Description
Verfahren zur Herstellung von neuen Derivaten von ercapto-carbonsäure
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Derivate von a-Mercaptk > - carbonsäure, welche insbesondere als Zwischenprodukte wertvoll sind, aber auch selber interessante biologische Eigenschaften besitzen.

Man stellt die Verbindungen der Formel I erfindungsgemäss her, indem man einen reaktionsfähigen Ester einer Hydroxylverbindung der Formel II

mit einem reaktionsfähigen Salz des l-Adamantanthiols umsetzt, und gegebenenfalls die erhaltene Verbindung der Formel I in die freie Säure überführt bzw. zu einem Salz hydrolysiert und gewünsch tenfalls die freie Säure bzw. das Salz in ein anderes Salz mit einer anorganischen oder organischen Base überführt.
Als reaktionsfähigen Ester von Verbindungen der Formel II seien beispielsweise entsprechende Bromide, Chloride, deren Methansulfonsäureester und p-Toluolfonsäureester genannt.
Als reaktionsfähige Salze des l-Adamantanthiols eignen sich insbesondere dessen gegebenenfalls in situ gebildete Alkalimetallsalze, das Silbersalz und die Quecksilbersalze.
Die Umsetzung wird vorzugsweise in einem geeigneten organischen Lösungsmittel wie z. B. Di äthylenglykoldimethyläther in der Wärme und zweckmässig unter Luftausschluss durchgeführt.
Die Hydrolyse erfolgt z. B. durch Erwärmen in einer alkanolischen Alkalilauge, wie auch in einem sauren Medium, z. B. wässrig-alkanolischer Salzsäure.
Das als Ausgangsstoff verwendete l-Adamantan- thiol wird z. B. durch Kochen des bekannten l-Bromadamantan mit Thioharnstoff in einem Gemisch von Eisessig und Bromwasserstoffsäure, gefolgt von der Hydrolyse des erhaltenen S-(l-Adamanthyl)-isothiur- onium-hydrobromids z. B. mittels stark verdünnter Natronlauge hergestellt. Das S-(l-Adamantyl)-iso- thiuronium-hydrobromid kann ferner z. B. auch durch Umsetzung des bekannten l-Adamantanols mit Thioharnstoff und Bromwasserstoffsäure erhalten werden.

R2 ist z. B. Wasserstoff oder einer der obengenannten niederen Alkylreste und R z. B. einer der obengenannten niederen Alkylreste, insbesondere ein Methyl- oder Äthylrest, wobei nachträglich der Rest R3 in erfindungsgemäss erhaltenen Verbindungen gegen ein Natrium-, Kalium-, Lithium-, Ammonium-, Athylammonium-, Triäthylammonium, Piperidinium-, Athanolammonium-, Diäthanolammonium- oder N,N Diäthyläthanolammonium-ion bzw. das Normaläquivalent eines Magnesium-, Calcium- oder Äthylendiammonium-ions ausgetauscht werden kann.
Die erfindungsgemäss herstellbaren Verbindungen der Formel I eignen sich als Zwischenprodukte zur Herstellung von Arzneimitteln, wie z. B. Estern und Amiden. Von den biologischen Eigenschaften, insbesondere der aus erfindungsgemäss erhaltenen Verbindungen der Formel I herstellbaren freien Säuren und der Salze, sei deren choleretische Wirksamkeit erwähnt.
In den nachfolgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1 a) 107 g (0,5 Mol) l-Brom-adamantan werden mit 76 g (1 Mol) Thioharnstoff und 250 ml 48 % iger Bromwasserstoffsäure in 500 ml Eisessig 3 Stunden unter Rückfluss gekocht. Dann wird das Reaktionsgemisch noch heiss auf etwa 1800 g Eis gegossen.
Das ausgeschiedene S-(l-Adamantyl)-isothiuronium- bromid wird abgenutscht und mit Wasser und dann mit Methylenchlorid gewaschen. Hierauf wird es mit 40 g Natriumhydroxyd in 1000 Teilen Wasser und 250ml Äthanol etwa 14 Stunden bei Raumtemperatur geschüttelt. Das Reaktionsprodukt wird abgenutscht, in Benzol gelöst, die Benzollösung mit Wasser gewaschen und eingedampft. Das als öl zurückbleibende l-Adamantanthiol kristallisiert beim Anreiben mit Pentan. Es kann durch Sublimation, z.
B. bei 1100 unter 12 mm Druck gereinigt werden und schmilzt dann bei 100-1020. b) Zu einer Lösung von 16,8 g (0,1 Mol) l-Adamantanthiol in 250 mi Diäthylenglykoldimethyl- äther wird bei einer Badtemperatur von 1500 unter Rühren in Stickstoffatmosphäre eine Suspension von 6 g (0,15 Mol) Natriumamid in Toluol (Gesamtvolumen 18 ml) zulaufen gelassen und das Gemisch drei Stunden unter kräftigem Rückfluss gekocht. Hierauf lässt man bei gleicher Temperatur 34 g = 23 ml (0,2 Mol) o-Bromessigsäureäthylester innerhalb 30 Minuten zutropfen und kocht noch etwa 14 Stunden weiter. Nach dem Abkühlen giesst man das Reaktionsgemisch auf etwa 1000 g Eis-Wasser-Gemisch und extrahiert mit Toluol.
Die Toluollösung wird mehrmals mit Wasser gewaschen und dann das Toluol im Rotationsverdampfer unter vermindertem Druck abgedampft.
Der als Öl zurückbleibende a.-(Adamant-l-ylthio)- essigsäure-äthylester wird gewünschtenfalls durch Destillation im Hochvakuum gereinigt. Unter 0,001 mm Druck geht er bel 117-1180 über. c) Zur Gewinnung der freien Säure kocht man den oben als Eindampfrückstand erhaltenen rohen Ester mit 20 g Natriumhydroxyd in 400 ml Athanol 5 Stunden unter Rückfluss. Das Äthanol wird abdestilliert, der Rückstand mit etwa 750 ml Wasser versetzt und die wässrige Phase durch Hyflo filtriert.
Das Filtrat wird mit Salzsäure angesäuert, das ausgeschiedene Öl in Benzol-Äther aufgenommen, die Lösung eingedampft, der Rückstand in 1000 ml 0,2n. Natronlauge gelöst, die Lösung mit Kohle geklärt und durch Hyflo filtriert. Beim Ansäuern mit Salzsäure fällt die a-(Adamant-l-ylthio)-essigsäure zunächst ölig aus, wird aber bald fest. Sie wird abgenutscht, mit viel Wasser neutral gewaschen und dann in Benzol gelöst. Die Benzollösung wird über Natriumsulfat getrocknet. Nach Abdampfen des Benzols bleibt die a-(Adamant-l-ylthio)-essigsäure als Öl zurück und kann z. B. aus HexanlPentan kristallisiert werden. Smp. 68-700.
Beispiel 2
Analog Beispiel lb erhält man unter Verwendung von 45 g (0,2 Mol) a - Bromcapronsäureäthylester den rohen a-(Adamant - 1 - ylthio)-capronsäureäthyl- ester. Dieser wird mit 20 g Natriumhydroxyd in 400 ml Äthanol 6 Stunden unter Rückfluss gekocht und das Äthanol abdestilliert. Der Rückstand wird in 250 ml Wasser gelöst und mittels Filtration durch Hyflo geklärt. Das Filtrat wird mit Salzsäure angesäuert, das ausgeschiedene Ö1 in Toluol aufgenommen und die Toluollösung mit Wasser gewaschen und eingedampft. Der Rückstand wird in Cyclohexan gelöst und diese Lösung mehrmals mit gesättigter Kaliumbicarbonatlösung gut durchgeschüttelt, bis kein Kohlendioxyd mehr freigesetzt wird.
Hierauf wird die Cyclohexanlösung mit Natriumcarbonatlösung extrahiert, die erhaltene Lösung nach Zusatz von Aktivkohle durch Hyflo filtriert, das Filtrat mib Salzsäure angesäuert und das ausgefallene Öl in Sther-Hexan aufgenommen.
Die Äther-Hexan-Lösung wird eingedampft und der Rückstand aus 80% dem Methanol unter längerem Stehenlassen bei-5 umkristallisiert. Die Kristalle von a-(Adamant-l-ylthio)-capronsäure werden mit 50 ml 80% igem Methanol von -100 angeteigt, abgenutscht und im Hochvakuum bei 550 getrocknet.
Smp. 74-760.
Beispiel 3
36 g (0,2 Mol) a-Brom-propionsäure-äthylester werden analog Beispiel lb) in den rohen a-(AdamantW l-ylthio)-propionsäure-äthylester übergeführt und dieser mit 16 g Natriumhydroxyd in 250 ml Äthanol etwa 14 Stunden unter Rückfluss gekocht. Der beim Abdampfen des Äthanols hinterbleibende feste Rückstand wird in 250 ml Wasser aufgenommen, die wässrige Lösung zur Entfernung von Neutralteilen mit Methylenchlorid extrahiert, mit Aktivkohle geklärt, filtriert und hierauf mit konz. Salzsäure angesäuert.
Die ausgeschiedene a-(Adamant - 1 - ylthio) - propion- säure wird abgenutscht, aus Cyclohexan und hierauf aus Benzin umkristallisierb und hierauf während 14 Stunden bei 500 im Hochvakuum getrocknet.
Smp. 142-1440.
Beispiel 4
Ausgehend von 39 g = 29,5 ml (0,2 Mol) a-Brombuttersäure-äthylester erhält man analog Beispiel 1b den rohen a- (Adamant-1-ylthio)-buttersäuremäthyl- ester. Dieser wird mit 16 g Natriumhydroxyd in 250 ml Äthanol 5 Stunden unter Rückfluss gekocht, das Athanol abdestilliert, der Rückstand in 500 ml Wasser gelöst, die Lösung mit Aktivkohle geklärt, durch Hyflo filtriert. Das Filtrat wird mit konz. Salzsäure angesäuert und hierauf mit Benzol extrahiert.
Die Benzollösung wird dreimal mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Die zurückbleibende a-(Adamant-l-ylthio)buttersäure wird aus etwa 300 ml Benzin umkristallisiert und bei 12 mm Druck getrocknet. Smp.
113-1140.
Beispiel 5
Analog Beispiel 1b wird unter Verwendung von 42 g (0,2 Mol) a-Brom-isovaleriansäure-äthylester der rohe a-(Adamant-l-ylthio)-isovaleriansäureäthyl- ester hergestellt und mit 20 g Natriumhydroxyd in 300 ml Äthanol 5 Stunden unter Rückfluss gekocht.
Das Athanol wird abedstilliert, der Rückstand in
1000 ml Wasser gelöst, die Lösung mit Aktivkohle geklärt, durch Hyflo filtriert und hierauf mit konz.
Salzsäure angesäuert. Die a < AdamanGl-ylthio)-iso- valeriansäure fällt anfangs schmierig aus, erstarrt aber bald. Sie wird abfiltriert, zerrieben, mit Wasser gewaschen und hierauf in Benzol gelöst. Die Benzollösung wird über Natriumsulfat getrocknet und eingedampft und der Rückstand unter Entfärbung mit Aktivkohle aus 250 ml Benzin und hierauf aus 300 ml Hexan umkristallisiert und 2 Stunden im Hochvakuum bei 900 getrocknet. Smp. 134-1350.
In analoger Weise erhält man unter Verwendung von 39 g (0,2 Mol) a-Brom-isobuttersäure-äthylester die a-(Adamant)- 1 -ylthio)-isobutters äure vom Smp.
141-1420 (aus Hexan). Ebenfalls in analoger Weise erhält man unter Verwendung von a-Brom-ss, ss- dimethylbuttersäure-äthylester die a - (Adamant - 1 - ylthio)-ss, ssdimethylbuttersäure vom Smp. 207-208 (aus Benzin).
Beispiel 6
Der analog Beispiel 1b unter Verwendung von 40 g (0,17 Mol) cr-Brom-cr-cyclopentyl-essigsäure- äthylester erhaltene rohe a- (Adamant-1-ylthio)- a- cyclopentyl-essigsäure-äthylester wird mit 20 g Natriumhydroxyd in 400 i Äthanol 5 Stunden unter Rückfluss gekocht. Das Äthanol wird abdestilliert und der Rückstand in 1000 ml Wasser gelöst. Die Lösung wird durch Hyflo filtriert, mit Kohlendioxyd bis zum pH 8 abgestumpft und unter Zusatz von Aktivkohle erneut filtriert. Das Filtrat wird mit konzentrierter Salzsäure bis pH 6 angesäuert, die ausgeschiedene Substanz in Benzol-Äther aufgenommen und die erhaltene Lösung dreimal mit je 100 ml Kaliumcarbonatlösung ausgeschüttelt.
Die Benzol Äther-Lösung wird eingedampft und der Rückstand unter Entfärbung mit Kohle zweimal aus etwa 250 ml Hexan umkristallisiert. Die so erhaltene a-(Adamans 1-ylthio) - a - cyclopentyl-essigsäure schmilzt bei 136 bis 1390.
Beispiel 7
34 g (0,2 Mol) l-Adamantanthiol in 500 ml warmen 95 %igem Äthanol werden zu 38 g (0,1 Mol) Bleizucker (Blei-II-acetatQrihydrat) in 500 ml des gleichen Lösungsmittels gegeben, wobei das intensiv gelbe Bleisalz ausfällt. Nach Abkühlen auf 0 wird filtriert und mit Äthanol gewaschen. Nach dem Trocknen des Niederschlages bei 500 im Hochvakuum beträgt die Ausbeute an Bleisalz 52 g.
34 g (0,063 Mol) dieses Salzes und 38 g (0,156 Mol) a - Brom - phenylessigshureäthylester werden in 750 ml absolutem Dioxan 60 Stunden unter Rückfluss erhitzt, gekühlt und filtriert. Das Filtrat wird ein gedampft, der dabei entstehende Rückstand in Hexan aufgenommen, durch Filtration von unlöslichem Schlamm abgetrennt, eingeengt und im Hochvakuum von iiberschüssigem Bromester befreit.
Der Destillationsrückstand (22 g) wird mit 100 ml Äthanol und 5 g Natriumhydroxyd 16 Stunden unter Rückfluss erhitzt. Nach Abdampfen des Äthanols wird in 500 ml Wasser gelöst, durch Hyflo filtriert und mit HCl angesäuert. Nach Extraktion des schmie rigen Niederschlages mit Ätlher/Benzol, Eindampfen und Extraktion des Rückstandes mit verdünnter Natriumbicarbonatlösung, Klärung mit Kohle und Ansäuern erhält man nach dem Umkristallisieren aus Cyclohexan und Hexan aAdamant- 1 -ylthio)-a-phe- nylessigsäure. Smp. 122-124 .
In analoger Weise erhält man unter Verwendung von Bromessigsäureäthylester die a - (Adamant- 1 - ylthio)-essigsäure. Smp. 68-700.
Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung von neuen Derivaten von a-Mercapto-carbonsäuren der Formel I EMI4.1 worin R1 Wasserstoff, einen aliphatischen, cycloali phatischen, aromatischen oder araliphatischen Rest, R2 Wasserstoff oder einen niederen Alkylrest, und R3 einen niederen Alkylrest bedeuten, dadurch gekennzeichnet, dass man einen reaktionsfähigen Ester einer Hydroxylverbindung der Formel II EMI4.2 mit einem reaktionsfähigen Salz des l-Adamantan- thiols umsetzt.UNTERANSPRÜCHE 1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man erhaltene Verbindungen der Formel I in die freie Säure überführt.2. Verfahren nach Patentanspruch und Unteranspruch 1, dadurch gekennzeichnet, dass man die erhaltene freie Säure der Formel I in ein Salz mit einer anorganischen oder organischen Base überführt.3. Verfahren nach Patentanspruch und den Unteransprüchen 1 und 2, dadurch gekennzeichnet, dass man ein erhaltenes Salz der Verbindung der Formel I in ein anderes mit einer anorganischen oder organischen Base überführt.
