Description
Verfahren zur Herstellung von neuen Derivaten des Sulfanilamids Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Derivate des Sulfanilamids. Verbindungen der allgemeinen Formel I, EMI1.1 in welcher R1 eine Alkylgruppe mit 3-5 Kohlenstoffatomen oder eine Cycloalkylgruppe von 5-7 Kohlenstoffatomen bedeutet, wobei eine Cycloalkylgruppe mit 5 Kohlen stoffatomen durch eine Methyl- oder eine Äthyl gruppe und eine Cycloalkylgruppe mit 6 Kohlen stoffatomen durch eine Methylgruppe substituiert sein kann, und ihre Additionssalze mit anorganischen oder organischen Säuren sind bisher nicht bekannt geworden. Wie nun gefunden wurde, besitzen diese Verbindungen wertvolle pharmakologische Eigenschaften, insbesondere das 1-Sulfanilyl-2-imino-3-butyl-, das 1 -Sulfani- lyl-2-imino-3-tert.butyl- und das 1-Sulfanilyl-2-imino-3- -cyclohexylimidazolidin weisen bei peroraler oder parenteraler Verabreichung hypoglykämische Wirkung auf, die sie als geeignet zur Behandlung der Zuckerkrankheit charakterisieren. Die hypoglykämische Wirkung wird an Standardversuchen an Warmblütern, z.B. an Kaninchen und Ratten, nachgewiesen. In den Verbindungen der allgemeinen Formel I kann R, als niedere Alkylgruppe z.B. die Propyl-, Isopropyl-, Butyi-, sek.-Butyl-, tert.Butyl-, Isobutyl-, Pentyl-, Isopentyl-, 2,2-Dimethyl-propyl-, 1 -Methyl-butyl-, 1-Äthyl -propyl-, 1,2-Dimethyl-propylgruppe sein. Ferner kann R, beispielsweise sein: als Cycloalkylgruppe die Cyclopentyl-, I-Methyl-cyclopentyl-, 2-Methylcyclopentyl-, 3 -Methyl-cyclopentyl-, 1 -Äthyl-cyclopentyl-, 2-Äthyl-cyclopentyl-, 3 -Äthyl-cyclopentyl-, Cyclohexyl-, 1 -Methyl-cy- clohexyl-, 2-Methyl-cyclohexyl-, 3-Methyl-cyclohexyl-, 4 -Methyl-cyclohexyl oder die Cycloheptylgruppe. Nach dem erfindungsgemässen Verfahren stellt man eine Verbindung der allgemeinen Formel I her, indem man eine Verbindung der allgemeinen Formel II, EMI1.2 in welcher R1, eine Alkenylgruppe von 3-5 Kohlenstoffatomen oder eine Cycloalkenylgruppe von 5-7 Kohlenstoffato men, wobei eine Cycloalkenylgruppe mit 5 Kohlen stoffatomen durch eine Methyl- oder Äthylgruppe und eine Cycloalkenylgruppe mit 6 Kohlenstoffato men durch die Methylgruppe substituiert sein kann, und X die Aminogruppe oder einen Rest bedeutet, der durch Hydrolyse, Reduktion oder reduktive Spal tung in die freie Aminogruppe übergeführt werden kann, reduziert und gegebenenfalls zur Bildung der freien Aminogruppe hydrolysiert, reduziert oder reduktiv spaltet und gegebenenfalls eine erhaltene Verbindung mit einer anorganischen oder organischen Säure in ein Additionssalz überführt. Die Reduktion wird vorzugsweise mit Wasserstoff in Gegenwart eines Katalysators in einem Lösungsmittel vorgenommen. Geeignete Katalysatoren sind z.B. Edelmetallkatalysatoren, wie z.B. Palladium, welches beispielsweise auf Kohle eingesetzt wird und ferner Raney Nickel. Als Lösungsmittel können z.B. Alkanole, wie Methanol oder Äthanol, ferner Dioxan verwendet wer den. Es ist vorteilhaft, die Reduktion bei normalem Druck und Raumtemperatur durchzuführen. Die anschliessende Umwandlung der Gruppe X des Reaktionsproduktes in die freie Aminogruppe, welche dieses in eine Verbindung der allgemeinen Formel 1 überführt, wird je nach der Art der Gruppe X durch eine Hydrolyse, Reduktion oder reduktive Spaltung vorgenommen. Durch Hydrolyse in die freie Aminogruppe überführbare Reste X sind beispielsweise Acylaminoreste, wie z.B. die Acetamidogruppe. Ferner sind solche Reste niedere Alkoxycarbonylaminoreste, wie z.B. die Äthoxycarbonylaminogruppe, Aryloxycarbonylaminoreste, wie der Phenoxycarbonylaminorest, oder Arylmethoxycarbonylaminoreste, wie der Benzylcarbonylaminorest, oder Reste von entsprechenden Thiokohlensäurederivaten. Weitere Beispiele sind substituierte Methylenaminoreste, wie z.B. die Benzylidenamino- oder die p-Dimethylamino benzylidenaminogruppe. Die Hydrolyse zur Freisetzung der Aminogruppe kann in saurem Medium, z.B. in me thanolischer Salzsäure, oder in verdünnter wässriger Salzsäure oder Schwefelsäure erfolgen oder, falls X durch einen Alkoxycarbonylaminorest verkörpert ist, auch unter milden alkalischen Bedingungen, z.B. mittels 1-n. oder 2-n. Natronlauge, vorgenommen werden. Ein Beispiel für einen durch Reduktion in die Aminogruppe überführbaren Rest X ist die Nitrogruppe und Beispiele für solche Reste, die durch reduktive Spaltung zur Aminogruppe führen, sind die Phenylazo- oder - Dimethylamino-phenylazogruppen. Die Reduktion dieser Reste kann allgemein katalytisch, z.B. mittels Wasserstoff in Gegenwart von Raney-Nickel, Palladium- oder Platin-Kohle, in einem inerten Lösungsmittel, wie z.B. Äthanol. erfolgen. Neben diesen kommen auch andere übliche Reduktionsverfahren in Betracht, beispielsweise die Reduktion von Nitrogruppen oder die reduktive Spaltung von Azogruppen mittels Eisen in Essigsäure oder Salzsäure. Ausgangsstoffe der allgemeinen Formel II können z.B. hergestellt werden, indem man von einem 1-Phenyl- sulfonyl-aziridin ausgeht, welches durch den Rest X substituiert ist und dieses Aziridin mit einem Alkenyl- oder Cycloalkenyl-amin zu einem entsprechenden durch den Rest X substituierten N-(2-Alkenylamino-äthyl)-bzw. N -(2-Cycloalkenylamino-äthyl)-benzolsulfonamid umsetzt und dieses mit Bromcyan kondensiert und cyclisiert. Vertreter der genannten Phenylsulfonyl-aziridine sind in der Literatur beschrieben [vgl. R. Lehmann et al, Bull. Soc. Chim. Belges 55, 52-97 (1946); C.A. 41, 5475 f (19cm7)1. Weitere Aziridine von diesem Typus können analog hergestellt werden. Die nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen der allgemeinen Formel I werden anschliessend gegebenenfalls in ihre Salze mit anorganischen sowie organischen Säuren übergeführt. Die Herstellung dieser Salze erfolgt z.B. durch Umsetzung der Verbindungen der allgemeinen Formel I mit der äquivalenten Menge einer Säure in einem geeigneten wässrigorganischen oder organischen Lösungsmittel, wie z.B. Methanol, Äthanol, Diäthyläther, Chloroform oder Methylenchlorid. Zur Verwendung als Arzneistoffe können anstelle der freien Verbindungen der allgemeinen Formel I deren pharmazeutisch annehmbare Salze mit Säuren eingesetzt werden. Geeignete Additionssalze sind z. B. Salze mit Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthansulfonsäure, p-Hydroxyäthansulfonsäure, Essigsäure, Milchsäure, Oxalsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Apfelsäure, Weinsäure, Citronensäure, Benzoesäure, Salicylsäure, Phenylessigsäure, Mandelsäure und Embonsäure. Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel I und von bisher nicht beschriebenen Zwischenprodukten näher, stellen jedoch keinesweg die einzige Ausführungsform derselben dar. Die Temperaturen sind in Celsiusgraden angegeben. Beispiel I a) 28,0 g 1-Sulfanilyl-2-imino-3-allyl-imidazolidin vom Smp. 158-1600 werden in 350 ml Dioxan mit 5,0% Iger Palladiumkohle bei Raumtemperatur und Normaldruck hydriert. Nachdem die Wasserstoffaufnahme beendet ist, wird vom Katalysator abfiltriert und das Filtrat eingedampft. Man erhält das l-Sulfanilyl-2-imino-3- -propyl-imidazolidin vom Smp. 164-1660 (aus Methanol). b) Die Ausgangsverbindung, das 1-Sulfanilyl-2-imino-3-allyl-imidazolidin, erhält man ausgehend vom 4' -(Aziridin-1-ylsulfonyl)-acetanilid über die Zwischenprodukte N1 - (2 - Allylamino - äthyl) - Nt- acetylsulfanilamid (Rohprodukt) und 49 4'-(2-Imino-3 -allyl-imidazolidin- 1 -yl- sulfonyl)-acetanilid vom Smp. 247-249 (aus Methanol). Beispiel 2 Analog Beispiel 1 a) erhält man ausgehend von 32,0 g 1 -Sulfanilyl-2-imino-3-(3-cyclohexen- 1 -yl)-imidazo- lidin vom Smp. 172-1730 das 1-Sulfanilyl-2-imino-3-cyclohexyl-imidazolidin vom Smp. 181-1830. Ferner werden analog erhalten: das 1 -Sulfanilyl-2-imino-3-butyl-imidazolidin vom Smp. 179-1810; das 1 -Sulfanilyl-2-imino-3-isobutyl-imidazolidin vom Smp. 146-1470, und das 1 -Sulfanilyl-2-imino-3 -pentyl-imidazolidin vom Smp. 192-1930.
Claims (1)
- PATENTANSPRUCHVerfahren zur Herstellung von neuen Derivaten des Sulfanilamids der allgemeinen Formel I, EMI2.1 in welcher R1 eine Alkylgruppe mit 3-5 Kohlenstoffatomen oder eine Cycloalkylgruppe von 5-7 Kohlenstoffatomen be deutet, wobei eine Cycloalkylgruppe mit 5 Kohlen stoffatomen durch eine Methyl- oder eine Äthylgrup pe und eine Cycloalkylgruppe mit 6 Kohlenstoffato men durch die Methylgruppe substituiert sein kann, und ihre Additionssalze mit anorganischen oder organischen Säuren, dadurch gekennzeichnet, dass man eine Verbindung der allgemeinen Formel II, EMI3.1 in welcher Rl' eine Alkenylgruppe von 3-5 Kohlenstoffatomen oder eine Cycloalkenylgruppe von 5-7 Kohlenstoffato men,wobei eine Cycloalkenylgruppe mit 5 Kohlen stoffatomen durch eine Methyl- oder Äthylgruppe und eine Cycloalkenylgruppe mit 6 Kohlenstoffato men durch die Methylgruppe substituiert sein kann, und X die Aminogruppe oder einen Rest bedeutet, der durch Hydrolyse, Reduktion oder reduktive Spaltung in die freie Aminogruppe übergeführt werden kann, reduziert und gegebenenfalls zur Bildung der freien Aminogruppe hydrolysiert, reduziert oder reduktiv spaltet und gegebenenfalls eine erhaltene Verbindung mit einer anorganischen oder organischen Säure in ein Additionssalz überführt.
