Es versteht sich, dass untergeordnete Abwandlungen in den Reaktionsbedingungen zur Durchführung der Reaktion erforderlich odler zweckmässig sein können, je nach der Natur des im Einzelfalle eingesetzten Reaktionspartners. So werden die 1, 2, 4-Benzothiadiazin-1, 1-di- oxyde (I) gemäss der Erfindung vorzugsweise durch Erhitzen des entsprechenden o-Sulfamylanilins (V) mit Orthoestern, Carbonsäuren, Säurehalogeniden, Säure- anhydniden oder a-Alkoxyaldehyd-acetalen hergestellt ;

Beispiel 1
2-Sulfamyl-4-chlor-anilin
Stuf.
Gruppe A : (1) 2, 4, 5-Trichlor-nitrobenzol ; (2) 2, 4, 5-Tribrom-nitrobenzol ; (3) 2, 4-Dichlor-5-trifluormethyl-nitrobenzol ; (4) 2, 5-Dichlor-4-trifluormethyl-nitrobenzol ; (5) 2-Chlor-4-trifluormethyl-nitrobenzol ; (6) 2-Chlor-4, 5-di-trifluormethyl-nitrobenzol ; (7) 2, 4-Dichlor-5-methyl-nitrobenzol ; (8) 2, 5-Dichlor-4-methyl-nitrobenzol ; (9) 2-Chlor-4-trifluormethyl-5-methyl- nitrobenzol ; (10) 2-Chlor-4-methyl-5-trifluormethyl- nitrobenzol ; (11) 2-Chlor-4-trifluormethyl-nitrobenzol ; (12) 2-Chlor-5-trifluormethyl-nitrobenzol ;
(13) 2, 4-Dibrom-nitrob, enzol ; (14) 2, 3, 5-Trichlor-nitrobenzol ; (15) 2, 5-Dichlor-3-methyl-nitrobenzol ; (16) 2, 4-Dichlor-3-methyl-nitrobenzol.
Gruppe B : (1) 2-Sulfamyl-4, 5-dichlor-anilin ; (2) 2-Sulfamyl-4, 5-dibrom-anilin ; (3) 2-Sulfamyl-4-chlor-5-trifluormethyl-anilin ; (4) 2-Sulfamyl-4-trifluormethyl-5-chlor-anilin ; (5) 2-Sulfamyl-4-trifluorrnethyl-anilin ; (6) 2-Sulfamyl-4, 5-di-trifluormethyl-anilin, (7) 2-Sulfamyl-4-chlor-5-methyl-anilin ; (8) 2-Sulfamyl-4-methyl-5-chlor-anilin ; (9) 2-Sulfamyl-4-trifluormethyl-5-methyl-anilin ; (1 0) 2-Sulfamyl-4-methyl-5-trifluormethyl-anilin ; (11) 2-Sulfamyl-4-trifluormethyI-anilin ; (1 2) 2-Sulfamyll-5-trifluormethyl-anilin ;
(13) 2-Sulfamyl-4-brom-anilin ; (14) 2-Sulfamyl-3, 5-dichlor-andlin ; (15) 2-Sulfamyl-3-methyl-5-chlor-anilin ; (16) 2-Sulfamyl-3-methyl-4-chlor-anilin.
Gruppe C : (1) 3-Methyl-6, 7-dichlor-1, 2, 4-benzothiadiazin 1, 1-dioxyd ; (2) 3-Methyl-6, 7-dibrom-1, 2, 4-benzothiadiazin
1, 1-dioxyd ; (3) 3-Methyl-6-trifluormethyl-7-chlor-1, 2, 4 benzothiadiazin-1, 1-dioxyd ; (4) 3-Methyl-6-chlor-7-trifluormethyl-1, 2, 4 benzothiadiazin-1, 1-dioxyd ; (5) 3-Methyl-7-trifluormethyl-1, 2, 4 benzothiadiazin-1, 1-dioxyd ; (6) 3-Methyl-6, 7-di-trifluormethyl-1, 2, 4 benzothiadiazin-1, 1-dioxyd ; (7) 3, 6-Dimethyl¯7-chlor-1, 2, 4 benzothiadiazin-1, 1-dioxyd ; (8) 3-Methyl-6-chlor-7-methyl-1, 2, 4 benzothiadiazin-1, 1-dioxyd ; (9) 3, 6-Dimethyl-7-trifluormethyl-1, 2, 4 benzothiadiazin-1, 1-dioxyd ;
(10) 3-Methyl-6-trifluormethyl-7-methyl-1, 2, 4 benzothiadiazin-1, 1-dioxyd ; (11) 3-Methyl-7-trifluormethyl-1, 2, 4 benzothiadiazin-1, 1-dioxyd ; (12) 3-Methyl-6-trifluormethyl-1, 2, 4 benzothiadiazin-1, 1-dioxyd ; (13) 3-Methyl-7-brom-l, 2, 4-benzothiadiazin
1, 1-dioxyd ; (14) 3-Methyl-6, 8-dichlor-1, 2, 4-benzothiadiazin 1, 1-dioxyd ; (15) 3-Methyl-6-chlor-8-methyl-1, 2, 4 benzothiadiazin-1, 1-dioxyd ;
(16) 3-Methyl-7-chlor-8-methyl-1, 2, 4 benzothiadiazin-1, 1-dioxyd,
3-Butyl-1, 2, 4-benzothiadiazin1, 1-dioxyden.
Beispiel 4 3-Butyl-6-chlor-7-methyl-1, 2, 4-benzothiadiazin
1, 1-dioxyd

Process for the production of new therapeutically active substances
It is known that certain groups of sulfonamide compounds show pronounced diuresis activity. For these substances, the name Sulònamid'Diureticas has been used, which is also used in the following.
Many of the well-known sulfonamide diuretics belong to the large class of benzothiadiazines, including their hydrogenation products, especially the diuretics from the chlorothiazide and hydrochlorothiazide series (e.g.
Chlorothiazide and hydrochlorothiazide itself, cyclopethiazide, hydroflumethiazide, benzhydroflumethiazide and trichloromethiazide) and certain 3-oxobenzothiadiazines, such as 6-chloro-7-sulfamyl-3-oxo-3, 4-dihydro-1, 2, 4-benzothiadiazine -1, 1-dioxide. Other classes of compounds in which valuable sulfonamide diuretics have been found are z. B. the 4-quinazolones and the isoindolines, here especially the 1-oxo-3- (3'-sulfamyl-4'-chlorophenyl) -3-hydroxy-isoindoline and its N-methyl derivative.
This list does not claim to be complete
There are also references to the literature that certain of these sulfonamide diuretics, in addition to their diuretic properties, have a weak antihypertensive effect in patients with abnormally high blood pressure. However, this antihypertensive effect is controversial. Insofar as it was confirmed, the authors concerned assumed a causal relationship between the diuretic and antihypertensive effects. In fact, it has not yet been possible to separate the two effects.
It has now been observed that an initially very specific desulfamylated analog of a sulfonamide diuretic, i.e. H. a compound which differs from the sulfonamide diuretic in question in that it does not have the-SOo NHe group, although it does not have the slightest diuretic activity (possibly even rather a weak antidiuretic effect), it is a very effective antihypertensive agent. It is particularly noteworthy that the antihypertensive effect also occurs in patients with normal blood pressure. Based on this observation, a number of desulfamylated analogs of very different sulfonamide diuretics were examined for their physiological effects, with similar results being obtained.
From this the fundamental new finding can be derived that the sulfamylated analogs of sulfonamide diuretics are potential antihypertensive agents without diuretic effects.
As to be expected, this rule is not without exception, but it enables the person skilled in the art, by means of simple series tests, to arrive at a large number of those which are effective in the specified manner. Connections to arrive.
Based on the above knowledge, the subject of the present patent is the development! a group of such substances with an isolated antihypertensive effect, namely substances which correspond to the following formula
EMI1.1
in which R is a lower, aliphatic or alicyclic
Hydrocarbon radical, X substitution in one of the positions 6, 7 or 8 by a halogen atom or the trifluoromethyl radical and Y substitution in one of the positions 6, 7 or 8 by a trifluoromethyl or lower alkyl group, a halogen atom or, if X, no 6 A permanent halogen atom is a hydrogen atom, including its non-toxic salts, in particular their alkali salts.
(It is well known that 1, 2, 4 benzothiadiazine-1, 1-dioxides can occur in two tautomeric forms, with the double bond of the heterocyclic part of the molecule being between atoms 2 and 3 or between atoms 3 and 4. Although throughout the present description only the last-mentioned form is written, it goes without saying that the other tautomer in each case is also included in the invention. According to the invention, the compounds defined above are prepared by adding a corresponding compound of the formula
EMI2.1
in which Z represents a hydrogen atom, a benzyl group or a lower aliphatic hydrocarbon radical, with a carboxylic acid of the formula R-COOH or with an ester, orthoester, imidoester, anhydride, amide,
Amidine or halide thereof condensed with ring closure.
The lower aliphatic or alicyclic hydrocarbon radicals used here are, for example, saturated lower, straight-chain, branched-chain and ring-shaped radicals with at most 5 carbon atoms in the manner of methyl, ethyl, propyl, isopropyl, cyclopropyl, n-butyl, tert-butyl -, cyclobutyl, amyl, isoamyl and cyclopentyl groups.
In the preferred processes described below for the preparation of the compounds in question, the starting materials used are X, Y-substituted o-sulfamylanilines, where X and Y have the meanings given above, with the proviso that they are each in one of the positions 3, 4 or 5 of the o-sulfamylaniline are bound.
Such X, Y-substituted o-sulfamylanilines can be prepared, for example, by converting a corresponding X, Y-substituted o-chloronitrobenzene (II) to the corresponding o-benzyl thioether (III) by treatment with a mixture of thiourea, benzyl chloride and alkali , which is then treated successively with chlorine in an aqueous-acidic medium and with ammonia, the corresponding nitrosulfonamide (IV) being formed. This is reduced, for example with iron in ammonium chloride solution, to the corresponding X, Y-substituted o-sulfamylaniline (V).
This general method of preparation can be represented by the following reaction scheme A:

<tb> <SEP> (IV) <SEP> (V)
<tb> (If the o-nitro-chlorobenzenes [II] in question are not known, they can be prepared analogously to known processes).
The o-sulfamylanilines (V) can be converted directly into the corresponding 1, 2, 4-benzothiadiazine-l, l-dioxides (I) or first converted to intermediate products, which in turn can be cyclized to the desired end products (I) .
According to the invention, the o-sulfamylaniline (V) is treated with a reactant in the manner of an orthoester, R-C (OR2) 3; a carboxylic acid, R-COOH, or one of its derivatives, namely an acid halide, anhydride, amide, amidine or simple lower alkyl ester or a corresponding imido ester, or, as explained in more detail below, with another, with regard to the finally resulting end product, chemically equivalent compound, e.g. B. an a-alkoxy-aldehyde-acetal, R30-CH (R ') - CH (OR2) 2, or a compound which is converted into one of the compounds mentioned under the prevailing reaction conditions.
In the case of all of these reactants, R denotes that hydrocarbon radical which should be in position 3 of the desired end product, and Ra and Ra denote lower alkyl groups. In the case of the reaction with the acetal, the grouping (R ') CH-denotes that hydrocarbon radical (less than a hydrogen atom) which ultimately appears as a 3-position substituent. (During the conversion, the substituent supplements its hydrogen requirement.)
In these reactions, all of the reaction partners mentioned are equivalent with regard to the end product ultimately formed.
The reaction, for example, of 2-sulfamyl-4-chloro-aniline with ethyl orthoacetate, with ce-methoxy-acetaldehyde-diethyl acetal or with acetic acid, acetyl chloride, acetic anhydride or ethyl acetate, leads to the formation of 3-methyl-7-chloro 1, 2, 4-benzothiadiazine 1, 1-dioxide.
It goes without saying that minor modifications in the reaction conditions may be necessary or useful for carrying out the reaction, depending on the nature of the reactant used in the individual case. Thus, the 1, 2, 4-benzothiadiazine-1, 1-dioxides (I) according to the invention are preferably prepared by heating the corresponding o-sulfamylaniline (V) with orthoesters, carboxylic acids, acid halides, acid anhydnides or a-alkoxyaldehyde acetals made;
but if carboxylic acids, acid halides or acid anhydrides are used as reactants, you generally need a catalyst to obtain the corresponding 1, 2, 4-benzothiadiazine-1, 1-dioxide directly without prior formation of intermediates, although one in some cases, the product can also be obtained directly by heating the reactants to much higher temperatures without using a catalyst. An optionally used catalyst can be basic, such. B. the alkali salts of the carboxylic acids in question or a tertiary amine of the type of pyridine, picolines or lutidines, or acidic, as in the case of per chloric acid.
If the o-sulfamylaniline (V) is reacted with an orthoester, this usually happens at elevated temperatures. In the absence of solvents, the temperature range is usually between about 40 C and the boiling point of the orthoester in question; Temperatures of about 80 to 130 ° C. are preferred. If the reaction is carried out in the presence of an inert organic solvent, the temperature is usually set at around the reflux temperature of the reaction mixture.
Dioxane or dimethylene glycol dimethyl ether solvents are preferred, but other inert solvents can also be used.
If a carboxylic acid is used as the reactant, the reaction is preferably carried out catalytically under pressure at temperatures in the range of about 130-200 ° C. (more preferably about 160-170 ° C.).
If an acid halide or anhydride is used, it is expedient to heat an excess of such a reactant with the relevant o-sulfamylaniline (V) in the presence of one of the abovementioned catalysts.
The chemically equivalent production methods above are illustrated by the following reaction scheme B:

<tb>
In the above scheme, R, X and Y are as given above. Meanings.
The orthoester of reaction path B (a) is written as ethyl ester, but it goes without saying that other common esters can be used in a similar manner without affecting the end product, since the alkoxy groups concerned are eliminated during the reaction.
Reaction partners and reactions have been described above by means of which X, Y-substituted 3-R-1, 2, 4-benzathiadiazine-1, l-dioxides (I) are obtained in essentially one-step processes. If the reaction conditions are modified slightly, various intermediate products are formed, which in turn can be converted into the same end product which is also directly obtainable. If, for example, the catalyst is omitted in the reaction between o-sulfamylaniline (V) and acid halide or anhydride, the corresponding N-acylated o-sulfamylaniline (VI) is initially obtained as an intermediate.
The last-mentioned embodiment is generally carried out in an inert aromatic hydrocarbon, such as benzene, toluene, xylene or the like, as the solvent, preferably at reflux temperature; however, other solvents such as dialkyl ethers, cyclic ethers or the like can also be used.
The acylated intermediate is isolated and heated above its melting point, during which it cyclizes to form the corresponding 1, 2, 4 benzothiadiazine-1, l-dioxide (I). An example of this is the reaction of 2-sulfamyl-4-chloro-aniline with acetyl chloride or acetic anhydride in refluxing toluene, with 2-sulfamyl-4-chloro-acetanilide being formed. When this intermediate product is heated above its melting point, 3-methyl-7-chloro-1, 2, 4-benzothiadiazine-1, 1-dioxide is formed.
It goes without saying that by modifying the o-sulfamylaniline used as starting material or the acylating agent, the various end products falling under the invention can easily be prepared.
If the solvent is exchanged for a tertiary amine, for example one from the pyridine series, in the reaction described above, a diacylated intermediate (VII) is formed, one acyl group on the aniline nitrogen atom and the other on the sulfamyl nitrogen atom. atom hangs. This diacylated intermediate cyclizes in a similar way by blouse heating above its melting point to give the corresponding 1,2,4-benzothiadiazine-1,1-dioxyde (I).
The latter two embodiments are illustrated by the following Reaction Scheme C:

According to reaction scheme A, the nitrosulfonamide (IV) is prepared by amination of a corresponding sulfamyl chloride with ammonia. Equivalent compounds are obtained by amination with a primary lower alkylamine or with benzylamine.
The starting materials according to the invention then contain the corresponding additional substituent which, however, is eliminated when the ring closes to form the 1,2,4-benzothiadiazine-1,1-dioxide (I). If, for example, benzylamine is used, the starting material (V) in reaction scheme C contains a benzyl substituent on the nitrogen atom of the sulfamyl group. As described, this substance is first acylated to an intermediate (VI) or (VII), and when it is heated, the benzyl group is split off again, and the same end product (I) is formed.
The same applies to those variants of Reaction Scheme B which relate to the reaction with carboxylic acids and their derivatives.
The reaction with orthoesters of modified starting material (V) with a benzyl or lower aliphatic hydrocarbon radical having at least three carbon atoms, preferably isopropyl, on the sulfamyl nitrogen atom is illustrated in reaction scheme D. This reaction takes place in two stages with the formation of an intermediate product (XIV) which, on heating, yields (I).

As mentioned, a suitable starting material can be condensed with a reactant which is converted into one of the abovementioned reactants under the prevailing reaction conditions.
To illustrate, the observation that z. B. compounds of the RCC13 type when heated with o-sulfamylanilines (V) in the presence of a strong base and an alcoholic solvent likely to convert into corresponding carboxylic acids, but in any case into compounds that react with the o-sulfamylaniline, so that you can, for example, by heating 2-sulfamyl-4-chloro-aniline with 1, 1, 1 trichloroethane finally leads to 3-methyl-7-chloro-1, 2, 4 benzothiadiazine-1, 1-dioxide.
The new compounds prepared according to the invention can be used for relieving and controlling essential hypertension, malignant hypertension and the like, as well as peripheral vascular diseases.
The effective dosage of the compounds will depend on the severity, stage, and details of each case. In general, the dosage for the average compounds of the present class is in the range from about 0.25 to 15 mg per kg of body weight and day, for the particularly effective preferred compounds in the range from 0.25 to 2.5 mg per kg and day.
The compounds prepared according to the invention can be used in the form of pharmaceutical preparations which contain the active ingredient mixed with a pharmaceutical carrier such as is suitable for enteral or parenteral administration. Such preparations are in solid form, for example tablets, capsules and suppositories, in liquid form, for example, elixirs, emulsions and injection solutions.
For the formulation of pharmaceutical preparations, substances that do not react with the active ingredients can be added, e.g. B. water, gelatin, lactose, starches, magnesium stearate, calcium carbonate, talc, vegetable oils, benzyl alcohols, rubber, poly alkylene glycols or petrolatum. The active ingredient in question is expediently present in the preparation in a weight proportion of 0.1 to 50%.
The pharmaceutical preparations can also contain further active ingredients, for example a therapeutically effective amount of a diuretic.
Production of the raw materials
example 1
2-sulfamyl-4-chloro-aniline
Step A: A mixture of 63 g benzyl chloride, 38 g thiourea, 3 drops of concentrated ammonium hydroxide solution and 250 ml 95% ethanol is allowed to reflux for 3 hours. It is then cooled and a solution of 96 g of 2,4-dichloro-nitrobenzene in 200 ml of ethanol is added. The mixture is heated to reflux and a solution of 70 g of potassium hydroxide in 500 ml of ethanol is added dropwise. The mixture is allowed to reflux for a further 2 hours, then cooled, the solid which has separated out is filtered off, washed with aqueous ethanol and dried. This leads to 2-benzylthio-4-chloronitrobenzene, which corresponds to the above formula III.
Stage B: 50 g of 2-benzylthio-4-chloronitrobenzene are suspended in 1000 ml of 33% strength aqueous acetic acid and gaseous chlorine is blown through the suspension, which has been cooled to about 0 to 5 ° C., for 2 hours. The mixture is then extracted three times with 400 ml of chloroform each time, the combined extracts are washed with water, the chloroform solution is dried over anhydrous sodium sulfate and filtered.
Stage C: The dried chloroform solution is evaporated to dryness, 400 ml of liquid ammonia are added to the residue, the mixture is stirred, the excess ammonia is allowed to evaporate and the residue is rubbed with hexane. A crystalline solid forms which, after further digestion, is filtered with water. Essentially pure 2-sulfamyl-4-chloronitrobenzene, which corresponds to the above formula (IV), is obtained in this way. For purification, it is recrystallized from aqueous methanol.
Stage D: A mixture of 4.4 g of ammonium chloride, 18 M methanol, 9 ml of water and 3.0 g of 2-sulfamyl-4-chloronitrobenzene is heated to reflux and 4 is added in portions over the course of about 11/2 hours , 4 g iron filings. The mixture is cooled and filtered, the filtrate is concentrated to a residue, this is digested with 15 ml of water and filtered off.
When the solid is recrystallized from aqueous methanol, essentially pure 2-sulfamyl-4-chloro-aniline which corresponds to the above formula (V) is obtained.
As can be seen by those skilled in the art, if the 2,4-dichloro-nitrobenzene of Example 1 is replaced by other correspondingly substituted 2-chloro-nitrobenzenes, corresponding other o-sulfamylanilines which can be used as starting materials can be obtained, essentially following the method described in Example 1 works.
For example, if the 2,4 dichloro-nitrobenzene is replaced by one of the 2-chloro-nitrobenzenes (II) belonging to the following group A, the corresponding intermediates are obtained by means of processes which essentially correspond to steps A to D above III and IV to the corresponding 2-sulfamyl-anilines (V), which are listed in group B.

If the ammonia of the amination stage C of Example 1 is replaced by a primary lower alkylamine or by benzylamine, the corresponding N-substituted sulfamylnitrobenzene is formed, from which a suitable starting material is also obtained by further reaction according to Stage D of Example 1.
The production of such a starting material is described below:
Example 2 2- (N-Isopropyl-sulfamyl) -4-chloro-aniline
Stage A: 50 ml of isopropylamine are allowed to act for 3 hours at room temperature on 10 g of 2-nitro-5-chloro-benzal-sulfonyl chloride, the reaction mixture is evaporated to dryness and the residue is crystallized from water to give N-isopropyl-5-chloro-2- nitro-benzene-sulfonamide.
Stage B: A mixture of 12.5 g of N isopropyl-5-chloro-2-nitro-benzenesulfonamide, 16.9 g of ammonium chloride, 140 ml of methanol and 70 ml of water is allowed to reflux while stirring continuously, and added in portions over the course of 11 / 2 hours 16.9 g iron filings and allow another 11/2 hours to reflux.
The mixture is then cooled, filtered, the filtrate is diluted with 350 ml of water, heated again to reflux, treated with activated charcoal, filtered, cooled and left to stand until crystals of 2-isopropyl-sulfamyl-4-chloro-aniline are formed filtered off.
The isopropylamine from stage A of Example 2 can be replaced by other primary lower aliphatic amines or benzylamine, the corresponding N-substituted starting materials being obtained under similar reaction conditions. Of course, other 2-nitro-benzenesulfonyl chlorides, for example the compounds listed above in group A, can be converted into the corresponding X, Y-substituted N-substituted sulfamyl-nitrobenzenes and in a similar manner after conversion according to stages A and B of Example 1 further converted into the corresponding starting materials.

A mixture of 5 g of 2-sulfamyl-4-chloro-aniline and 15 ml of ethyl orthoacetate is heated to 100 to 110 ° C. for 1 hour, cooled and the solids are filtered off. Recrystallization from aqueous ethanol gives 3-methyl-7-chloro-1, 2, 4-benzothia-diazine-1, 1-dioxide (melting point = 325 to 327 ° C.).
By replacing the 2-sulfamyl-4-chloro-aniline of this example with equivalent amounts of the substances listed in group B and then treating essentially as in Example 3, the corresponding compounds of the following group C are obtained.
Group C: (1) 3-methyl-6, 7-dichloro-1, 2, 4-benzothiadiazine 1, 1-dioxide; (2) 3-methyl-6, 7-dibromo-1, 2, 4-benzothiadiazine

In the above reaction, the ethyl orthoacetate can be replaced by equivalent amounts of other saturated lower aliphatic orthoesters, the corresponding 3-substituted 1, 2, 4 benzothiadiazine-1, l-dioxides being formed. If, for example, the ethyl orthoacetate is replaced by equivalent amounts of ethyl orthopropionate, ethyl orthobutyrate or ethyl orthovalerate, the corresponding 3-ethyl, 3-propyl or 3-butyl-1, 2, 4-benzothiadiazin1, 1-dioxyden.

Recrystallization from aqueous ethanol gives 3-buty1-6-chloro-7-methyl-1,2,4-benzothiadiazine-1,1-dioxide.
Again, when using compounds from group B above, instead of 2-amino-4-methyl-5-chlorobenzenesulfonamide, the corresponding X, Y-substituted 3-butyl-1,2,4-benzothiadiazine 1,1-dioxydes are obtained. In a similar way, the valeric acid and its sodium salt can be replaced by other saturated lower alkanecarboxylic acids and their sodium salts, whereby the corresponding 3-substituted substances are obtained.
