Abstract
ANTI-INFLAMMATORY QUINAZOLINE DERIVS. N3A- Of formula (I):- (where R degree = NO2, NH2, NH (1-4C) alkyl, N (1-4C)alkyl 2, or N-morpholino; R=H, F, Cl, Br, (1-5C) alkyl, or (1-4C) o-alkyl, with the proviso that R is not equal H if R degree =NO2 or NH2; R'=(1-5C) alkyl, allyl or propargyl with the proviso that R'=alkyl if R degree=NH alkyl; and R2=Ph or C6H3(Y)n, where Y=H, F, Cl, Br, (1-4C) alkyl, CF3, or (1-4C) o-alkyl, and n=1 or 2 with the proviso that not more than one CF3 group is present) and salts of I, are useful anti-inflammatories at 20-2000 mg/day. - They are prepared by a variety of standard routes.

worin R1 , R, R1' und R" obige Bedeutung besitzen, mit einem Alkylcarbamat, worin die Alkylgruppe 1-5 Kohlenstoffatome besitzt, in Gegenwart einer kataly tischen Menge Lewis-Säure umsetzt.
Die Umsetzung wird zweckmässigerweise bei Tem peraturen zwischen 160 und 200 C während einer bis 4 Std. in einem inerten organischen Lösungsmittel, beispielsweise o-Dichlorbenzol, in Gegenwart einer ka talytischen Menge einer Lewis-Säure, beispielsweise Zinkchlorid, durchgeführt. Das inerte organische Lö sungsmittel kann hierbei durch einen Überschuss des Carbamats ersetzt werden.
Die erfindungsgemäss erhaltenen Verbindungen der allg. Formel I werden nach erfolgter Umsetzung aus den entsprechenden Reaktionsgemischen auf an sich bekann te Weise, beispielsweise durch Filtrieren, Extraktion, Eindampfen der entsprechenden Lösungen, Salzbildung der basisch substituierten Verbindungen der allg, For mel I usw., isoliert und gegebenenfalls anschliessend auf an sich bekannte Weise, beispielsweise durch Umkri stallisation, gereinigt.
Aus den erhaltenen Salzen können die basisch sub stituierten Verbindungen der allg. Formel I durch Be handlung mit wässrigen Alkalimetallhydroxid-Lösungen freigesetzt werden. Die erfindungsgemäss erhaltenen basisch substituier ten Verbindungen der allg. Formel I können gegebenen falls anschliessend durch Umsetzung mit geeigneten Säu ren in ihre Säureadditionssalze übergeführt werden.
Die als Ausgangsverbindungen verwendeten Verbin dungen der all-. Formel III können auf an sich be kannte Weise hergestellt werden. Falls Verbindungen der all-. Formel III, d.i. o-Alkylaminobenzophenone, hergestellt werden sollen, worin die Alkylgruppe eine verzweigte Alkylgruppe darstellt und die Verzweigung am Kohlenstoffatom stattfindet, das direkt an das Stick stoffatom gebunden ist, insbesondere, falls man o-Iso- propylaminobenzophenon herstellen will, ist es günstig, entsprechend substituierte 2-Aminobenzophenone di rekt mit entsprechenden Alkyljodiden oder -bromiden in an sich bekannter Weise zu alkylieren. Die Umset- zun- erfolgt vorzugsweise in Gegenwart einer Base, z.B.
eines Alkalicarbonats, und in einem organischen Lö sungsmittel, wie Dioxan, Benzol oder Toluol. Zweck- mässigerweise arbeitet man bei Temperaturen zwischen 70 und 140 C, vorzugsweise 80 - 100 C. Verbindungen der allg. Formel III, die einen 5-Nitro-Substituenten be sitzen, können vorzugsweise durch Umsetzung des ent sprechenden 5-Nitro-2-chlorbenzophenons mit einem ge eigneten Amin (R1'-NH2) in Gegenwart eines geeigneten Katalysators, beispielsweise eines Gemisches von Kup fer und Kupferschlorid, hergestellt werden.
Diejenigen Verbindungen der allg. Formel I, worin R1 für eine N-Dialkylamino- oder N-Morpholino-Grup- pe steht, können ebenfalls in Form ihrer Säureaddi tionssalze erhalten werden. Für die Bildung der Säuread- ditionsalze haben sich die Chlorwasserstoffsäure, Fumar- säure, Maleinsäure, Ameisensäure, Essigsäure, Sulfon- säure und Malonsäure als geeignet erwiesen.
Die erfindungsgemäss erhaltenen Verbindungen der allg. Formel I besitzen ausserordentlich günstige phar- makodynamische Eigenschaften. Insbesondere besitzen diese Verbindungen, wie den Resultaten beim Carraghen- induzierten Ödemtest bei Ratten zu entnehmen ist, eine ausserordentlich günstige entzündungshemmende Wir kung. Die täglich zu verabreichende Menge soll zwi schen 20 und 2000 g betragen, die vorzugsweise in meh reren (2-4) täglichen Dosen von 5-1000 mg oder in Re- tardform verabreicht werden.
Die erfindungsgemäss hergestellten Verbindungen körnen als Arzneimittel allein oder in entsprechenden Arzneiformen für orale oder parenterale Verabreichung verwendet werden. Zwecks Herstellung geeigneter Arz neiformen werden diese mit anorganischen oder orga nischen, pharmakologisch indifferenten Hilfsstoffen ver arbeitet.
Diejenigen Verbindungen der allg. Formel I, worin R1 eine N-Dialkylamino- oder N-Morpholino-Gruppe bedeutet, können ebenfalls in Form ihrer Säureadditions salze verwendet werden, wobei die Salze die gleiche Ak tivität besitzen wie die freien Basen.
Bevorzugt werden diejenigen Verbindungen der allg. Formel I, worin R1' für eine Isopropylgruppe steht. Falls in der all-. Formel I R, R1 oder Y für eine Alkyl- bzw. Alkoxygruppe stehen, besitzen die Alkyl- bzw. die Alkoxygruppen jeweils 1-3 bzw. 1 oder 2 Koh lenstoffatome.
Ausgangsverbindungen, deren Herstellung nicht be schrieben wird, sind entweder bekannt oder können auf an sich bekannte Weise hergestellt werden. Verfahren zur Herstellung dieser Ausgangsverbindungen sind in den belgischen Patenten Nrn. 714 568; 723 041, 728 869 sowie 735 344 beschrieben.
In den nachfolgenden Beispielen, welche die Ausfüh rung des Verfahrens erläutern, die Erfindung aber in keiner Weise einschränken sollen, erfolgen alle Tem peraturangaben in Celsiusgraden und sind korrigiert.
<I>Beispiel 1</I> 1-Isopropyl-7-methyl-6-nitro-4-phenyl- -2(1H)-chinazolinon a) 2-Isopropylamino-4-methyl-5-nitro-benzophenon Ein Gemisch aus 15g 2-Chlor-4-methyl-5-nitroben- zophenon, 700 mg Kupferpulver, 700m- Kupferchlorid, 15 ml Äthanol und 15 ml flüssigem Isopropylamin wird während 20 Stunden am Rückfluss zum Sieden erhitzt. Danach werden 200m1 Methylenchlorid zugefügt, vom anorganischen Material abfiltriert und das Filtrat mit 100m1 Wasser gewaschen.
Die organische Phase wird über Natriumsulfat getrocknet, filtriert und im Vakuum zur Trockne eingedampft, wobei das 2-Isopropylamino- -4-methyl-5-nitro-benzophenon als Rückstand erhalten wird.
b) 1-Isopropyl-7-methyl-6-nitro-4-phenyl-2(IH)- -chinazolinon Ein Gemisch aus 37g 2-Isopropylamino-4-methyl-5- -nitro-benzophenon, 45 g Urethan und 1,7 g Zinkchlorid wird während 4 Stunden im Ölbad auf 180-200 C er hitzt. Danach wird auf Raumtemperatur abgekühlt und mit 220m1 Methylenchlorid versetzt. Das erhaltene Ge misch wird filtriert und das Filtrat 2mal mit je 110m1 Wasser gewaschen. Danach wird die organische Phase über Natriumsulfat getrocknet, filtriert und im Vakuum zur Trockne eingedampft.
Der Rückstand wird aus Äthylacetat umkristallisiert, wobei man das 1-Isopropyl- -7-methyl-6-nitro-4-phenyl-2(1H)-chinazolinon vom Smp. 192-194 C erhält.
<I>Beispiel 2</I> In zu Beispiel 1 analoger Weise gelangt man unter Verwendung entsprechender Ausgangsprodukte zu fol genden Verbindungen: 2 a) 7-Dimethylamino-l-isopropyl-4-phenyl-2(1H)- -chinazolinon; Smp. 181-183 C.
2 b) 6-Dimethylamino-l-isopropyl-7-methyl-4-phenyl- -2(1H)-chinazolinon; Smp. 184-186 C.
2 c) 6-Dimethylamino-l-isopropyl-4-phenyl-2(1H)- -chinazolinon; Smp. 167-168 C.
2 d) 6-Diisopropylamino-l-isopropyl-4-phenyl-2(1H)- -chinazolinon; Smp. 225 C (Zers.).
2 e) 6-(N-Äthyl-N-isopropyl)-amino-l-isopropyl-4- -phenyl-2(1H)-chinazolinon; Smp. 165 C.
2 f) 1-Isopropyl-6-morpholino-4-phenyl-2(1H)-china- zolinon; Smp. 172-175 C.

The compounds of general formula I obtained according to the invention have extraordinarily favorable pharmacodynamic properties. In particular, as can be seen from the results of the Carraghen-induced edema test in rats, these compounds have an extremely favorable anti-inflammatory effect. The amount to be administered daily should be between 20 and 2000 g, which are preferably administered in several (2-4) daily doses of 5-1000 mg or in retarded form.
The compounds prepared according to the invention can be used as medicaments alone or in corresponding medicament forms for oral or parenteral administration. In order to produce suitable medicament forms, these are processed with inorganic or organic, pharmacologically indifferent auxiliaries.
Those compounds of the general formula I in which R1 is an N-dialkylamino or N-morpholino group can also be used in the form of their acid addition salts, the salts having the same activity as the free bases.
Those compounds of the general formula I are preferred in which R1 'is an isopropyl group. If in the all-. Formula I R, R1 or Y represent an alkyl or alkoxy group, the alkyl or the alkoxy groups each have 1-3 or 1 or 2 carbon atoms.
Starting compounds, the preparation of which is not described, are either known or can be prepared in a manner known per se. Processes for the preparation of these starting compounds are described in Belgian Patent Nos. 714 568; 723 041, 728 869 and 735 344.
In the following examples, which explain the execution of the process but are not intended to restrict the invention in any way, all temperatures are given in degrees Celsius and have been corrected.
<I> Example 1 </I> 1-Isopropyl-7-methyl-6-nitro-4-phenyl- -2 (1H) -quinazolinone a) 2-Isopropylamino-4-methyl-5-nitro-benzophenone A mixture of 15 g of 2-chloro-4-methyl-5-nitrobenzophenone, 700 mg of copper powder, 700 mg of copper chloride, 15 ml of ethanol and 15 ml of liquid isopropylamine are refluxed for 20 hours. Then 200 ml of methylene chloride are added, the inorganic material is filtered off and the filtrate is washed with 100 ml of water.
The organic phase is dried over sodium sulfate, filtered and evaporated to dryness in vacuo, the 2-isopropylamino--4-methyl-5-nitro-benzophenone being obtained as a residue.
b) 1-Isopropyl-7-methyl-6-nitro-4-phenyl-2 (IH) - -quinazolinone A mixture of 37g 2-isopropylamino-4-methyl-5--nitro-benzophenone, 45 g urethane and 1, 7 g of zinc chloride is heated to 180-200 C in an oil bath for 4 hours. It is then cooled to room temperature and mixed with 220 ml of methylene chloride. The mixture obtained is filtered and the filtrate is washed twice with 110 ml of water each time. The organic phase is then dried over sodium sulfate, filtered and evaporated to dryness in vacuo.
The residue is recrystallized from ethyl acetate, 1-isopropyl-7-methyl-6-nitro-4-phenyl-2 (1H) -quinazolinone with a melting point of 192-194 ° C. being obtained.
<I> Example 2 </I> In a manner analogous to Example 1, using appropriate starting materials, the following compounds are obtained: 2 a) 7-dimethylamino-1-isopropyl-4-phenyl-2 (1H) - -quinazolinone; 181-183 C.
2 b) 6-dimethylamino-1-isopropyl-7-methyl-4-phenyl--2 (1H) -quinazolinone; 184-186 C.
2 c) 6-dimethylamino-1-isopropyl-4-phenyl-2 (1H) - -quinazolinone; M.p. 167-168 C.
2 d) 6-diisopropylamino-1-isopropyl-4-phenyl-2 (1H) - -quinazolinone; M.p. 225 C (dec.).
2 e) 6- (N-ethyl-N-isopropyl) -amino-1-isopropyl-4-phenyl-2 (1H) -quinazolinone; M.p. 165 C.
2 f) 1-isopropyl-6-morpholino-4-phenyl-2 (1H) -quinazolinone; 172-175 C.
2 g) 1-isopropyl-7-methyl-6-morpholino-4-phenyl--2 (1H) -quinazolinone; M.p. 218-221 C.
2 h) 7-chloro-6-dimethylamino-1-isopropyl-4-phenyl--2 (1H) -quinazolinone; 160-163 C.
