Verfahren zur Herstellung von 5-Fluor-uracil Gegenstand der Erfindung ist ein Verfahren zur Herstellung von 5-Fluor-uracil, das dadurch gekenn zeichnet ist, dass man ein Alkalisalz der Fluoressig- säure mit einem Alkylierungsmittel umsetzt, den er haltenen Fluoressigsäureester in Gegenwart eines Alkalimetalls oder einer Alkalimetallverbindung als Kondensationsmittel mit einem Ameisensäureester kondensiert,
das erhaltene Salz des Hydroxymethy- len-fluoressigsäureesters mit einem Äther von Isothio- harnstoff oder von Pseudoharnstoff kondensiert und das erhaltene, in 2-Stellung die entsprechende Äther gruppe tragende 5-Fluor-uracil hydrolysiert.
In der ersten Stufe des erfindungsgemässen Ver fahrens wird ein Alkalisalz, vorzugsweise das Na trium- oder Kaliumsalz der Fluoressigsäure, mit einem Alkylierungsmittel umgesetzt. Es ist zweck mässig, als Alkylierungsmittel einen Alkylester einer anorganischen Mineralsäure, wie z. B. Diäthylsulfat, Methylbromid oder Äthyljodid, zu verwenden. Die Reaktion kann sowohl mit als auch ohne Lösungs mittel durchgeführt werden.
Vorzugsweise wird das erhaltene Salz der Fluoressigsäure mit Diäthylsulfat erhitzt und der entstandene Fluoressigsäureäthylester zwecks Reinigung destilliert.
Zweckmässig werden bei der die zweite Stufe des erfindungsgemässen Verfahrens darstellenden Kon densation die Alkylester der Fluoressigsäure einer seits und der Ameisensäure anderseits mit gleichen Alkylgruppen verwendet. Als Kondensationsmittel können z. B. Kalium oder Natrium oder Alkali metallalkoholate, wie z. B. Kaliumäthanolat oder Natriummethanolat, verwendet werden. Der erhaltene Fluoressigsäureester wird zweckmässig zusammen mit dem Ameisensäureester und dem Kondensationsmittel in einem wasserfreien inerten Lösungsmittel, wie z.
B. Diäthyläther oder Toluol, erhitzt. Die erhaltenen Verbindungen können ohne weitere Reinigung für die nächste Verfahrensstufe verwendet werden.
In der dritten Stufe des erfindungsgemässen Ver fahrens wird das erhaltene Alkahmetallsalz des ent sprechenden Hydroxymethylen-fluoressigsäureesters, wie gesagt, mit einem Äther von Isothiohamstoff oder von Pseudohamstoff cyclisiert. Bevorzugte Äther sind die Alkyläther, wie z.
B. der Methyl- oder Äthyläther, und der Benzyläther des Isothiohamstoffes respektive des Pseudoharnstoffes. Die Reaktionwird unterwasser freien Bedingungen zweckmässig in Gegenwart eines wasserfreien inerten Lösungsmittels, wie z. B. Methanol oder Äthanol, durchgeführt. Es wird empfohlen, das erhaltene Salz des Hydroxymethylen-fluoressigsäure- esters in frisch bereitetem Zustande, vorzugsweise innerhalb von zwei bis drei Tagen nach Herstellung, zu verwenden.
Zweckmässig wird das Salz des Hy- droxymethylen-fluoressigsäureesters zusammen mit einem Säureadditionssalz des Äthers von Isothio- harnstoff resp. Pseudoharnstoff gleichzeitig mit der zur Neutralisierung der Mineralsäure berechneten Menge Alkali erhitzt.
Die erhaltenen entsprechenden 2-Äther oder 2-Thioäther des 5-Fluor-uracils kön nen in üblicher Weise durch Umkristallisieren aus inerten Lösungsmitteln oder durch Sublimation im Vakuum gereinigt werden.
Die Hydrolyse des erhaltenen 2-Thioäthers von 2-Thio-S-fluor-uracil erfolgt z. B. durch Erhitzen mit einer wässerigen Lösung einer Mineralsäure, wie z. B. konzentrierte Chlorwasserstoffsäure. Das anfallende 5-Fluor-uracil kann z. B. durch Umkristallisieren aus inerten Lösungsmitteln oder durch Sublimation im Vakuum, gereinigt werden.
Der erhaltene Äther des 2-Oxy-5-fluor-uracils wird ebenfalls z. B. durch Erhitzen mit einer wässe rigen Lösung einer Mineralsäure, z. B. konzentrier ter wässeriger Chlorwasserstoffsäure, hydrölysiert. Zur Reinigung kann das gebildete 5-Fluor-uracil aus einem inerten Lösungsmittel umkristallisiert oder im Vakuum sublimiert werden.
Das nach dem erfindungsgemässen Verfahren er hältliche 5-Fluor-uracil zeigt saure Eigenschaften und lässt sich mit Basen in die entsprechenden Salze über führen. Es ist vorteilhaft, solche Basen zu verwen den, welche zu nicht toxischen Salzen führen, z. B. Alkalimetallhydroxyde, Erdalkalimetallhydroxyde, Ammoniak oder nicht toxische organische Basen, wie z. B. Äthanolamin.
Das erfindungsgemäss erhaltene 5-Fluor-uracil be sitzt wertvolle pharmakologische Eigenschaften, ins besondere keimtötende, antimetabolische und ge- schwürhemmende Wirkungen.
<I>Beispiel 1</I> Eine Mischung von 200 g trockenem Natrium salz der Fluoressigsäure und 442 g Diäthylsulfat wer den 31/2 Stunden unter Rückfluss gekocht. Nach Destillation der Reaktionsmischung erhält man 177,3 g rohen Äthylester der Fluoressigsäure mit einem Siedepunkt bei 116-120 . Nach nochmaliger Destillation durch eine Fraktionierkolonne erhält man den zwischen 114 und 118 siedenden Äthyl- ester der Fluoressigsäure.
In einen mit Rührer, Tropftrichter und Rückfluss- kühler versehenen Kolben gibt man 880 ml absoluten Diäthyläther und 47,6 g in 5 mm Stücke geschnit tenes Kalium. Unter Rühren werden 220 ml abso lutes Äthanol zugetropft und bis zur vollständigen Auflösung des Kaliums am Rückfluss erhitzt. In die eisgekühlte Lösung trägt man innerhalb von 21/2 Stunden unter Rühren und Kühlen 135 g des erhal tenen Fluoressigsäureäthylesters und 96,4 g von frisch destilliertem Ameisensäureäthylester ein.
Wenn alles eingetragen ist, wird unter Kühlen noch eine Stunde gerührt und dann das Reaktionsgemisch über Nacht stehengelassen. Der gebildete kristalline Rückstand wird abgenutscht, mit Diäthyläther ge waschen und im Vakuum getrocknet. Man erhält das Kaliumsalz des Hydroxymethylen-fluoressigsäure- äthylesters.
Eine Mischung von 103,6 g frisch hergestelltem h aliumsalz des Hydroxymethylen - fluoressigsäure- äthylesters, 83,4 g des Sulfats von S-Methyl-isothio- harnstoff und 32,5g Natriummethanolat wird unter Rühren in 1500 ml absolutem Methanol unter Rück fluss erhitzt. Nach vorübergehender Lösung wird eine Fällung beobachtet. Nach zweistündigem Erhitzen unter Rückfluss wird das Reaktionsgemisch im Va kuum zur Trockne eingedampft und der Rückstand mit 280 ml Wasser behandelt.
Das entstandene Ge misch wird durch Kohle filtriert und das entfärbte Filtrat durch Zusatz von 37o/oiger Chlorwasserstoff säure schwach kongosauer gestellt. Die gebildeten Kristalle werden abfiltriert, mit Wasser sulfatfrei ge waschen und bei 100 getrocknet. Man erhält den rohen S-Methyl-thioäther von 2-Thio-5-fluor-uracil vom Schmelzbereich 202-221 .
Nach dem Lösen in 2035 ml siedendem Äthylacetat und Abkühlen auf -20 schmilzt das Produkt bei 230-237 . Nach weiterem Umkristallisieren aus Wasser steigt der Schmelzpunkt auf 241-243 .
Eine Lösung von 10 g des so erhaltenen S-Methyl- thioäthers von 2-Thio-S-fluor-uracil vom Schmelz punkt 230-237 in 150 ml 37o/oiger wässeriger Chlorwasserstoffsäure wird unter Stickstoff vier Stunden unter Rückfluss erhitzt. Das Reaktions gemisch wird im Vakuum abgedampft und der bräun liche kristalline Rückstand aus Wasser umkristalli siert. Das Produkt wird durch Sublimation im Va kuum bei etwa 190 /0,1 mm weitergereinigt. Man erhält farbloses oder rötlich kristallines 5-Fluor-uracil vorn Schmelzpunkt 282-283 (unter Zersetzung).
<I>Beispiel 2</I> In einen mit Rührer, Rückflusskühler, Tropftrich ter, Thermometer und Zuführrohr, durch welches ein ständiger Stickstoffstrom zugeführt wird, versehenen Kolben gibt man 880 ml Toluol und 47,6 g Kalium. Die Mischung wird auf etwa 80 erhitzt, bis das Kalium schmilzt. Durch Rühren wird das Metall fein verteilt. 190 ml Äthanol werden so zugetropft, dass die Mischung leicht siedet. Wenn alles eingetra gen ist, wird weitererhitzt, bis alles Kalium in Lö sung gegangen ist.
Etwa 250 ml des Toluol-Äthanol- Gemisches werden abgedampft und die Lösung in einem Eisbad gekühlt, wobei ein Teil des gebildeten Kaliumäthanolats ausfällt. Nach Zugabe von 146 g über Calciumcarbonat destilliertem Ameisensäure methylester erhält man eine farblose Lösung. Unter Kühlen und Rühren gibt man innerhalb von 2 Stun den 135 g des nach Beispiel 1 erhaltenen Fluoressig- säureäthylesters zu. Die Reaktionsmischung wird eine weitere Stunde in einem Eisbad gerührt und dann bei Zimmertemperatur über Nacht stehengelassen.
Die beinahe farblosen Kristalle werden abfiltriert, mit Äther gewaschen und im Vakuum bei Zimmer temperatur getrocknet. Man erhält das Kaliumsalz des Hydroxymethyl,n-fluoressigsäureäthylesters.
Zu einer Lösung von 6,7g Natrium in 720 ml absolutem Äthanol gibt man in einen mit Rührer und Rückflusskühler versehenen Kolben 50 g des Kaliumsalzes des Hydroxymethylen-fluoressigsäure- äthylesters und 53,6 g des Bromids von S-Äthyl-iso- thioharnstoff. Nach vorübergehender Lösung findet Kristallisation statt. Die Mischung wird noch zwei Stunden unter Rühren und Rückfluss auf einem Wasserbad erhitzt.
Der Alkohol wird im Vakuum abgedampft und der Rückstand in 140 ml Wasser gelöst. Die Lösung wird durch Kohle filtriert, in einem Eisbad gekühlt und mit 22 ml konzentrierter Chlorwasserstoffsäure kongosauer gestellt. Die kristal line Fällung wird abfiltriert, mit Wasser chlorfrei ge waschen und bei 100^ getrocknet. Man erhält den 2-Äthvl-thioäther des 2-Thio-5-fluor-uracils vom Schmelzpunkt 175-179 .
Nach Umkristallisieren aus heissem Essigsäureäthvlester erhält man Kristalle vom Schmelzpunkt 189-190 . Durch Verseifung nach den Angaben von Beispiel 1 erhält man das 5-Fluor- uracil.
<I>Beispiel 3</I> Einer Mischung von 17,3 g von nach den An gaben in Beispiel 2 frisch hergestelltem Kaliumsatz von Hydroxymethylen-fluoressigsäureäthylester, 18,5 g Bromid von S-Äthyl-isothioharnstoff und einer Lö sung von 2,3g Natrium in 250 ml absolutem Äthanol wird unter Rühren und Rückfluss zwei Stunden er hitzt. Die Reaktionsmischung ist eine Suspension von gelblichen Kristallen und wird im Vakuum zur Trockne eingedampft.
Der Rückstand wird in 80 ml warmem Wasser gelöst, die Lösung durch Kohle fil triert, das Filtrat eisgekühlt und durch Zusatz von konzentrierter Chlorwasserstoffsäure schwach kongo sauer gestellt. Die ausgefallenen Kristalle werden ab filtriert, mit Wasser bromidfrei gewaschen und bei 100 getrocknet. Man erhält den S-Äthyl-thioäther von 2-Thio-5-fluor-uracil, der zwischen 164 und 175 schmilzt. Nach Reinigung durch Umkristallisa tion aus Toluol weist das reine Produkt einen Schmelzpunkt von 181-185 auf.
Nach weiterem Umkristallisieren aus Essigsäureäthylester steigt der Schmelzpunkt auf 192-193 .
Eine Lösung von 0,52 g des erhaltenen Thio- äthers in 10 ml konzentrierter wässeriger Chlor wasserstoffsäure wird 41/2 Stunden unter Rückfluss gekocht. Die Reaktionsmischung wird im Vakuum abgedampft und der kristalline Rückstand aus 4 ml Wasser umkristallisiert. Die erhaltenen Kristalle wer den nacheinander mit kaltem Wasser und mit Metha nol gewaschen. Man erhält 5-Fluor-uracil vom Schmelzpunkt 281-282 (unter Zersetzung). Nach Sublimation im Vakuum (0,1 mm, 190 Badtempe- ratur) steigt der Schmelzpunkt auf 282-283 .
<I>Beispiel 4</I> Eine Lösung von 11 g des nach Beispiel 1 er haltenen Kaliumsatzes von Hydroxymethylen-fluor- essigsäureäthylester, 7,05g Hydrochlorid von O-Me- thyl-pseudoharnstoff und 3,78g Natriummethanolat in 200 ml Methanol werden zwei Stunden unter Rühren und Rückfluss erhitzt.
Die entstehende trübe Lösung wird im Vakuum zur Trockne eingedampft, der Rückstand in 18 ml Wasser aufgenommen, die Lösung von ungelöstem Material abfiltriert, in Eis gekühlt und durch Zusatz von 2,2 ml konzentrierter Chlorwasserstoffsäure kongosauer gestellt. Die gebil deten Kristalle werden abfiltriert und nacheinander mit Wasser und mit Diäthyläther gewaschen. Man erhält den 2-Methyläther von 5-Fluor-uracil vom Schmelzpunkt 190-192 .
Nach zweimaligem Um kristallisieren aus Wasser steigt der Schmelzpunkt auf 206-207 .
50 g des so erhaltenen 2-Methyläthers von 2-Oxy-5-fluor-uracil werden vier Stunden in einer Stickstoffatmosphäre mit 5 ml konzentrierter wässe riger Chlorwasserstoffsäure erhitzt. Die Lösung wird zur Trockne eingedampft, der Rückstand mit Wasser aufgenommen und wieder eingedampft. Man erhält 5-Fluor-uracil vom Schmelzpunkt 282-283 .

It is convenient to use an alkyl ester of an inorganic mineral acid, such as. B. diethyl sulfate, methyl bromide or ethyl iodide to use. The reaction can be carried out either with or without a solvent.
The obtained salt of fluoroacetic acid is preferably heated with diethyl sulfate and the resulting ethyl fluoroacetate is distilled for purification.
In the condensation which is the second stage of the process according to the invention, the alkyl esters of fluoroacetic acid on the one hand and formic acid on the other hand with the same alkyl groups are expediently used. As a condensing agent, for. B. potassium or sodium or alkali metal alcoholates, such as. B. potassium ethanolate or sodium methoxide, can be used. The fluoroacetic ester obtained is expediently together with the formic acid ester and the condensing agent in an anhydrous inert solvent, such as.
B. diethyl ether or toluene heated. The compounds obtained can be used for the next process stage without further purification.
In the third stage of the process according to the invention, the alkali metal salt obtained of the corresponding hydroxymethylene fluoroacetic acid ester, as said, is cyclized with an ether of isothiourea or of pseudohamea. Preferred ethers are the alkyl ethers, such as.
B. the methyl or ethyl ether, and the benzyl ether of isothiourea or pseudourea. The reaction is conveniently carried out under water-free conditions in the presence of an anhydrous inert solvent such as e.g. B. methanol or ethanol performed. It is recommended to use the obtained salt of the hydroxymethylene fluoroacetic acid ester in a freshly prepared state, preferably within two to three days after preparation.
The salt of the hydroxymethylene fluoroacetic acid ester is expediently used together with an acid addition salt of the ether of isothiourea, respectively. Pseudourea heated at the same time as the amount of alkali calculated to neutralize the mineral acid.
The corresponding 2-ethers or 2-thioethers of 5-fluoro-uracil obtained can be purified in the usual manner by recrystallization from inert solvents or by sublimation in vacuo.
The hydrolysis of the 2-thioether obtained from 2-thio-S-fluoro-uracil is carried out, for. B. by heating with an aqueous solution of a mineral acid, such as. B. concentrated hydrochloric acid. The resulting 5-fluoro-uracil can, for. B. by recrystallization from inert solvents or by sublimation in vacuo.
The obtained ether of 2-oxy-5-fluoro-uracils is also z. B. by heating with an aqueous solution of a mineral acid such. B. concentrated aqueous hydrochloric acid, hydrolysed. For purification, the 5-fluorouracil formed can be recrystallized from an inert solvent or sublimed in vacuo.
The 5-fluorouracil obtainable by the process according to the invention shows acidic properties and can be converted into the corresponding salts with bases. It is advantageous to use those bases which lead to non-toxic salts, e.g. B. alkali metal hydroxides, alkaline earth metal hydroxides, ammonia or non-toxic organic bases, such as. B. ethanolamine.
The 5-fluorouracil obtained according to the invention possesses valuable pharmacological properties, in particular germicidal, antimetabolic and anti-ulcer effects.
<I> Example 1 </I> A mixture of 200 g of dry sodium salt of fluoroacetic acid and 442 g of diethyl sulfate are refluxed for 31/2 hours. After distillation of the reaction mixture, 177.3 g of crude ethyl ester of fluoroacetic acid with a boiling point of 116-120 are obtained. After another distillation through a fractionating column, the ethyl ester of fluoroacetic acid boiling between 114 and 118 is obtained.
880 ml of absolute diethyl ether and 47.6 g of potassium cut into 5 mm pieces are placed in a flask equipped with a stirrer, dropping funnel and reflux condenser. 220 ml of absolute ethanol are added dropwise with stirring and the mixture is refluxed until the potassium has completely dissolved. 135 g of the ethyl fluoroacetate obtained and 96.4 g of freshly distilled ethyl formate are added to the ice-cold solution within 21/2 hours, with stirring and cooling.
When everything has been entered, the mixture is stirred for a further hour while cooling and the reaction mixture is then left to stand overnight. The crystalline residue formed is filtered off with suction, washed with diethyl ether and dried in vacuo. The potassium salt of the ethyl hydroxymethylene fluoroacetate is obtained.
A mixture of 103.6 g of freshly prepared halium salt of hydroxymethylene - fluoroacetic acid ethyl ester, 83.4 g of the sulfate of S-methyl isothiourea and 32.5 g of sodium methoxide is heated under reflux in 1500 ml of absolute methanol with stirring. After temporary dissolution, precipitation is observed. After refluxing for two hours, the reaction mixture is evaporated to dryness in vacuo and the residue is treated with 280 ml of water.
The resulting mixture is filtered through charcoal and the decolorized filtrate is made weakly acidic to Congo by adding 37% hydrochloric acid. The crystals formed are filtered off, washed free of sulfate with water and dried at 100. The crude S-methyl thioether of 2-thio-5-fluoro-uracil with a melting range of 202-221 is obtained.
After dissolving in 2035 ml of boiling ethyl acetate and cooling to -20, the product melts at 230-237. After further recrystallization from water, the melting point rises to 241-243.
A solution of 10 g of the S-methyl thioether of 2-thio-S-fluorouracil with a melting point of 230-237 in 150 ml of 37% aqueous hydrochloric acid is refluxed under nitrogen for four hours. The reaction mixture is evaporated in vacuo and the brownish crystalline residue is recrystallized from water. The product is further purified by sublimation in a vacuum at around 190 / 0.1 mm. Colorless or reddish crystalline 5-fluorouracil with a melting point of 282-283 (with decomposition) is obtained.
<I> Example 2 </I> 880 ml of toluene and 47.6 g of potassium are placed in a flask equipped with a stirrer, reflux condenser, dropping funnel, thermometer and feed tube through which a constant stream of nitrogen is fed. The mixture is heated to about 80 until the potassium melts. The metal is finely divided by stirring. 190 ml of ethanol are added dropwise so that the mixture boils gently. When everything has been entered, heating continues until all of the potassium has dissolved.
About 250 ml of the toluene-ethanol mixture are evaporated and the solution is cooled in an ice bath, some of the potassium ethanolate formed precipitating out. After adding 146 g of methyl formate distilled over calcium carbonate, a colorless solution is obtained. With cooling and stirring, 135 g of the ethyl fluoroacetate obtained according to Example 1 are added over the course of 2 hours. The reaction mixture is stirred for an additional hour in an ice bath and then left to stand at room temperature overnight.
The almost colorless crystals are filtered off, washed with ether and dried in vacuo at room temperature. The potassium salt of hydroxymethyl, n-fluoroacetic acid ethyl ester is obtained.
50 g of the potassium salt of ethyl hydroxymethylene-fluoroacetate and 53.6 g of the bromide of S-ethyl-isothiourea are added to a solution of 6.7 g of sodium in 720 ml of absolute ethanol in a flask equipped with a stirrer and reflux condenser. After temporary dissolution, crystallization takes place. The mixture is heated on a water bath with stirring and reflux for a further two hours.
The alcohol is evaporated off in vacuo and the residue is dissolved in 140 ml of water. The solution is filtered through charcoal, cooled in an ice bath, and acidified to Congo with 22 ml of concentrated hydrochloric acid. The crystalline precipitate is filtered off, washed free of chlorine with water and dried at 100 ^. The 2-ethyl-thioether of 2-thio-5-fluoro-uracil is obtained with a melting point of 175-179.
After recrystallization from hot ethyl acetate, crystals with a melting point of 189-190 are obtained. The 5-fluorouracil is obtained by saponification as described in Example 1.
<I> Example 3 </I> A mixture of 17.3 g of the potassium set of hydroxymethylene fluoroacetic acid ethyl ester freshly prepared according to the information in Example 2, 18.5 g of bromide of S-ethyl isothiourea and a solution of 2, 3g of sodium in 250 ml of absolute ethanol is heated under reflux and stirring for two hours. The reaction mixture is a suspension of yellowish crystals and is evaporated to dryness in vacuo.
The residue is dissolved in 80 ml of warm water, the solution is filtered through charcoal, the filtrate is ice-cooled and made weakly acidic to Congo by adding concentrated hydrochloric acid. The precipitated crystals are filtered off, washed bromide-free with water and dried at 100. The S-ethyl thioether of 2-thio-5-fluoro-uracil, which melts between 164 and 175, is obtained. After purification by recrystallization from toluene, the pure product has a melting point of 181-185.
After further recrystallization from ethyl acetate, the melting point rises to 192-193.
A solution of 0.52 g of the thioether obtained in 10 ml of concentrated aqueous hydrochloric acid is refluxed for 41/2 hours. The reaction mixture is evaporated in vacuo and the crystalline residue is recrystallized from 4 ml of water. The obtained crystals who washed the successively with cold water and with Metha nol. 5-fluorouracil is obtained with a melting point of 281-282 (with decomposition). After sublimation in a vacuum (0.1 mm, 190 bath temperature) the melting point rises to 282-283.
<I> Example 4 </I> A solution of 11 g of the potassium set of hydroxymethylene fluoroacetic acid ethyl ester obtained according to Example 1, 7.05 g of the hydrochloride of O-methyl pseudourea and 3.78 g of sodium methoxide in 200 ml of methanol are used heated for two hours with stirring and reflux.
The resulting cloudy solution is evaporated to dryness in vacuo, the residue is taken up in 18 ml of water, the solution is filtered off from undissolved material, cooled in ice and acidified to Congo by adding 2.2 ml of concentrated hydrochloric acid. The formed crystals are filtered off and washed successively with water and with diethyl ether. The 2-methyl ether of 5-fluorouracil is obtained with a melting point of 190-192.
After recrystallizing twice from water, the melting point rises to 206-207.
50 g of the 2-methyl ether of 2-oxy-5-fluoro-uracil thus obtained are heated for four hours in a nitrogen atmosphere with 5 ml of concentrated aqueous hydrochloric acid. The solution is evaporated to dryness, the residue is taken up in water and evaporated again. 5-Fluorouracil is obtained with a melting point of 282-283.
