worin R6 Methoxy oder Hydroxy bedeutet, kann, falls erwünscht, in ein Säuresalz übergeführt werden.
Die niederen Alkylreste R2, R3 und R5 in obigen Formeln stellen z. B. solche mit 1-4 Kohlenstoffatomen dar, wie die Methyl-, Athyl-, Propyl-, Isopropyl-oder Butylgruppe. Athylen und Isopropylen sind Beispiele eines divalenten Kohlenwasserstoff- radikals X. Besonders interessante Verbindungen werden erhalten, wenn in den substituierten Isochinolinen I R2 MethyI, R3 Wasserstoff, R4 je Methoxy in den Stellungen 6 und 7 und X Äthylen bedeutet.
Die Ausgangsverbindungen der allgemeinen Formel I sind zum Teil bekannt und können z. B. dadurch hergestellt werden, dal3 man ein im aromatischen Ring entsprechend substituiertes ss-Phenäthyl- amin, welches in a-StelIungeine Alkylgruppe tragen kann, mit einer entsprechenden, gegebenenfalls in a-und bzw. oder, 8-Stellung alkylierten Phenylpropionsäure kondensiert, das gebildete Säureamid zum entsprechenden l-Phenylalkyl-3, 4-dihydro-isochino- lin-Derivat cyclisiert, letzteres reduziert und das entstandene substituierte Tetrahydroisochinolin am Stickstoffatom alkyliert.
l-Phenäthyl-2-methyl-6, 7-dimethoxy-1, 2, 3, 4- tetrahydro-isochinoline, deren Phenylgruppe durch 1 oder 2 Halogenatome substituiert ist, wie l- (4-Ch ! or-phenäthyl)- oder 1-(3, SDichIor-phenäthyl)-2-methyl-6, 7 dimethoxy-l, 2, 3, 4-tetrahydro-dsochinolin, stellen bevorzugte Ausgangsmaterialien dar.
In der ersten Stufe des erfindungsgemässen Verfahrens werden als Quaternisierungsmittel z. B. niedere Alkylhalogenide, wie Methyljodid, Athylbromid, Propyljodid oder Dialkylsulfate, wie Dimethylsulfat oder Diäthylsulfat, verwendet. Die Quaternisierung mit Methylgruppen liefernden Aktivierungsmitteln ist bevorzugt. Die Quaternisierung wird zweckmässiger- weise in einem geeigneten organischen Lösungsmittel, wie Aceton oder Essigester, durchgeführt. Eine geeignete Ausführungsform besteht z. B. darin, dass man die Ausgangsverbindungen der allgemeinen Formel I mit einer äquivalenten Menge des Alkylierungsmittels mehrere Stunden bei Raumtemperatur stehenlässt.
Die quatemären Salze fallen dabei in der Regel aus und stellen meist kristalline Verbindungen dar.
Anstelle der so gewonnenen quatemären Salze können auch die daraus freigesetzten quaternären Basen durch Erhitzen in die Verbindungen der Formel II übergeführt werden. Die quaternÏren Basen können aus den quaternären Salzen durch Behandlung mit Silberoxyd, vorzugsweise in alkoholisch- wässriger Lösung, erhalten werden. Das nach Abfiltrieren der unlöslichen Silbersalze und Einengen des Lösungsmittelgemisches erhaltene, meist ölige quaternäre Hydroxyd wird anschliessend der Pyrolyse unterworfen, wodurch unter Ringöffnung die Verbindungen der allgemeinen Formel II gebildet werden. Als Pyrolysetemperatur wird zweckmässigerweise eine Temperatur angewandt, welche zwischen 100 und 200 liegt.
Bei einer Temperatur von 150 bis 170 hat sich die Pyrolyse als besonders gut durchführbar erwiesen. Die direkte tYberführung der quatemären Salze in deren Abbauprodukte der Formel II durch Erhitzen der ersteren kann in : alkalisch-wϯrigem Milieu, z. B. durch mehrstün- diges Kochen in 10 % iger wässriger Natronlauge unter Rückfluss, durchgeführt werden.
Für die Hydrierung der Kohlenstoff-Doppelbin- dung in der Seitenkette der Abbauprodukte der Formel II kann man verschiedene Reduktionsmethoden anwenden, von denen jedoch die katalytische Hy- drierung in Gegenwart eines Metallkatalysators, wie z. B. Raney-Nickel, Platinoxyd, Palladiumkohle usw., besonders geeignet ist. Sowohl die freie Base wie auch geeignete Säureadditionssalze, z. B. deren Hydrohalogenide, können hydriert werden, wobei man zweckmässig in einem geeigneten Lösungsmittel, wie einem Alkohol, z. B. Methanol oder Athanol, oder wässrigem Alkohol, Essigester oder Wasser, arbeitet. Die Hydrierung findet schon bei Raumtemperatur und Normaldruck statt.
Die Hydrierung von Verbindungen der Formel II, worin R eine Benzyloxygruppe darstellt, führt unter Debenzylie- rung zu Produkten der Formel III, worin Rs eine Hydroxygruppe bedeutet. Diese gleichzeitige Debenzylierung geschieht in der Regel in Gegenwart geeigneter Katalysatoren, z. B. Palladium oder palladiumhaltiger Katalysatoren, wie Palladiummoor, Palladiumkohle usw.
Die erfindungsgemäss erhältlichen tertiären Amine der Formel III stellen in den meisten Fällen farblose bis leicht gelbgefärbte Ole dar, die im Hochvakuum unzersetzt destilliert werden können. Sie können durch Zugabe einer anorganischen Säure, wie z. B. Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure usw., oder einer organischen Säure, wie z. B. Fumarsäure, Weinsäure, Zitronen- säure, Methansulfonsäure usw. in deren Salze über- geführt werden.
Die Verfahrensprodukte besitzen zum mindesten ein asymmetrisches Kohlenstoffatom. Dieses entspricht dem Kohlenstoff-Ringatom Ci des Isochinolinkernes der Ausgangsverbindungen I. Falls X eine verzweigte Kohlenstoffkette bedeutet, können weitere Asymmetriezentren entstehen. Verwendet man als Ausgangssubstanz eine rac. Verbindung, erhält man ein rac. Endprodukt. Oft ist es wünschenswert, die Endprodukte in Form ihrer optischen Antipoden herzustellen. Dies kann z. B. dadurch geschehen, dass man optisch aktive Ausgangsmaterialien verwendet, oder dal3 man unter Verwendung von rac.
Ausgangsmaterial eine Auftrennung in die optischen Antipoden, zu beliebigem Zeitpunkt nach erfolgtem Hofmannschem Abbau vornimmt. Eine solche Auftrennung kann nach an sich bekannten Methoden erfolgen, z. B. durch fraktionierte Kristallisation der Salze mit einer optisch aktiven Säure, wie D-Weinsäure, Dibenzoyl-D-Weinsäure oder D-Campher- sulfonsäure.
Die Verfahrensprodukte sowie deren Salze besitzen analgetische, spasmolytische, hustenlindernde bzw. blutdrucksenkende Wirkungen. Sie können als Heilmittel in Form pharmazeutischer Präparate Verwendung finden, welche sie oder ihre Salze in Mi schung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen inerten Trägermaterial enthalten. Die pharmazeutischen Präparate können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Beispiel I
10 g 1- (4-Chlorphenäthyl)-2-methyl-6, 7-di methoxy-1, 2, 3, 4-tetrahydro-isochinolin werden in
150 ml Aceton unter Erwärmen gelöst und mit 4, 1 g Methyljodid versetzt. Nach dem Stehen über Nacht wird vom sich gebildeten Niederschlag abfiltriert und derselbe aus Methanol-Äther umgelöst. Man erhält 12, 4 g des Methojodides vom Schmelzpunkt 196-198 . UV-Absorptionsmaxima (in Äthanol) bei 280 und 284 m?; ? = 3680 und 3710.
12, 4 g 1-(4-ChlorphenÏthyl)-2,2-dimethyl-6, 7-dimethoxy-1, 2, 3, 4-tetrahydro-isochinoliniumjodid werden in 150 ml Methanol gelöst. Zu der Lösung gibt man 15 ml Wasser und 13 g Silberoxyd und schüttelt die Mischung während 4 Stunden bei Raum- temperatur. Man filtriert, engt das Filtrat im Wasserstrahlvakuum ein und erhitzt den öligen Rückstand während 30 Minuten auf 150-170 Badtemperatur. Nach dem Erkalten nimmt man in Ather auf und extrahiert die basischen Anteile mit 2n Schwefelsäure. Durch Zusatz von verdünnter Na tronlauge bis zur alkalischen Reaktion des Schwefel- säureextraktes werden die basischen Anteile ausge- fällt und in Äther aufgenommen.
Der nach dem Trocknen und Einengen der ätherischen Lösung erhaltene Rückstand liefert nach der Destillation im Hochvakuum 8, 8 g
1-(2-Vinyl-4, 5-dimethoxy-phenyl)-1- dimethylamino-3- (4-chlor-phenyl)-propan in Form eines hellgelben ) les vom Siedepunkt 163 bis 165¯/0, 015 mm. UV-Absorptionsmaxima bei 260 und 292 m, u; ? = 13 760 und 3870 (in Athanol).
Das durch Umsatz mit Methyljodid erhältliche Methojodid zeigt einen Schmelzpunkt von 138 bis 139 . Das D-Camphersulfonat der rac. tertiÏren Base schmilzt bei 202-203 .
Zwecks Reduktion der Vinylgruppe werden 2, 0 g der obigen tertiären Base in 50 ml Methanol gelöst und über 40 mg Platinoxyd hydriert. Nach dem Abfiltrieren des Katalysators wird das Filtrat eingeengt und der Rückstand im Hochvakuum destil liert. Man erhält 1, 8 g
1-(2-Athyl-4, 5-dimethoxy-phenyl)-1-dimethyl- amino-3- (4-chlor-phenyl)-propan vom Siedepunkt 155-157¯/0, 015 mm. W-Absorp- tionsmaximum bei 284 mu ; e = 3810 (in Athanol).
Das in Acetonlösung bereitete D-Camphersulfonat der rac. Base schmilzt bei 208-209 .
Beispiel 2
Wird (+)-1-(4-Chlor-phenÏthyl)-2-methyl-6, 7dimethoxy-1, 2, 3, 4-tetrahydro-isochinolin [Siede- punkt 196-198 /0, 06 mm ; [a] 25D = 17,9¯ (c = 1, 0 in Methanol) ; hergestellt aus dem Racemat nach den Angaben in Helv. Chim. Acta 43, 1472 (1960)] den in Beispiel I beschriebenen Reaktionsfolgen unterworfen, so erhält man folgende Produkte (+)-1- (4-Chlor-phenäthyl)-2, 2-dimethyl
6, 7-dimethoxy-1, 2, 3, 4-tetrahydro isochinoliniumjodid vom Schmelzpunkt 98-100 (aus Wasser) ; [a] D = + 16 (c = 1 in Methanol).
(-)-l-(2-Äthyl-4,5-dimethoxy-phenyI)-l- dimethylamino-3- (4-chlor-phenyl)-propan, welches bei einer Badtemperatur von 200 /0, 02 mm überdestilliert. [a] 22D =-12, 4 (c = 1, 4inMethanol).
Das in Acetonlösung bereitete (+)-Camphersu] fonat schmilzt bei 164-165 ; [a] r3 = + 16, 4 (c = 1 in Methanol).
Beispiel 3
Wird 1-(3, 4-Dichlor-phenäthyl)-2-methyl-6, 7-dimethoxy-1, 2, 3, 4-tetrahydro-isochinolin den in Beispiel 1 beschriebenen Reaktionsfolgen unterworfen, so erhält man folgende Produkte : 1- (3, 4-Dichlor-phenäthyl)-2, 2-dimathyl-6, 7 dimethoxy-1, 2, 3, 4-tetrahydro-isochinolinium- jodidvom Schmelzpunkt 180-181¯ (aus Methanol/¯ther).
UV-Absorptionsmaximum bei 282 m, u; ? = 4010 (in Athanol). l-(2-VinyI-4,5-dimethoxy-phcnyl)-l-dimethyl- amino-3- (3, 4-dichlor-phenyl)-propan vom Siedepunktl89-191 /0, lmm. W-Absorptions- maxima bei 262 und 295 m?; ? = 13 100 und 3810 (m ¯thanol).
1- (2-Athyl-4, 5-dimethoxy-phenyl)-1-dimethyl amino-3- (3, 4-dichlor-phenyl)-propan vom Siedepunkt 190-192 /0, 07 mm. UV-Absorp- tionsmaxima bei 282 und 288 m, u (Schulter) ; s = 3890 und 3180 (in Äthanol). Das D-Camphersulfonat der rac. Base schmilzt bei 207-208 .
Beispiel 4
Wird (+)-1- (3, 4-Dichlor-phenathyl)-2-metliyl- 6, 7-dimethoxy-1, 2, 3, 4-tetrahydro-isochinolin [Schmelzpunkt 55-56 ; [a] 26 = + 11, 0 (c = 1, 0 in Methanol] der in Beispiel 1 beschriebenen Reak- tionsfolgen unterworfen, so erhält man folgende Produkte
1-(2-VinylA, 5-dimethoxy-phenyl)-1-dimethyl- amino-3-(3, 4-dichlor-phenyl)-propan vom Siedepunkt 175-179 /0, 01 mm ; [a] 24D = -12,5 (c = 1,0 in Methanol).
(-)-1-(2-¯thyl-4,5-dimethoxy-phenyl)-1 dimethylamino-3- (3, 4-dj. chlor-phenyl)-propan vom Siedepunkt 176-178 /0, 01 mm ; [a] =-13, 3 (c = 1, 0 in Methanol).
Beispiel S
Wird 1- (4-Chlor-phenäthyl)-2, 3-dimethyl-6, 7-di- methoxy-1, 2, 3, 4-tetrahydro-isochinolin den in Beispiel 1 beschriebenen Reaktionsfolgen unterworfen, so erhält man
1-(2-n-Propyl-4,5-dimethoxy-phenyl)-1 dimethylamino-3- (4-chlor-phenyl)-propan vom Siedepunkt 155-160 /0, 01 mm ; W-Absorp- tionsmaxima (in Athanol) bei 278 und 283 m?; je 3390.
Beispiel 6
Wird 1 - PhenÏthyl-2-methyl-6, 7-dimethoxy1, 2, 3, 4-tetrahydro-isochinolin den in Beispiel 1 be schriebenen Reaktibnsfolgen unterworfen, so erhält man folgende Produkte : l-PhenäthyI-2, 2-dimethyl-6, 7-dimethoxy-

Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung von tertiären Aminen der Formel EMI4.1 worin Ri Fluor, Chlor oder Brom, R2 und R5 niederes Alkyl und R3 Wasserstoff oder niederes Alkyl, R6 Methoxy oder Hydroxy, m den Wert 0, 1, 2 oder 3 und n den Wert 1, 2 oder 3 bedeuten und X ein divalentes Kohlenwasserstoffradikal mit zwei oder mehr Kohlenstoffatomen darstellt, dessen Valenzen durch 2 Kohlenstoffatome voneinander getrennt sind, dadurch gekennzeichnet, dass man eine Isochinolinverbindung der Formel EMI4.2 worin R4 Methoxy oder Benzyloxy bedeutet, mit einem einen niederen Alkylrest abgebenden Alkylierungsmittel umsetzt, das gewonnene quaternäre Salz bzw.die aus diesem, freigesetzte quaternäre Base erhitzt und die erhaltene Verbindung der Formel EMI4.3 unter gleichzeitiger ¯berf hrung einer allfällig vorhandenen Benzyloxygruppe R. in die Hydroxygruppe, hydriert.UNTERANSPRUCHE 1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man eine erhaltene Base in ein Sauresalz iiberfiihrt.2. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass racemisches oder optisch aktives Ausgangsmaterial verwendet wird.3. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man racemisches Ausgangsmaterial verwendet und die Auftrennung in die optischen Antipoden zu einem beliebigen Zeitpunkt nach erfolgter ¯berf hrung des quaternären Salzes bzw. der quaternären Base in die Verbindung der Formel II vornimmt.4. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man racemisches oder (+)-1-(4-Chlor-phenÏthyl)-2-methyl-6, 7 dimethoxy-1, 2, 3, 4-tetrahydro-isochinolin oder racemisches oder (+)-1- (3, 4-Dichlor-phenäthyl)-2-methyl-6, 7 dimethoxy-1, 2, 3, 4-tetrahydro-isochinolin mit einem Methylierungsmittel quaternisiert.
