Description
Verfahren zur Herstellung neuer sekundärer Amine Gegenstand der Erfindung ist ein Verfahren zur Herstellung von sekundären Aminen der Formel EMI1.1 worin Rj einen Kohlenwasserstoffrest aliphatischen Charakters mit 3-7 Kohlenstoffatomen, R2 einen Kohlenwasserstoffrest und n eine niedere ganze Zahl, mindestens aber 2 bedeuten. Besonders sind zu nennen die Verbindungen der Formel EMI1.2 worin Ri und R, zusammen mindestens 5 Kohlenstoffatome, vorzugsweise 7-10 Kohlenstoffatome enthalten, und n für eine Zahl von 2-4 steht, und vor allem Verbindungen der Formel EMI1.3 worin Ri und R2 zusammen mindestens 5 Kohlenstoffatome, vorzugsweise 7-10 Kohlenstoffatome enthalten. Der Kohlenwasserstoffrest aliphatischen Charakters R1 mit 3-7 C-Atomen ist beispielsweise ein Alkylrest wie Propyl, Isopropyl, Butyl, sek.-Butyl, Pentyl, Isopentyl, Hexyl oder Heptyl, ein Cycloalkylrest, wie Cyclopentyl oder Cyclohexyl, oder ein ungesättigter Rest, wie Allyl, Cyclopentenyl oder Cyclohexenyl. Der Kohlenwasserstoffrest R2 ist zweckmässig ebenfalls aliphatischen Charakters, vor allem ein aliphatischer oder cycloaliphatischer Kohlenwasserstoffrest mit 1-10, insbesondere 3-6 Kohlenstoffatomen, vorzugsweise ein niederer gerader oder verzweigter Alkylrest, z. B. Methyl, Athyl, Propyl, Isopropyl, Allyl, Methallyl, Butyl, sek.-Butyl, Pentyl oder Hexyl oder auch ein Cycloalkylrest, z. B. Cyclopentyl oder Cyclohexyl. Das Symbol n steht vorzugsweise für eine Zahl von 2-4, insbesondere für 2. Die neuen Verbindungen, ganz besonders diejenigen der Formel EMI1.4 worin Ri einen Butyl-oder Pentylrest und R2 einen Propyl-oder Butylrest bedeuten, und ihre Salze, besitzen gute lokalanästhetische Eigenschaften und sollen dementsprechend als Heilmittel verwendet werden. In erster Linie sind hier zu erwähnen das N-[ss- (m-n-Pentyloxy-phenoxy)-äthyl]-isopropylamin, das N- [p)- (m-n-Pentyloxy-phenoxy)-dthyl]-isobutylamin und das N- [- (m-n-Butoxy-phenoxy)-äthyl]-n-butyl- amin und ihre Salze. Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man eine Verbindung der Formel EMI1.5 worin X eine reaktionsfähige veresterte Hydroxylgruppe bedeutet, mit einem Amin der Formel H2N-R2 kondensiert. Als Reste X kommen insbesondere mit starken anorganischen oder organischen Säuren, z. B. Halogenwasserstoffsäuren oder organischen Sulfonsäuren, wie p-Toluolsulfonsäure, veresterte Hydroxylgruppen in Frage. Je nach der Arbeitsweise erhält man die neuen Verbindungen in Form ihrer Basen oder Salze. Aus den Salzen können in an sich bekannter Weise die freien Aminbasen gewonnen werden. Von letzteren wiederum lassen sich durch Umsetzung mit Säuren, die zur Bildung therapeutisch verwendbarer Salze geeignet sind, Salze gewinnen, wie z. B. der Halogenwasserstoffsäuren, Schwefelsäure, Salpetersäure, Phos phorsäure, Rhodanwasserstoffsäure, Essigsäure, Pro pionsäure, Oxalsäure, Malonsäure, Weinsäure, Bern- steinsäure, Äpfelsäure, Methansulfonsäure, AthansulfonsÏure, OxyÏthansulfonsÏure, Benzol-oder Toluolsulfonsäure oder von therapeutisch wirksamen Säuren. Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden gewonnen werden. Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer PrÏparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale, parenterale oder topicale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten. In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben. Beispiel 1 13, 6 g (0, 05 Mol),-(m-n-Butoxy-phenoxy)-äthyl- bromid und 7, 3 g (0, 1 Mol) n-Butylamin werden 11/2 Stunden auf 120 erhitzt. Nach dem Abkuhlen nimmt man das Reaktionsgemisch in 100 cm3 Chloroform auf und schüttelt die Lösung zunächst mit 25 cm3 n-Natronlauge und dann mit Wasser aus. Die Chloroformlösung hinterlässt nach dem Trocknen über wasserfreiem Natriumsulfat und anschliessendem Eindampfen im Wasserstrahlvakuum N- [13- (m-n-Butoxy- phenoxy)-äthyl]-n-butylamin als gelbes 01. Dieses wird mit einer 2n-Lösung von Chlorwasserstoff in Essigester neutralisiert, die Lösung eingedampft und das erhaltene Hydrochlorid aus Athanol/ Äther zweimal umkristallisiert. Farblose Kristalle vom F. 157-15der Formel EMI2.1 Das als Ausgangsstoff verwendete i- (m-n-Butoxy- phenoxy)-äthylbromid wird z. B. wie folgt erhalten : Zu einer Lösung von 11, 5 g (0, 5 Mol) Natrium in 250 cm3 absolutem Athanol lässt man 83 g (0, 5 Mol) m-Butoxy-phenol, welches in 100 cm3 absolutem Athanol aufgelöst worden ist, zufliessen. Zu dem auf 0 abgekühlten Gemisch werden 282 g (1, 5 Mol) Athylenbromid zugegeben. Unter kräftigem R hren kocht man den Kolbeninhalt unter Rückfluss, bis er neutral reagiert (2-3 Stunden). Nach dem Abkühlen filtriert man von entstandenem Natriumbromid ab und engt im Wasserstrahlvakuum ein. Der Rückstand wird in 200 cm3 Chloroform aufgenommen, zweimal mit je 50 cm3 2n-Natronlauge ausgezogen, dann mit Wasser neutralgewaschen und schliesslich über wasserfreiem Natriumsulfat getrocknet. Nach dem Abdampfen des Lösungsmittels destilliert man den Rückstand im Wasserstrahlvakuum. Das erhaltene ss-(m-n-But- oxy-phenoxy)-äthylbromid der Formel EMI2.2 ist ein hellgelbes Íl vom Kpt2 : 157-160 . Beispiel 2 11, 0 g (0, 043 Mol) ss-(m-n-Propoxy-phenoxy)- äthylbromid und 11, 8 g (0, 2 Mol) n-Propylamin werden in 20 cm3 Athanol 12 Stunden unter Rückfluss gekocht. Man dampft im Wasserstrahlvakuum ein, versetzt mit 50 cm3 n-Natronlauge und extrahiert mit Chloroform. Beim Eindampfen der Chloroform-Auszüge erhält man das N- [/3- (m-n-Propoxy-phenoxy)- äthyl]-n-propylamin der Formel EMI2.3 als gelbliches Ö1, welches durch Neutralisieren mit Salzsäure in Essigester direkt in das Hydrochlorid übergeführt wird : Aus Äthanol-Essigester farblose Kristalle vom F. 148-150 . Das als Ausgangsstoff verwendete ss-(m-n-Propoxy- phenoxy)-äthylbromid wird z. B. wie folgt erhalten : 83, 8 g (0, 55 Mol) Resorcin-mono-n-propyläther gibt man zu einer Lösung von 12, 8 g (0, 56 Mol) Natrium in 400 cm3 Athanol. Man erhitzt zum Sieden und lässt unter kräftigem R hren 310 g (1, 65 Mol) Athylenbromid einfliessen. Der Kolbeninhalt wird nun so lange unter Rückfluss erhitzt, bis er neutral reagiert (2-3 Stunden). Nach dem Abkühlen auf 0 filtriert man das ausgeschiedene Natriumbromid ab und engt das Filtrat im Wasserstrahlvakuum ein. Der R ckstand wird in 200 cm3 Chloroform aufgenommen, zweimal mit je 50 cm3 2n-Natronlauge ausgezogen, dann mit Wasser neutralgewaschen und schliesslich über wasserfreiem Natriumsulfat getrocknet. Nach dem Entfernen des Lösungsmittels destilliert man den Rückstand im Hochvakuum. Das P- (m-n-Propoxy- phenoxy)-äthylbromid der Formel EMI3.1 ist ein fast farbloses 01 vom Kpo, 02: 75-83¯. Die hier und für die Herstellung von in nachfolgenden Beispielen genannten Ausgangsstoffen verwendeten neuen Resorcinäther werden z. B. wie folgt erhalten : Zu einer Lösung von 23 g (1 Mol) Natrium in 400 cm3 Athanol lässt man 110 g (1 Mol) Resorcin in 300 cm3 Athanol einfliessen. Man rührt das Gemisch 30 Minuten, fügt dann 200 g (1, 18 Mol) n-Propyljodid hinzu und kocht unter Rückfluss und Rühren 3 Stunden. Das Lösungsmittel wird abgedampft, der dunkle Rückstand in Chloroform aufgenommen, mit verdünnter Salzsäure und Wasser extrahiert und über wasserfreiem Natriumsulfat getrocknet. Nach dem Abdampfen des Lösungsmittels wird der Rückstand im Hochvakuum an einer Vigreux-Hickman-Kolonne fraktioniert. Man erhält so den Resorcin-mono-n-propyläther als hellgelbes 01 vom Kpo, 07 : 80-89 ; n 24 D = 1, 5232. Auf die gleiche Weise erhält man ausgehend von 23 g (1, 0 Mol) Natrium und 110 g (1, 0 Mol) Resorcin, die man in 400 cm3 Athanol zur Reaktion bringt, durch Umsetzen mit 165 g (1, 2 Mol) Isobutylbromid das m-Isobutoxy-phenol als farblose Flüssigkeit vom Kpo o6 80-87 . Wenn man 110 g (1, 0 Mol) Resorcin in eine Lösung von 23 g (1, 0 Mol) Natrium in 400 cms Athanol einträgt und anschliessend wie oben beschrieben mit 180 g (1, 0 Mol) n-Amylbromid umsetzt, so erhält man das m-Pentyloxy-phenol vom Kpos06 100-105 . Schliesslich erhält man auf gleiche Weise durch Umsetzmg von 110 g (1, 0 Mol) Resorcin mit 23 g (1, 0 Mol) Natrium und anschliessende Reaktion mit 145 g (1, 20 Mol) Allylbromid das m-Allyloxy-phenol vom Kip,, : 145-150 . Beispiel 3 Aus 11, 0 g (0, 043 Mol) #- (m-n-Propoxy-phen- oxy)-äthylbromid und 14, 6 g (0, 2 Mol) Isobutylamin erhält man auf die im Beispiel 2 beschriebene Weise das N-[ss-(m-n-Propoxy-phenoxy)-äthyl]-isobutylamin der Formel EMI3.2 als gelbes 01, welches in der im Beispiel 1 angegebenen Weise direkt in das Hydrochlorid übergeführt werden kann : Aus Äthanol-Essigester farblose Kristalle vom F. 139-140 . Beispiel 4 Aus 11, 0 g (0, 043 Mol) ss-(m-n-Propoxy-phen- oxy)-äthylbromid und 17, 4 g (0, 2 Mol) n-Pentylamin wird nach dem im Beispiel 2 beschriebenen Verfahren das N-[ss-(m-n-Propoxy-phenoxy)-äthyl]- pentylamin der Formel EMI3.3 hergestellt und in das Hydrochlorid übergeführt. Aus Äthanol-Essigester farblose Kristalle vom F. 148 bis 150 . Beispiel 5 12, 0 g (0, 044 Mol) ¯-(m-n-Butoxy-phenoxy) äthylbromid und 9, 0 g (0, 2 Mol) Äthylamin werden nach Zugabe von 50 cm3 Athanol 14 Stunden im geschlossenen Rohr auf 100 erhitzt. Die Aufarbeitung erfolgt gleich wie im Beispiel 2 angegeben und führt zum N- [/3- (m-n-Butoxy-phenol-äthyl]-äthylamin der Formel EMI3.4 dessen Hydrochlorid nach Umkristallisieren aus Athanol-Essigester bei 160-162 schmilzt. Beispiel 6 Aus 12, 0 g (0, 044 Mol) #- (m-n-Butoxy-phenoxy)- äthylbromid und 11, 8 g (0, 02 Mol) n-Propylamin erhält man nach dem im Beispiel 2 beschriebenen Verfahren das N-[¯-(m-n-Butoxy-phenoxy)-Ïthyl]-n-propylamin der Formel EMI4.1 als gelbes 01. Das in üblicher Weise hergestellte Hydrochlorid bildet aus Athanol-Essigester farblose Schuppen vom F. 147, 5 . Beispiel 7 Aus 12, 0 g (0, 044 Mol) #- (m-n-Butoxy-phenoxy)- äthylbromid und 14, 2 g (0, 2 Mol) Methallylamin erhÏlt man nach dem im Beispiel 2 beschriebenen Verfahren das N- [/3- (m-n-Butoxy-phenoxy)-äthyl]-Nmethallylamin der Formel EMI4.2 als farblose Fl ssigkeit, Kp0.05: 125-128¯. Sein Hydrochlorid kristallisiert aus Äthanol-Essigester in farblosen Schuppen vom F. 109-111 . Beispiel 8 Aus 6, 1 g (0, 022 Mol) ¯-(m-n-Butoxy-phenoxy) äthylbromid und 7, 3 g (0, 1 Mol) Isobutylamin erhält man nach dem im Beispiel 2 beschriebenen Verfahren das N- [ :- (m-n-Butoxy-phenoxy)-äthyl]-N-isobutyl- amin der Formel EMI4.3 als bräunliches 01. Hydrochlorid : Aus Essigester farblose Kristalle vom F. 137-138 . Beispiel 9 Aus 8, 0 g (0, 029 Mol) ss-(m-n-Butoxy-phenoxy)- äthylbromid und 14, 6 g (0, 2 Mol) tert.-Butylamin erhÏlt man nach dem im Beispiel 2 beschriebenen Verfahren das N- [- (m-n-Butoxy-phenoxy)-äthyi]-N-tert.- butylamin der Formel EMI4.4 als hellbraunes 01. Das auf übliche Weise hergestellte Hydrochlorid schmilzt nach Umkristallisieren aus Essigester-Ather bei 108¯. Beispiel 10 Die Reaktion von 8, 0 g (0, 029 Mol) ¯-(m-n-Butoxy-phenoxy)-äthylbromid mit 17, 4 g (0, 2 Mol) n-Pentylamin führt nach dem im Beispiel 2 beschriebenen Verfahren zum N- [P- (m-n-Butoxy-phenoxy)- äthyl]-N-n-pentylamin der Formel EMI4.5 dessen Hydrochlorid nach dem Umkristallisieren aus Essigester bei 142-144 schmilzt. Beispiel 11 12, 5 g (0, 046 Mol) ss-(m-Isobutoxy-phenoxy)-äthyl- bromid und 11, 8 g (0, 2 Mol) n-Propylamin führen nach dem im Beispiel 2 wiedergegebenen Verfahren zum N-[ss-(m-Isobutoxy-phenoxy)-äthyl]-N-n-propyl- amin der Formel EMI5.1 dessen Hydrochlorid, farblose Nadeln aus Essigester Athanol, bei 138-140 schmilzt. Das als Ausgangsstoff verwendete #- (m-Isobut- oxy-phenoxy)-äthylbromid wird z. B. wie folgt erhalten : 75, 0 g (0, 45 Mol) m-Isobutoxy-phenol werden durch Zugabe zu einer Lösung von 10, 4 g (0, 45 Mol) Natrium in 400 cm3 Athanol ins Natriumsalz übergeführt und mit 255 g (1, 35 Mol) Athylenbromid auf analoge Weise wie im Beispiel 2 angegeben in das ss-(m-Isobutoxy-phenoxy)-äthylbromid der Formel EMI5.2 verwandelt, welches als hellgelbes Ö1 vom K 06:@ 82-90 anfällt. Beispiel 12 Auf gleiche Weise wie im Beispiel 2 angegeben führt die Reaktion zwischen 12, 5 g (0, 046 Mol) /- (m-Isobutoxy-phenoxy)-äthylbromid und 20 cm3 n-Butylamin zum N-[ss-(m-Isobutoxy-phenoxy)-äthyl]- N-n-butylamin der Formel EMI5.3 Das Hydrochlorid schmilzt nach dem Umkristallisieren aus Essigester bei 136-138 . Beispiel 13 Die Reaktion von 12, 5 g (0, 046 Mol) ss-(m-Iso- butoxy-phenoxy)-äthylbromid mit 20 cm3 Isobutylamin ergibt nach den gemäss Beispiel 2 angewandten Reaktionsbedingungen das N-[ss-(m-Isobutoxy-phen- oxy)-äthyl]-N-isobutylamin der Formel EMI5.4 als bräunliches 01. Hydrochlorid : Aus Essigester farblose Nadeln vom F. 140-142 . Beispiel 14 Aus 12, 0 g (0, 043 Mol) P- (m-n-Pentyloxy-phen- oxy)-äthylbromid und 11, 8 g (0, 2 Mol) n-Propylamin erhält man nach der im Beispiel 2 beschriebenen Methode das N-[¯-(m-n-Pentyloxy-phenoxy)-Ïthyl]-N-npropylamin der Formel EMI5.5 als hellgelbes 01. Hydrochlorid : Aus Athanol-Essigester farblose Schuppen vom F. 146-147 . Das als Ausgangsstoff verwendete ss-(m-n-Pentyl- oxy-phenoxy)-äthlbromid wird z. B. wie folgt erhalten : 120 g m-n-Pentyloxy-phenol werden mit 15, 4 g (0, 67 Mol) Natrium in 400 cm3 Athanol ins Natriumsalz übergeführt und anschliessend mit 376 g (2, 0 Mol) Athylenbromid gleich wie im Beispiel 2 be schrieben zur Reaktion gebracht. Das Produkt, ss-(m- n-Pentyloxy-phenoxy)-äthylbromid der Formel EMI6.1 ist ein farbloses 01 vom Kpo, 08 : 102-112 . Beispiel 15 Aus 10, @ g (0, 035 Mol) ¯-(m-n-Pentyloxy-phen oxy)-äthylbromid und 14, 6 g (0, 2 Mol) n-Butylamin erhält man nach dem im Beispiel 2 beschriebenen Verfahren das N- [N- (m-n-Pentyloxy-phenoxy)-äthyl]- n-butylamin der Formel EMI6.2 dessen Hydrochlorid nach Umkristallisation aus Methanol-Essigester bei 151-153 schmilzt. Beispiel 16 Aus 12, 0 g (0, 043 Mol) ss-(m-n-Pentyloxy-phen- oxy)-äthylbromid und 14, 6 g (0, 2 Mol) Isobutylamin erhält man nach dem im Beispiel 2 beschriebenen Verfahren das N-[ss-(m-n-Pentyloxy-phenoxy)-äthyl]- isobutylamin der Formel EMI6.3 Hydrochlorid : Farblose Kristalle aus Athanol-Essigester vom F. 135-136 . In analoger Weise kann man das N-[¯-(m-n-But oxy-phenoxy)-äthyl]-methylamin, Kpo, HO-112", erhalten. Hydrochlorid : F. 138-139 (aus Athanol- Essigester). Beispiel 17 Aus 16, 0 g (0, 062 Mol) ss-(m-Allyloxy-phenoxy)- äthylbromid und 30 cm3 n-Butylamin erhÏlt man nach dem im Beispiel 2 beschriebenen Verfahren das N-[¯ (m-Allyloxy-phenoxy)-äthyl]-n-butylamin der Formel EMI6.4 als farbloses 01 vom Kpo : 118-125 . Hydrochlorid : Farblose Kristalle aus Essigester vom F. 140-142 . Das als Ausgangsstoff verwendete j6- (m-AllyIoxy- phenoxy)-äthylbromid wird z. B. wie folgt erhalten : 34, 6 g (0, 23 Mol) m-Allyloxy-phenol werden in 100 cmS absolutem Athanol gelost und durch Zugabe von 5, 4 g (0, 23 Mol) Natrium in das Natriumsalz übergeführt. Man lässt 130 g (0, 69 Mol) Athylenbromid einfliessen und verfährt im übrigen gleich wie im Beispiel 2 beschrieben. Das Produkt, #- (m-Allyloxy- phenoxy)-äthylbromid der Formel EMI6.5 ist eine gelbliche Flüssigkeit vom Kpoo8 : 102-110 .
Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung der neuen sekundären Amine der Formel EMI6.6 worin R1 einen Kohlenwasserstoffrest aliphatischen Charakters mit 3-7 Kohlenstoffatomen, R2 einen Kohlenwasserstoffrest und n eine niedere ganze Zahl, mindestens aber 2, bedeuten, dadurch gekennzeichnet, dass man eine Verbindung der Formel EMI6.7 worin X eine reaktionsfähige veresterte Hydroxylgruppe bedeutet, mit einem Amin der Formel H2N-R2 kondensiert.UNTERANSPRUCHE 1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass der Rest X eine mit einer Halogenwasserstoffsäure oder mit einer organischen Sulfonsäure veresterte Hydroxylgruppe bedeutet.2. Verfahren nach Patentanspruch und Unteranspruch 1, dadurch gekennzeichnet, dass man von Verbindungen ausgeht, worin R, einen Alkylrest mit 3-7 Kohlenstoffatomen und R2 einen Alkylrest mit 3-6 Kohlenstoffatomen bedeuten.3. Verfahren nach Patentanspruch und Unteranspruch 1, dadurch gekennzeichnet, dass man von Verbindungen ausgeht, worin Ri n-Butyl und R2 n-Butyl bedeuten.4. Verfahren nach Patentanspruch und den Unteransprüchen 1-3, dadurch gekennzeichnet, dass man erhaltene freie Amine in die Säureadditionssalze überführt.5. Verfahren nach Patentanspruch und Unteransprüchen 1-3, dadurch gekennzeichnet, dass man erhaltene Säureadditionssalze in die freien Amine überführt.
