  Verfahren zur Herstellung von Dibenzocycloheptenen
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von neuen 5H-Dibenzo (a,d)-cycloheptenen und   10,11 -Dihydro-5H-dibenzo-    (a,d)-cycloheptenen und deren Salze, welche am Kohlenstoff der 5-Stellung durch einen Aminopropylrest substituiert sind.



   Diese Verbindungen können zur Behandlung von Geisteskrankheiten verwendet werden, da sie als Mittel gegen Depressionszustände bzw. zur Aufheiterung des Gemüts und als psychische Energiespender wirksam sind. Für solche Fälle kann die tägliche Dosis 5 bis 250 mg betragen, welche vorzugsweise in Teildosen eingenommen wird. Diese Verbindungen werden vorzugsweise in Form ihrer mit Säuren erhältlichen Salze verabreicht.



   Die nach dem erfindungsgemässen Verfahren herstellbaren Dibenzocycloheptene haben folgende Formel

   Die punktierte Linie zwischen den Stellungen 10 und 11 bedeutet, dass die Verbindungen an dieser Stelle gesättigt oder ungesättigt sein können, wobei die gesättigte Verbindung als die entsprechende 10,11 Dihydroverbindung bezeichnet wird.



   Das erfindungsgemässe Verfahren zur Herstellung von Dibenzocycloheptenen der angegebenen Formel ist nun dadurch gekennzeichnet, dass man ein tertiäres Amin der Formel

 mit einem Cyanhalogenid zur Reaktion bringt, wobei eine Verbindung der Formel

 entsteht, die man hierauf hydrolisiert.  



   Das erfindungsgemässe Verfahren kann beispielsweise wie folgt durchgeführt werden, wobei für den Rest R vorzugsweise eine Methyl- oder   Äthylgruppe    steht: Das tertiäre Amin wird in einem nicht-hydroxylischen Lösungsmittel gelöst, beispielsweise Benzol oder Äther, die Lösung langsam in eine Cyanhalogenidlösung im gleichen Lösungsmittel einfliessen gelassen, wobei sich bildendes Alkylhalogenid entweicht.



  Nach beendeter Reaktion wird die basische Komponente durch Waschen mit verdünnter Salzsäure entfernt und das Cyanamid durch Verdampfen des Lösungsmittels isoliert. Hierauf erfolgt Hydrolyse in einem sauren oder alkalischen Medium zum sekundären Amin.



   Beispiel 1
Herstellung von   10,11-Dihydro-   
5-(3-methylaminopropyl)-5H-dibenzo(a,d)cyclohepten
A. Eine Lösung von 0,0834 Mol   10,11-Dihydro-    5-(3-dimethyl-aminopropyl)-5H-dibenzo(a,d)cyclohepten in 61 ml Benzol wird einer gerührten Lösung von 9,80 g (0,0917 Mol) Bromcyan in 38 ml Benzol tropfenweise zugefügt. Man rührt weiter während 3 Stunden bei Raumtemperatur und filtriert. Das Filtrat, zusammen mit der Benzolwäsche des Niederschlages, wird auf einem Dampfbad unter vermindertem Druck zur Trockne verdampft, so dass Lösungsmittel und überschüssiges Bromcyan abgetrennt werden. Hierauf wird der Rückstand aus rohem Cyanamid in Benzol gelöst, die Lösung zur Entfernung von Resten unver änderten Amins mit verdünnter Salzsäure und dann mit Wasser gewaschen.

   Nun wird die   Benzollösung    unter vermindertem Druck verdampft und man erhält das Endprodukt A als gelbes   Öl.   



   B. Das erhaltene ölige Cyanamid wird mit 500 ml Eisessig, 330 ml Wasser und 50 ml konzentrierter Salzsäure während 16 Stunden zum Rückfluss erhitzt, wobei Hydrolyse und Decarboxylierung stattfindet.



  Man engt auf ein kleines Volumen durch Verdampfen unter reduziertem Druck ein, löst den Rückstand in   250ml    Wasser, das 1 ml konzentrierte Salzsäure enthält, wäscht die wässrige Lösung mit Benzol und macht durch Zusatz eines Überschusses an Kaliumkarbonat alkalisch. Die dadurch freigewordene Base wird in   Äther    aufgenommen und   die Atherlösung    über wasserfreiem Kaliumkarbonat getrocknet. Nach Verdampfung des ethers auf einem Dampfbad unter vermindertem Druck erhält man als Rückstand das entsprechende Cyclohepten in Form eines Öles.



   Die freie Base kann in das Oxalat übergeführt werden, indem man sie in warmem absolutem Äthanol löst, eine äquimolare Menge an in Äthanol gelöste Oxalsäure zufügt und durch   Ätherzusatz    das Oxalat zur Ausfällung bringt. Das oxalatsaure Salz wird zweckmässig aus einer Mischung von Methanol und Äthylacetat umkristallisiert.



   Die Hydrolyse kann aber auch wie folgt unter alkalischen Bedingungen durchgeführt werden:
2,0 g (0,007 Mol) des gemäss A erhaltenen Cyanamids werden unter Rühren in eine heisse Lösung von 25 g Kaliumhydroxyd in 38 ml abs. Methanol gegeben.



  Man erwärmt die Mischung unter Rühren während 45 Stunden auf Rückflusstemperatur, verdünnt hierauf mit Wasser und   nimmt    das sich dabei ausscheidende gelbe Öl in Benzol auf. Dann wird die Benzollösung mit Wasser gewaschen und nach Trocknung mit Natriumsulfat eingedampft, bis ein gelber öliger Rückstand zurückbleibt. Nun wird die erhaltene Base in n Salzsäure aufgenommen und die neutrale Substanz mit   Äther    extrahiert. Nach erneutem Behandeln mit Alkali wird die erhaltene Base nochmals in Äther aufgenommen.



  Nach Verdampfung des ethers erhält man die Base   10, l-Dlhydro- 5 - (3 -methylaminopropyl) -5H-dlbenzo      (a,d)cyclohepten,    welche wie oben angegeben in das Oxalat übergeführt werden kann.



   Das salzsaure Salz kann unmittelbar aus der rohen freien Base gewonnen werden oder wie folgt aus dem Oxalat: Man versetzt eine wässrige Lösung des Oxalats mit Lithiumhydroxyd und nimmt die gebildete Base in Benzol auf. Nach Verdampfung des Benzols bleibt die Base praktisch in reiner Form zurück. Nun behandelt man die Base mit äthanolischer Salzsäure und fällt das gebildete salzsaure Salz durch Zusatz von Äther. Nach Umkristallisation aus einer   ithanol-l2ither-Mischung    schmilzt das Produkt oberhalb   216     C.



   Beispiel 2
Herstellung von 5-(3-Methylaminopropyl)   5H-dibenzo(a,d)cyclohepten   
Verfährt man gemäss Beispiel 1 A unter Verwendung einer äquivalenten Menge 5-(3-Dimethylaminopropyl)-5H-dibenzo   (a,d)cyclohepten    anstelle von   10, 11-Dihydro   5-(3-Dimethylaminopropyl)-5H-diben-    zo(a,d)cyclohepten, so erhält man das entsprechende Cyanamid. Nach Hydrolyse gemäss Beispiel 1 B erhält man   5-(3 -Methylaminopropyl)-5H-dibenzo    (a,d)cyclohepten. Letztere Verbindung kann unmittelbar durch Behandlung mit äthanolischer Salzsäure gemäss Beispiel 1 B in das salzsaure Salz übergeführt werden und zeigt nach   Umkristallisation    aus einer Isopropylalko  hol-Äther-Mischung    einem Smp. von   166,5-167,5"    C.



   Analyse für   C10H21N.      HCl    berechnet:   76,11 0/o    C;   7,40 3/o    H;   4,670/c    N; gefunden:   7614 0h    C;   7,30 0/o    H;   4,64 0/o    N.



   Die Herstellung der dafür verwendeten Ausgangsstoffe kann nach folgender Methode erfolgen:
A.   1, 08 g      (0,0442 g-Atom)    Magnesiumspäne werden in einen 200 ml fassenden 3-Halskolben versehen mit Rührer,   Tropfrichter    und Rückflusskühler mit Trockenrohr eingefüllt. Während der Reaktion strömt trockener Stickstoff durch das Reaktionsgefäss. Nun gibt man einen   Jodkristall    und dann 10   ml    trockenes Tetrahydrofuran zu, fügt hierauf   3 mol    einer   bereils-    ten   3-Dimethylaminopropyl-magnesiumchlorid-Lösung    hinzu und erhitzt zum Rückfluss auf dem Dampfbad.



  Dazu giesst man tropfenweise eine Lösung von 5,37 g   (0,0442 Mol)    3-Dimethylaminopropylchlorid in   35 mg    trockenem Tetrahydrofuran und achtet darauf, dass die Mischung dauernd unter Rückfluss siedet. Nach beendeter Zugabe wird noch während 2 Stunden zum Rückfluss erhitzt, wobei sich der grösste Teil des Magnesiums auflöst.



   Anstelle von   3 -Dimethylaminopropylmagnesium-    chlorid zur Auslösung der Reaktion kann man   Äthyl-    bromid in einer Menge von   0,05 Molen    pro Mol Magnesium verwenden.



   B. Die nach dem Verfahrensschritt A erhaltene   Grignardiösung    wird auf Raumtemperatur abgekühlt und unter Rühren einer Lösung von   5, 05 g    (0,0221 Mol)   5-Chlor-5H-dibenzo(a,d) cyclohepten    in   25 mol    trockenem Tetrahydrofuran tropfenweise zugesetzt,  wobei von aussen derart gekühlt wird, dass die Raumtemperatur praktisch eingehalten wird. Nach beendetem Zusatz wird die Mischung während 15 Minuten zum Rückfluss erhitzt. Hierauf wird das Tetrahydrofuran unter reduziertem Druck bei einer Wasserbadtemperatur von   50-60     C abdestilliert. Die sich ergebende sirupöse Mischung löst man in 75 ml Benzol und fügt hierauf unter Rühren tropfenweise   15ml    Wasser zu.



  Man   dekantiert    die Benzolschicht von der gelatinösen Fällung ab, welche noch 3 mal mit je 20   ml    siedend heissem Benzol extrahiert wird. Die vereinigten Benzolextrakte werden mit Wasser gewaschen und dreimal mit je 15 ml 3 n-Salzsäure geschüttelt; man setzt Natriumhydroxyd bis zur alkalischen Reaktion zu, wobei die gelbe ölige Base ausfällt, welche in Hexan aufgenommen wird. Man wäscht die vereinigten Hexanextrakte mit Wasser und dampft das Hexan ab. Es werden   4, 61 g    eines gelben Öles erhalten. Durch Behandeln mit einer Mischung von 15 ml abs. Alkohol, 15 ml abs. Äther und 1,8   ml    einer trockenen 10,28 n-Chlorwasserstofflösung in abs. Alkohol wird die Base in das salzsaure Salz übergeführt.

   Durch Verdünnen der Lösung mit 25 ml abs.   J3ither    wird die Kristallisation des Produktes ausgelöst, welches in einer Ausbeute von 4,17 g mit einem Smp. von   1861900    C erhalten wird. Nach Umkristallisation aus einer Mischung von abs. Alkohol und abs. Äther steigt der Smp. auf 1911930 C.



   Analyse für   C20H23N. HCI    berechnet:   76,53 0/o      C;      7,71 0/0    H;   4,46 0/0    N; gefunden:   76,230/0      C;      7,83 0/0    H;   4,45 0/0    N.



   Beispiel 3
Herstellung von   10,1 1-Dihydro-5-(3-methylamino-    propyl)   SH-dibenzo(a,d)cyclohepten   
Gemäss Beispiel 1 A wird   10,11-Dihydro-5-(3-      dimethylaminopropyl)-SH-dibenzo (a, d) cyclohepten    in das entsprechende Cyanamid übergeführt. Hierauf wird das Cyanamid (5,03 g) zusammen mit 60 ml Eisessig, 40 ml Wasser und 8   ml    konzentrierter Salzsäure während 16 Stunden zum Rückfluss erhitzt und verdampft die Reaktionsmischung durch Erhitzung unter vermindertem Druck zur Trockne. Umkristallisation des Rückstandes aus Isopropylalkohol ergibt 4,48 g (86   O/o)    des gewünschten Hydrochlorids mit einem Smp. von   174-1750    C.



   Analyse für   Ct9H23N. HC1    berechnet:   75,600/0    C;   8, 01  /o    H;   4,64 0/0    N; gefunden:   75,89 0/o    C;   8,05 0/0    H;   4,61 0/0    N.



   Der in diesem Beispiel verwendete Ausgangsstoff kann nach dem Verfahrensschritt A und B gemäss dem Verfahren im Anschluss an Beispiel 2, jedoch unter Verwendung von   5-Chlor- 10,1    11-dihydro-5H-diben  zo(a,d) cyclohepten    als Ausgangsverbindung, hergestellt werden.



   Beispiel 4
Herstellung von   3-Chlor-1 0,11      -dihydro-5-       (3-methylarninopropyi)-5H-dibenzo(a,d)cyclohepten
3 -Chlor-1 0,1 1-dihydro5-(3 -dimethylaminopro-      pyl)-dibenzo(a,d)cyclohepten    wird gemäss Beispiel 1 A in das entsprechende Cyanamid übergeführt. Nach Hydrolyse gemäss Beispiel 1 B erhält man das gewünschte Produkt, welches durch Behandeln mit äthanolischer Salzsäure unmittelbar in das salzsaure Salz übergeführt werden kann. Nach Umkristallisation aus einer   Isopropylalkohol-Äther-Mischung    schmilzt das salzsaure Salz bei   192, 5-193, 5     C (Zers.).



   Analyse für   CtDH22NC1.      HCl    berechnet: 67,85    /o    C;   6,89 0/0    H;   4,17 0/0    N; gefunden:   68,      Olo    C; 6,94   O/o    H; 4,13   O/o    N.



   Das in diesem Beispiel als Ausgangsstoff verwendete   3-Chlor-10, 11-dihydro-5-(3-dimethylaminopropyl)      -5H-dibenzo(a,d)cyclohepten    kann nach den Verfahrensschritten A und B, wie im   Anschluss    gemäss Beispiel 2 angegeben ist, hergestellt werden, indem man von   3,5-Dichlor-10,1    1-dihydro-5H-dibenzo(a,d)cyclohepten ausgeht.



   Beispiel 5    Gemäss    Verfahren nach Beispiel 4, jedoch ausgehend von der   10,11-ungesättigten    Verbindung 3,5  Dichlor-5H-dibenzo(a,d)cyclohepten,    erhält man das entsprechende 3-Chlor-5-(3-methyl-aminopropyl)-5Hdibenzo(a,d)cyclohepten.   



  
 



  Process for the preparation of dibenzocycloheptenes

A. After evaporation of the ether on a steam bath under reduced pressure, the corresponding cycloheptene is obtained as a residue in the form of an oil.



   The free base can be converted into the oxalate by dissolving it in warm absolute ethanol, adding an equimolar amount of oxalic acid dissolved in ethanol and causing the oxalate to precipitate by adding ether. The oxalate acid salt is expediently recrystallized from a mixture of methanol and ethyl acetate.



   The hydrolysis can also be carried out under alkaline conditions as follows:
2.0 g (0.007 mol) of the cyanamide obtained according to A are poured into a hot solution of 25 g of potassium hydroxide in 38 ml of abs. Given methanol.



  The mixture is heated to reflux temperature for 45 hours while stirring, then diluted with water and the yellow oil which separates out is taken up in benzene. The benzene solution is then washed with water and, after drying with sodium sulfate, evaporated until a yellow oily residue remains. The base obtained is then taken up in n hydrochloric acid and the neutral substance is extracted with ether. After renewed treatment with alkali, the base obtained is again taken up in ether.



  After evaporation of the ether, the base 10,1-Dlhydro-5- (3-methylaminopropyl) -5H-dlbenzo (a, d) cycloheptene is obtained, which can be converted into the oxalate as indicated above.



   The hydrochloric acid salt can be obtained directly from the crude free base or from the oxalate as follows: Lithium hydroxide is added to an aqueous solution of the oxalate and the base formed is taken up in benzene. After the benzene has evaporated, the base remains in practically pure form. The base is now treated with ethanolic hydrochloric acid and the hydrochloric acid salt formed is precipitated by adding ether. After recrystallization from an ithanol-l2ither mixture, the product melts above 216 C.



   Example 2
Preparation of 5- (3-methylaminopropyl) 5H-dibenzo (a, d) cycloheptene
If one proceeds according to Example 1 A using an equivalent amount of 5- (3-dimethylaminopropyl) -5H-dibenzo (a, d) cycloheptene instead of 10, 11-dihydro 5- (3-dimethylaminopropyl) -5H-dibenzo (a , d) cycloheptene, the corresponding cyanamide is obtained. After hydrolysis according to Example 1B, 5- (3-methylaminopropyl) -5H-dibenzo (a, d) cycloheptene is obtained. The latter compound can be converted directly into the hydrochloric acid salt by treatment with ethanolic hydrochloric acid according to Example 1B and, after recrystallization from an isopropyl alcohol-ether mixture, has a melting point of 166.5-167.5 "C.



   Analysis for C10H21N. HCl calculated: 76.11 ° C; 7.40 3 / o H; 4,670 / c N; found: 7614 ° C; 7.30 0 / o H; 4.64 0 / o N.



   The raw materials used for this can be manufactured using the following method:
A. 1.08 g (0.0442 g atom) of magnesium shavings are poured into a 200 ml 3-necked flask equipped with a stirrer, dropping funnel and reflux condenser with a drying tube. During the reaction, dry nitrogen flows through the reaction vessel. An iodine crystal and then 10 ml of dry tetrahydrofuran are then added, 3 mol of a prepared 3-dimethylaminopropyl magnesium chloride solution are then added and the mixture is heated to reflux on the steam bath.



  A solution of 5.37 g (0.0442 mol) of 3-dimethylaminopropyl chloride in 35 mg of dry tetrahydrofuran is then poured in dropwise, and it is ensured that the mixture refluxes continuously. After the addition has ended, the mixture is heated to reflux for a further 2 hours, during which time most of the magnesium dissolves.



   Instead of 3-dimethylaminopropyl magnesium chloride to initiate the reaction, ethyl bromide can be used in an amount of 0.05 moles per mole of magnesium.



   B. The Grignard solution obtained in process step A is cooled to room temperature and, while stirring, a solution of 5.05 g (0.0221 mol) of 5-chloro-5H-dibenzo (a, d) cycloheptene in 25 mol of dry tetrahydrofuran is added dropwise, cooling from the outside in such a way that the room temperature is practically maintained. When the addition is complete, the mixture is refluxed for 15 minutes. The tetrahydrofuran is then distilled off under reduced pressure at a water bath temperature of 50-60.degree. The resulting syrupy mixture is dissolved in 75 ml of benzene and then 15 ml of water are added dropwise with stirring.



  The benzene layer is decanted from the gelatinous precipitate, which is extracted 3 more times with 20 ml of boiling hot benzene each time. The combined benzene extracts are washed with water and shaken three times with 15 ml of 3N hydrochloric acid each time; sodium hydroxide is added until an alkaline reaction occurs, the yellow oily base precipitating, which is taken up in hexane. The combined hexane extracts are washed with water and the hexane is evaporated. 4.61 g of a yellow oil are obtained. By treating with a mixture of 15 ml abs. Alcohol, 15 ml abs. Ether and 1.8 ml of a dry 10.28 N hydrogen chloride solution in abs. Alcohol, the base is converted into the hydrochloric acid salt.

   By diluting the solution with 25 ml abs. With this, the crystallization of the product is triggered, which is obtained in a yield of 4.17 g with a melting point of 1861 900 ° C. After recrystallization from a mixture of abs. Alcohol and abs. Ether the m.p. rises to 1911930 C.



   Analysis for C20H23N. HCI calculated: 76.53 ° C; 7.71 0/0 H; 4.46 0/0 N; found: 76.230 / 0 C; 7.83 0/0 H; 4.45 0/0 N.



   Example 3
Preparation of 10.1 1-dihydro-5- (3-methylaminopropyl) SH-dibenzo (a, d) cycloheptene
According to Example 1A, 10,11-dihydro-5- (3-dimethylaminopropyl) -SH-dibenzo (a, d) cycloheptene is converted into the corresponding cyanamide. The cyanamide (5.03 g) is then refluxed for 16 hours together with 60 ml of glacial acetic acid, 40 ml of water and 8 ml of concentrated hydrochloric acid and the reaction mixture is evaporated to dryness by heating under reduced pressure. Recrystallization of the residue from isopropyl alcohol gives 4.48 g (86 O / o) of the desired hydrochloride with a melting point of 174-1750 C.



   Analysis for Ct9H23N. HC1 calculated: 75,600 / 0 C; 8.01 / o H; 4.64 0/0 N; found: 75.89 0 / o C; 8.05 0/0 H; 4.61 0/0 N.



   The starting material used in this example can be prepared according to process steps A and B according to the process following Example 2, but using 5-chloro-10,1 11-dihydro-5H-diben zo (a, d) cycloheptene as the starting compound, getting produced.



   Example 4
Preparation of 3-chloro-1 0.11-dihydro-5- (3-methylaminopropyi) -5H-dibenzo (a, d) cycloheptene
3 -Chlor-1 0.1 1-dihydro5- (3-dimethylaminopropyl) -dibenzo (a, d) cycloheptene is converted into the corresponding cyanamide according to Example 1A. After hydrolysis according to Example 1B, the desired product is obtained, which can be converted directly into the hydrochloric acid salt by treatment with ethanolic hydrochloric acid. After recrystallization from an isopropyl alcohol-ether mixture, the hydrochloric acid salt melts at 192.5-193.5 ° C. (decomp.).



   Analysis for CtDH22NC1. HCl calculated: 67.85 / o C; 6.89% H; 4.17 0/0 N; found: 68, Olo C; 6.94 O / o H; 4.13 o / o N.



   The 3-chloro-10,11-dihydro-5- (3-dimethylaminopropyl) -5H-dibenzo (a, d) cycloheptene used as starting material in this example can be prepared according to process steps A and B, as indicated in Example 2 below , be prepared by starting from 3,5-dichloro-10,1 1-dihydro-5H-dibenzo (a, d) cycloheptene.



   Example 5 According to the method of Example 4, but starting from the 10,11-unsaturated compound 3,5 dichloro-5H-dibenzo (a, d) cycloheptene, the corresponding 3-chloro-5- (3-methyl-aminopropyl) is obtained -5Hdibenzo (a, d) cycloheptene.
