Verfahren zur Herstellung von Flavon-7-oxy-essigsäureamiden
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von neuen Flavon-7-oxyessigsäureamiden der Formel

in welcher R und R1 Wasserstoff, gegebenenfalls substituierte Alkyle mit 1-5 C-Atomen, substituierte oder nicht substituierte Cycloalkyle bzw. Cycloalkylalkyle, Aralkyle, substituierte oder unsubstituierte Aryle oder substituierte Pyrazolone bedeuten, oder zusammen mit dem N-Atom einen gegebenenfalls substituierten Piperidin-, Morpholin- oder Pyrrolidinring, ist dadurch gekennzeichnet, dass Flavon-7-oxy-essigsäure oder funktionelle Derivate davon mit entsprechenden Aminen zur Umsetzung gebracht werden.
Es wurde gefunden, dass diese neue Körperklasse nach an sich bekannten Arbeitsmethoden der Carbonsäureamidierung aus Flavon-7-oxy-essigsäure oder ihren funktionellen Derivaten, wie Flavon-7-oxyessigsäurehalogeniden oder entsprechenden Ester durch Umsatz mit entsprechenden Aminen oder ihren funktionellen Derivaten zugänglich ist.
Die neue Körperklasse ist durch Umsatz von Fla von-7- oxy-essigsäurealkylestern oder Flavon-7-oxyessigsäure-halogeniden mit Aminen nach an sich bekannten Arbeitsmethoden entweder direkt oder in einem geeigneten Lösungs- bzw. Verdünnungsmittel zugänglich. Als Flavon -7- oxy-essigsäurealkylester können Flavon-7-oxy-essigsäuremethyläthyl-, -propylester, allgemein Ester mit niederen Alkoholen in Betracht kommen. Um die Amidierung zu beschleunigen, können geringe, katalytisch wirkende Zusätze von Alkalimetallen beigefügt werden.
Als Flavon-7-oxy-essigsäure-halogenide sind die entsprechenden Chlor- und Bromderivate zweckmässig. Die neuen Verbindungen zeichnen sich durch ausserordentlich geringe Giftigkeit und bemerkenswerte pharmakodynamische Eigenschaften aus. Einige sind ausserdem sehr leicht in Wasser löslich.
Diese Verbindungen sind als Heilmittel bzw. Zwischenprodukte zur Herstellung von Heilmitteln verwendbar.
Die Herstellung von Flavon-7-oxy-essigsäurealkylestern sowie Flavon-7-oxy-essigs äure-alkylamino- alkanolestern ist bekannt. Diese Ester zeichnen sich durch eine gute coronardilatorische Wirkung aus. Die Wirkung dieser Ester ist allerdings kurz, weil sie durch Verseifung leicht wieder in die praktisch wenig wirksame Flavon-7-oxy-essigsäure und Alkohole gespalten werden.
Demgegenüber besitzt die erfindungsgemäss herstellbare Körperklasse im allgemeinen folgende Vorteile: a) geringe Giftigkeit, b) zum Teil noch bessere coronardilatorische Wir kung als die Ester, c) hohe Beständigkeit und Stabilität und damit lang dauernde protahierte Wirksamkeit.
Der Nachweis der grösseren Stabilität der neuen Körperklasse ist leicht zu erbringen. Man rührt je 3 g der Ester, welche nach dem britischen Patent Nr. 803 372 erhalten werden können, und derjenigen der Flavon-7-oxy-essigsäureamide, die nach dem er findungsgemässen Verfahren dargestellt wurden, in einer wässrigen oder alkoholisch-wässrigen Suspension, welche etwa 0,3 bis 1 % an Mineralsäure enthält, 30 bis 100 Minuten in einem Thermostat bei 30 bis 370 C. Die Aufarbeitung ergibt eine weitgehende Hydrolyse der Ester, während die erfindungsgemäss erhaltenen Amide quantitativ unverseift zurückerhalten werden.
Die nachfolgende Tabelle gibt in Daten die Über- legenheit und Vorteile der neuen Körperklasse an.
Als Vergleichskörper wurden Khellin und Flavon-7oxy-essigsäureäthylester getestet.

Die Bestimmung der Toxizität erfolgte durch intraperitoneale Applikation an weissen Mäusen. Die coronardilatorische Wirkung wurde nach der Methode nach Langendorff (Pflügers Arch. Bd. 61, S. 219 [1895]) in der modifizierten Form nach Morawitz und A. Jahr (Zbl. physiol. 26, S. 465 [1912] durchgeführt.
Die verlängerte und gleichbleibende Wirksamkeit der neuen erfindungsgemässen Körperklasse gegen über dem Flavon-7-oxy-essigsäure-alkylamino-alkanolester lässt sich in einem Vergleich der Wirkung an folgenden Körpern zeigen: Flavon-7-oxy-essigsäure- (N-ss-dimethylamino äthyl)-ester-hydrochlorid (Versuchskörper Fla XXX) und Flavon-7- oxy-essigsäure-(ss-diäthylaminoäthyl)- amid (Beispiel 15, Versuchspräparat Fla XX).
Beide Körper sind leicht löslich in Wasser. Sie werden zu je 4 mg/l in einer Ringer-Lookelösung gelöst und der Coronardurchfluss in der Versuchsanordnung nach Langendorff-Morawitz gemessen. In den nachfolgenden Kurven wurden die Mittelwerte aus je fünf Versuchs ansätzen gegeben. Die Lösungen wurden einmal frisch für die Versuche hergestellt, dann fünf Tage stehengelassen, und an jedem Tage wurden die Lösungen zwei Stunden im Thermostaten gerührt. Die Abszisse gibt die Zeit der Perfussion in Minuten und die Ordinate des Coronardurchflusses in ml/min. an (Siehe Kurve der Zeichnung).
Hieraus ist zu ersehen, dass die mit zu der neuen Körperklasse gehörende Versuchssubstanz Fla XX nicht allein erheblich stärker wirksam, sondern auch länger wirksam und praktisch gleichbleibend in der Wirkung ist, da sie ja nicht verseift wird.

6,2 g Flavon-7-oxy-essigs äureäthylester werden mit 6 ml wasserfreiem Diäthylamin (Überschuss) in 50 ml abs. Methanol 5-6 Stunden unter Rückfluss gekocht, dann wird Alkohol zur Hälfte eingedampft.
Die Lösung wird mit Wasser verdünnt und angesäuert, wobei sich das neue Diäthylamid sofort als farbloses Kristallmehl abscheidet; es kann aus Alkohol oder Aceton gut umkristallisiert werden. Smp.: 126/280 C.

It has been found that this new body class can be obtained from flavone-7-oxy-acetic acid or its functional derivatives, such as flavone-7-oxyacetic acid halides or corresponding esters, by reaction with corresponding amines or their functional derivatives, using known working methods of carboxy-amidation.
The new body class is accessible through conversion of Fla-7-oxy-acetic acid alkyl esters or flavone-7-oxy-acetic acid halides with amines by working methods known per se, either directly or in a suitable solvent or diluent. As flavone-7-oxy-acetic acid alkyl ester, flavone-7-oxy-acetic acid methyl ethyl or propyl ester, generally esters with lower alcohols, can be considered. In order to accelerate the amidation, small, catalytically active additives of alkali metals can be added.
The corresponding chlorine and bromine derivatives are useful as flavone-7-oxy-acetic acid halides. The new compounds are characterized by extremely low toxicity and remarkable pharmacodynamic properties. Some are also very easily soluble in water.
These compounds can be used as medicinal products or as intermediates for the production of medicinal products.
The production of flavone-7-oxy-acetic acid alkyl esters and flavone-7-oxy-acetic acid-alkylaminoalkanol esters is known. These esters are distinguished by a good coronary dilatory effect. However, the effect of these esters is short because they are easily split back into the practically ineffective flavone-7-oxy-acetic acid and alcohols by saponification.
In contrast, the body class that can be produced according to the invention generally has the following advantages: a) low toxicity, b) in some cases even better coronary dilatory effects than the esters, c) high resistance and stability and thus long-lasting protracted effectiveness.
Proof of the greater stability of the new body class is easy to provide. 3 g each of the esters, which can be obtained according to British Patent No. 803 372, and those of the flavone-7-oxy-acetic acid amides, which were prepared according to the process according to the invention, are stirred in an aqueous or alcoholic-aqueous suspension, which contains about 0.3 to 1% of mineral acid, 30 to 100 minutes in a thermostat at 30 to 370 ° C. The prolonged and constant effectiveness of the new body class according to the invention against the flavone-7-oxy-acetic acid-alkylamino-alkanol ester can be shown in a comparison of the effects on the following bodies: Flavone-7-oxy-acetic acid- (N-ß-dimethylamino ethyl ) ester hydrochloride (test specimen Fla XXX) and flavone-7-oxy-acetic acid (ß-diethylaminoethyl) amide (Example 15, test preparation Fla XX).
Both bodies are easily soluble in water. They are each dissolved at 4 mg / l in a Ringer-Look solution and the coronary flow is measured in the Langendorff-Morawitz experimental setup. In the following curves, the mean values from five test approaches were given. The solutions were freshly prepared once for the experiments, then left to stand for five days, and each day the solutions were stirred for two hours in the thermostat. The abscissa gives the time of perfusion in minutes and the ordinate of the coronary flow in ml / min. (see curve in drawing).
