EMI1.14
<Desc/Clms Page number 2>
Derartige Verbindungen (IV) erhält man z.
B. durch Überführung von Lactamen der Formel:
EMI2.8

Process for the preparation of 6-basic substituted morphanthridines The invention relates to a process for the preparation of 6-basic substituted morphanthridines of the formula:
EMI1.1
as well as acid addition salts thereof. In formula I, R denotes an alkylene group with preferably at most 5 carbon atoms, R1 is hydrogen or alkyl, and R2 and R3 are identical or different and represent hydrogen, alkyl or together an ethylene group, in the latter case R1 in addition to the meanings mentioned can also have the meaning of hydroxyalkyl or alkoxyalkyl. R4 and R5 are identical or different and are hydrogen or halogen. The basic substituent in the 6-position is preferably a 4-methyl-1-piperazinyl group.
Any substituents in the benzene nuclei are preferably in the 3- or 8-position.
The compounds (I) mentioned are obtained according to the invention if a nitrilium or ammonium cation of the formula:
EMI1.8
wherein R4 and R5 have the meaning given above, containing reaction mixture with an amine of the formula:
EMI1.14
<Desc / Clms Page number 2>
wherein R, R1, R2 and R3 have the meaning given, is converted.
The nitrilium or imonium cations of the formula II can be used as dissociation products of compounds of the formula:
EMI2.1
be understood in which R4 and R5 have the meaning given above and X is a halogen atom, the sulfhydryl group or an optionally activated alkoxy or alkylthio group, eg. B. represents a p-nitrobenzyl group. Such compounds (IV) are obtained, for.
B. by converting lactams of the formula:
EMI2.8
wherein R4 and R5 have the meanings given above, into the thiolactams, if desired with subsequent alkylation of the latter, or by reacting the lactams (V) with a halogenating agent such as phosphorus oxychloride or phosphorus pentachloride, preferably in the presence of catalytic amounts of dimethylaniline or Dimethylformamide. The lactams (V) are in turn z. B. obtainable by ring closure of corresponding o-isocyanatodiphenylmethane with aluminum chloride.
Depending on the chemical nature of the radical X and any substituents R4 and R5, the compounds IV in the reaction mixtures obtained are more or less strongly dissociated into the nitrilium or imonium cations, so that the reaction mixtures can be used directly for the reaction with the amine Formula III can be used.
Some of the compounds of the formula IV prepared in this way or in another way can be isolated in undissociated form and then give when dissolving in a suitable, preferably polar solvent, optionally with heating and in the presence of the amine of the formula III, which also. can serve as a solvent, the desired nitrifium or imonium cations (II). Reaction mixtures containing cations of the formula II can also be used, for.
B. by intramolecular Ritter reaction (attack of a nitrile group on a phenyl cation) in o-cyanodiphenylmethanes, by Beckmann rearrangement of optionally suitably substituted anthrone oxime or by Schmidt reaction of optionally suitably substituted anthrone with hydrazoic acid. However, if unsymmetrically substituted anthrone oxime or anthrone is used as the starting point, the last two reactions mentioned lead to isomer mixtures which, if necessary, have to be separated subsequently.
Anions of sulfuric acid, toluenesulfonic acid, phosphoric acid, hydrofluoric acid, hydrofluoric acid etc.
The bases obtained in the manner described are in most cases crystallizable, otherwise they can be distilled without decomposition in a high vacuum and form with inorganic and organic acids, for example hydrochloric acid, hydrobromic acid, sulfuric acid, nitric acid, phosphoric acid, acetic acid, oxalic acid, tartaric acid, toluenesulfonic acid and the like in water stable addition salts, in which form the products can also be used.
The bases obtained in the manner described and their acid addition salts are new compounds which are used as active ingredients in medicaments or as intermediates for the preparation of such use. In particular, the products come into consideration as neuroplegics, neuroleptics and analgesics. Some of them are suitable for the treatment of psychotic states. This efficacy manifests itself pharmacologically in a strong reduction in motility in mice, which can be associated with a catalepatic effect. The motility damping is determined by measuring the running activity according to the method of Caviezel and Baillod [Pharm. Acta Helv. 33, 469 (1958)].
The running activity values of some products according to the invention and their toxicity are compared in Table I below with the corresponding numbers for chlorpromazine.

Example 1 A mixture of 4.9 g of 5,6-dihydro-6-oxo-morphanthridine, 37 ml of phosphorus oxychloride and 1.5 ml of dimethyl aniline is heated to reflux for 3 hours.
The viscous oil obtained by evaporating the reaction mixture in vacuo at 60 ° C. is diluted with 20 ml of absolute dioxane and, after adding 30 ml of N-methylpiperazine, heated to reflux for 4 hours. The clear solution obtained is concentrated to dryness at 60 ° C. in vacuo. The residue is distributed between ether and ammonia water. The ethereal solution is separated, with water
<Desc / Clms Page number 3>
washed water and then extracted with 1N acetic acid.
The acetic acid extract is mixed with ammonia water and then extracted with ether. The ethereal solution is washed with water, dried over sodium sulfate, filtered through alumina and evaporated. The residue is crystallized from ether / petroleum ether and recrystallized from acetone / petroleum ether. 6.0 g (88% of theory) 6- (4-methyl-1-piperazinyl) -morphanthridine with a melting point of 138 to 138.5 ° C. are obtained.
The 5,6-dihydro-6-oxo-morphanthridine used as starting material is expediently lightened in the following way: 30.2 g of o-amimodiphenylmethane are dissolved in 65 ml of absolute toluene and 140 ml of a 20 is left at 0 to -10 ° C. with stirring Add dropwise% phosgene solution in toluene. The milky mixture is heated to reflux temperature over a period of 30 minutes while phosgene is slowly passed through, which is maintained for about 20 minutes. Dry nitrogen is passed into the boiling reaction mixture over a period of 10 minutes with vigorous stirring. After evaporation of the solvent, 29.7 g (86% of theory) of o-isocyanatodiphenylmethain with a boiling point of 169 ° C./12 Torr are obtained by vacuum distillation.
21.1 g of aluminum chloride are heated in 110 ml of o-dichlorobenzene to 80 ° C. and a solution of 29.7 g of o-isocyanatodiphenylmethane in 60 ml of o-dichlorobenzene is added dropwise with stirring, the mixture heating to 120 ° C. This temperature is maintained for 1 hour with stirring. After cooling, the reaction mixture is poured into 200 ml of 2N hydrochloric acid, a brown precipitate being formed. After steam distillation, the residue is isolated by filtration and crystallized from acetone / water. 28.6 g (97% of theory) of 5,6-dihydro-6-oxo-morphanthridine with a melting point of 201-203 C. are obtained. Example 2 33.5 g of 5,6-dihydro-6-oxo-morphanthridine are mixed with 10 Poured ml of N, N-dimethylaniline and, after adding 300 ml of phosphorus oxychloride, refluxed for 4 hours.
The reaction mixture is evaporated in a vacuum. The residue is suspended in absolute xylene. The residue obtained by evaporating this suspension in vacuo is taken up in ether and poured onto ice / water. The ethereal phase is separated off and washed three times with dilute hydrochloric acid, the Balisaurs wash water being backwashed with ether. The combined ether phases are washed successively with water, sodium bicarbonate solution, water and saturated sodium chloride solution, dried over sodium sulfate, treated with activated charcoal, filtered through clay and largely concentrated. The addition of petroleum ether gives 30.2 g (83% of theory) of 6-chloromorphane-thridine in the form of pale yellowish prisms with a melting point of 149-151 ° C.
7.5 g of 6-chloromorphanethridine obtained in this way are refluxed for 4 hours in 100 ml of absolute xylene with 15 ml of N, N-dimethyiaminoethylamine. The reaction mixture is mixed with water and concentrated sodium hydroxide solution. The aqueous phase is separated off and washed with ether, and the ether used for washing is combined with the xylene phase. The organic phase is extracted exhaustively with dilute hydrochloric acid. The combined acidic extracts are washed with ether, made alkaline with concentrated sodium hydroxide solution and then extracted with ether. The ethereal extract is washed with water and saturated sodium chloride solution, dried over sodium sulfate, treated with activated charcoal and evaporated. The residue is taken up in petroleum ether.
The solution is filtered through clay and largely evaporated. When the concentrated solution cools down, white, prismatic-tabular crystals form. 7.2 g of 6- (s-dimethylaminoethylamino) -morphanthridin with a melting point of 92-94 ° C. are obtained.
In a similar manner as in the above-mentioned examples, the products listed in Table II below are obtained from appropriate starting materials. R, R1, R2, R3, R4 and R5 therein have the meaning given earlier. In the right column, Ae means acetone, ether, CH chloroform, Me methanol and Pe petroleum ether.
