Verfahren zur Herstellung von Benzothiadiazin-1,1-dioxyden Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Benzothiadiazin-1,1-dioxyden, welche im Benzoring eine Sulfamylgruppe enthalten. Die Strukturformel der neuen Verbindungen, welche nach diesem Verfahren erhalten werden, wird unten angeführt.
Es ist dabei aber zu beachten, dass bei jenen Verbindungen, welche an den beiden Stickstoff atomen im Benzothiadiazin-Kern keine andern Sub- stituenten als Wasserstoff aufweisen, die Doppelbin dung tautomer ist, das heisst entweder zwischen den Atomen in den Stellungen 2 und 3 oder zwischen den Atomen in den Stellungen 3 und 4 vorliegt. Bei jenen Verbindungen, welche am Stickstoff-Atom in 4,Stellung einen Substituenten aufweisen, befindet sich die ungesättigte Bindung zwischen den Stellun gen 2 und 3.
Somit kann den Verbindungen folgende Strukturformel I zugeschrieben werden, die in 2,3- oder in 3,4-Stellung eine Doppelbindung aufweist:

Darin bedeutet R, Wasserstoff, ein Halogen, wie Chlor, Brom oder Fluor, eine niedrige Alkylgruppe, vorzugsweise mit 1-5 C-Atomen, eine niedrige Alkoxygruppe, ebenfalls vorzugsweise mit 1-5 C- Atomen im Alkylteil des Radikals, oder die Nitro- gruppe;

Die oben genannten Verbindungen, welche nach dem erfindungsgemässen Verfahren erhalten werden, sind wertvolle Chemotherapeutika, in erster Linie auf Grund ihrer diuretischen und/oder natriureti- schen Eigenschaften. Die Verbindungen können so wohl oral in Form von Tabletten oder Kapseln und dergleichen, als auch, gelöst in einem verdünnten alkalischen Medium oder in Polyäthylenglykol- Lösung, durch Injektion verabreicht werden.
Das erfindungsgemässe Verfahren zur Herstellung der Verbindungen der Formel I ist dadurch gekenn zeichnet, dass man ein Disulfamylanilin der Formel II

B. ein Ameisensäureamid, wie Formamid oder Dimethylformamid, oder ein Essig- säureamid, wie Acetamid oder Dimethylacetamid, vorzugsweise unter Erhitzen auf 150-200 C, zur Umsetzung bringt.
Für die erfindungsgemässe Reak tion verwendet man mindestens molare Äquivalente der Disulfamylanilin-Verbindung und des Amids; in der Praxis wendet man jedoch im allgemeinen das Amid im überschuss an, da es als Lösungsmittel wirkt; gewünschtenfalls kann aber auch ein anderes Lösungsmittel für die Reaktionsteilnehmer verwen det werden.
Das als Ausgangsstoff verwendete Disulfamyl- anilin kann in situ durch Einleiten von trockenem Ammoniakgas in eine Lösung des entsprechenden Disulfonylchlorids, nötigenfalls oder gewünschtenfalls in Gegenwart eines Lösungsmittels, unter nachfol gendem Entfernen von überschüssigem Ammoniak, hergestellt werden.
Zur Ammonolyse werden min destens vier molare Äquivalente Ammoniak benötigt, doch wird im allgemeinen, wie oben ausgeführt, ein überschuss angewendet.
Die erfindungsgemässe Ringbildung wird vorzugs weise in Gegenwart eines sauren Katalysators, wie Ammoniumchlorid, Schwefelsäure, Sulfonsäure, einem Säureharz, beispielsweise einem sulfonierten Poly- styrolharz (es eignet sich hierzu das unter der Han delsbezeichnung IR 120 von The Rohm & Haas Company oder das unter der Handelsbezeichnung Dowex 50 von The Dow Chemical Company auf den
Markt gebrachte Produkt), durchgeführt. Im all gemeinen genügt eine Spur von Säurekatalysator, doch können auch 1% und mehr Katalysator mit Erfolg zur Beschleunigung der Geschwindigkeit, mit welcher das Disulfamylanilin-Derivat acyliert wird, verwendet werden.
<I>Beispiel 1</I> Man erhitzt eine Lösung von 5,72 g (0,02 Mol) 5-Chlor-2,4-disulfamylanilin und 4,5g Ammonium chlorid in 50 cm3 Dimethylformamid eine Stunde lang auf 180-190 C. Die Lösung wird dann gekühlt und mit Wasser verdünnt, wobei man 6-Chlor-7- sulfamyl-1,2,4-benzothiadiazin-1,1-dioxyd erhält, wel ches abgetrennt, mit Wasser gewaschen und getrock net wird. Nach dem Kristallisieren aus verdünntem Dimethylsulfoxyd erhält man das Produkt in Form von farblosen Nadeln, Schmelzpunkt 347-352 C.
<I>Beispiel 2</I> Man erhitzt eine Lösung von 12 g 5-Chlor-2,4- disulfamylanilin in 50 cm3 Formamid eine Stunde lang auf 170-180 C. Das Reaktionsgemisch wird dann gekühlt und mit Wasser verdünnt zur Aus fällung von 6-Chlor-7-sulfamyl-1,2,4-benzothiadiazin- 1,1-dioxyd. Das Produkt wird abfiltriert, mit Wasser gewaschen und getrocknet. Nach dem Umkristallisie- ren aus verdünntem Dimethylformamid erhält man ein Produkt in Form von farblosen Nadeln mit dem Schmelzpunkt 242,5-343 C.
<I>Beispiel 3</I> Man leitet eine Stunde lang trockenes Ammoniak gas in eine Lösung von 12 g 5-Chloranilin-2,4-disul- fonylchlorid in 50 cm3 Formamid ein. Eine Probe des Reaktionsprodukts wird analysiert, indem man das Formamid mit 300/uigem Natriumhydroxyd hydrolysiert und zur Ausfällung des 5-Chlor-2,4- disulfamylanilins, welches einen Schmelzpunkt von 257,5-259,5 C besitzt, ansäuert.
Das obige Reaktionsgemisch wird von überschüs sigem Ammoniak befreit, indem man die Lösung un ter Vakuum und unter Rühren auf ungefähr 600C erhitzt. Wenn die Ammoniak-Entwicklung aufhört, erhitzt man eine Stunde lang auf 170 C. Das Reak tionsgemisch wird dann gekühlt und mit Wasser ver dünnt zur Ausfällung von 6-Chlor-7-sulfamyl-1,2,4- benzothiadiazin-1,1-dioxyd. Durch potentiometrische Titration wird eine Reinheit von 98,9 0/a * ermittelt, mit einem pka von 6,7, welcher für das Produkt charakteristisch ist.
Das Ultraviolett-Spektrum zeigt ein Maximum bei 225 mp, ein Minimum bei 247 m,if, ein weiteres Maximum bei 279 m/i, und einen Wende punkt bei 318 m/s; das Spektrum ist identisch mit demjenigen einer Vergleichsprobe.
<I>Beispiel 4</I> Man gibt 12,0 g 5-Chlor-2,4-disulfamylanilin und 4,5 g Ammoniumchlorid zu 48 cm3 Formamid und erhitzt eine .Stunde lang auf 170 C. Man giesst das Reaktionsgemisch in 5 Volumteile Wasser, lässt eine Stunde lang bei 5 C stehen, filtriert, wäscht gut mit Wasser und trocknet an der Luft.
Man erhält 9,8 g (79% der Theorie) 6-Chlor-7-sulfamyl-1,2,4- benzothiadiazin-1,1-dioxyd mit einer Reinheit von 89,90/0 *, ermittelt durch potentiometrische Titra- tionen.
<I>Beispiel S</I> Man erhitzt ein Gemisch aus 0,02 Mol 5-Chlor- 2,4-disulfamylanilin, 0,1 cm3 konzentrierter Schwefel säure und 0,1 Mol Dimethylacetamid eine Stunde lang auf etwa 180-190 C. Man kühlt das Reaktions gemisch ab, fügt Wasser zu und sammelt das so gebildete 3-Methyl-6-chlor-7-sulfamyl-1,2,4-benzo- thiadiazin-1,1-dioxyd, welches mit Wasser gewaschen und getrocknet wird. Das Produkt wird durch Um kristallisieren aus verdünntem Dimethylsulfoxyd ge reinigt.
<I>Beispiel 6</I> Man erhitzt ein Gemisch aus 0,1 Mol 5-Chlor- 2,4-disulfamylanilin und 0,2 Mol Acetamid eine Stunde lang auf 180-190 C. Die Lösung wird dann abgekühlt und mit Wasser versetzt, und das dabei gebildete 3-Methyl-6-chlor-7-sulfamyl-1,2,4-benzo- thiadiazin-1,1-dioxyd wird gesammelt, mit Wasser gewaschen und getrocknet. Das Produkt wird durch Umkristallisieren aus verdünntem Dimethylsulfoxyd gereinigt.
