Verfahren zur Herstellung von Steroiden Vorliegende Erfindung bezieht sich auf ein Ver fahren zur Herstellung von 6-Alkyl(oder 6-Aryl)- 11ss - oxy(oder 11- oxo) - 17a - oxy-4-pregnen-3,20- dionen, das durch folgende Formeln erläutert wird:

B. Methyl, Äthyl, Phenyl, Tolyl, Naphthyl, vorzugsweise aber Methyl, bedeutet.
Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass man ein 6-Alkyl(oder 6-Aryl)- llss-oxy(oder 11-oxo)-17a-oxy-21-jod-4-pregnen- 3,20-dion mit einem enthalogenierenden Mittel be handelt. Die Reaktion kann z. B. mit einem Reduk tionsmittel, wie Zinkstaub, Natriumthiosulfat, Na trium- oder Kaliumbisulfit und dergleichen, in wässrig organischem Lösungsmittelgemisch durch geführt werden.
Anschliessend kann ein so erhaltenes 6-Alkyl(oder 6-Aryl)-llss,17a-dioxy-4-pregnen-3,20- dion gewünschtenfalls mit einem Oxydationsmittel, wie Chromsäureanhydrid, N-Bromacetamid, N-Brom- succinimid oder dergleichen, zum 6-Methyl-17a-oxy- 4-pregnen-3,11,20-trion oxydiert werden.
Insbesondere bevorzugt man die Herstellung der hochaktiven 6a-Epimeren der oben genannten Ver bindungen. Die erfindungsgemäss erhältlichen Verbindungen 6 - Methyl -11,8,17a - dioxy-4-pregnen-3,20-dion und 6-Methyl-17a-oxy-4-pregnen-3,11,20-trion besitzen, speziell in Form ihrer 6a-Epimeren, eine starke physiologische Wirksamkeit, deren Wirkungsbereich von den in der Natur zu findenden Adrenocortical- hormonen, wie Hydrocortison und Cortison, ab weicht.
Dies trifft speziell für den Mineral- und Wasserstoffwechsel zu. Die Verbindungen bewirken eine Abgabe von Salz und Wasser, was sie besonders für die Behandlung chronisch kongestiver Herz schwächen und der Lebercyrrhose, des nephrotischen Syndroms und der Eclampsia und Praeeclampsia ge eignet macht.
Die neuen synthetischen Corticosteroid- hormone besitzen auch entzündungshemmende, gluco- corticoide, anästhetische, wachstumshemmende sowie corticoide Wirkung. Die entzündungshemmende Wir kung ist bei allen erfindungsgemäss hergestellten Steroiden bemerkenswert. Ferner wirken sie auf Ge bärmutter, Eierstock und Nebennieren.
Die Steroide können zu Präparaten für orale, parenterale und äusserliche Applikation verarbeitet werden. Man kann sie auch mit allen bekannten Antibiotika kombinieren, wobei eine gegenseitige Ergänzung und Potenzierung der Eigenschaften er zielt wird.
Da sie einen starken Salzverlust bewirken, kann man sie auch mit andern Steroiden, wie Cortison und Hydrocortison, kombinieren, um die Salzreten- tion der letzteren abzuschwächen.
Die 6 - Alkyl (oder 6 - Aryl) -11,8,17a - dioxy - 4- pregnen-3,20-dione sowie deren 11-Keto-analoga sind auch wertvolle Ausgangsmaterialien zur Herstel lung anderer physiologisch wichtiger Verbindungen. Diese daraus herstellbaren 9a-Halogenderivate haben beispielsweise glucocorticoide und entzündungs hemmende Wirkung und können anstelle der halogen freien Verbindungen in pharmazeutischen Präparaten verwendet werden.
Die als Ausgangsverbindungen für das vorlie gende Verfahren verwendeten 6-Alkyl(oder 6-Aryl)- llss-oxy(oder 11-oxo)-17-oxy-4-pregnen-21-jod-3,20- dione können aus den gemäss Schweiz. Patentschrift Nr. 353737 erhältlichen Verbindungen, wie in den Präparationen 1 bis 5 erläutert, hergestellt werden.
Bei der Durchführung des Verfahrens kann das rohe oder gereinigte 6-Alkyl(oder 6-Aryl)-11/3,17a- dioxy-21-jod-4-pregnen-3,20-dion mit einem Reduk tionsmittel, wie Zinkstaub in Essigsäure, Natrium- oder Kaliumthiosulfat oder Natriumbisulfit, umgesetzt werden. Bei einer bevorzugten Ausführungsform wird die 21-Jodverbindung mit Zinkstaub in Essigsäure aufgeschlämmt und gerührt.
Der Zinkstaub ist ge wöhnlich in einem überschuss von 5 bis 10 Mol- äquivalenten pro Moläquivalent Steroid vorhanden. Die Reaktionszeit ist nicht kritisch und liegt gewöhn lich zwischen 5 Minuten und 1 bis 2 Stunden.
Nach Beendigung der Reaktion wird überschüssiges Zink durch Filtration entfernt, das Filtrat neutralisiert und das 6-Alkyl(oder 6-Aryl)-llss,17a-dioxy-4-pregnen- 3,20-dion nach üblichen Methoden, wie Umkristalli- sieren oder Extraktion, isoliert und falls erforderlich durch Umkristallisieren oder Chromatographie weiter gereinigt.
Wenn man für die Enthalogenierung Na- triumbisulfit oder Natriumthiosulfat verwendet, so arbeitet man in der Regel bei Zimmertemperatur unter ständigem Rühren in wässriger Essigsäure lösung.
Die fakultative Oxydation des 6-Alkyl(oder 6- Aryl)-llss,17a-dioxy-4-pregnen-3,20-dions erfolgt im allgemeinen in üblicher Weise, z. B. mit der berech neten Menge oder einem kleinen überschuss von 10 bis 309/o Chromsäureanhydrid in Essigsäure, oder mit einem Halogenamid oder -imid, z.
B. N-Brom- acetamid, N-Chlorsuccinimid oder N-Bromsuccin- imid in Pyridin, Dioxan oder andern Lösungsmitteln. Nach Beendigung der Oxydation wird das Oxyda tionsmittel zerstört, sei es durch Zugabe von Methyl- oder Äthylalkohol oder von Bisulfit. Dann wird das 6 - Alkyl(oder 6 - Aryl) -17a -oxy-4-pregnen-3,11,20- trion, falls dies erforderlich scheint,
in üblicher Weise, z. B. durch Extraktion mit in Wasser nicht löslichen Lösungsmitteln, wie Methylenchlorid, Äthy- lenchlorid, Chloroform, Tetrachlorkohlenstoff, Äther, Benzol, Toluol oder chromatographisch gereinigt.
Präparation <I>1</I> Eine Lösung von 1 g (2,65 Millimol) 6a-Methyl- hydrocortison (siehe Schweiz. Patentschrift Nummer 353737) in 7 ml Pyridin wird auf 0 gekühlt und mit 0,3 ml Methansulfonylchlorid versetzt, wonach man bei 0 bis 5 2 Stunden stehenlässt.
Man verdünnt mit Wasser und extrahiert dreimal mit 25 ml Methylen- chlorid. Die Extrakte werden vereinigt, mit kalter verdünnter Salzsäure (bis zum pH 2 bis 3 der wäss- rigen Phase) gewaschen, dann wiederum mit kalter Natriumbicarbonatlösung und Wasser und schliess lich mit wasserfreiem Natriumsulfat getrocknet.
Nach Eindampfen des Methylenchloridextraktes bei ver mindertem Druck erhält man das 6a - Methyl- 1 lss,17a,21-trioxy - 4 - pregnen-3,20-dion-21-methan- sulfonat (6 - Methylhydrocortison-21-methansulfonat) als weisses glasartiges Produkt.
Präparation <I>2</I> Das rohe Methansulfonat nach Präparation 1 wird in 15 ml Aceton gelöst und mit einer Lösung von 1 g Natriumjodid in 10 ml Aceton versetzt. Man erhitzt unter Rühren 15 Minuten am Rückfluss, drosselt die Wärmezufuhr und konzentriert unter vermindertem Druck zur Trockne.
Man erhält 6a- Methyl-1 lss,17a-dioxy-21-jod-4-pregnen-3,20-dion. Präparation <I>3</I> In gleicher Weise wie in Präparation 1 wird 6a Äthyl-hydrocortison in Pyridin gelöst und mit Toluol- sulfonylchlorid in das 6a-Äthyl-llss,17a,21-trioxy-4- pregnen-3,20-dion-21-toluolsulfonat (6a-Äthyl-hydro- cortison-21-toluolsulfonat) übergeführt.
Präparation <I>4</I> In der gleichen Weise wie in Präparation 2 er hält man beim Erhitzen von 6a-Äthyl-llss,17a,21- trioxy-4-pregnen-3,20-dion-21-toluolsulfonat mit Na- triumjodid in Aceton das 6a-Äthyl-llss,17a-dioxy- 21-jod-4-pregnen-3,20-dion.
Präparation <I>5</I> In gleicher Weise wie in Präparation 1 erhält man durch Behandlung von 6a-Methylcortison mit Me- thansulfonylchlorid in Pyridinlösung 6a-Methyl- 17a,21 - dioxy - 4 - pregnen-3,11,20-trion-21-methan- sulfonat, das beim Kochen am Rückfluss mit Kalium jodid in Aceton gemäss Präparation 2 das 6a-Me- thyl-17a-oxy-21-jod-4-pregnen-3,11,
20-trion liefert. <I>Beispiel 1</I> Das rohe 6a-Methyl-llss,17a-dioxy-21-jod-4- pregnen-3,20-dion, hergestellt nach Präparation 2, wird in 15 ml Essigsäure suspendiert und 45 Minuten gerührt. Dann setzt man 1 g Zinkstaub zu und rührt weitere 15 Minuten. Vom überschüssigen Zinkstaub wird abfiltriert, das Filtrat mit Methylenchlorid ver dünnt und mit Wasser und kalter Natriumbicarbonat- lösung gewaschen, bis alle Essigsäure neutralisiert ist.
Nach dem Trocknen über Natriumsulfat wird die Lösung an 75 g Florisil chromatographiert, wobei man Fraktionen zu 200 ml wie folgt abtrennt: 14 Fraktionen Aceton Skellysolve B mit 10% Aceton- gehalt; eine Fraktion mit 100'% Aceton.
Die Fraktionen 8 bis 12 werden vereinigt und eingedampft. Man erhält 760 mg Kristalle, die nach dem Umkristallisieren aus Aceton-Skellysolve Bin zwei Fraktionen 650 mg 6a-Methyl-11ss,17a-dioxy- 4-pregnen-3,20-dion vom Smp. 203 bis 210 liefern.
Analyse: berechnet für C22H3204: C: 73,30 H: 8,95 gefunden: C: 73,19 H: 8,59 Zur Umwandlung der 11-Oxy- in die 11-Oxo- Gruppe kann man wie folgt verfahren: Man stellt eine Mischung aus 0,5 g 6a-Methyl- llss,17a-dioxy-4-pregnen-3,20-dion, <B><I>0,15</I></B> g Chrom säure, 10 ml Eisessig und 0,5 ml Wasser her, die gerührt und dann 8 Stunden bei Zimmertemperatur gehalten wird. Dann giesst man in 50 ml Eiswasser, neutralisiert mit verdünnter Natronlauge und sammelt den Niederschlag auf dem Filter.
Nach dreimaligem Umkristallisieren aus Äthylacetat und Skellysolve B erhält man 6a-Methyl-17a-oxy-4-pregnen-3,11,20- trion.
<I>Beispiel 2</I> Das gemäss Präparation 4 erhaltene 6a-Äthyl- 11ss,17a-dioxy-21-jod-4-pregnen-3,20-dion wird in 50%iger wässriger Essigsäure gelöst. Dazu gibt man überschüssiges Natriumbisulfit und rührt eine Stunde bei Zimmertemperatur. Dann giesst man in Wasser und extrahiert mit Methylenchlorid. Die Extrakte werden vereinigt, mit Wasser,
Bicarbonat und wieder mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Man erhält das rohe 6a-Äthyl- llss,17a-dioxy-4-pregnen-3,20-dion, das durch Um kristallisieren aus Aceton-Skellysolve B gereinigt wird.
Zur Umwandlung der 11-Oxy- in die 11-Oxo- Gruppe kann man wie folgt verfahren: Zu 200 mg 6a-Äthyl-llss,17a-dioxy-4-pregnen-4 3,20-dion in 8 ml Methanol gibt man 0,2 ml Pyridin, 0,4 ml Wasser und 150 mg N-Bromacetamid. Man hält 20 Stunden bei Zimmertemperatur und setzt dann 25 ml verdünnte Natriumsulfitlösung zu, um überschüssiges N-Bromacetamid zu zerstören.
Dann wird unter vermindertem Druck konzentriert, bis kräftige Kristallisation einsetzt. Man kühlt auf 0 und sammelt nach einer Stunde die Kristalle auf dem Filter. Nach Umkristallisieren aus Skellysolve B erhält man reines 6a-Äthyl-17a-oxy-4-pregnen- 3,11,20-trion.
<I>Beispiel 3</I> Behandelt man das gemäss Präparation 5 erhal tene Produkt nach dem Verfahren des Beispiels 1 mit Zink in Essigsäure, so wird 6a-Methyl-17a-oxy- 4-pregnen-3,11,20-trion erhalten.

A 6-alkyl (or 6-aryl) -llss, 17a-dioxy-4-pregnen-3,20-dione obtained in this way can then, if desired, be treated with an oxidizing agent such as chromic anhydride, N-bromoacetamide, N-bromosuccinimide or the like, be oxidized to 6-methyl-17a-oxy-4-pregnen-3,11,20-trione.

It is then concentrated under reduced pressure until vigorous crystallization begins. It is cooled to 0 and after one hour the crystals are collected on the filter. After recrystallization from Skellysolve B, pure 6a-ethyl-17a-oxy-4-pregnen-3,11,20-trione is obtained.
<I> Example 3 </I> If the product obtained according to preparation 5 is treated by the method of Example 1 with zinc in acetic acid, 6a-methyl-17a-oxy-4-pregnen-3,11,20-trione is obtained receive.
