Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass man ein Steroid der Formel

in der R, eine Acyl-, Alkyl-, Aryl- oder Aralkyl- gruppe bedeutet, in Gegenwart von Ammoniak oder einem aliphatischen primären Amin mit 1 bis 6 Kohlenstoffatomen mit einem Alkalimetall behandelt.
Die neuen Verbindungen besitzen sehr kräftige Hemmwirkung gegen Geschlechtshormone (gonad- inhibiting effects) und zeigen anabolische, andro- genische und progrestative Eigenschaften. Die Ausgangsstoffe können wie folgt erhalten werden: 44-3-Hydroxy-19-nor-steroide können durch Reduktion der entsprechenden 3-Keto-Verbindungen erhalten werden.
Diese Reduktion kann mit Hilfe eines der üblichen Reduktionsmittel, wie einem Alkaliborhydrid, einem Aluminiumisopropoxyd in Isopropanol durchgeführt werden.
Die Verätherung kann durchgeführt werden, indem man das 44-3-Hydroxy-19-nor-steroid in einem organischen Lösungsmittel mit einem Alkohol in Gegenwart einer anorganischen Säure, wie Salz säure, behandelt. In manchen Fällen kann der Alkohol gleichzeitig auch als Lösungsmittel dienen.
Man kann die Verätherung auch durchführen, indem man die 3-Hydroxy-Verbindung mit einem Kohlen wasserstoffhalogenid, wie Triphenylmethylhalogenid, in Gegenwart von z. B. Pyridin behandelt.
Die Äthergruppe kann sein: eine Alkoxy-, Aryl- oxy- oder Aralkoxygruppe, z. B. eine Methoxy-, Äthoxy-, fsopropoxy-, Pentoxy-, Phenyloxy-, Benzoxy- oder Triphenylmethoxygruppe.
Die Veresterung kann vorgenommen werden, indem man die d4-3-Hydroxy-Verbindung mit einer Säure, vorzugsweise in Gegenwart eines Dehydrie- rungsmittels, wie Äthoxyacetylen, behandelt, oder indem man mit einem Säureanhydrid, gegebenenfalls in Gegenwart eines geeigneten Lösungsmittels, wie Pyridin, umsetzt. Man kann die Veresterung auch mittels eines Säurechlorids, vorzugsweise in Gegen wart einer tertiären Base, wie Pyridin oder Chinolin, vornehmen.
Das erfindungsgemässe Abspalten des Substituen- ten in 3-Stellung wird vorzugsweise in einem geeigne ten Lösungsmittel, beispielsweise einen niederen ali- phatischen Äther, wie Dimethyläther, Methyläthyl- äther, Diäthyläther sowie Dioxan oder Tetrahydro- furan, vorgenommen.
Das bei der Reaktion anzuwendende Alkali- metall kann Lithium, Natrium oder Kalium sein. Als vorteilhaft hat sich Lithium erwiesen.
Als Beispiele für niedere aliphatische primäre Amine seien erwähnt: Methylamin, Äthylamin, n - Propylamin, Isopropylamin, n - Butylamin und n-Amylamin.
Wenn die Abspaltung in Gegenwart eines niede ren aliphatischen primären Amins vorgenommen wird, wird die fragliche Reaktion z. B. beim Siede punkt dieses Amins durchgeführt. Wenn jedoch der Siedepunkt dieses Amins 0 übersteigt, ist es ratsam, die Reduktion unter 0 durchzuführen.
Gewünschtenfalls können in erhaltenen Verbin dungen freie Hydroxylgruppen verestert werden.
Die Veresterung wird vorzugsweise mit einer ali- phatischen, aromatischen oder araliphatischen Car- bonsäure mit 1 bis 10 Kohlenstoffatomen oder mit deren funktionellen Derivaten durchgeführt, z.
B. mit Essigsäure, Propionsäure, Buttersäure, Valeriansäure, Capronsäure, Isocapronsäure, Bernsteinsäure, Wein säure, Cyclopentylessigsäure, ss-Cyclopentylpropion- säure, Cyclohexylessigsäure, "-Cyclohexylbuttersäure, Phenylessigsäure, ss - Phenylpropionsäure, Benzoe- säure, Glycin,
Ahnin und Phenylalanin.
<I>Beispiel 1</I> Zu einer Lösung von 3,0 g 44-11ss,17ss-Di- hydroxy - 3 - keto - 17a - methyl-19 -nor-androsten in 150 ml Methanol werden 1,4g Natriumborhydrid gegeben. Nach 20minutigem Stehenlassen wird das Reaktionsgemisch mit Eisessig neutralisiert und dann im Vakuum auf 25 ml eingeengt. Der Rückstand wird in 200 ml Wasser gegossen, worauf das 44 - 3,1 lss,17ss - Trihydroxy - 17a - methyl - 19 -nor- androsten auskristallisiert.
Die abgenutschten Kristalle werden in 112 ml Methanol gelöst, worauf 1,6 ml 360/0ige Salzsäure dieser Lösung zugefügt werden. Die resultierende Lösung wird eine Stunde bei Raumtemperatur ge rührt, worauf man das Reaktionsgemisch mit Na triumbicarbonat neutralisiert und auf 25 ml einengt. Der Rückstand wird in 200 ml Wasser gegossen, dann werden die entstandenen Kristalle von d4-3- Methoxy -11,8,17ss - dihydroxy -17a - methyl-19 -nor- androsten abgesaugt und anschliessend aus Aceton umkristallisiert.
2,3 g dieser Verbindung werden in 25 ml trockenem Äther gelöst, worauf eine Lösung von 2 g Lithium in 90g Äthylamin tropfenweise dieser Lö sung zugeführt wird. Nach 10minutigem Rühren wird der überschuss von Lithium durch langsames Zugeben von 15 ml Methanol zu dem Reaktions gemisch entfernt. Nach dem Entfärben des Reak tionsgemisches wird das Äthylamin im Vakuum ab gedampft. Der Rückstand wird in Eiswasser auf genommen, worauf die Mischung mehrmals mit Äther extrahiert wird. Die gesammelten Äther extrakte werden gewaschen, und danach der Äther im Vakuum verdampft.
Der Rückstand wird zwi- schen 70% Methanol und Petroläther verteilt, die Petrolätherschicht abgetrennt, getrocknet und schliess lich zur Trockne eingedampft. Nach dem Umkristal- lisieren aus Petroläther stellt der Rückstand das 44-1 lss,17ss-Dihydroxy-17a-methyl-19-nor-androsten dar.
<I>Beispiel 2</I> In analoger Weise wie in Beispiel 1 beschrieben, wird das 44-3-Keto-Ilss,17ss-dihydroxy-17a-vinyl- 19-nor-androsten zu der entsprechenden 3-Hydroxy- Verbindung reduziert, die dann mit Isopropanol und Salzsäure in die entsprechende 3-Isopropoxyverbin- dung umgewandelt wird.
Nach dem Verfahren des Beispiels 1 wird diese Verbindung mit einer Lösung von Natrium in Me- thylamin behandelt, wodurch das J4-llss,17ss-Di- hydroxy-17a-vinyl-19-nor-androsten erhalten wird. <I>Beispiel 3</I> 2,5 g d4-3,20,21-Tripropionyloxy-19-nor-pregnen wird in 120 ml absolutem Äther gelöst. Danach wird der Lösung eine Lösung von 0,9 g Natrium in 25 ml Methylamin zugegeben. Die blaue Lösung wird an schliessend 2 Stunden bei -15 C gerührt, wonach 18 ml absolutes Äthanol dem Reaktionsgemisch zugefügt werden.
Dann verdampft man das Methyl- amin, verdünnt den Rückstand mit Wasser und extrahiert das Gemisch einige Male mit Äther. Die gesammelten Ätherextrakte werden mit Wasser ge waschen, über Natriumsulfat getrocknet und zur Trockne eingedampft. Der Rückstand wird aus Me thanol und anschliessend aus Petroläther umkristalli siert, wodurch man das J4-20,21-Dipropionyloxy- 19-nor-pregnen erhält.
<I>Beispiel 4</I> 2,1 g d4-3-Methoxy-20-hydroxy-19-nor-pregnen wird in 120 ml absolutem Äther gelöst, worauf 120 ml flüssiges Ammoniak und dann 0,9 g in kleine Stücke geschnittenes Natrium zugegeben werden. Die blaue Lösung wird anschliessend zwei Stunden lang bei -35 gerührt, dann mit 80 ml absolutem Me thanol versetzt. Danach wird das Ammoniak ver dampft, der Rückstand mit Wasser verdünnt und das Gemisch einige Male mit Äther extrahiert. Die gesammelten Ätherextrakte werden mit Wasser ge waschen, über Natriumsulfat getrocknet und zur Trockne eingedampft.
Der Rückstand wird aus Methanol und Petrol- äther umkristallisiert, wobei man das 44-20-Hydroxy- 19-nor-pregnen erhält.
<I>Beispiel 5</I> 2,75g NaBH4 werden zu einer Lösung von 5,5 g d4-17-Hydroxy-3-keto-19-nor-androsten in 200 ml Methanol bei Raumtemperatur gegeben. Nach 20- minutigem Stehen wird die Reaktionsmischung mit Eisessig neutralisiert und dann im Vakuum auf 25 ml eingeengt. Der Rückstand wird in 200 ml Wasser gegossen, wonach das 44-3,17-Dihydroxy-19-nor- androsten auskristallisiert. Die abgenutschten Kristalle werden in 15 ml Pyridin gelöst, worauf 10,2 g Essigsäureanhydrid dieser Lösung zugegeben werden.
Das Reaktions gemisch wird 20 Minuten bei Raumtemperatur ge halten und dann mit 20 ml Wasser versetzt.
Das Gemisch wird zur Trockne eingedampft und dann der Rückstand aus Methanol umkristallisiert; man erhält das 44-3,17-Diacetoxy-19-nor-androsten.
2 g der so hergestellten Verbindung werden in 25 ml trockenem Äther gelöst, worauf dieser Lösung tropfenweise eine Lösung von 2 g Lithium in 90 g Athylamin zugegeben wird. Nach 10minutigem Rühren wird der überschuss von Lithium durch lang sames Zugeben von 15 ml Methanol zu dem Reak tionsgemisch entfernt. Nachdem das Reaktions gemisch entfärbt ist, wird das Äthylamin im Vakuum abgedampft. Der Rückstand wird in Eiswasser auf genommen, worauf die Mischung mehrmals mit Äther extrahiert wird. Die gesammelten Äther extrakte werden gewaschen, worauf der Äther im Vakuum entfernt wird.
Der Rückstand wird zwischen 70% Methanol und Petroläther verteilt, die Petrol- ätherschicht abgetrennt, getrocknet und schliesslich zur Trockne eingedampft.
Beim Auskristallisieren aus Petroläther erhält man aus dem Rückstand das J4-17ss-Acetoxy-19-nor-androsten. Die Verseifung dieser Verbindung durch Behandlung mit einer ver dünnten methanolischen Natriumhydroxydlösung er gibt das 44-17ss-Hydroxy-19-nor-androsten vom Schmelzpunkt 98 C.
<I>Beispiel 6</I> Zu einer Lösung von 5,8 g J4-3-Keto-llss-di- hydroxy-19-nor-androsten in 200 ml Methanol wer den 2,79g Natriumborhydrid bei Raumtemperatur zugegeben. Nach 20minutigem Stehen wird mit Eis essig neutralisiert und im Vakuum auf 25 ml ein geengt.
Der Rückstand wird in 200 ml Wasser gegossen, worauf das A4 - 3,1 lss,17ss - Trihydroxy - 19 - nor- androsten auskristallisiert.
Die abgesaugten Kristalle werden in 25 ml Pyri- din gelöst, worauf 30,3g Cyclopentylpropionsäure- anhydrid der Lösung zugegeben werden. Das Reak tionsgemisch wird 30 Minuten zum Rückfluss erhitzt, worauf 25 ml Wasser zugegeben werden. Die Mi schung wird anschliessend zur Trockne eingedampft und dann der Rückstand aus Methanol-Petroläther umkristallisiert. Man erhält das 44-3,llss"17ss-Tri- hydroxy-19 -nor- androsten-3,17ss-dicyclopentyl-pro- pionat.
2 g der so hergestellten Verbindung werden in 25 ml trockenem Äther gelöst, worauf diese Lösung tropfenweise einer Lösung von 2 g Lithium in 90 g Äthylamin zugefügt wird. Nach 10minutigem Rühren wird der überschuss an Lithium durch langsames Zugeben von 15 ml Methanol entfernt. Nachdem die Reaktionsmischung entfärbt ist, wird das Äthylamin im Vakuum entfernt. Der Rückstand wird in Eis wasser aufgenommen, worauf die Mischung mehr mals mit Äther extrahiert wird. Die gesammelten Ätherextrakte werden gewaschen, worauf man den Äther im Vakuum abdampft.
Der Rückstand wird zwischen 70% Methanol und Petroläther verteilt, die Petrolätherschicht abgetrennt, getrocknet und schliesslich zur Trockne eingedampft.
Beim Aus- kristallisieren aus Petroläther erhält man aus dem Rückstand das d4 -11fl,17ss - Dihydroxy -19 - nor- androsten-17ss-cyclopentylpropionat. Die Verseifung dieser Verbindung durch Behandlung mit einer ver dünnten methanolischen Natriumhydroxylösung führt zum 44-llss,17ss-Dihydroxy-19-nor-androsten.
<I>Beispiel</I> Entsprechend der in Beispiel 1 beschriebenen Weise wird das 44-17ss-Hydroxy-17a-methyl-3-keto- 19-nor-androsten zu der entsprechenden 3-Hydroxy- Verbindung mit Natriumborhydrid reduziert.
4,9 g dieser Verbindung werden dann in 105 ml Athanol gelöst, worauf 2,8 ml konzentrierte Salz säure zugefügt werden. Die Behandlung dieses Reak tionsgemisches erfolgt wie in Beispiel 1 beschrieben, wonach man das 44-3-Athoxy-17ss-hydroxy-17a- methyl-19-nor-androsten erhält.
3 g dieser Verbindung werden in 125 ml abso lutem Äther gelöst. Zu dieser Lösung werden 120 ml flüssiges Ammoniak und anschliessend 0,9 g in kleine Stücke geschnittenes Lithium gegeben. Die blaue Lösung wird dann 2 Stunden lang bei -35 C ge rührt, wonach 20 ml absolutes Äthanol bei der glei chen Temperatur zugegeben werden, so dass die blaue Farbe verschwindet.
Das Ammoniak wird ver dampft, der Rückstand mit Wasser verdünnt und das resultierende Gemisch mehrmals mit Äther extrahiert. Die gesammelten Ätherextrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet und zur Trockne eingedampft.
Der Rückstand wird zwischen Petroläther und 7011/o Methanol (1 : 1) verteilt, die Petrolätherschicht abgetrennt, der Petroläther zur Hälfte seines Volumens abgedampft, worauf die Kri- stallisation des d4-17 ,6-Hydroxy-17a-methyl-19-nor- androstens stattfindet. Diese Verbindung hat einen Schmelzpunkt von 147-151 C.
Genau nach dem in diesem Beispiel beschriebe nen Verfahren wird das 44-17ss-Hydroxy-17a-äthyl- 3-keto-19-nor-androsten zu dem d4-17ss-Hydroxy- 17a-äthyl-19-nor-androsten über die 3-Methoxy- Verbindung reduziert.

The new compounds have a very powerful inhibiting effect against sex hormones (gonad-inhibiting effects) and show anabolic, androgenic and progressive properties. The starting materials can be obtained as follows: 44-3-Hydroxy-19-nor-steroids can be obtained by reducing the corresponding 3-keto compounds.
This reduction can be carried out with the help of one of the usual reducing agents, such as an alkali borohydride, an aluminum isopropoxide in isopropanol.
The etherification can be carried out by treating the 44-3-hydroxy-19-nor-steroid in an organic solvent with an alcohol in the presence of an inorganic acid such as hydrochloric acid. In some cases, the alcohol can also serve as a solvent at the same time.

The resulting solution is stirred for one hour at room temperature, whereupon the reaction mixture is neutralized with sodium bicarbonate and concentrated to 25 ml. The residue is poured into 200 ml of water, then the resulting crystals of d4-3-methoxy -11,8,17ss-dihydroxy -17a-methyl-19-nor androstene are suctioned off and then recrystallized from acetone.
2.3 g of this compound are dissolved in 25 ml of dry ether, whereupon a solution of 2 g of lithium in 90 g of ethylamine is added dropwise to this solution. After stirring for 10 minutes, the excess lithium is removed by slowly adding 15 ml of methanol to the reaction mixture. The residue is distributed between 70% methanol and petroleum ether, the petroleum ether layer is separated off, dried and finally evaporated to dryness. After recrystallization from petroleum ether, the residue represents 44-1 lss, 17ss-dihydroxy-17a-methyl-19-nor-androstene.
<I> Example 2 </I> In a manner analogous to that described in Example 1, the 44-3-keto-Ilss, 17ss-dihydroxy-17a-vinyl-19-nor-androstene is reduced to the corresponding 3-hydroxy compound which is then converted into the corresponding 3-isopropoxy compound with isopropanol and hydrochloric acid.
Following the procedure of Example 1, this compound is treated with a solution of sodium in methylamine, whereby the J4-llss, 17ss-dihydroxy-17a-vinyl-19-nor-androstene is obtained. <I> Example 3 </I> 2.5 g of d4-3,20,21-tripropionyloxy-19-nor-pregnen are dissolved in 120 ml of absolute ether. <I> Example 4 </I> 2.1 g of d4-3-methoxy-20-hydroxy-19-nor-pregnen is dissolved in 120 ml of absolute ether, whereupon 120 ml of liquid ammonia and then 0.9 g into small pieces cut sodium can be added. The blue solution is then stirred for two hours at -35, then treated with 80 ml of absolute methanol. The ammonia is then evaporated, the residue is diluted with water and the mixture is extracted a few times with ether. The residue is recrystallized from methanol and petroleum ether, giving 44-20-hydroxy-19-nor-pregnen.
<I> Example 5 </I> 2.75 g of NaBH4 are added to a solution of 5.5 g of d4-17-hydroxy-3-keto-19-nor-androstene in 200 ml of methanol at room temperature. The reaction mixture is kept at room temperature for 20 minutes and then treated with 20 ml of water.
The mixture is evaporated to dryness and then the residue is recrystallized from methanol; 44-3,17-diacetoxy-19-nor-androstene is obtained.
2 g of the compound thus prepared are dissolved in 25 ml of dry ether, whereupon a solution of 2 g of lithium in 90 g of ethylamine is added dropwise to this solution. After stirring for 10 minutes, the excess lithium is removed by slowly adding 15 ml of methanol to the reaction mixture. After the reaction mixture is decolorized, the ethylamine is evaporated in vacuo. The residue is taken up in ice water, whereupon the mixture is extracted several times with ether. The collected ether extracts are washed, after which the ether is removed in vacuo.
The residue is partitioned between 70% methanol and petroleum ether, the petroleum ether layer is separated off, dried and finally evaporated to dryness.
When crystallizing out from petroleum ether, the J4-17ss-acetoxy-19-nor-androsten is obtained from the residue. The saponification of this compound by treatment with a dilute methanolic sodium hydroxide solution gives 44-17ss-hydroxy-19-nor-androsten with a melting point of 98 C.
<I> Example 6 </I> 2.79 g of sodium borohydride are added at room temperature to a solution of 5.8 g of J4-3-keto-IIIss-dihydroxy-19-nor-androstene in 200 ml of methanol. After standing for 20 minutes, it is neutralized with ice cream and concentrated in vacuo to 25 ml.
The residue is poured into 200 ml of water, whereupon the A4 - 3.1 lss, 17ss - trihydroxy - 19 - norandrosten crystallizes out.
The extracted crystals are dissolved in 25 ml of pyridine, whereupon 30.3 g of cyclopentylpropionic anhydride are added to the solution. The reaction mixture is refluxed for 30 minutes, after which 25 ml of water are added. The mixture is then evaporated to dryness and then the residue is recrystallized from methanol-petroleum ether. The 44-3, 11ss "17ss-trihydroxy-19-norandrostene-3,17ss-dicyclopentyl propionate is obtained.
2 g of the compound thus prepared are dissolved in 25 ml of dry ether, whereupon this solution is added dropwise to a solution of 2 g of lithium in 90 g of ethylamine. After stirring for 10 minutes, the excess lithium is removed by slowly adding 15 ml of methanol. After the reaction mixture is decolorized, the ethylamine is removed in vacuo. The residue is taken up in ice water, whereupon the mixture is extracted several times with ether. The collected ether extracts are washed, whereupon the ether is evaporated in vacuo.
The residue is distributed between 70% methanol and petroleum ether, the petroleum ether layer is separated off, dried and finally evaporated to dryness.
Crystallization from petroleum ether gives d4 -11fl, 17ss-dihydroxy-19-nor- androsten-17ss-cyclopentylpropionate from the residue. The saponification of this compound by treatment with a dilute methanolic sodium hydroxide solution leads to 44-llss, 17ss-dihydroxy-19-nor-androsten.
<I> Example </I> Corresponding to the manner described in Example 1, the 44-17ss-hydroxy-17a-methyl-3-keto-19-nor-androstene is reduced to the corresponding 3-hydroxy compound with sodium borohydride.
4.9 g of this compound are then dissolved in 105 ml of ethanol, whereupon 2.8 ml of concentrated hydrochloric acid are added. This reaction mixture is treated as described in Example 1, after which 44-3-ethoxy-17ss-hydroxy-17a-methyl-19-nor-androstene is obtained.
3 g of this compound are dissolved in 125 ml of absolute ether. 120 ml of liquid ammonia and then 0.9 g of lithium cut into small pieces are added to this solution. The blue solution is then stirred for 2 hours at -35 C, after which 20 ml of absolute ethanol are added at the same temperature so that the blue color disappears.
The ammonia is evaporated, the residue diluted with water and the resulting mixture extracted several times with ether. The collected ether extracts are washed with water, dried over sodium sulfate and evaporated to dryness.
The residue is partitioned between petroleum ether and 7011 / o methanol (1: 1), the petroleum ether layer is separated off, the petroleum ether is evaporated to half its volume, whereupon the d4-17,6-hydroxy-17a-methyl-19-nor is crystallized - androstens takes place. This compound has a melting point of 147-151 C.
Exactly according to the method described in this example, the 44-17ss-hydroxy-17a-ethyl-3-keto-19-nor-androstene becomes the d4-17ss-hydroxy-17a-ethyl-19-nor-androstene via the 3 -Methoxy- compound reduced.
