Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von 1-ss-Phenylpropioloyl-3-indolylessigsäure- estern, welche Zwischenprodukte für die Herstellung der 1 -Cinnamoyl-3-indolylessigsäure bzw. deren Derivaten der folgenden Formel:

worin Rl ein Wasserstoffatom, ein Halogenatom, einen Alkylrest mit 1 bis 4 Kohlenstoffatomen, einen Alkoxyrest mit 1 bis 4 Kohlenstoffatomen, R2 und R4, die gleich oder verschieden sein können, Wasserstoffatome oder Alkylreste mit 1 oder 2 Kohlenstoffatomen und R3' ein Wasserstoffatom, einen Benzylrest oder einen Alkylrest mit 1 bis 4 Kohlenstoffatomen bedeuten, sind.
Die Verbindungen der obigen Formel sind wertvolle Arz- neimittel.
Die Erfindung bezieht sich somit auf ein neues Verfahren für die Herstellung von 1-ss-Phenylpropioloyl-3-indolylessig- säureestern, welche der folgenden Formel entsprechen:

Das neue Verfahren zur Herstellung dieser l-p-Phenylpro- pioloyl-3-indolylessigsäure und deren Derivate der obigen Formel (l1 besteht darin, dass man ein 3-1ndolylessigsäurede- rivat der Formel:

worin X ein Halogenatom darstellt, behandelt und das so erhaltende 1-ss-Phenylpropioloyl-3-indolylacetat gegebenenfalls in die entsprechende rreie Säure überîührt.
Das erfindungsgemässe Verfahren verläuft glatt und liefert die gewünschten Produkte in hoher Ausbeute, weswegen es sich beim vorliegenden Verfahren um ein für die Praxis vorteilhaftes Verfahren handelt.
Die 1-ss-Phenylpropioloyl-3-indolylessigsäureester der Formel (I) können dadurch erhalten werden, dass man ein 3-Indolylessigsäurederivat der Formel (II) mit einem p-Phenylpro- piolylhalogenid der Formel (III) in Gegenwart eines Kondensationsmittels umsetzt. Bevorzugte Beispiele solcher Kondensationsmittel sind Natrium, Kalium, Lithium, Natriumhydrid, Natriumamid, Kaliumamid, Lithiurnamid, Lithiumhydroxyd, Natriumhydroxyd, Kaliumhydroxyd usw. Die Umsetzung erfolgt in Gegenwart eines Lösungsmittels, wie z.B. Benzol, Toluol, Xylol, Pyridin. Dimethylformamid, Äthyläther, Tetrahydrofuran, Dioxan, Dimethoxyäthan, Hexan usw. Die Umsetzung kann bei Zimmertemperatur oder gewünschtenfalls unter Erhitzen oder unter Kühlen durchgeführt werden.
Die Reaktionstemperatur liegt vorzugsweise zwischen O"C und 15"C.
Die so erhaltenen Alkylester oder Benzylester der l-y-Phe- nylpropioloyl-3-indolylessigsäure lassen sich in an sich bekannter Weise, beispielsweise gemäss der in der belgischen Patentschrift Nr. 698 378 beschriebenen Methode, in die freie Säure überführen.
Nach dem obigen Verfahren lassen sich die folgenden l-P- -Phenylpropioloyl-3-indolylessigsäurederivate herstellen: 1 --Phenylpropioloyl-2-methyl-5-methoxy-3-indolylessigsäure äthylester, I- -frPhenylpropioloyl-2methy1-5-chlor3 -indolylessigsa ure- äthylester.

Die 1 -P-Phenylpropioloyl-3 -indolylessigsäureester der Formel (I) und die entsprechenden freien Säuren, wie man sie erfindungsgemäss erhält, sind neue Verbindungen, welche leicht in 1 -Cinnamoyl-3 -indolylessigsäurederivate der Formel:

worin R1, R2, R3, und R4 die obigen Bedeutungen haben, übergeführt werden können, indem man die l-p-Phenylpro- pioloyl-3-indolylessigsäurederivate in Gegenwart eines metallischen Katalysators reduziert. Die so erhaltene 1-Cinnamoyl- -3-indolylessigsäure und deren Derivate der Formel (1V) besitzen starke entzündungshemmende und analgetische Wirkungen bei ausserordentlich niedriger Toxizität.
Die vorliegende Erfindung wird ausführlich durch das folgende Beispiel erläutert.
Beispiel
Zu einer Lösung von 10 g 2-Methyl-5-methoxy-3-indolylessigsäureäthylester in 70 cm3 Dimethylformamid gibt man 1,5 g Natriumhydrid hinzu. Das Reaktionsgemisch wird während 2 Stunden bei 45 bis 50"C gerührt. Dann wird das Gemisch mit 6,5 g p-Phenylpropioloylchlorid bei 0 bis 5"C unter Rühren versetzt. Anschliessend rührt man das Reaktionsgemisch während 2 Stunden bei 0 bis 50C und hierauf während 3 Stunden bei 55 bis 60"C. Hierauf wird Wasser dem Reaktionsgemisch zugefügt, worauf man das Gemisch dreimal mit jeweils 100 cm3 Benzol extrahiert.
Die Benzolschichten werden vereinigt und mit Wasser gewaschen und über Magnesiumsulfat getrocknet. Das Lösungsmittel wird unter vermindertem Druck entfernt, wobei man ein viskoses Öl erhält. Äthanol wird hierauf dem öl hinzugefügt, wobei man gelbe Kristalle erhält, welche aus dem 1-p- -Phenylpropioloyl-2-methyl- 5-methoxy- 3 -indolylessigsäure- äthylester vom Smp. 121 bis 122"C bestehen.

The process according to the invention runs smoothly and provides the desired products in high yield, which is why the present process is a process which is advantageous in practice.
The 1-ß-phenylpropioloyl-3-indolylacetic acid esters of the formula (I) can be obtained by reacting a 3-indolylacetic acid derivative of the formula (II) with a p-phenylpropiolyl halide of the formula (III) in the presence of a condensing agent. Preferred examples of such condensing agents are sodium, potassium, lithium, sodium hydride, sodium amide, potassium amide, lithium amide, lithium hydroxide, sodium hydroxide, potassium hydroxide, etc. The reaction is carried out in the presence of a solvent such as e.g. Benzene, toluene, xylene, pyridine. Dimethylformamide, ethyl ether, tetrahydrofuran, dioxane, dimethoxyethane, hexane, etc. The reaction can be carried out at room temperature or, if desired, with heating or cooling.
The reaction temperature is preferably between 0 "C and 15" C.
The alkyl esters or benzyl esters of l-y-phenylpropioloyl-3-indolylacetic acid thus obtained can be converted into the free acid in a manner known per se, for example according to the method described in Belgian Patent No. 698,378.

wherein R1, R2, R3 and R4 have the above meanings, can be converted by reducing the l-p-phenylpropioloyl-3-indolylacetic acid derivatives in the presence of a metallic catalyst. The 1-cinnamoyl-3-indolylacetic acid thus obtained and its derivatives of the formula (IV) have strong anti-inflammatory and analgesic effects with extremely low toxicity.
The present invention is illustrated in detail by the following example.

1.5 g of sodium hydride are added to a solution of 10 g of ethyl 2-methyl-5-methoxy-3-indolylacetate in 70 cm3 of dimethylformamide. The reaction mixture is stirred for 2 hours at 45 to 50 "C. 6.5 g of p-phenylpropioloyl chloride are then added to the mixture at 0 to 5" C. while stirring. The reaction mixture is then stirred for 2 hours at 0 ° to 50 ° C. and then for 3 hours at 55 ° to 60 ° C. Water is then added to the reaction mixture, whereupon the mixture is extracted three times with 100 cm3 of benzene each time.
The benzene layers are combined and washed with water and dried over magnesium sulfate. The solvent is removed under reduced pressure to give a viscous oil. Ethanol is then added to the oil, yellow crystals being obtained which consist of ethyl 1-p- -phenylpropioloyl-2-methyl-5-methoxy-3-indolylacetate with a melting point of 121 to 122 ° C.
