Verfahren zur Herstellung von Phosphonsäureestern und Thionophosphonsäureester
Eine Anzahl von Alkyl-thionophosphonsäure- estern zeichnet sich durch gute insektizide Wirkungen aus (vergleiche z. B. deutsche Auslegeschrift Num- mer 1032 247). Oft jedoch wird diese gute insekti- zide Wirkung dadurch beeinträchtigt, dass die wirksamsten Vertreter gleichzeitig auch bei Warmblütern besonders toxisch sind.
Es wurde die überraschende Beobachtung gemacht, dass solche niederen Alkan-thionophosphonsäureester [B. A. Arbusow und N. P. Bogonowceva, Z. obsc. Chim. 27 (1957) 2360] weitgehend entgiftet sind, in denen die Alkylgruppe eine Substitution durch einen Alkylmercaptorest erfahren hat.
Folgende Gegenüberstellung möge dies erläutern :

R und Ri stehen in diesen Formeln für Alkylreste, R2 fiir beliebige Alkyl-, Aryl-, Aralkyl-oder hetero- cyclische Reste.
Gegenstand der Erfindung ist nun ein Verfahren zur Herstellung von Phosphonsäureestern oder von Thionophosphonsäureestern, das dadurch gekennzeichnet ist, dass Alkylmercapto-alkan-phosphonsäure- bzw.-thionophosphonsäuredihalogenide mit Alkoholen in die entsprechenden Alkylmercapto-alkan- phosphon-bzw. thionophosphonsäure-O-alkylesterhalogenide übergeführt und diese Alkylmercapto alkan-phosphon-bzw.-thionophosphonsäureesterhalo- genide mit OH-bzw. SH-gruppenhaltigen Verbindungen zur Umsetzung gebracht werden.
Einfache Alkylmercapto-alkan-phosphonsäureester, das heisst Verbindungen der Formel II entsprechende Stoffe mit einer P = O-anstelle einer P = S Gruppierung, in welchen R. und R2 einfache Alkylreste sind, sind in der Literatur teilweise beschrieben. Man kann diese Verbindungen leicht durch Umsetzung von a-Chlormethyl-phosphonsäureestern mit beliebigen Mercaptanen herstellen [siehe B. A.
Arbusow und N. P. Bogonowceva, Z. obsc. Chim. 27 (1957) 2360] wie an folgendem Formelschema ersichtlich ist.

Die nach den genannten Methoden leicht herstellbaren Alkylmercapto-alkan-phosphonsäureester können nach bekannten Verfahren mit Salzsäure in die entsprechenden freien Säuren übergeführt werden. Diese können dann ohne besondere Isolierung mit chlorierenden Mitteln, wie z. B.
Phosphorpentachlorid oder Thionylchlorid in die noch nicht be schriebenen Alkylmercapto-alkan-phosphonsäuredichloride übergeführt werden :

Mit der berechneten Menge eines beliebigen Alkoholes in Gegenwart z. B. einer tertiären Base, können diese PhosphonsäurePdihalogenide in die entsprechenden Halbesterchloride folgender Zusammensetzung :

Die Alkylmercapto-alkan-phosphonsäure-dichlor- ide, z. B. (III), können auch mit Phosphorpentasulfid so umgesetzt werden, dass die entsprechenden Alkylmercapto-alkan-thionophosphonsäure-dichloride entstehen. [M. I. Kabachnik und N. M. Godovikow, Ber. d. Akad. d. Wiss. d. UdSSR 110, 217-219 (1956)]. Diese werden dann ebenfalls in die Halbesterhalogenide übergeführt :

Die Halbesterchloride (z. B. IV) können nun ebenfalls in bekannter Weise mit Alkoholen oder Phenolen oder Mercaptanen in Gegenwart von Säurebindemitteln zur Umsetzung gebracht werden.
Die auf diese Weise entstehenden neuen Ester zeichnen sich durch hervorragende insektizide Wirkungen aus. Hinsichtlich der Warmblüter-Toxizität sind sie gegenüber den vergleichbaren Methyl-phosphonsäureestern wesentlich günstiger, wie beispielsweise aus folgender Tabelle hervorgeht :

Die folgenden Beispiele erläutern die mannigfachen Möglichkeiten des neuen Verfahrens.
Beispiel 1 a) Herstellung des Ausgangsmaterials :
213 g Athylmercapto-methan-phosphonsäure-di- äthylester (Kp. 187 ) werden mit 500 ml konzentrierter Salzsäure 15 Stunden unter Rückfluss zum Sieden erhitzt. Die Lösung wird im Vakuum eingedampft. Man erhält auf diese Weise 152 g Athyl mercapto-methan-phosphonsäure. 78 g (0, 5 Mol) Athylmercapto-methan-phosphonsäure werden in 300 ml Tetrachlorkohlenstoff angeschlämmt und unter Rühren bei 35 nach und nach mit 218 g Phosphorpentachlorid versetzt. Das Reaktionsprodukt wird eine Stunde zum Sieden erwärmt und anschlie ssend fraktioniert. Man erhält auf diese Weise 73 g Athylmercapto-methan-phosphonsäure-dichlorid vom Kp.1 85¯.
Ausbeute 76% der Theorie. 386 g (2 Mole) Athylmercapto-methan-phosphonsäure-dichlorid werden mit 94 g Phosphorpentasulfid 2 Stunden unter Ruhren auf 120 erwärmt. Dann destilliert man aus dem Reaktionsprodukt unter Vakuum das gebildete ¯thylmercapto-methan-thionophosphonsÏure-dichlorid ab. Man erhält 255 g vom Kp. 1 85-88¯. Ausbeute 61 % der Theorie. b) 209 g (1 Mol) Athylmercapto-methan-thiono phosphonsäure-dichlorid werden in 1000 ml Benzol gelöst. Unter starkem Rühren gibt man bei gleichzeitiger Aussenkiihlung bei 0 eine Natriumäthylat- lösung hinzu, die 1 Mol Natrium gelöst enthält.
Das Reaktionsprodukt wird anschliessend noch so lange bei Zimmertemperatur nachgerührt, bis es neutral reagiert. Anschliessend gibt man das Reaktionsprodukt in Eiswasser und trennt die benzolische Lösung ab. Die Benzollösung wird mit Natriumsulfat getrocknet und fraktioniert. Man erhält auf diese Weise 190 g Athylmercapto-methan-thionophosphonsäure- O-äthylesterchlorid vom Kp. 192-93 . Ausbeute 87 % der Theorie. c) 23 g (0, 15 Mol) 3-Methyl-4-methylmercaptophenol werden in 150 ml Benzol gelöst. Anschlie ¯end gibt man zu dieser Lösung eine Natriumäthylat- lösung, die 0, 15 Mol Natrium gelöst enthält.
Bei 60 gibt man dann unter gutem Rühren 33 g Athyl- mercapto-methan-thiono-phosphonsäure-O-äthylester- chlorid hinzu. Man lϯt noch 2 Stunden bei 60 riihren. Anschliessend gibt man das Reaktionsprodukt in Eiswasser. Das ausgeschiedene Íl wird in Benzol aufgenommen. Die Benzollösung wird getrennt, mit Wasser gewaschen und mit Natriumsulfat getrocknet. Nach dem Abdestillieren des Lösemittels im Vakuum erhält man 49 g des neuen Esters folgender Konstitution :

Zu der Benzolsung gibt man eine Natrium-äthylatlösung, die 0, 15 Mol Natrium entspricht. Anschliessend gibt man unter weiterem gutem Rühren bei 60 33 g Athyl mercapto-methan-thionophosphonsäure-O-äthylester- chlorid hinzu. Man hält das Reaktionsprodukt noch eine Stunde bei 60 und arbeitet dann wie im vorhergehenden Beispiel auf.
Es werden auf diese Weise 39, 6 g des neuen Esters als schwachbraunes, was serunlösliches Ö1 erhalten. Aubeute 82% der Theorie.
Berechnet für Mol 322 : S 29, 8% P 9, 6 %
Gefunden : S 28, 4% P 9, 6%
Die mittlere Toxizität an der Ratte per os beträgt 50 mg/kg.
Spinnmilben werden mit 0, 01 % igen Lösungen zu 100% abgetötet. 0, 1% ige Lösungen haben eine 100 % ige systemische Wirkung bei Blattläusen.
Beispiel 3

Spinnmilben werden mit 0, 01 % igen Lösungen sicher abgetötet. Raupen werden mit 0, 1 % igen Lösungen zu 100% getötet.

Anschliessend lässt man bei 60 44 g Athyl- mercapto-methan-thionophosphonsäure-O-äthylester- chlorid zutropfen. Man erwÏrmt das Reaktionspro- dukt noch eine Stunde auf 60 und arbeitet dann wie in Beispiel 1 auf.
Man erhält auf diese Weise 61 g des neuen Esters als gelbliches, wasserunlösliches 01.
Ausbeute 93 % der Theorie.

Spinnmilben werden mit 0, 01 % igen Lösungen 100 % ig abgetötet.
Beispiel 5 Äthylmercapto-methan-thionophosphonsäure methylester-chlorid a) In die Lösung von 293 g Äthylmercapto- methan-thionophosphonsäuredichlorid (1, 4 Mol) in 1, 5 1 Benzol wird bei 0 unter Rühren eine Natriummethylatlösung eingetropft, die 1, 4 Mol Natrium gelöst enthält. Man lässt das Gemisch 3 Stunden bei Raumtemperatur nachreagieren und verrührt es dann mit 1, 5 1 Wasser. Die Benzolphase wird abgetrennt, mit Natriumsulfat getrocknet und im Vakuum vom Lösungsmittel befreit. Der Rückstand ergibt nach Vakuum-Destillation 246 g einer fast farblosen Flüs- sigkeit vom Kp. 194 . Ausbeute 86 % der Theorie.

b) 20, 9 g p-Nitrophenol (0, 15 Mol) wurden in 250 cm3 Benzol angeschlämmt und durch Zutropfen von 34, 5 g Natriummethylatlösung (= 0, 15 Mol Na) in das Salz übergeführt. Bei 60 wurden unter Rühren 30, 8 g Athylmercapto-methan-thionophosphon- säuremethylesterchlorid (0, 15 Mol) zugetropft. Das Filtrat wurde mit Bicarbonatlösung und zweimal mit Wasser gewaschen, getrocknet und im Vakuum vom Lösungsmittel befreit. Als Rückstand hinterblieben 37, 0 g eines gelblichen bles, welches beim längeren Stehen langsam durchkristallisierte. Ausbeute 80% der Theorie.

Bei 60 werden dann unter Rühren 22, 0 g des obigen Methylester-chlorids zugetropft. Nach zweistündigem Rühren bei 60 wird abgekühlt und filtriert. Das Filtrat wird mit Bicarbonatlösung und Wasser gewaschen und getrocknet.
Nach restloser Entfernung des Lösungs- mittels im Vakuum erhält man 31, 0 g des neuen Esters als gelbliches Íl (92% der Theorie).
Beispiel 13

a) 91, 5 g Äthylmercapto-methyl-phosphonsäuredichlorid werden, gelöst in 500 cm3 Petroläther, unter heftigem Rühren bei 0 tropfenweise mit einer Mischung aus 23, 0 g ¯thanol, 75, 0 g Diäthylanilin und 200 cms Petroläther versetzt. Nach der Zugabe lässt man die Mischung noch 3-4 Stunden bei Raumtemperatur rühren. Nach Filtration wird das Lösungs- mittel im Vakuum abdestilliert und der Rückstand im Vakuum destilliert. Man erhÏlt auf diese Weise 60, 4 g ¯thylmercapto - methyl-phosphonsÏure-Ïthylester-chlorid als farblose Flüssigkeit vom Kp. 1 92¯ (60% der Theorie).
b) 24, 1 g p-Chlorthiophenol-natrium werden in 100 cm3 ¯thanol gelöst und bei 25 unter Rühren tropfenweise mit 29, 5 g des obigen Äthylesterchlorids versetzt. Nach einstündigem Rühren bei Raumtemperatur wird das Salz durch Zusatz von 100 cm3 Wasser in Lösung gebracht und das Reaktionsprodukt mit 100 cm3 Chloroform ausgeschüt- telt. Die organische Phase wird gewaschen, getrocknet und im Vakuum vom Lösungsmittel befreit. Man erhält auf diese Weise 38, 5 g des neuen Esters als gelbliches Íl (85 % der Theorie).

SH-gruppenhaltigen Verbindungen zur Umsetzung gebracht werden.
Process for the preparation of phosphonic acid esters and thionophosphonic acid esters
A number of alkyl thionophosphonic acid esters are characterized by good insecticidal effects (cf., for example, German Auslegeschrift No. 1032 247). However, this good insecticidal effect is often impaired by the fact that the most effective representatives are also particularly toxic in warm-blooded animals.
The surprising observation was made that such lower alkanethionophosphonic acid esters [B. A. Arbusow and N. P. Bogonowceva, Z. obsc. Chim. 27 (1957) 2360] are largely detoxified, in which the alkyl group has been substituted by an alkyl mercapto radical.

The following examples explain the many possibilities of the new process.
Example 1 a) Production of the starting material:
213 g of ethylmercapto-methane-phosphonic acid-diethyl ester (boiling point 187) are refluxed with 500 ml of concentrated hydrochloric acid for 15 hours. The solution is evaporated in vacuo. In this way, 152 g of ethyl mercapto-methane-phosphonic acid are obtained. 78 g (0.5 mol) of ethyl mercapto-methane-phosphonic acid are suspended in 300 ml of carbon tetrachloride, and 218 g of phosphorus pentachloride are gradually added while stirring at 35. The reaction product is heated to the boil for one hour and then fractionated. In this way, 73 g of ethylmercapto-methane-phosphonic acid dichloride with a boiling point of 85¯ are obtained.
Yield 76% of theory. 386 g (2 moles) of ethyl mercapto-methane-phosphonic acid dichloride are heated to 120 with 94 g of phosphorus pentasulfide for 2 hours while stirring. The ¯thylmercapto-methane-thionophosphonic acid dichloride formed is then distilled off from the reaction product under vacuum. 255 g of bp 1 85-88¯ are obtained. Yield 61% of theory. b) 209 g (1 mol) of ethylmercapto-methane-thionophosphonic acid dichloride are dissolved in 1000 ml of benzene. A sodium ethylate solution containing 1 mol of dissolved sodium is added with vigorous stirring and simultaneous external cooling at 0.
The reaction product is then stirred at room temperature until it reacts neutrally. The reaction product is then added to ice water and the benzene solution is separated off. The benzene solution is dried with sodium sulfate and fractionated. In this way, 190 g of ethylmercapto-methane-thionophosphonic acid-O-ethyl ester chloride of bp 192-93 are obtained. Yield 87% of theory. c) 23 g (0.15 mol) of 3-methyl-4-methylmercaptophenol are dissolved in 150 ml of benzene. A sodium ethylate solution which contains 0.15 mol of sodium in dissolved form is then added to this solution.
At 60, 33 g of ethyl mercapto-methane-thiono-phosphonic acid-O-ethyl ester chloride are added with thorough stirring. The mixture is left to stir at 60 for another 2 hours. The reaction product is then added to ice water. The excreted oil is taken up in benzene. The benzene solution is separated, washed with water and dried with sodium sulfate. After the solvent has been distilled off in vacuo, 49 g of the new ester of the following constitution are obtained:

The reaction product is kept at 60 for an hour and then worked up as in the previous example.
In this way 39.6 g of the new ester are obtained as a pale brown oil which is insoluble in serum. Yield 82% of theory.

Spider mites are 100% killed with 0.01% solutions. 0.1% solutions have a 100% systemic effect on aphids.

A sodium ethylate solution containing 0.2 mol of dissolved sodium is added to the solution. Then 44 g of ethyl mercapto-methane-thionophosphonic acid-O-ethyl ester chloride are added dropwise at 60. The reaction product is heated to 60 for a further hour and then worked up as in Example 1.
In this way, 61 g of the new ester are obtained as yellowish, water-insoluble oil.
Yield 93% of theory.

Example 5 Ethylmercapto-methane-thionophosphonic acid methyl ester chloride a) In the solution of 293 g of ethyl mercapto-methane-thionophosphonic acid dichloride (1.4 mol) in 1.5 l benzene, a sodium methylate solution is added dropwise at 0 while stirring, the 1.4 mol of sodium contains dissolved.
