Dibenzo a,d cyclo-heptene derivs. as pharms. The cmpd. are of gen. formula: where Z is -CH=CH- or -CH2-CH2- and R is H, amino or salts formed by reaction with mineral or organic acids. Used as antidepressants, tranquillisers, sedatives, anticonvulsants, analgesics and diuretics.

  Verfahren zur Herstellung neuer, in Stellung 5 substituierter SH-Dibenzo(a,d)cycloheptene
Die Erfindung betrifft ein Verfahren zur Herstellung neuer, in Stellung 5 substituierter 5H-Dibenzo[a,d]cycloheptene der Formel I,

 worin Z für -CH=CH- oder   -CH2-CH2- steht.   



   Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man Verbindungen der Formel II,

 worin Z obige Bedeutung hat, bei erhöhter Temperatur alkalisch hydrolysiert.



   Das Verfahren wird zweckmässigerweise bei erhöhten Temperaturen, beispielsweise   30-120     C, vorzugsweise 50 bis   100"    C, durchgeführt. Zur alkalischen Hydrolyse lassen sich an sich bekannte Mittel verwenden, vorzugsweise Alkalihydroxide, wie Natrium- oder Kaliumhydroxid. Man arbeitet zweckmässigerweise in einem inerten organischen Lösungsmittel. Bevorzugt werden dabei leicht mit Wasser mischbare Lösungsmittel, insbesondere niedere Alkohole, wie Äthanol.



   Die Verbindungen der Formel I können in an sich bekannter Weise isoliert und gereinigt werden.



   Zu den Ausgangsprodukten der Formel II kann man gelangen, indem man Verbindungen der Formel III,

 worin Z obige Bedeutung besitzt und R' für Alkyl mit 1 bis 4 Kohlenstoffatomen steht, mit Hydrazin umsetzt und die erhaltenen Verbindungen der Formel IV,

 worin Z obige Bedeutung hat, umlagert und cyclisiert.



   Zur Herstellung von Verbindungen der Formel IV arbeitet man zweckmässigerweise bei Temperaturen zwischen 40 und   1500    C, vorzugsweise 70 und   1200    C. Als Reaktions medium wird mit Vorteil ein Überschuss an Hydrazin verwendet, man kann jedoch auch in Gegenwart inerter Lösungsmittel arbeiten. Die Isolierung und Reinigung der erhaltenen Verbindungen erfolgt in an sich bekannter Weise.



   Die Herstellung der Verbindungen der Formel II aus Verbindungen der Formel IV erfolgt unter Curtius-Umlagerung und anschliessender Cyclisierung. Sie wird vorzugsweise in einem sauren wässrigen Medium unter Verwendung eines Metallnitrits, vorzugsweise eines Alkalinitrits, wie Natriumoder Kaliumnitrit, durchgeführt. Das saure Medium wird vorzugsweise unter Verwendung einer niederen Carbonsäure geschaffen, wie Essigsäure, die gleichzeitig als weiteres Lösungsmittel dient. Die Verbindungen der Formel II können in an sich bekannter Weise isoliert und gereinigt werden.  



   Die Verbindungen der Formel III sind bekannt und können beispielsweise aus den entsprechenden Dibenzo[a,d]cyclohepten-5-onen hergestellt werden. Diejenigen Verbindungen der Formel III, bei denen Z für -CH=CH- steht, lassen sich beispielsweise ausgehend von   Dibenzo[a,d]cyclohepten-    5-on nach der bekannten Reformatsky-Reaktion herstellen, wie dies in Beispiel 1 beschrieben ist. Zu den Verbindungen der Formel III, bei denen Z   für -CH2-CH2- steht    und R' ein verzweigtes Alkyl mit 34 Kohlenstoffatomen bedeutet, gelangt man durch Umsetzung entsprechender   10,1 1-Dihy-    drobenzo[a,d]cyclohepten-5-one mit einem entsprechend verzweigtkettigen Essigsäureester in Gegenwart einer starken Base, wie Diäthylaminomagnesiumbromid.



   Die freien Basen der Verbindungen der Formel I können gegebenenfalls in Säureadditionssalze überführt werden und umgekehrt.



   Die Verbindungen der Formel I weisen interessante pharmakodynamische Eigenschaften auf. Sie wirken insbesondere depressiv auf das Zentralnervensystem (Orloff et al., Proc.



  Soc. Exp. Biol. 70, 254 [1949] sowie Toman et al., J. Neurophysiol. 9, 231 [1946]), und können daher als Anticonvulsiva verwendet werden. Sie besitzen jedoch auch noch andere pharmakodynamische Eigenschaften. Die Verbindung des später beschriebenen Beispiels 2 wirkt in verschiedener Weise auf das Zentralnervensystem. Sie wirkt als Tranquilizer und/ oder muskelentspannend (Dunham et al., J. Am. Pharm.



     Assoc. 46,    208 [1957]. Sie wirkt ferner diuretisch (R. Aston, Tox. and Appl. Pharm. 1, 277 [1959] sowie analgetisch (Okuns modification of Seigmunds  Whrithing-Method , J. Pharm. and Exper. Therap. 139, 107 [1963]). Die Verbindung des Beispiels 1 wirkt schliesslich als Tranquilizer und/oder Sedativum. Die Verbindungen der Formel I werden zweckmässigerweise in Dosen von etwa 30 bis 800 mg verabreicht, was vorzugsweise in mehreren täglichen Dosen zwischen etwa 8 und 400 mg oder in Retardform erfolgt. Zur Herstellung geeigneter Arzneiformen werden sie mit üblichen Hilfs- und Zusatzstoffen verarbeitet. Sie können entweder in Form ihrer freien Basen oder ihrer Säureadditionssalze verwendet werden, deren Wirksamkeit den freien Basen entspricht.

  Geeignete Säureadditionssalze sind beispielsweise die Fumarate, Hydrochloride, Maleate, Formlate, Acetate, Sulfonate, Methansulfonat oder Malonate.



   Beispiel 1    5-Aminomethyl-5H-dibenzo[a,d]cyclohepten-5-ol    a) 5-Hydroxy-dibenzo[a,d]cyclohepten-5-essigsäure-äthylester
Eine Suspension von 8,35 g aktiviertem Zink in 120 ml absolutem Benzol wird bei Raumtemperatur mit einer Lösung von 10,3 g Dibenzo[a,d]cyclohepten-5-on und 8,35 g Äthylbromacetat in 50 ml absolutem Benzol versetzt. Nachdem etwa 1/4 dieser Lösung zugesetzt ist, wird das Gemisch unter Rückfluss bis die Reaktion schwächer wird erhitzt und sodann mit dem Rest der Lösung versetzt, wobei das Reaktionsgemisch auf Rückflusstemperatur gehalten wird. Nach Beendigung der Zugabe wird weitere 4 Stunden unter Rückfluss erhitzt. Das Gemisch wird dann zu verdünnter Essigsäure zugesetzt und zweimal mit Äthylacetat extrahiert. Die organischen Phasen werden mit Wasser gewaschen, getrocknet und im Vakuum eingedampft.

  Nach Umkristallisieren des Rückstandes aus Diäthyläther/Pentan erhält man den 5-Hydroxy   dibenzo[a,d]cyclohepten-5-essigsäureäthylester    vom Smp.



     93-99     C.



  b)   5-Hydroxy-dibenzo[a,d]cyclohepten-5-essigsäurehydrazid   
Ein Gemisch aus 4,0 g 5-Hydroxy-dibenzo[a,d]cyclohepten-5-essigsäureäthylester und 8 ml wasserfreiem Hydrazin wird unter Rühren zwei Stunden auf 80 bis   85"    C gehalten, worauf man es 19 Stunden bei Raumtemperatur stehenlässt.



  Das Gemisch wird sodann unter Hochvakuum bei   100"    C zur Trockne eingedampft, der Rückstand in heissem Äthanol gelöst, mit Aktivkohle behandelt und auskristallisiert, wobei man das   5-Hydroxy-dibenzo [a,d]cyclohepten-5 -essigsäure-    hydrazid vom Smp.   192-194     C erhält.



  c) Spiro   [5H-dibenzo[a,d]cyclohepten-5-oxazolin-2-on]   
In einen Scheidetrichter werden 3,1 g 5-Hydroxy-dibenzo[a,d]cyclohepten-5-essigsäurehydrazid, 20 ml Wasser, 5 g Eis, 2 ml Essigsäure und eine Lösung von 1,6 g Natriumnitrit in 3 ml Wasser gegeben. Das Gemisch wird 10 Minuten geschüttelt, mit weiteren 2 ml Essigsäure versetzt und weitere 15 Minuten geschüttelt. Das erhaltene Gemisch wird mit Benzol extrahiert und die Benzolschicht mit Wasser, dann mit Natriumbicarbonat-Lösung und schliesslich mit Natriumchlorid-Lösung gewaschen. Die Benzolschicht wird auf einem Dampfbad zur Trockne eingedampft, und nach Umkristallisieren des Rückstandes aus   95 %igem    Äthanol erhält man das   Spiro[5H-dibenzo[a,d]cyclohepten-5-oxazolin-2-on]    vom Smp. 3050 C (Zers.   S      270     C).



  d)   5-Aminomethyl-5H-dibenzo[a,d]cyclohepten-5-ol   
Ein Gemisch aus 1,0 g   Spiro[5H-dibenzo[a,d]cyclohep    ten-5-oxazolin-2-on], 2,0 g Kaliumhydroxid und 30 ml   95 %-    igem Äthanol wird etwa 15 Stunden unter Rückfluss erhitzt.

 

  Das Gemisch wird im Vakuum zur Trockne eingedampft und der Rückstand mit 2n Salzsäure neutral gestellt. Das erhaltene Gemisch wird mit Diäthyläther extrahiert, die Ätherphase zweimal mit Wasser und einmal mit Natriumchlorid Lösung gewaschen, getrocknet und im Vakuum eingedampft, wobei man nach Umkristallisieren aus Diäthyläther das 5   Aminomethyl-5H-dibenzo[a,d]cyclohepten-5 -ol    vom Smp.



     1171200 C    erhält.



   Beispiel 2   5 -Aminomethyl-lO,l l-dihydro-5H-dibenzo[a,d]cyclohepten-    5-ol
Analog Beispiel 1 und unter Verwendung geeigneter Ausgangsprodukte in entsprechenden Mengen gelangt man zur Titelverbindung, die bei   147-149     C schmilzt.

   The process is expediently carried out at elevated temperatures, for example 30-120 ° C., preferably 50 to 100 ° C. For the alkaline hydrolysis, known agents can be used, preferably alkali hydroxides, such as sodium or potassium hydroxide. It is expedient to work in an inert organic Solvents, solvents which are readily miscible with water, in particular lower alcohols such as ethanol, are preferred.



   The compounds of the formula I can be isolated and purified in a manner known per se.

   The preparation of compounds of the formula IV is expediently carried out at temperatures between 40 and 1500 ° C., preferably 70 and 1200 ° C. The reaction medium used is advantageously an excess of hydrazine, but it is also possible to work in the presence of inert solvents. The compounds obtained are isolated and purified in a manner known per se.



   The compounds of the formula II are prepared from compounds of the formula IV with Curtius rearrangement and subsequent cyclization. It is preferably carried out in an acidic aqueous medium using a metal nitrite, preferably an alkali nitrite such as sodium or potassium nitrite. The acidic medium is preferably created using a lower carboxylic acid, such as acetic acid, which also serves as a further solvent. The compounds of the formula II can be isolated and purified in a manner known per se.



   The compounds of the formula III are known and can be prepared, for example, from the corresponding dibenzo [a, d] cyclohepten-5-ones. Those compounds of the formula III in which Z is -CHCHCH- can be prepared, for example, starting from dibenzo [a, d] cyclohepten-5-one by the known Reformatsky reaction, as described in Example 1. The compounds of the formula III in which Z is -CH2-CH2- and R 'is a branched alkyl having 34 carbon atoms are obtained by reacting corresponding 10.1 1-dihydrobenzo [a, d] cycloheptene-5- one with an appropriately branched-chain acetic acid ester in the presence of a strong base such as diethylaminomagnesium bromide.

A suspension of 8.35 g of activated zinc in 120 ml of absolute benzene is treated at room temperature with a solution of 10.3 g of dibenzo [a, d] cyclohepten-5-one and 8.35 g of ethyl bromoacetate in 50 ml of absolute benzene. A mixture of 4.0 g of 5-hydroxydibenzo [a, d] cycloheptene-5-acetic acid ethyl ester and 8 ml of anhydrous hydrazine is kept at 80 to 85 ° C. with stirring for two hours, after which it is left to stand for 19 hours at room temperature.

  d) 5-aminomethyl-5H-dibenzo [a, d] cyclohepten-5-ol
A mixture of 1.0 g of spiro [5H-dibenzo [a, d] cycloheptene-5-oxazolin-2-one], 2.0 g of potassium hydroxide and 30 ml of 95% ethanol is refluxed for about 15 hours.

 

  The mixture is evaporated to dryness in vacuo and the residue is made neutral with 2N hydrochloric acid. The resulting mixture is extracted with diethyl ether, the ether phase washed twice with water and once with sodium chloride solution, dried and evaporated in vacuo, the 5 aminomethyl-5H-dibenzo [a, d] cyclohepten-5-ol from being recrystallized from diethyl ether M.p.
