Die Erfindung betrifft ein Verfahren zur Herstellung von neuem racemischem oder optisch aktivem 1 -(2'-Nltrilophenoxy)-2-hydroxy-3 -isopropyl- ammopropan der Formel I

Die Ausgangsverbindungen sind z. T. bereits bekannt, z. T. können sie nach üblichen Verfahren gewonnen werden, wobei man zumeist von 1-(2'-Nitrilophenoxy)-2,3-epoxypropan ausgeht. Diese Epoxyverbindung entsprechend der Formel II kann durch Umsetzung von 2-Hydroxy-benzonitril mit Epichlorhydrin in Anwesenheit von Natronlauge dargestellt und durch Reaktion mit der entsprechenden Halogenwasserstoffsäure in die Halogenhydrine der Formel II umgewandelt werden.
Die erfindungsgemäss hergestellte neue Verbindung besitzt an der -CHOH-Gruppierung ein asymmetrisches Kohlenstoffatom und kommt daher in Form des Racemats wie auch der optisch aktiven Antipoden vor. Die optisch aktiven Verbindungen können erhalten werden, indem man entweder von optisch aktiven Ausgangsverbindungen ausgeht oder das erhaltene Racemat auf übliche Weise, beispielsweise mittels Dibenzoylweinsäure oder Bromcamphersulfonsäure, in die optischen Antipoden spaltet.
Die erfindungsgemäss hergestellte Verbindung kann in üblicher Weise in ihre physiologisch verträglichen Säureadditionssalze überführt werden. Geeignete Säuren sind beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Methansulfonsäure, Maleinsäure, Essigsäure, Oxalsäure, Milchsäure, Weinsäure oder S-Chlortheophyllin.
Die erfindungsgemäss hergestellte Verbindung der Formel I (bzw. deren physiologisch verträgliche Säureadditionssalze) hat wertvolle therapeutische, insbesondere fi-adrenolytische Eigenschaften und kann daher beispielsweise zur Behandlung oder Prophylaxe von Erkrankungen der Herzkranzgefässe und zur Behandlung von Herzarrhythmien, insbesondere von Tachycardien, an Menschen eingesetzt werden.
Die Verbindung hat sich bezüglich der bradycardischen Eigenwirkung und bezüglich der aludrinantagonistischen Wirkung [Aludrin= 1 -(3',4'-Dihydroxyphenyl) 1 -hydroxy-
2-isopropylaminoäthan] im Tierexperiment am Meerschweinchen als wesentlich wirksamer und auch weniger toxisch erwiesen als das bekannte l-(1iNaphthoxy)-2-hydroxy-3-isopropyl- aminopropan. Überdies tritt die bradycardische Wirkung schon bei sehr niedrigen Dosen ein und lässt sich durch höhere Dosierung nicht mehr über einen bestimmten Grenzwert hinaus steigern, was auf ein Fehlen der unerwünschten cardiodepressiven Komponente der bradycardischen Eigenwirkung hindeutet.
Die Verbindung zeigt zudem eine starke antiarrhythmische Wirkung bei Strophantin-G-Intoxikationen. Auch die blutdrucksenkenden Eigenschaften der Verbindung sind therapeutisch interessant. Die orale Resorption ist ebenfalls besser als bei bekannten ss-Adrenolytica.
Die Einzeldosis der erfindungsgemäss hergestellten Verbindung liegt bei 1-150 mg; vorzugsweise bei 5-50 mg (oral) bzw. 1-20 mg (parenteral).
Die galenische Verarbeitung der erfindungsgemäss hergestellten Verbindung zu den üblichen Anwendungsformen wie Lösungen, Emulsionen, Tabletten, Dragees oder Depotformen kann in bekannter Weise unter Heranziehung der dafür gebräuchlichen galenischen Hilfs-, Träger-, Spreng-, Binde-, thberzugs- oder Schmiermittel, Geschmacksstoffe, Süssungsmittel, Mittel zur Erzielung eines Depoteffekts oder Lösungsvermittler geschehen. Die erfindungsgemäss hergestellte Verbindung ist auch für die Kombination mit anderen pharmakodynamisch wirksamen Stoffen wie Coronardilatatoren oder Sympathicomimetica geeignet.
Beispiel I 1 -(2'-Nitrilophenoxy)-2-hydroxy-3 -isopropyl- aminopropan HCl
26,2 g (0,15 Mol) 1-(2'-Nitrilophenoxy)-2,3-propan- epoxid werden in 150 ml Äthanol gelöst, 23,6 g (0,4 Mol) Isopropylamin zugegeben und nach 10stün- digem Stehen bei Raumtemperatur 3 Stunden am Rückfluss zum Sieden erhitzt. Das Lösungsmittel wird dann im Vakuum abdestilliert, der Rückstand mit verdünnter Salzsäure digeriert und vom Unlöslichen abgesaugt.
Die saure wässrige Phase wird mit Natronlauge alkalisch gestellt, die ölig ausfallende Base mehrmals mit Äther extrahiert, die organische Phase über Natriumsulfat getrocknet und der Äther abdestilliert. Es verbleiben 26 g rohes 1-(2'-Nitrilophenoxy)-2-hydroxy-3-isopropyl- aminopropan, das aus Essigester unter Zugabe von Petroläther umkristallisiert wird. Das Kristallisat wird in Alkohol gelöst, mit ätherischer HC1 versetzt und das sich abscheidende kristalline Hydrochlorid abfiltriert.
Fp.: 133 bis 1350 C.
Beispiel 2
23,4 g (0,1 Mol) racemisches 1-(2'-Nitrilophenoxy)2-hydroxy-3-isopropylaminopropan werden in 125 ml Methanol gelöst, eine Lösung von 38,6 g (0,1 Mol) (-)-Di-O,O-p-Toluylweinsäure in 125 ml Methanol zugegeben und mehrere Tage bei 200 C stehengelassen.
Dann werden die kristallinen Anteile abgesaugt. Das Filtrat wird mit 125 ml Wasser versetzt und wiederum stehengelassen. Es scheiden sich langsam grosse Kristalle ab, die schliesslich isoliert werden. Sie schmelzen bei 166168 C, ihr Drehwert beträgt [a] 2D = -100 .
5 g des (-)-Di-O,O-p-Toluyltartrates werden mit 100 ml Äther und 50 ml 1n Natronlauge kräftig geschüttelt und der Äther abgetrennt. Die organische Phase wird mit Wasser neutral gewaschen und über MgSO4 getrocknet. Der Äther wird abfiltriert und zum Filtrat ätherische HC1 gegeben. Das ausfallende Kristallisat wird aus Äthanol unter Zugabe von Ather um kristallisiert. Ausbeute 1,5 g, [a]9D = -28,1 , Fp. des Hydrochlorids: 131 bis 1330 C.

The compound prepared according to the invention can be converted into its physiologically tolerable acid addition salts in the customary manner. The compound has with regard to the bradycardic intrinsic effect and with regard to the aludrin-antagonistic effect [Aludrin = 1 - (3 ', 4'-dihydroxyphenyl) 1 -hydroxy-
2-isopropylaminoethane] in animal experiments on guinea pigs has been shown to be significantly more effective and also less toxic than the well-known 1- (1iNaphthoxy) -2-hydroxy-3-isopropylaminopropane.
