Description
Verfahren zur Herstellung von Phenylpropylamin-derivaten Gegenstand der Erfindung ist ein Verfahren zur Herstellung von basisch substituierten Phenylpropan Derivaten der Formel EMI1.1 worin R1 einen Phenyl-, Methoxyphenyl-oder 2-Pyridyl-rest, R2 Wasserstoff oder die Methylgruppe, R2 und Rq Alkylgruppen mit höchstens 4 Kohlenstoffatomen bedeuten, und deren Salze, sowie Verfahren zu ihrer Herstellung. Man erhält diese Herz- und Blutdruckwirksamen Verbindungen, indem man Verbindungen der Formel EMI1.2 worin R Wasserstoff oder die Benzylgruppe bedeutet, mit Verbindungen der Formel EMI1.3 worin X ein Sauerstoffatom oder die Gruppe EMI1.4 bedeutet, kondensiert und gleichzeitig oder anschliessend reduziert und anschliessend eine gegebenenfalls vorhandene N-Benzylgruppe abspaltet und die erhaltenen basischen Verbindungen gegebenenfalls durch Behandlung mit anorganischen. oder organischen Säuren in ihre Salze überführt. Das Verfahren zur Herstellung der Verbindungen der Formel I besteht darin, dass man Amine der Formel II mit Athylenoxyden de Formel III umsetzt, wobei die Reaktion vorteilhaft in Gegenwart eines Lösungsmittels, z. B. Äthanol, und bei erhöhter Temperatur durch geführt wird. Als Äthylenoxyde kommen beispielsweise in Frage: 1-(3, 4-Dimethylphenyl)-äthylenoxyd, l-(3 ,4-Dimethylphenyl)-2-methyl-äthylenoxyd, 1-(3 ,4-Diäthylphenyl)-äthylenoxyd, 1-(3,4-Diäthylphenyl)-2-methyl-äthylenoxyd. Eine weitere Herstellungsmöglichkeit für die bean spruchten Verbindungen, ausgehend von Aminen der Formel II, besteht darin, dass man diese in Gegenwart von Ketonen oder Aldehyden der Formel IV der Re duktion unterwirft. Als solche Ketone bzw. Aldehyde kommen beispielsweise in Betracht: 1-(3 ,4-Dimethylphenyl)-1 ,2-dioxo-propan, 1 -(3 ,4Diäthylphenyl)-1 2-dioxo-propan, 1 -(3 ,4Dimethylphenyl)-1 2-dioxo-äthan, 1-(3 ,4-Diäthylphenyl)-1 2-dioxo-äthan, 1(3 ,4-Dimethylphenyl)-glykol-(2)-aldehyd, 1-(3,4-Dimethylphenyl)-1-hydroxy-2-oxo-propan, 1-(3,4-Diäthylphenyl)-glykol-(2)-aldehyd, 1-(3,4-Diäthylphenyl)-1-hydroxy-2-oxo-propan. Die Reduktion erfolgt vorteilhaft durch katalytische Hydrierung äquimolarer Mengen beider Verbindungen in Gegenwart eines indifferenten Lösungsmittels. Als Katalysatoren verwendet man Metalle der 8. Gruppe des periodischen Systems, vorzugsweise Edelmetalle. Als Lösungsmittel eignen sich die für die Hydrierung üblichen Lösungsmittel, wie Alkohole, wässrige Alkohole oder Wasser. Es können auch Nickelkatalysatoren verwendet werden. Ebenso kann man auch zunächst die Oxo-verbindungen der Formel IV mit den Aminen der Formel II kondensieren und das Kondensat tionsprodukt anschliessend mit nascierendem Wasserstoff, z. B. mit Aluminiumamalgam und Alkohol, Natriumamalgam, Lithiumaluminiumhydrid oder Natriumborhydrid, reduzieren. Die Reduktion ist auch elektrolytisch durchführbar. Bei der Kondensation reagiert vorzugsweise die der Methylgruppe benachbarte Ketogruppe bzw. die Aldehydgruppe mit dem Amin; die andere Oxogruppe kann entweder gleichzeitig oder nachträglich in an sich bekannter Weise zur Hydroxylgruppe reduziert werden. Als Amine, die als Ausgangsstoffe der Formel II für das erfindungsgemässe Verfahren eingesetzt werden können, kommen beispielsweise in Frage: 3, 3-Diphenylpropylamin, 3- (O-Methoxy-phenyl)-3 -phenyl-propylamin, 3-(O-Hydroxy-phenyl)-3 -phenyl-propylamin, 3-(m-Methoxy-phenyl)-3 -phenyl-propylamin, 3-(m-Hydroxy-phenyl)-3-phenyl-propylamin, 3-(p-Methoxy-phenyl)-3-phenyl-propylamin, 3-(p-Hydroxy-phenyl)-3-phenyl-propylanun, 3-Phenyl-3-pyridyl-(2)-propylamin. Nach der gleichen Methode können auch solche -Hydroxyketone oder -aldehyde der Formel IV eingesetzt werden, in denen die Hydroxygruppe durch einen Alkyl-, Benzyl- oder Acylrest geschützt ist, der anschliessend in üblicher Weise abgespalten werden kann. Die Verfahrens erzeugnisse können als basische Verbindungen mit Hilfe von anorganischen oder organischen Säuren In entsprechende Salze übergeführt werden. Als anorganische Säuren kommen beispielsweise in Betracht: Halogenwasserstoffsäure wie chlorwasser- stoffsäure und Bromwasserstoffäure sowie Schwefelsäure, Phosphorsäure und Amidosulfonsäure. Als organische Säuren seien beispielsweise genannt: Essigsäure, Propionsäure, Milchsäure, Glykolsäure, Gluconsäure, Maleinsäure, Bernsteinsäure, Weinsäure, Benzoesäure, Salicylsäure, Zitronensäure, Acetursäure, Oxyäthan sulfonsäure und Athylendiamintetraessigsäure. Die Verfahrensprodukte sind wertvolle Arzneimittel und weisen eine sehr gute Herz- und Kreislaufwirkung auf. So führt z. B. die Verabreichung von 1-(3',4'-Dk methylphenyl)-2- (m - methoxy - diphenylpropylamino)- propanol-(l) im Versuch am isolierten Meerschweinchenherzen nach Langendorff bei einer einmaligen Injektion von 5 r zu einer Zunahme der Coronardurchströmung von der gleichen Grössenordnung, wie sie auch durch Verabreichung von 5 y der bekannten Verbindungen l-Phenyl-2-[3', 3'-diphenylpropyl-(1')-amino]- propan erzielt wird. Der Vorteil der neuen Verfahrensprodukte besteht jedoch vor allem darin, dass sie neben ihrer coronargefässerweiternden Wirkung eine ausgezeichnete ss-sym- pathicolytische Wirkung aufweisen. ss-Sympathicolytisch wirksame Substanzen hemmen die Wirkung von Adrenalin und Isopropylnoradrenalin, die in einer Steigerung der Herzfrequenz und der Kontraktionskraft des Herzens besteht. Adrenalin und Isopropylnoradrenalin steigern indirekt über die genannten Wirkungen aber auch direkt den Herzstoffwechsel. Eine zum ungeeigneten Zeitpunkt einsetzende Aktivierung des sympathischen Nervensystems, wofür die beiden Substanzen als Modelle dienen, kann beim Herzen zum Sauerstoffmangel führen, besonders wenn die Koronargefässe Einengungen (durch Verkalkung) aufweisen. Eine ss-sympathicolytische Wirkung schützt das Herz vor solchem starken Anstieg des Stoffwechsels. So zeigt z. B. das 1-(3',4'LDimethylphenyl)-2-(m- methoxy - diphenylpropylamino) - propanol - (1) an der Trachealkette nach Castillo mit 20 y auf 50 cm3 Tyrode lösung eine Verminderung der Wirkung von 3 y Isopropylnoradrenalin um etwa 50%. Im Vergleich dazu sind zur Erzielung der gleichen Wirkung von der bekannten Verbindung l-Phenyl 2-[3', 3'-diphenylpropyl- (l')-amino]-propan 500 y/50 cm3 Tyrodelösung g erforder- lich. Die Verfahrensprodukte weisen ausser der Herzund Kreislaufwirkung zusätzlich eine günstige Blutdrucksenkung auf. So senkt z. B. das 1-(3',4'-Dimethylphenyl)-2 [phenyl-pyridyl-(2")-propylamino]-propanol-(1)-hydrochlorid bei Ratten, die einen Nierenhochdruck haben, den Blutdruck stark und langanhaltend. Der mittlere Blutdruck betrug bei 11 Hochdruckratten durchschnittlich 192 mm Hg. 2 Stunden nach der subcutanen Gabe von 40 mg/kg des Verfahrensproduktes war der Blutdruck auf 136 mm Hg abgesunken, und 24 Stunden nach der Injektion betrug er 168 mm Hg, hatte also den Ausgangswert noch nicht wieder erreicht. Das gleiche Verfahrensprodukt zeigt ausserdem eine starke ss-sympathicolytische Wirkung. Es zeigt z. B. an der Trachealkerte nach Castiüo mit nur 2,5 y auf 50 cm3 Tyrodelösung eine Verminderung der Wirkung von 5 r Isopropylnordrenalin um etwa 50%. Am isolierten Meerschweinchenherzen nach Langendorff verminderte das gleiche Verfahrensprodukt bei Dauerinfusion von 0, 1 y pro Minute die durch 0,1 y Isopropylnoradrenalin bewirkte Zunahme der Herzfrequenz um ein Drittel. Am Meerschweinchenherzen nach Langendorff wurde durch 15 y des Verfahrensproduktes die Coronardurchströmung deutlich erhöht. Durch die Kombination der drei erwähnten Wirkungen zeichnen sich die Verfahrensprodukte durch besondere Vorteile gegenüber bisher bekannten Verbind dungen aus. Die Verfahrensprodukte sind zur Behandlung der Hypertonie besonders günstig, denn gerade bei Hypertonikern ist eine Herzkranzgefäss erweiternde Wirkung von Vorteil, da das Herz des Hochdruckkranken eine stark erhöhte Muskelmasse aufweist, die ernährt werden muss. Ausserdem neigt der Hypertoniker zu Arteriosklerose der Herzkranzgefässe, deren Folgen evtl. durch eine Erweiterung der kleinen, nicht verkalkenden Gefässe kompensiert werden kann. Beispiel 1 9,9 g y-Phenyl y-pyridyl-(2)-propylamin (Kp. o. 143 bis 1460) und 8,9 g 1-(3,4-Dimethylphenyl)-1-hydroxy- aceton [hergestellt aus dem l-Benzyloxy-1-(3, 4-dime- thyl-phenyl)-aceton durch Entbenzylieren in Methanollösung mit Wasserstoff/Palladium bei Zimmertemperatur in theoretischer Ausbeute; Fp. nach Umkristaüisie- ren aus Cyclohexan 99-100 ] werden zusammen in 50 cm3 Benzol gelöst, wobei sich die Lösung leicht erwärmt. Das Benzol wird im Vakuum abdestilliert, der Rückstand in Methanol gelöst und Natriumborhydrid in kleinen Portionen eingetragen, wobei eine lebhafte Reaktion eintritt. Man lässt etwa 20 Minuten stehen, säuert mit verdünnter Salzsäure an und destilliert das Methanol im Vakuum ab. Der Rückstand wird mit verdünnter Natronlauge und Äther geschüttelt. Die ätherische Phase wird abgetrennt, mit Kaliumcarbonat getrocknet und der Äther abdestilliert. Der Rückstand wird in wenig Methanol gelöst und 1n Salzsäure zugegeben. Nach Abdestillieren des Lösungsmittels und Umkristalli- sieren des Rückstandes aus Aceton/Äther werden 11,3 g 1-(3', 4'-Dimethyl-phenyl)-2-[phenyl-pyridyl-(2") propylamino]-propanol-(1)-hydrochlorid vom Schmp. 196-197 erhalten. Beispiel 2 Ein Gemisch aus 7,4 g 1-(3',4'-Dimethyl-phenyl)- äthylenoxyd (dargestellt nach Beilsteins Handbuch der Chemie, 4. Auflage, Bd. XVII, I. 23, Kp. lo 105-108 ) und 10,6 g γ,γ-Diphenyl-propylamin in 100 cm Äthanol wird 4 Stunden unter Rückfluss erhitzt. Nach beendeter Reaktion wird die Lösung eingeengt und mit 1n Salzsäure versetzt. Nach Abdestillieren des restlichen Lösungsmittels und Umkristallisieren des Rückstandes aus abs. Alkohol/Äther werden 5,6 g 1-(3',4'-Dimethylphenyl)-2-[3",3"-diphenyl-propyl (1")-amino]-äthanol-(1)-hydrochlorid vom Fp. 134-135 erhalten.
Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung von Phenylpropylaminderivaten der Formel EMI3.1 worin R1 einen Phenyl, Methoxyphenyl-oder Pyridyl (2)-rest, R2 Wasserstoff oder die Methylgruppe, R3 und R4 Alkylgruppen mit höchstens 4 Kohlenstoffatomen bedeuten, und deren Salze, dadurch gekennzeichnet, dass man Verbindungen der Formel EMI3.2 worin R Wasserstoff oder die Benzylgruppe bedeutet, mit Verbindungen der Formel EMI3.3 worin X ein Sauerstoffatom oder die Gruppe EMI3.4 bedeutet, kondensiert und gleichzeitig oder anschliessend reduziert und anschliessend eine gegebenenfalls vorhandene N-Benzylgruppe abspalltet.UNTERANSPRUCH Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man die erhaltenen basischen Verbindungen durch Behandlung mit anorganischen oder organischen Säuren in ihre Salze überführt.
