion of cis to trans compounds are, for. B. iodine or eosin under the action of light absorbed during the reaction.
The invention is particularly important in order to produce tachysterol or tachystero13 or the esters thereof from precalciferol, or from precalciferol, or from the esters thereof.
It is very important that the solvent does not have an acidic reaction during the reaction. It was found that even the formation of traces of acid has a detrimental effect on the yield of Tachysterol. For this reason, it is advisable to add a compound to the solvent used that is capable of binding any acids that may be generated.
Such compounds suitable for this purpose are e.g. B. tertiary amines that do not inactivate the catalyst, such as pyridine, collidine and quinoline.
Since the solvents used could also form an acid under the action of the catalyst, it is advisable to choose the solvent with some care. Particularly suitable for this are hydrocarbons, individually or in a mixture, e.g. B. petroleum ether, ligroin, gasoline. In addition, very good results were achieved using benzene, toluene and cyclohexane.
Ether can also be used, e.g. B. diethyl ether, and further carbon disulfide can be used. If the conversion of precalciferol into tachysterol takes place in a liquid that was mentioned in the previous section and no acid binding agent is added, it is advisable to keep the concentration of precalciferol low, i.e. less than 1 mg of precalciferol per 100 cm3 of solvent, otherwise the so-called isotachysterol is formed.

It has been shown that the inventive conversion of precalciferol into tachysterol is taken little impaired by the presence of products that are ent in the formation of precalciferol, z. B. the by-products, the irradiation of a 3-hydroxy-delta-5,7-sterol.
For the preparation of a tachysterol or an ester thereof, one can therefore use both of the crude precalciferol containing. Product as well as precalciferols or the esters of the same, which are subjected to purification.
In order to produce the purest possible tachysterol or an ester thereof, a precalciferol as pure as possible or an ester thereof should preferably be selected as the starting material. The easiest way to produce precalciferol is in the pure state using the Velluz method.
On the other hand, the greatest possible yield of tachysterol, calculated according to 3-hydroxy-delta-5,7-sterol, is achieved if the crude irradiation product of a 3-hydroxy-5,7-sterol is subjected to the influence of the catalyst without intermediate purification.
Namely, both a precalciferol and a tachysterol are generated during the irradiation reaction. As a result of the conversion of precalciferol into tachysterol by means of a catalyst, the maximum production of tachysterol is calculated according to 3 hydroxy-delta-5,7-sterol or an ester thereof,
achieved as a starting product.
If the crude irradiation product of a 3-hydroxy-delta-5,7-sterol is used as the starting product for the production of a tachysterol, it is possible to carry out the catalytic conversion of the precalciferol produced into a tachysterol in the same solvent that was used during the irradiation was used; at least if the solution is suitable for allowing the catalytic conversion to take place in it. Such solvents are e.g. B. benzene, cyclohexane or diethyl ether.
The person skilled in the art does not need to present any particular difficulties in cleaning the raw tachysterol. If the tachysterol is obtained by treating the raw irradiation product with the catalyst, so that the raw tachysterol will still contain 3-hydroxy-delta-5,7-sterol, the latter compound can easily be recrystallized or deposited with digitonin removed.
The tachysterol can be isolated from the rest by converting the compound into an easily crystallizable ester, e.g. B. an ester of 3,5-dinitro-4-methyl-benzoic acid.
The conversion of precalciferol into tachysterol cannot be explained with certainty. On the basis of the results achieved according to the invention, it can be assumed that a precalciferol, contrary to what was previously assumed, has the following cis structure:

From this formula it can be seen that a precalciferol can be regarded as a cis isomer of a tachysterol, which has a trans structure.
<I> Examples </I> 1. By alkaline hydrolysis of 3,5-dinitrobenzoic acid ester of previtamin D2 (Ve11uz et al. Bulletin Soc. Chim. 2949, page 501 ff.), Previtamin D2 became more colorless, more resinous Substance with an ultraviolet absorption maximum at 2630 A received. 199 mg of this was dissolved in 100 cm3 petroleum ether (boiling range 40 to 60).
A solution of 0.25 mg of iodine in 0.1 cubic meters of petroleum ether was added to this solution. The mixture was exposed to an incandescent lamp of 200 W in a nitrogen atmosphere. After 10 minutes, a spectrophotometric examination showed that the ultraviolet absorption spectrum of the solution had the characteristic shape for tachysterol, i.e. with an absorption maximum at 2810 A.
2. A 0.2% solution of Ergosterot in ethanol was exposed to ultraviolet light of wavelengths longer than 2650 Å.
The irradiation was continued until 35% of the ergosterol was converted. Most of the unconverted ergosterol was removed from the solution obtained in this way, which not only contained not converted ergosterol, but also a large amount of previtamin D2 and a small amount of Lumistero12 and Tachystero12,
that the solution was quickly evaporated to a small volume at room temperature, whereupon the remainder crystallized at -5 C and the precipitated ergosterol was filtered off. The filtrate obtained in this way was evaporated dry in vacuo. 1.5 mg of the remainder were dissolved in 100 cm 3 of cyclohexane. This solution had an ultraviolet absorption maximum at 2630 Å; E i% of the solute is 193.
0.02 mg of iodine in 0.15 cm3 of cyclohexane was added to the cyclohexane solution. After exposure for 10 minutes in a nitrogen atmosphere with a 200 W lamp, the ultraviolet absorption spectrum was measured. It was found that it had a maximum at 2800 A (egg% of the solute = 302), which indicated the formation of tachystero- l2 from previtamin D2.
3. 500 mg previtamin D2 was dissolved in 30 cm- 'diethyl ether to which 10 mg pyridine had been added. 10 mg of iodine were dissolved in the solution, whereupon the mixture was kept in diffuse daylight for 40 minutes in a glass flask in which the air had been replaced by nitrogen. At the end of this time, the mixture was shaken with a dilute thiosulfate solution in a separatory funnel under nitrogen to remove the iodine.
The ethereal solution was distilled off after drying, the remainder was dissolved in a few cm3 of benzene, which was distilled off in vacuo to remove traces of moisture. The remainder was dissolved in 5 cm3 of anhydrous benzene, whereupon 3 cm3 of anhydrous pyridine and then a solution of 0.5 g of 3,5-dinitro-4-methyl-benzoyl chloride in benzene were added,
sealed against moisture and in a nitrogen atmosphere. After the mixture had been kept at room temperature for 18 hours, Tachystero12 Dimtro-methyl-benzoic acid ester was isolated by adding diethyl ether and water to the reaction mixture and washing the ethereal extract with dilute acid, aqueous bicarbonate solution, a saline solution and water, whereupon the extract was dried and subjected to vacuum distillation.
The ester was recrystallized from acetone and was found according to the mixed melting point and the X-ray photograph of powdered preparations - identical to Tachysterol2 esters produced in another way.
The tachystero12 to be obtained therefrom by alkaline hydrolysis has an ultraviolet absorption spectrum in an ether solution which is identical to that of a tachystero12 preparation produced in another way with an ultraviolet absorption maximum of 2810 A (E1% = 700). The infrared absorption spectrum is also identical to that of a well-known Tachystero12 preparation.
4. 2 mg previtamin D2 was dissolved in 100 cm3 cyclohexane. About 0.25 cc of a saturated solution of eosin in cyclohexane was added to the solution. The solution was exposed to direct sunlight in a glass flask.
After 30 minutes, the ultraviolet absorption spectrum was determined. The maximum at 2630 Å of previtamin D2 was shifted to 2700-2800 Å, from which the formation of tachysterol2 was evident.
