Title derivs. of formula (I) (where R is methyl, ethyl or propyl and R2 is H, unbranched 1-4C alkyl, isopropyl or allyl) and their acid addition salts, which are useful in the treatment of tension and anxiety states, are prepd. by reducing a cpd. (I; R1 = R3) (where R3 is an alkanoyl residue corresponding to R1) with diborane (e.g. generated from NaBH4 and BF3 etherate) in an ether type solvent and opt. converting the product into an acid addition salt.

  Verfahren zur Herstellung von neuen Azepinderivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Azepinderivate der allgemeinen Formel I,

 in welcher   Rt    die Methyl-,   Äthyl-    oder die Propylgruppe R2 Wasserstoff, eine niedere unverzweigte Alkylgruppe mit 1-4 Kohlenstoffatomen, eine Isopropyl- und die Allylgruppe bedeuten sowie ihrer Additionssalze mit anorganischen oder organischen Säuren.



   Solche Verbindungen, insbesondere Verbindungen, deren Rest R2 eine niedere Alkylgruppe bedeutet, sowie die Additionssalze solcher Verbindungen, besitzen interessante pharmakologische Eigenschaften und einen hohen therapeutischen Index. Sie wirken bei peroraler, rektaler und parenteraler Verabreichung zentraldämpfend, z. B. vermindern sie die Motilität, potenzieren die Wirkung von Narcotica, antagonisieren die Wirkung von Amphetamin, wirken beim  test de la traction , wirken antiemetisch, serotoninantagonistisch an der Rattenpfote, analgetisch im Strecktest und senken die Körpertemperatur. Ferner weisen sie Antihistamin-Wirkung auf. Diese Wirkungsqualitäten, welche durch ausgewählte Standardversuche [vg. R. Domenjoz und W. Theobald, Arch.Int.Pharmacodyn. 120, 450 (1959), W. Theobald et al., Arzneimittelforsch. 17, 561 (1967) und W. Theobald und R.

  Domenjoz, Arzneimittelforsch. 8, 18 (1958)] erfasst werden, charakterisieren die Verbindungen als geeignet zur Behandlung von Spannungs- und Erregungszuständen.



   In den Verbindungen der allgemeinen Formel I ist R2 beispielsweise als Alkylgruppe: die Methyl-, Äthyl-, Propyl-, oder die Butylgruppe.



   Verbindungen der allgemeinen Formel I stellt man her, indem man eine Verbindung der allgemeinen Formel II,

 in welcher   R2    die unter Formel I angegebene Bedeutung und   R3    einen entsprechenden Alkanoylrest bedeu  tet, mittels Diboran, in einer ätherartigen Flüssigkeit reduziert und die erhaltene Verbindung der allgemeinen Formel I gegebenenfalls in ein Additionssalz mit einer anorganischen oder organischen Säure überführt.



   Als Reaktionsmedium dient beispielsweise Tetrahydrofuran, Dioxan, Methylenglykoldimethyläther oder Diäthylenglykoldimethyläther. Die Reaktionstemperatur liegt vorzugsweise zwischen Raumtemperatur und ca.   100",    und die Reaktionsdauer zwischen ca. 30 Minuten und 25 Stunden. Das Diboran wird z. B. aus Bortrifluirid-ätherat und Natriumborhydrid entweder in einer separaten Apparatur entwickelt und in das Reaktionsgemisch eingeleitet oder in situ gebildet.



   Ausgangstoffe der allgemeinen Formel II, in denen   R3    als Alkanoylrest die Formyl-, Acetyl- oder Propionylgruppe bedeuten kann, werden dargestellt, indem man   5-Alkanoyl-10,1 1-bis-brommethyl-5H-dibenz[b,-    f]azepinverbindungen der allgemeinen Formel III

 mit einem Amin der allgemeinen Formel IV,

 worin R2 die angegebene Bedeutung hat, umsetzt.



   Die   5-Alkanoyl-1 0,11 -bis-brommethyl-5H-dibenz-    [b,f]azepin-verbindung der allgemeinen Formel III wird z. B. erhalten, wenn man das 10,11-Dimethyl   5H-dibenz[b,f]azepin    (vgl. Geigy A.G., U.S. Patentschrift No.   3 130 191)    mit einem Alkanoylchlorid zum   5-Alkanoyl-10,11-dimethyl-5H-dibenz[b,f]    azepin acyliert und das Reaktionsprodukt mit N-Brom-succinimid bromiert.



   Die nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen der allgemeinen Formel I werden anschliessend gewünschtenfalls in üblicher Weise in ihre Additionssalze mit anorganischen und organischen Säuren übergeführt. Beispielsweise versetzt man eine Lösung einer Verbindung der allgemeinen Formel I in einem organischen Lösungsmittel mit der als Salzkomponente gewünschten Säure oder mit einer Lösung derselben. Vorzugsweise wählt man für die Umsetzung organische Lösungsmittel, in denen das entstehende Salz schwer löslich ist, damit es durch Filtration abgetrennt werden kann. Solche Lösungsmittel sind z. B.



  Methanol, Aceton, Methyläthylketon, Aceton- Äthanol,   Methanol-Äther    oder   .3ithanol-laither.   



   Zur Verwendung als Arzneistoffe können anstelle freier Basen pharmazeutisch annehmbare Säureadditionssalze eingesetzt werden, d. h. Salze mit solchen Säuren, deren Anionen bei den in Frage kommenden Dosierungen nicht toxisch sind. Ferner ist es von Vorteil, wenn die als Arzneistoffe zu verwendenden Salze gut kristallisierbar und nicht oder wenig hygroskopisch sind. Zur Salzbildung mit Verbindungen der allgemeinen Formel I können z. B. die Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthansulfonsäure, ss-Hydroxyäthansulfonsäure, Essigsäure, Äpfelsäure, Weinsäure, Citronensäure, Milchsäure, Oxalsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Benzoesäure, Salicylsäure, Phenylessigsäure, Mandelsäure und Embonsäure verwendet werden.



   Die neuen Wirkstoffe werden, wie weiter vorne erwähnt, peroral, rektal oder parenteral verabreicht. Die Dosierung hängt von der Applikationsweise, der Spezies, dem Alter und von dem individuellen Zustand ab.



  Die täglichen Dosen der freien Basen oder von pharmazeutisch annehmbaren Salzen derselben bewegen sich zwischen 0,1 mgikg und 10,5 mg/kg für Warmblüter. Geeignete Doseneinheitsformen, wie Dragees, Tabletten, Suppositorien oder Ampullen enthalten vorzugsweise 5-200 mg eines erfindungsgemässen Wirkstoffes oder eines pharmazeutisch annehmbaren Salzes derselben.



   Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel I und von bisher nicht beschriebenen Zwischenprodukten näher, sollen jedoch den Umfang der Erfindung in keiner Weise beschränken. Die Temperaturen sind in Celsiusgraden angegeben.



   Beispiel 1 a) 0,64 g Diboran, frisch hergestellt aus 2,6 g Natriumborhydrid und 13 g Bortrifluorid-ätherat (G.



  Zweifel and G.C. Brown, Organic Reactions, vol.



  XIII, S.32) werden mit Stickstoff in eine Lösung von 13,5 g   2-Äthyl-8-acetyl-1,2,3,8-tetrahydrodibenzo [b,f]-    pyrrolo[3,4-d]azepin in 70 ml abs. Tetrahydrofuran eingeleitet, wobei durch Eiskühlung die Temperatur zwischen 0 und   S"gehalten    wird. Anschliessend wird noch eine Stunde bei   0-5"    und eine weitere Stunde bei   20-25     gerührt. Hierauf tropft man unter Eiskühlung 30 ml 100/oige Natriumphosphatlösung zu und dampft anschliessend das Tetrahydrofuran im Rotationsverdampfer ab. Der Rückstand wird nun während einer Stunde mit 300 ml 4 n Salzsäure unter Rückfluss zum Sieden erwärmt und nach dem Abkühlen auf   20O    mit konz. Natronlauge phenolphtalein-alkalisch gestellt und die ausgefallene Base mit Äther extrahiert. 

  Die   ätheri-    sche Lösung wäscht man mit Wasser, trocknet sie über Kaliumcarbonat und engt sie auf ein kleines Volumen ein, wobei das 2,8-Diäthyl-1,2,3,8-tetrahydrodibenzo [b,f]pyrrolo [3,4-d] azepin auskristallisiert. Das Rohprodukt wird aus Äthanol umkristallisiert, Smp. 100   102O.    5,8 g der erhaltenen Base löst man in 40 ml abs.



  Aceton und gibt   4 mol    5n abs. äthanolische Salzsäure zu, worauf das Hydrochlorid auskristallisiert; Smp. nach dem Umkristallisieren aus Isopropanol   267-271     (Zers.).  



   Der Ausgangsstoff, das 5-Acetyl-10,11-bis-brom   methyl-5H-dibenz[b,f]azepin,    wird wie folgt hergestellt: b) Zu einer Lösung von 98 g 10,11-Dimethyl-SHdibenz[b,f]azepin (vgl.   Geigy    A.G., USA Patentschrift No. 3 130 191) vom Smp.   131-132 in    295 ml Toluol lässt man innerhalb 30 Minuten unter Rühren 39,5 g Acetylchlorid zutropfen. Man kocht anschliessend das Reaktionsgemisch 5 Stunden unter Rückfluss, dampft es im Vakuum ein und löst den Rückstand in Äther.



  Die ätherische Lösung wird mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Der Rückstand, der aus Petroläther umkristallisiert wird, liefert das   5-Acetyl-10,1 1-dimethyl-5H-dibenz[b,f]azepin    vom Smp.   109-111 ;    c) 101 g   5-Acetyl-10,11-dimethyl-5H-dibenz[b,f] a-    zepin werden in einem Liter Tetrachlorkohlenstoff gelöst und mit 138 g N-Brom-succinimid versetzt. Unter Rühren und Belichten mit zwei 200 Watt-Lampen oder einer UV-Lampe wird das Gemisch zum Sieden erwärmt. Man hält solange im Sieden, bis alles N-Bromsuccinimid umgesetzt ist. Hierauf kühlt man das Reaktionsgemisch auf   0     ab und lässt 200 ml Wasser zulaufen.

  Das ausgefallene kristalline   5-Acetyl-lO,l l-bis-brommethyl-SH-dibenz[b,f]    azepin vom Smp.   175-176     wird abgesaugt;
Analog zu Beispiel   la)    werden erhalten: a) das 2,8-Dimethyl-1,2,3,8-tetrahydrodibenzo   [b,f]pyrrolo[3,4-d]azepm,    SmP.   120-121 ,    Oxalat, Smp.   230-232 ;    b) das   2,-Athyl-8-methyll,2,3,8-tetrahydro-    dibenzo[b,f]pyrrolo[3,4-d] azepin, Oxalat, Smp.   208-210     (aus Methanol), c) das   2-Allyl-8-methyl-1,2,3,8-tetrahydro-    dibenzo   [b,f]pyrrolo[3,4-d]azepm,    Smp.   137-139"(aus    abs. Äthanol), Methansulfonat, Smp.   194-296     (aus abs. Äthanol);

  ;
Beispiel 2 a) Analog Beispiel   1a)    wird hergestellt:
Aus 8 g   2-Sithyl-8-formyl-1,2,3,8-tetrahydro-      dibenzo[b,f]pyrrolo[3,4-d]azepin;    Smp.   107-109     (aus Essigester); Hydrochlorid: Smp.   239-2430    (aus Isopropanol); Ausbeute 6 g   (78 /o    der Theorie).



   Das als Ausgangsprodukt benötigte   2-Äthyl-8-formyl-1 ,2,3,8-tetrahydro- dibenzo [b,f] pyrroloi3,4-d] azepin    wird wie folgt hergestellt: b) 10 g 2-Äthyl-1,2,3,8-tetrahydro   dibenzo[b,f]pyrrolo[3,4-d]azepin    und 100 ml 990/oige Ameisensäure werden unter Rühren während 2 Stunden am Rückfluss gekocht. Anschliessend wird die überschüssige Ameisensäure im Vakuum aus dem Reaktionsgemisch abdestilliert. Den Rückstand löst man in Wasser und fällt mit konz. Ammoniak die Base, welche in   Äther    aufgenommen wird. Die Ätherlösung wird mit Wasser gewaschen, über Kaliumcarbonat getrocknet und eingedampft. 

  Der Rückstand, das   2-Äthyl-8-formyl-1 ,2,3,8-tetrahydro-      dibenzo [b,f] pyrrolo [3,4-d]azepin    ist ein viskoses öl, Rohprodukt, 10 g der erhaltenen rohen Base werden in   30ml    abs. Aceton gelöst und mit   6,9 mol    5n abs. äthanolsicher Salzsäure versetzt, worauf das Hadrochlorid auskristallisiert. Nach dem Umkristallisieren aus Methanol schmilzt die Substanz bei   245-250     (Zersetzung).

   The 5-alkanoyl-1 0.11 -bis-bromomethyl-5H-dibenz- [b, f] azepine compound of the general formula III is z. B. obtained if the 10,11-dimethyl 5H-dibenz [b, f] azepine (cf. Geigy AG, US Pat.    Example 1 a) 0.64 g of diborane, freshly prepared from 2.6 g of sodium borohydride and 13 g of boron trifluoride etherate (G.

The residue, which is recrystallized from petroleum ether, gives 5-acetyl-10,1-1-dimethyl-5H-dibenz [b, f] azepine with a melting point of 109-111; c) 101 g of 5-acetyl-10,11-dimethyl-5H-dibenz [b, f] azepine are dissolved in one liter of carbon tetrachloride, and 138 g of N-bromosuccinimide are added. The mixture is heated to the boil with stirring and exposure to two 200 watt lamps or a UV lamp. The mixture is kept at the boil until all the N-bromosuccinimide has been converted. The reaction mixture is then cooled to 0 and 200 ml of water are run in.

  The precipitated crystalline 5-acetyl-10, l-bis-bromomethyl-SH-dibenz [b, f] azepine with a melting point of 175-176 is filtered off with suction;

PATENTANSPRUCH PATENT CLAIM Verfahren zur Herstellung von neuen Azepinderivaten der allgemeinen Formel I, EMI3.1 in welcher Rt die Methyl-, Äthyl- oder die Propyl-gruppe, R2 Wasserstoff, eine niedere unverzweigte Alkylgruppe mit 1-4 Kohlenstoffatomen, eine Isopropyl- und die Allyl-gruppe bedeuten sowie ihre Additionssalze mit anorganischen oder organischen Säuren dadurch gekennzeichnet, dass man eine Verbindung der allgemeinen Formel II, EMI3.2 in welcher R2 die unter Formel I angegebene Bedeutung und R3 einen entsprechenden Alkanoylrest bedeuten, mittels Diboran, in einer ätherartigen Flüssigkeit reduziert und die erhaltenen Verbindungen der allgemeinen Formel I gegebenenfalls in ein Additionssalz mit einer anorganischen oder organischen Säure überführt.
