Verfahren zur Herstellung von heterocyclischen Verbindungen
Die vorliegende Erfindung betrifft die Herstellung von heterocyclischen Verbindungen mit dem Ringgerüst der Formel I

wobei die Kohlenstoffatome des Benzolringes Bestandteil eines weiteren Ringes sein können, worin X ein Sauerstoff- oder vorzugsweise ein Schwefelatom bedeutet, welches in 3-Stellung eine freie oder funktionell abgewandelte Carboxylgruppe oder eine Acylgruppe und in 4-Stellung eine Oxo-, Thiono- oder Iminogruppe enthalten, sowie Salzen von solchen Verbindungen.
Eine funktionell abgewandelte Carboxylgruppe ist z. B. eine Cyangruppe, sowie eine Carbamoyl- oder Hydrazinocarbonylgruppe, worin die Stickstoffatome gegebenenfalls, z. B. durch Niederalkyl-, wie Methyloder ;Tthylgruppen, mon oder poly-, z. B. disubstituiert sein kann, in erster Linie aber eine veresterte Carboxylgruppe, insbesondere eine Carbo-niederalkoxygruppe, wie z. B. eine Carbomethoxy- oder Carbäthoxygruppe. Ein Acylrest ist z. B. eine Alkanoylgruppe, insbesondere eine Niederalkanoyl-, in erster Linie eine Acetyl-, sowie eine Formyl-, Propionyloder n-Butyrylgruppe.
Die neuen Verbindungen können auch weitere Sub- stituenten enthalten. So können sie z. B. in 2-Stellung durch einen aliphatischen Rest, insbesondere einen nie deraliphatischen Kohienwasserstoffrest, wie eine Niederalkyl-, z. B. Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-33ut1- eder Tsobutylgruppe, oder eine Niederalke -nyl-, z. B. Allyl- oder Methallylgruppe, oder durch einen cycloaliphatischen Kohlenwasserstoffrest, wie eine -Cycloalkyl- oder Cyclo alkyl-niederalkylgruppe mit 3-8, vorzugsweise 5-6 Ringkohlenstoffatomen, z. B.
eine Cyclopentyl-, Cyclohexyl-, Cyclopentyl-methyloder Cyclohexyl-methylgruppe, substituiert sein. Der Substituent in 2-Stellung kann auch einen aromatischen Rest, wie eine Phenylgruppe, oder einen araliphatischen Rest, wie eine Phenyl-niederalkyl-, z. B.
Benzylgruppe, bedeuten, wobei der aromatische Ring Substituenten, wie z. B. Niederalkylreste, Niederalk oxy-, z. B. Methoxy-, Athoxy- oder Propyloxygruppen, Halogen-, z. B. Fluor-, Chlor- oder Bromatome, Trifluormethylgruppen oder Nitrogruppen enthalten kann.
Der aromatische Teil des Moleküls kann unsubstituiert oder durch einen oder mehrere gleiche oder verschiedene Substituenten, welche irgendeine der zur Substitution geeignete Stelle einnehmen können, substituiert sein. Substituenten sind z. B. Niederalkylreste, wie die oben erwähnten Gruppen, Niederalkenylgruppen, wie die oben erwähnten Reste, Niederalkoxygruppen, wie die oben erwähnten Gruppen, Niederalkylendioxy-, wie Methylendioxygruppen, Aryloxy-, wie Phenyloxygruppen, Aryl-niederalkoxy-, wie Phenyl-nieder alkoxy-, z. B. Benzyloxygruppen, Niederalkylmercapto-, z. B. Methylmercaptogruppen, Halogenatome, wie die oben erwähnten Halogenatome, Trifluormethyl-, Nitrc, freie oder substituierte Aminogruppen, wie Amino-, Niederalkylamino, z. B.
Methylamino-, Äthylamino- oder Isopropylamino-, Di-niederalkylamino-, z. B. Dimethylamino- oder Diäthylamino-, Acylamino-, wie Niederalkanoylamino-, z.B. Acetylaminoreste, Carbamoylamino- oder Thiocarbamoylaminogruppen, wie N-unsubstituierte oder N-substituierte, z. B. N-Phenyl- oder N-Niederalkyl-substituierte Carb amoylaminow oder Thiocarbamoylamino-, oder Sulfonylamino-, z. B. Fhenylsulfonylarninogruppen, freie oder funktionell abgewandelte Carboxylgruppen, wie die oben erwähnten Reste, freie oder funktionell abgewan- delte Sulfogruppen, wie z. B. Sulfamoylgruppen, oder Aryl- oder Aryl-niederalkyl-, z. B. Phenyl-, Benzyloder Phenyläthylgruppen, oder bivalente aliphatische Reste, z.
B. bivalente aliphatische Kohlenwasserstoffreste, welche vorzugsweise benachbarte Stellungen des aromatischen Teils des Moleküls substituieren können, wie Niederalkylenreste, vorzugsweise mit 4 Kohlenstoffatomen, z.B. der 1,4-Butylenrest, oder Niederalkenylenreste, vorzugsweise mit 4 Kohlenstoffatomen, z. B. der 1- oder 2-Buten-1,4-ylen- oder insbesondere der 1,3-Butadien-1,4-ylenrest, wobei solche Reste, insbesondere Aryl-, wie Phenylgruppen, oder bivalenten Reste, in erster Linie ein 1,3-Butadien-1,4-ylenrest, in der oben erwähnten Weise substituiert sein können.
Die neuen Verbindungen der vorliegenden Erfindung besitzen wertvolle pharmakologische Eigenschaften. Ausser ihren Wirkungen gegen Pilze, insbesondere gegen Trichophyton-Arten, zeigen sie Antivirus-Wirkungen z. B. gegen Sindbis-Virus, die mittels Experimenten an Versuchstieren, wie Hamstem nachgewiesen werden können, und in erster Linie Antiinfluenza-Wirkungen, die besonders prophylaktischer Natur sein können, z. B. gegen das Influenza-PR8-Virus, welche mittels Experimenten an Versuchstieren, wie Mäusen, bei subkutaner oder oraler Verabreichung nachgewiesen werden können. Sie können deshalb als pharmakologisch, insbesondere antiviral wirksame Mittel, verwendet werden, wobei sie vor oder nach einer entsprechenden Infektion angewendet werden können.
Ferner können sie auch als Zwischenprodukte in der Herstellung von anderen wertvollen, insbesondere von pharmakologisch aktiven Verbindungen dienen.
Besonders wertvoll in bezug auf ihre Antivirus-, wie Antiinfluenza-Wirkungen, sind Verbindungen der Formel II

Im Hinblick auf ihre ausgeprägte Antivirus-Wirkungen, insbesondere ihre Antiinfluenza-Wirkungen gegen das PPs8-Virus, welche durch Experimente mit Versuchstieren, wie Mäusen nachgewiesen werden können, sind als besonders wertvoll zu nennen das 3-Carboxy-7-nitro-4-oxo 4H-pyrimido [2,1-b] benzthiazol, oder dessen Niederalkyl-, z. B.
Methyl- oder Äthylester, oder Alkalimetallsalze, das 7-N-(4-Acetylamino-phenylsulfonyl)- amino-3-carb äthoxy4-oxo- 4H-pyrimido [2 1-b] benzthiazol, das 3-Carbäthoxy-7-N-(3-chlorpheny] carbamoyl)-amino-4-oxo-4H- pyrimido[2,1-b]benzthiazol oder das S-Sithoxy-3-carbäthoxy-4-oxo-4H- pyrimido[2,1-b]benzthiazol, sowie das 3-Carboxy-8-methyl-4-oxo 4H-pyrimido [2,1-b]benzthiazol, oder dessen Alkalimetallsalze, als auch das 1 0-Carbäthoxy-1 1-oxo-1 lH-naphtho- [1', 2':4,5]thiazolo [3,2-a]pyrimidin oder Säureadditionssalze von solchen Verbindungen;
diese Verbindungen zeigen bei subkutaner oder oraler Verabreichung einer täglichen Dosis von etwa 0,05 bis etwa 0,2 g/kg hervorragende Antivirus-Wirkungen.
Die neuen Verbindungen werden in an sich bekannter Weise hergestellt, indem man eine Verbindung mit dem Ringsystem der Formel V

worin mindestens eine der Orthostellungen des carbocyclischen Rings unsubstituiert ist und welche in 5-Stellung des heterocyclischen Rings eine freie oder funktionell abgewandelte Carboxylgruppe oder eine Acylgruppe und in 6-Stellung eine Oxo-, Thiono- oder Iminogruppe enthalten, ringschliesst.
Der Ringschluss von Ausgangsstoffen mit dem Ringsystem der Formel V kann in an sich bekannter Weise durchgeführt werden, z. B. nach der Hugen schoffschen Synthese, wie durch Behandeln mit einem Halogen, insbesondere mit Brom, sowie mit Chlor, oder mit einem Schwefel-Halogen-haltigen Kondensationsmittel, insbesondere Sulfurylchlorid, sowie Schwefelmonochlorid, vorzugsweise in Gegenwart eines geeigneten Lösungsmittels, wie Chloroform, Athylendi- chlorid, Chlorbenzol, Nitrobenzol, oder Essigsäure, wenn notwendig unter Kühlen oder Erhitzen und/oder in der Atmosphäre eines Inertgases.
In erhaltene Verbindungen können innerhalb des vorgenannten Rahmens Substituenten eingeführt, entfernt und/oder abgeändert werden. So kann z. B. eine funktionell abgewandelte Carboxylgruppe in an sich bekannter Weise in eine freie oder in eine andere funktionell abgewandelte Carboxylgruppe übergeführt werden, z.B. durch Hydrolyse (eine veresterte Carboyl- gruppe z. B. durch Behandeln mit einem basischen oder sauren Mittel), Alkoholyse, Umesterung (wenn notwendig, in Gegenwart eines Umesterungsmittels, wie eines Alkalimetallalkoholats), sowie Amidierung (eine veresterte Carboxylgruppe oder eine Chlorcarbonylgruppe z. B. durch Behandeln mit Ammoniak oder einem Amin) oder Behandeln mit einem Hydrazin (eine veresterte Carboxylgruppe z. B. durch Behandeln mit Hydrazin oder Hydrazinhydrat).
Eine freie Carboxylgruppe kann in eine funktionell abgewandelte Carboxylgruppe, z. B. in eine veresterte Carboxylgruppe durch Verestern (wie Behandeln mit einem Alkohol in Gegenwart eines Kalaysators, wie einer Säure, oder mit einer Diazoverbindung), oder in eine Carbamoyl- oder Cyangruppe, z. B. durch Dehydratisieren eines Ammo niumsalzes, umgewandelt werden. Eine Iminogruppe kann hydrolytisch, z. B. durch Behandeln mit wässriger Säure, wie verdünnter Salzsäure, in die Oxogruppe übergeführt werden. Ferner lässt sich eine Oxogruppe z.B. durch Behandeln mit einem Schwefel-einführenden Reagens, wie Phosphorpentasulfid, in eine Thionogruppe umwandeln.
Ferner können erhaltene Verbindungen der vorliegenden Erfindung in an sich bekannter Weise, z.B.
durch Behandeln mit einem Gemisch von konzentrierter oder rauchender Salpetersäure und konzentrierter Schwefelsäure, nitriert werden, wobei man im aromatischen Teil substituierte Nitro-, insbesondere Mononitro- oder Dinitroderivate, erhält; so führt z. B. Nitrierung von 3-Carbäthoxy-4-oxo-4H-pyrimido [2,1-b]benzthiazol zum 3 -Carbäthoxy-7-nitro-4-oxo4H-pyrimido [2, 1-b]benzthiazol.
In nitro-substituierten Verbindungen kann die Nitrogruppe durch Behandeln mit geeigneten Reduktionsmitteln, z.B. durch Behandeln mit Metall-5äure-Kom- binationen, oder mit Wasserstoff in Gegenwart von geigneten Katalysatoren, wie Nickel-, Platin- oder Palladium-Katalysatoren, oder mittels anderen zur Reduktion einer Nitrogruppe geeigneten Verfahren in eine Aminogruppe überführt werden; durch Behandeln mit Wasserstoff in Gegenwart eines Platinoxyd-Katalysators erhält man z. -B. aus dem 3-Carbäthoxy-7-nitro-4-oxo- 4H-pyrimido[2,1-b]benzthiazol das 7-Amino-3-carbäthoxy-4-oxo4H-pyrimido [2,1-b]benzthiazol.
Aminoverbindungen können substituiert, z. B. niederalkyliert werden, wobei man mono- oder disubstituierte Amino-, z. B. Mono- oder Di-niederalkylaminoverbindungen, erhält, oder in andere Derivate umgewandelt werden; so kann man z. B. Aminoverbindungen .mit reaktionsfähigen Estern von Alkoholen, wie Estern von Alkoholen mit Halogenwasserstoffsäuren oder organischen Sulfonsäuren, umsetzen oder mit Carbonylgruppen enthaltenden Verbindungen, wie Aldehyden, z.B.
Benzaldehyd oder substituierten Benzaldehyden, sowie Ketonen behandeln und Schiff'sche Basen erhalten, welche durch Reduktion z. B. mit einem geeigneten Hydrid-Reduktionsmittel, wie Natriumborhydrid, oder mit katalytisch aktiviertem Wasserstoff, reduziert werden können. Aminogruppen kann man ferner z. B.
durch Behandeln mit reaktionsfähigen funktionell abgewandelten Carbon- oder Sulfonsäuren, wie Halogeniden, Anhydriden oder Estern davon, acylieren. Säureanhydride sind ebenfalls die inneren Anhydride von Carbamin- oder Thiocarbaminsäuren, d.h. Isocyanate oder Isothiocyanate, wie Niederalkyl-, Aryl- oder Arylniederalkyl-isocyanate und -isothio-cyanate, z. B. Niederalkyl-isocyanate, wie Methyl-, Äthyl-, Propyl- oder Butylisocyanat Phenyl- oder substituierte, besonders Niederalkyl- oder Halogen-substituierte Phenyliso-cyanate, oder Benzylisocyanat; Acylierung von Aminoverbindungen mit solchen Mitteln führt zu den entsprechenden Harnstoff- und Thioharnstoffderivaten.
Die Ausgangsstoffe können in an sich bekannter Weise hergestellt werden. So lassen sich Ausgangsstoffe mit dem Ringsystem der Formel V z. B. herstellen, wenn man ein Arylisocyanat oder Arylisothiocyanat mit einem Äthenylamin, dessen ss-Kohlenstoffatom eine reaktionsfähige funktionell abgewandelte Carboxylgruppe und eine funktionell abgewandelte Carboxylgruppe oder eine Acylgruppe aufweist, umsetzt und den erhaltenen N-Aryl-N'-äthenyl-harnstoff oder thioharnstoff, in welchem die Aethenylgruppe in ss-Stellung eine reaktionsfähige funktionell abgewandelte Carboxylgruppe und eine funktionell abgewandelte Carboxylgruppe oder eine Acylgruppe aufweist, z. B. durch Behandeln mit Essigsäureanhydrid, in das erwünschte Ausgangsmaterial überführt.
Die Erfindung umfasst ebenfalls diejenigen Abänderungen des vorliegenden Verfahrens wonach ein Ausgangsmaterial in Form eines unter den Reaktionsbedingungen erhältlichen rohen Reaktionsgemisches oder in Form eines Salzes verwendet wird.
Vorzugsweise werden solche Ausgangsstoffe verwendet, welche zu den im vorstehenden als besonders wertvoll bezeichneten Verbindungen führen.
Je nach den RealcEionsbedingungen erhält man die neuen Verbindungen in freier Form oder in Form ihrer Salze, welche ebenfalls von der vorliegenden Erfindung umfasst werden. So erhält man z. B. basische, neutrale, saure oder gemischte Salze, wobei gegebenenfalls He mi., Mono-, Sesqui- oder Polyhydrate geblldet -werden können. Die Salze der neuen Verbindungen können in an sich bekannter Weise in die freien Verbindungen sowie in andere Salze übergeführt werden, Säureadditionssalze z. B. durch Behandeln mit basischen Mitteln, wie alkalischen Reagenzien oder Ionenaustauschern, Salze mit Basen durch Behandeln mit sauren Mitteln, wie Säuren. Eine freie Aminosäureverbindung -kann Salze, insbesondere nicht-toxische Salze mit Basen, z. B.
Alkalimetall-, Erdalkalimetall- oder Ammoniumsalzen, z. B. durch Behandeln mit Metallhydroxyden, insbesondere Alkalimetall- oder Erdalkalimetall-, wie Natrium-, Kalium- oder Calciumhydroxyden, oder mit Alkalimetallcarbonaten, wie Natrium- oder Kaliumcarbonat, mit Ammoniak oder mit organischen Aminen, sowie mit Säuren bilden. Säureadditionssalze sind vorzugsweise therapeutisch verwendbare, nicht-toxische Säureadditionssalze, z. B. solche mit anorganischen Säuren, wie Halogenwasserstoffsäuren, wie Salzsäure oder Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Salpetersäure oder Perchlorsäure, oder mit organischen Säuren, wie aliphatischen, alicyclischen, aromatischen oder heterocyclischen Carbon- oder Sulfonsäuren, z. B.
Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Apfel-, Wein-, Zitronen-, Malein-, Hydroxymalein-, Brenztrauben-, Phenylessig-, Benzoe-, p-Amino-benzoe-, Anthranil-, p-Hydroxy-benzoe-, Salicyl-, p-Aminosalicyl-, Embon-, Methansulfon-, Sithansulfon-, Hydroxyäthansulfon-, Äthylensulfon-, Halogenbenzolsufon-, Toluolsulfon-, Naphtalinsulfon-, N-Cyclohexylsulfamin- oder Sulfanilsäure, Methionin, Tryptophan, Lysin oder Arginin, sowie Ascorbinsäure.
Die oben erwähnten sowie andere Salze der neuen Verbindungen, wie z. B. ihre Pikrate, können gegebenenfalls zur Reinigung der erhaltenen freien Verbindungen, wobei man die freie Verbindung in ein Salz davon umwandelt, letzteres isoliert und die freie Verbindung daraus wieder herstellt, oder zu deren Charakterisierung verwendet werden. Infolge der engen Beziehungen zwischen den neuen Verbindungen in freier Fom und in Form ihrer Salze sind im vorausgegangenen und nachfolgend unter den freien Verbindungen oder den Salzen sinn- und zweckmässig gegebenenfalls auch die entsprechenden Salze bzw. freien Verbindungen zu verstehen.
Die neuen Verbindungen können in Form von Medikamenten, z. B. in Form von pharmazeutischen Präparaten, verwendet werden. In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
Ein Gemisch von 4,88 g 5-Carbäthoxy-2-mercapto-6-oxo1-phenyl-1,6-dihydro-pyrimidin in 50 ml trockenem Chloroform und 4 g Brom in 100 ml trockenem Chloroform wird während 6 Stunden am Rückfluss gekocht. Das Lösungsmittel wird dann unter vermindertem Druck eingedampft und der Rückstand in Wasser aufgenommen. Die wässrige Lösung wird mit 2-n. wässrigem Natriumhydroxyd basisch gestellt; das erhaltene feste Material wird abfiltriert, mit Wasser gewaschen und aus einem Gemisch von Dimethylformamid, Äthanol und Wasser umkristallisiert; das so erhaltene 3 -Carbäthoxy-4-oxo-4H pyrimido [2,1-b]benzthiazol der Formel

schmilzt bei 145-146 .
Das Ausgangsmaterial kann wie folgt erhalten werden:
Ein Gemisch von 13,5 g Phenylisothiocyanat und ls,l g Aminomethylen-malonsäure-diäthylester wird während 12 Stunden bei 160-1700 erhitzt, dann abgekühlt und mit Benzol trituiert. Das feste Material wird abfiltriert und aus Benzol kristallisiert, wobei man den N-Phenyl-N'-(2,2-dicarbäthoxyvinyl)-thioharnstoff erhält, der ohne weitere Reinigung verarbeitet wird.
Ein Gemisch von 17,1 g N-Phenyl-N'-(2,2-dicarb äthoxy-vinyl)-thioharnstoff und 100 ml Essigsäureanhydrid wird während 24 Stunden am Rückfluss gekocht. Die Lösung wird konzentriert, abgekühlt und filtriert; der Filterrückstand wird mit einer kleinen Menge Athanol gewaschen und aus Äthanol kristallisiert; das so erhaltene 5-Carbäthoxy-2-mercapto-6-oxo 1-phenyl- 1 ,6-dihydro-pyrimidin wird ohne weitere Reinigung weiterverarbeitet.

Isocyanates or isothiocyanates, such as lower alkyl, aryl or aryl lower alkyl isocyanates and isothio-cyanates, e.g. B. lower alkyl isocyanates, such as methyl, ethyl, propyl or butyl isocyanate, phenyl or substituted, especially lower alkyl or halogen-substituted phenyl isocyanates, or benzyl isocyanate; Acylation of amino compounds with such agents leads to the corresponding urea and thiourea derivatives.
The starting materials can be prepared in a manner known per se. So starting materials with the ring system of the formula V z. B. produce if an aryl isocyanate or aryl isothiocyanate with an ethenylamine, the ss-carbon atom has a reactive, functionally modified carboxyl group and a functionally modified carboxyl group or an acyl group, and the resulting N-aryl-N'-ethenyl urea or thiourea, in which the ethenyl group in the ss position has a reactive, functionally modified carboxyl group and a functionally modified carboxyl group or an acyl group, e.g. B. by treatment with acetic anhydride, converted into the desired starting material.
The invention also includes those modifications of the present process according to which a starting material is used in the form of a crude reaction mixture obtainable under the reaction conditions or in the form of a salt.
It is preferred to use those starting materials which lead to the compounds identified above as being particularly valuable.
Depending on the reaction conditions, the new compounds are obtained in free form or in the form of their salts, which are also included in the present invention. So you get z. B. basic, neutral, acidic or mixed salts, where appropriate He mi., Mono-, Sesqui- or polyhydrates - can be formed. The salts of the new compounds can be converted into the free compounds as well as into other salts in a manner known per se, acid addition salts z. B. by treating with basic agents such as alkaline reagents or ion exchangers, salts with bases by treating with acidic agents such as acids. A free amino acid compound can be salts, especially non-toxic salts with bases, e.g. B.
Alkali metal, alkaline earth metal or ammonium salts, e.g. B. by treatment with metal hydroxides, especially alkali metal or alkaline earth metal, such as sodium, potassium or calcium hydroxides, or with alkali metal carbonates, such as sodium or potassium carbonate, with ammonia or with organic amines, and with acids. Acid addition salts are preferably therapeutically useful, non-toxic acid addition salts, e.g. B. those with inorganic acids such as hydrohalic acids such as hydrochloric or hydrobromic acid, sulfuric acid, phosphoric acid, nitric acid or perchloric acid, or with organic acids such as aliphatic, alicyclic, aromatic or heterocyclic carboxylic or sulfonic acids, eg. Ant, vinegar, propionic, amber, glycolic, milk, apple, wine, lemon, maleic, hydroxymalein, pyruvic, phenyl acetic, benzoin, p-amino-benzoic, anthranil -, p-Hydroxybenzenesulfon-, salicyl-, p-aminosalicyl-, Embon-, methanesulfon-, sithanesulfon-, hydroxyethanesulfon-, ethylensulfon-, halobenzenesulfon-, toluenesulfon-, naphthalenesulfon-, N-Cyclohexylsulfamin- or methanesulfanils Tryptophan, lysine or arginine, as well as ascorbic acid.
The above-mentioned and other salts of the new compounds, such as. B. their picrates can optionally be used to purify the free compounds obtained, converting the free compound into a salt thereof, isolating the latter and reconstituting the free compound therefrom, or to characterize them. As a result of the close relationships between the new compounds in free form and in the form of their salts, in the preceding and in the following the free compounds or salts are to be understood, meaningfully and appropriately, also the corresponding salts or free compounds.
The new compounds can be in the form of drugs, e.g. B. in the form of pharmaceutical preparations can be used. In the following examples the temperatures are given in degrees Celsius.
example 1
A mixture of 4.88 g of 5-carbethoxy-2-mercapto-6-oxo1-phenyl-1,6-dihydro-pyrimidine in 50 ml of dry chloroform and 4 g of bromine in 100 ml of dry chloroform is refluxed for 6 hours. The solvent is then evaporated under reduced pressure and the residue is taken up in water. The aqueous solution is with 2-n. made basic with aqueous sodium hydroxide; the solid material obtained is filtered off, washed with water and recrystallized from a mixture of dimethylformamide, ethanol and water; the 3-carbethoxy-4-oxo-4H pyrimido [2,1-b] benzothiazole of the formula obtained in this way

The starting material can be obtained as follows:
A mixture of 13.5 g of phenyl isothiocyanate and ls.1 g of aminomethylene malonic acid diethyl ester is heated for 12 hours at 160-1700, then cooled and triturated with benzene. The solid material is filtered off and crystallized from benzene, giving the N-phenyl-N '- (2,2-dicarbethoxyvinyl) thiourea, which is processed without further purification.
A mixture of 17.1 g of N-phenyl-N '- (2,2-dicarb ethoxy-vinyl) -thiourea and 100 ml of acetic anhydride is refluxed for 24 hours. The solution is concentrated, cooled and filtered; the filter residue is washed with a small amount of ethanol and crystallized from ethanol; the 5-carbethoxy-2-mercapto-6-oxo 1-phenyl-1, 6-dihydro-pyrimidine obtained in this way is processed further without further purification.
