  Verfahren zur Herstellung von     Diazinen       Gegenstand der     Erfindung    ist ein neues Verfahren  zur Herstellung von     1,4-Diazinverbindungen    der Formel

  Das neue     Verfahren    ist dadurch     gekennzeichnet,dass     man     eine    Verbindung der Formel

          Cycloalkyl,        Cycloalkenyl,          Cycloalkyl-alkyl,        Cyclo@alkenyl-alkyl,        Aryl,        Aralkyl    oder       Aralkenyl-reste    in Frage.     Als        Substituenten    dieser Reste  seien z.

   B. genannt für     aliphatische    Reste     Hydroxy,        Alk-          oxy,    Halogen, wie     Chlor    oder     Brom,        Amino,        Alkyl-          amino    oder     Dialkylamino,    für     cycloaliphatische    Reste  z. B.     Alkyl,    und für aromatische Reste z.

  beliebiger    Stellung  verbundene     Propyl,        Butyl,        Pentyl    oder     Hexylreste,    als       Alkenylreste    insbesondere     Allyl,        als        Cycloalkylreste    ins  besondere     Cyclopentyl,        Cyclohexyl    oder     Cycloheptyl,    als       Gycloalkenylreste        insbesondere        Cyclopentyl    oder     Cyclo-          hexenyl,

          als        Arylreste        insbesondere        Phenylreste    und als       Aralkyl-    oder     Aralkenylreste    insbesondere     Benzyl-,          a-Phenyl-äthyl-,        ss-Phenyl        äthyl-        oder        Phenyl-vinylreste          zu    nennen.  



  Der oben genannte Rest R' ist z. B.     ein:        gegebenen-          falls    substituierter     Kohlenwasserstoffrest,    wie     .er    oben für  R beschrieben ist. Weiter     kann    er beispielsweise auch  ein     Pyridyl-,        Chinolyl-    oder     Thienylrest        sein.     



  Der     cyclische    Rest

         kann    auch weitere     Substituenten        aufweisen.    Ist er ein       alicyclischer    Rest, so kann er z. B. noch     Alkylgruppen     tragen, z. B. die oben     angegebenen.    Ist er aromatischer  oder     heterocyclischer        Natur,    so     kommen    z. B. die oben  für     Phenylreste        angegebenen        Substituenten    in Frage.

   Für       Pyrimidylreste    sind insbesondere     Aminograppen    oder       Hydroxy-    oder     Mercaptogruppen    zu erwähnen, so dass

  Die     verfahrensgemäss    als     Ausgangsstoffe    zu verwen  denden Verbindungen     HY        sind        Ammoniak    oder pri  märe oder vorzugsweise sekundäre     Amine,    wobei als       Substituenten    z.

   B. die für R     aufgezählten        gegebenenfalls          substituierten        Kohlenwasserstoffreste,        insbesondere          Alkylreste,    oder auch     Alkylenreste,    in denen die     Alk-          ylenkette    durch     Heteroatome    unterbrochen sein kann,  wie     Oxa-    oder     Aza-alkylenreste,    z.

   B.     Butylen-(1,4),          Pentylen    (1,5),     Hexylen-(1,5),        Hexylen-(1,6),    Heptylen-    (2,6),     3-Oxa-    oder     Aza-pentylen-(1,5),    3     Methyl-,        3-          Äthyl    oder     3-Hydroxyäthyl-3"aza-pentylen-(1,5),        3-Aza-          hexylen(1,6)    oder     4-Methyl-4-aza-heptylen-(2,6),        zu          nennen        sind.     



       Bevorzugte        Ausgangsstoffe        der    eingangs gezeigten  Formel     sind        insbesondere        o-Dichloracetylamino-aniline,          4-Amino-5-dichloracetylamino-pyrimidine    und     4-Amino-          3,dichloracetylamino-pyrazole.     



  Die     Umsetzung    mit     Ammoniak    oder     Aminen    erfolgt       vorzugsweise    durch     Erhitzen,    in An- oder     Abwesenheit     von     Lösungsmitteln,    wie neutralen     Lösungsmitteln,    z. B.       Alkoholen        oder        gegebenenfalls    im     überschuss    des     be-          treffenden        Amins,    das     dann    gleichzeitig als Lösungsmit  tel dient.  



       Als        Dehydrierungsmittel    verwendet man beispiels  weise Schwefel oder Kaliumpermanganat. Die     Dehydrie-          rung        kann        auch        mit    Selen oder     mit    Sauerstoff, z.

   B. mit  Sauerstoff     in    Gegenwart     eines        Katalysators    wie Platin,  oder mit     Wasserstoffsuperoxyd        durchgeführt        werden.    Die       Dehydrierung        kann    nach     Isolierung    oder ohne Isolie  rung des nach der 1.     Stufe        erhaltenen    Zwischenproduk  tes     durchgeführt    werden.

   Setzt man     das        Dehydrierungs-          mittel    gleichzeitig     mit        Ammoniak    oder dem Amin ein,  so     verwendet    man vor     allem    Schwefel als     Dehydrierungs-          mittel.    Die     Dehydrierung        erfolgt        vorzugsweise    in Anwe  senheit eines     Lösungs.    oder     Verdünnungsmittels    wie  z. B. eines Alkohols, wie z.

   B. eines niederen     Alkanols,     oder Wasser oder einem flüssigen     organischen        Amin,    wie       einem        Dialkylamin    oder     einem        Alkylenimin    oder einem       Oxaalkylenimin.     



  Die     Ausgangsstoffe        sind.    bekannt     oder    lassen sich  nach     an.    sich bekannten Methoden     gewinnen.    Sie können       gegebenenfalls    auch in     Form        ihrer        Hydrate        oder    Salze       eingesetzt    werden.  



  Die     verfahrensgemäss    erhaltenen, eingangs definier  ten     Diazine    sind zum Teil bekannt. Sie     sind        wertvolle          Zwischenprodukte,    insbesondere     zur    Herstellung von       Heilmitteln.    Verbindungen dieser     Art    sind     aber    auch von  Nutzen wegen ihrer     pharmakologischen        Wirkungen,    ins  besondere wegen ihrer     diuretischen    und     Natrium-aus-          scheidenden    Wirkung.

   Sie     können        dementsprechend    als       Medikamente        in    der     Human-    oder Veterinärmedizin       Verwendung        finden.     



  Besonders     wertvoll        in        dieser        Beziehung    sind die       neuen,        Verbindungen    der     Formel

    worin R die eingangs gegebene Bedeutung hat und Y  für eine     Aminogruppe,    z.

   B. eine     unsubstituierte    oder  eine wie oben für die     Amine    angegeben     substitnierte          Aminogruppe,    steht und     R1    und     R2        Wasserstoff    oder       gegebenenfalls        substituierte        Kohlenwasserstoffreste    be  deuten, z. B. solche der eingangs genannten     Art,    und  ihre Salze.  



  Die     Endstoffe    des Verfahrens können je nach den       Reaktionsbedingnungen    und     Ausgangsstoffen    in     freier       RTI ID="0002.0234" WI="9" HE="4" LX="1142" LY="2395">  Form.    oder in Form ihrer     Salze    erhalten     werden.    Die  Salze der neuen     Verbindungen    können in an sich be  kannter Weise in     die    freien Verbindungen     übergeführt     werden, z. B.     Säureadditionssalzedurch        Reaktion        mit     einem basischen Mittel.

   Anderseits     können    .gegebenen  falls erhaltene freie Basen mit     anorganischen    oder orga  nischen Säuren     Salze        bilden.        Zur    Herstellung von     Säure-          additionssalzen    werden     insbesondere    therapeutisch ver  wendbare Säuren     verwendet,    z. B.

   Halogenwasserstoff-           säuren,    beispielsweise     Salzsäure    oder Bromwasserstoff  säure,     Perchlorsäure,    Salpetersäure oder     Thiocyansäure,     Schwefel- oder Phosphorsäuren, oder organische     Säuren,     wie Ameisen-, Essig-,     Propion-,    Glykol-, Milch-,     Brenz-          trauben    ,     Oxal-,        Malon-,    Bernstein-,     Malein    ,     Fum,ar-,     Apfel-, Wein-, Zitronen-,     Ascorbin-,

          Hydroxymalein-          oder        Dihydroxymaleinsäure,        Benzoe-,        Phenylessig-,          4-Amino-benzoe-,        4-Hydroxy-benzoe-,        Anthranil-,     Zimt-, Mandel-,     Salicyl-,        4-Amino-salicyl-,        2-Phenoxy-          benzoe-    oder     2-Acetoxy-benzoesäure,        Embonsäure,          Methansulfon-,        Äthansulfon-,        Hydroxyäthansulfon-,

            Benzolsulfon-,        p-Toluol-sulfon-,        Naphthalinsulfon-        oder          Sulfanilsäure,    oder     Methionin,        Tryptophan,        Lysin    oder       Arginin.    Dabei können Mono- oder     Polysalze    vorliegen.  Die     Salze    können auch zur     Reinigung    der freien Basen  dienlich sein.  



  Die     pharmakologisch    wertvollen, neuen Verbindun  gen können als     Heilmittel    in Form von pharmazeutischen  Präparaten verwendet werden, welche diese Verbindun  gen zusammen mit     pharmazeutischen,    organischen     oder          anorganischen,    festen     oder    flüssigen Trägerstoffen, die  für     enterale,    z. B.     orale,    oder     parenterale    Gabe     geeignet     sind, enthalten können.  



  Sie können auch in der Tiermedizin, z. B. in einer       ,der    oben     gemannten    Formen, oder bei der Aufzucht und  Ernährung von Tieren in Form von     Futtermitteln    oder  von     Zusatzmitteln    für Tierfutter verwendet werden.  



  In den folgenden Beispielen sind die     Temperaturen     in Celsiusgraden angegeben.    <I>Beispiel 1</I>       Zu.    7,28 g     2-Methyl-4-amino-5-dichloracetylamino-          pyrimidin        dichloracetat    und 0,64 g Schwefel werden  40     cm3        Morpholin    zugegeben. Dabei     steigt    die Tempera  tur rasch an.     Anschliessend    erhitzt man noch während  5 Minuten in einem Bad von 100 .

   Nach dem Abkühlen  stellt man mit 2 n     Salzsäure    auf     pH    = 7,0, wobei sich  das     2-Methyl-6-hydroxy-7-morpholino-pteridin    der For  mel

    vom F. 290-292  ausscheidet. Nach     Umkristallis,ieren     aus Alkohol steigt der Schmelzpunkt     auf    291-293 .  



  Das als Ausgangsstoff verwendete     2-Methyl-4-amino-          5-dichloracetylamino-pyrimidin-dichloracetat    wird z. B.  wie folgt hergestellt:  37,2 g     2-Methyl-4,5=diamino-pyrimidin    werden mit  115 cm-'     Dichloressigsäure        unter    leichtem Vakuum wäh  rend 1 Stunde in     einem        Ölbad    von 100  belassen.

   Nach  dem Abkühlen versetzt man die Reaktionslösung mit       Natriumhydrogencarbonatlösung    und festem     Bicarbonat     bis ein     pH    von 1,8 erreicht ist.     Hierauf    scheidet sich das       2-Methyl-4-amino-5-,dichloracetylamino-pyrimid        ndi-          chloracetat    :der Formel

  <I>Beispiel 2</I>  0,91 g     2-Methyl-4-arnina-5-dichloracetylamino-pyr-          imidin-dichloracetat    werden mit 5     cm3        Morpholin    in  einem Ölbad von 100  während 5 Minuten erhitzt.

   Nach  dem Abkühlen versetzt man mit Wasser und stellt mit 2 n       Salzsäure    auf     pH    = 7, wobei sich das     2-Methyl-6-hydr-          oxy-7-morpholino-7,8-dihydro-pterid@in    der Formel

  0,25 g     2-Methyl-6-hydroxy-7-morpholino-7,8-di-          hydro-pteridin    und 32 mg Schwefel     werden    mit 2     cm3          Morpholin    während 5 Minuten in einem     Ölbad.    von 100   erwärmt.    Beispiel 2) werden     in    einer Lösung  von 112 mg     Kaliumhydroxyd    in 16     cm3    Wasser gelöst.  Dazu gibt man     tropfenweise    unter Rühren 16,6     cm3     0,2 n     Kahumpermanganatlösung.    Nach dem     Zutropfen     brennt man das     Mangandioxyd    ab, stellt die alkalische  Lösung auf     pH    = 7 und     engt    stark ein. Dabei scheidet  sich das oben beschriebene     2-Methyl6-hydroxy-7-          morpholin.o-pteridin    vom F.<B>289-291'</B> ab.  



  <I>Beispiel 4</I>  1,82 g     2-Methyl-4-amino-5-dichloracetylamino-pyr-          imi-din-dichloracetat    und 0,16 g Schwefel werden mit  10     cm3        Piperüdin    versetzt. Dabei steigt die Temperatur  rasch an.     Anschliessend        erhitzt    man noch während 5 Mi  nuten in einem Ölbad von 100 . Nach dem Abkühlen  stellt man mit 2 n     Salzsäure    auf     pH    = 7, wobei sich  das 2     Methyl-6-hydroxy-7-piperidino-pteridin    der For  mel

Dabei steigt die       Temperatur    rasch an.     Anschliessend        erhitzt    man noch  während 5 Minuten in einem Ölbad von 100 . Nach dem  Abkühlen stellt man mit 2 n Salzsäure auf     pH    = 7,  wobei sich das     2-Methyl-6-hydroxy-7-hexamethylen-          imino-pteridin    der Formel
