Die so erhaltenen Verbindungen führt man, falls gewünscht, in ein ungiftiges Salz, vorzugsweise in ein Alkalimetallsalz, über.
Die erfindungsgemäss erhältlichen Verbindungen haben schwach sauren Charakter, und ihre Salze können daher nach für die Herstellung von Salzen starker Basen mit schwachen Säuren wohlbekannten Methoden hergestellt werden. Obwohl die Alkalisalze durch Eindampfen alkalischer Lösungen erfindungsgemässer Verbindungen erhalten werden können, wird es vorgezogen, nicht-wässrige Medien zu verwenden. Z.B. wird durch Zusammenmischen einer alkoholischen Lösung einer erfindungsgemäss erhältlichen Verbindung mit einer alkoholischen Lösung, welche eine stöchiometrische Menge eines Alkalialkoxids enthält, nach Abdampfen des Lösungsmittels das entsprechende Alkalisalz erhalten.
Die Dehydrierung einer Verbindung der allgemeinen Formel II kann nach bekannten Methoden durch Behandlung mit einem entsprechenden Dehydrierungsmittel, vorzugsweise Kaliumpermanganat, erreicht werden. Die Dehydrohalogenierung kann ebenfalls durch Behandlung mit bekannten Dehydrohalogenierungsmitteln durchgeführt werden.
Der Ausdruck Cyclopentenyl , so wie er in dieser Beschreibung und den anhängenden Ansprüchen gebraucht wird, bedeutet solche Cyclopentenyl-Radikale, deren Doppelbindung nicht in Konjugation mit derjenigen des 1,2,4-Benzothiadiazin-1,1-dioxid-kernes ist, d.h. ein 2'-Cyclopentenyloder ein 3'-Cyclopentenyl-Radikal. Abgesehen vom Cyclopentenyl-Radikal in Stellung 3 sind die hier beschriebenen Verbindungen im heterocyclischen Teil des Kernes vorzugsweise unsubstituiert.
Unter den erfindungsgemäss hergestellten Verbindungen sind jene bevorzugt, welche in den Stellungen 5 und 8 Wasserstoff tragen, besonders 3-(3'-Cyclopentenyl)-6,7-dichlor-2H- 1 ,2,4-benzothia- diazin- 1,1 -dioxid,3-(3-Cyclopentenyl)-6-trifluor- methyl-7-chior-2H- 1 ,2,4-benzothiadiazin- 1,1 1-dioxid und 3-(3'-Cyclopentenyl)-6-chlor-7-trifluormethyl 2H- 1,2,4-benzothiadiazin- 1, 1-dioxid.
Es ist bekannt, dass ein 1,2,4-Benzothiadiazin-1,1-dioxid in zwei tautomeren Formen vorkommen kann: In der einen Form ist die Doppelbindung zwischen den Atomen der Stellungen 3 und 4, in der anderen zwischen den Stellungen 2 und 3. Die vorliegende Erfindung umfasst beide tautomeren Strukturen.
Die erfindungsgemäss erhältlichen Verbindungen können zur Behandlung von zu hohem Blutdruck verwendet werden.
Sie sind hochschmelzende, weisse kristalline Festkörper, die in Wasser praktisch unlöslich sind. Sie sind jedoch in wässrigem Alkali und in anderen polaren Lösungsmitteln löslich, so in niedrig aliphatischen Alkoholen.
Es ist bekannt, dass gewisse 1,2,4-Benzothiadiazin-1,1dioxide auf das periphere Gefässystem im Sinne eines ausgeprägten anti-hypertensiven Effektes wirken. Man hatte erwartet, dass solche Verbindungen sich eines weiten Anwendungsbereiches in der Linderung und Bekämpfung der essentiellen Hypertonie, der malignen Hypertonie, der Bürgerschen Krankheit, der Raynaudschen Krankheit und dergleichen erfreuen würden. Unglücklicherweise jedoch zeigten die bisher untersuchten Verbindungen auch Elektrolytretention und hyperglykämische Eigenschaften, die ihre allgemeine Verwendbarkeit bei der Linderung und Bekämpfung der Hypertonie ernstlich schmälerten. Im Gegensatz dazu zeigen die erfindungsgemäss herstellbaren Verbindungen noch stärkere antihypertensive Wirkung ohne gleichzeitige Elektrolytretention und hyperglykämische Nebenwirkungen.
Zur Ermittlung der hypotensiven und hyperglykämischen Eigenschaften sowie der Elektrolytretention der erfindungsgemäss erhältlichen Verbindungen wurden verschiedene pharmakologische Verfahren durchgeführt. So wurde zunächst die hypotensive Stärke dieser Verbindungen ermittelt, indem Gruppen hypertensiver Ratten (präpariert durch Einpflanzung von Desoxycorticosteronacetat-Pillen und Ernährung auf Salzbasis) stark variierte Dosen verabreicht wurden. Mittels statistischer Auswertung der Wirkungen dieser Dosenreihe hat man die minimale wirksame Dosis für antihypertensive Wirksamkeit bestimmt. Auf der Grundlage dieses Testes und anderer Standard-Auswertungsverfahren, wie des Standard-in-vitro-Aorten-Streifentests, hat man festgestellt, dass die erfindungsgemäss erhältlichen Verbindungen im Vergleich zu den bekannten Verbindungen grössere Wirksamkeitswerte zeigen.
In ähnlicher Weise hat man auch bei der Auswertung nach geeigneten pharmakologischen Methoden festgestellt, dass die erfindungsgemäss herstellbaren Verbindungen im Vergleich zu bekannten 1,2,4-Benzothiadiazin-1,1-dioxiden wesentlich geringere Elektrolytretention und hyperglykämische Eigenschaften zeigen. So sieht man bei der Analyse der pharmakologischen Testergebnisse, dass die erfindungsgemäss zugänglichen Verbindungen eine wesentliche Verbesserung bei der Linderung und Bekämpfung der Hypertonie bedeuten.
Ganz unerwartet hat man auch gefunden, dass die blutdrucksenkende Wirkung der genannten 3-Cyclopentenyl
1,2,4-benzothiadiazin-1,1-dioxide erheblich länger anhält und stärker ist als die der entsprechenden 3-Cyclopentyl-1,2,4 benzothiadiazin- ,l-dioxide.
Es wurde festgestellt, dass 3-Cyclopentenyl-1,2, 4-benzothiadiazin- 1, 1-dioxide, welche einen Nitrooder einen Methyl-Substituenten anstelle eines 6oder 7-ständigen Chlor-, Brom- oder Trifluormethyl- substituenten haben, Verbindungen ergeben, deren Pharmakologie nicht wesentlich beeinträchtigt ist.
Zur Erfindung gehört also auch die Herstellung von Verbindungen, wie z.B. 3-(3'-Cyclopentenyl)-6-chlor-7 methyl- 1,2,4-benzothiadiazin- 1, 1-dioxid und 3-(3' Cyclopentenyl)-6-nitro-7-chlor- 1,2,4-benzothiadiazin-1, 1-dioxid.
Die wirksame Dosierung erfindungsgemässer Verbindungen hängt von der Schwere, dem Stadium und den Einzelheiten eines jeden Falles ab und wird von dem die Diagnose stellenden und behandelnden Arzt festgelegt. Im allgemeinen stellt eine Gabe von 0,03 bis etwa 1 mg pro kg Körpergewicht pro Tag den Dosierungsbereich für die vorliegende Verbindungsklasse dar.
Die erfindungsgemäss erhältlichen Verbindungen können in Form pharmazeutischer Zubereitungen angewandt werden, die das aktive Ingredienz in Mischung mit einem pharmazeutischen Trägerstoff enthalten, wie er für enterale oder parenterale Verabreichung geeignet ist. Solche Zubereitungen können in fester, beispielsweise als Tabletten, Kapseln oder Suppositorien, oder in flüssiger Form, beispielsweise als Elixiere, Emulsionen und Injektionslösungen, vorliegen.
Für die Rezeptur pharmazeutischer Zubereitungen kann man Stoffe zusetzen, die mit den aktiven Ingredienzien nicht reagieren, so z.B. Wasser, Gelatine, Lactose, Stärke, Magnesiumstearat, Calciumcarbonat, Talkum, Pflanzenöle, Benzylalkohole, Gummi, Polyalkylenglycole oder Vaseline. Das betreffende aktive Ingredienz liegt in der Zubereitung zweckmässig in einem Gewichtsanteil von 0,1 bis 50% vor.
Beispiel 1
Herstellung von 3-(3'-Cyclopentenyl)-6,7-dichloro
2H-1,2,4-benzothiadiazin-1, 1-dioxid
3,36 g 3-(3'-Cyclopentenyl)-6,7-dichloro-3,4-dihy dro-2H- 1,2,4-benzothiadiazin- 1, 1-dioxid werden in einer Lösung von 3,6 g Silbernitrat in 1 ml Ethanol und 10 ml Wasser suspendiert. Zu dieser Suspension wird unter Rühren eine Lösung von 1,6 g NaOH in 10 ml Wasser hinzugefügt und die Reaktionsmischung wird 16 Stunden bei Zimmertemperatur weitergerührt. Darauf wird die Mischung mit verdünnter Salpetersäure angesäuert und die Feststoffe abfiltriert. Die Feststoffe werden mit Aceton extrahiert und die Extrakte eingedampft. Durch Abkühlen erhält man die gewünschte Verbindung (Schmp. 344-346 C).
Beispiel 2
Herstellung von 3-(3'-Cyclopentenyl)-6,7-dichloro
2H-1,2,4-benzothiadiazin-1, 1-dioxid
Eine Lösung von 6,5 g 3-(1'-Chlor-3'-cyclopentenyl) 6,7-dichlor-2H-3,4-dihydro-1,2,4-benzothiadiazin- 1, 1-dioxid und 1,4 g Kaliumzyanid wird 3 Stunden unter Rückfluss erhitzt. Das gebildete Kaliumchlorid wird abfiltriert und das Filtrat abgekühlt. Die gewünschte Verbindung kristallisiert aus und kann durch Umkristallisieren aus Methanol gereinigt werden.

UNTERANSPRÜCHE
1. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man eine Verbindung der im Patentanspruch angegebenen allgemeinen Formel II, in der A wie in dem Patentanspruch definiert ist und Rl 2'- oder 3 '-Cyclopentenyl bedeutet, mit einem Dehydrierungsmittel behandelt.
2. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man eine Verbindung der in dem Patentanspruch angegebenen allgemeinen Formel II, in der A wie im Patentanspruch I definiert ist und Rl ein 1'-halogeniertes 2- oder 3/- Cyclopentenyl bedeutet, mit einem Dehydrohalogenierungsmittel behandelt.
3. Verfahren nach Unteranspruch 1, dadurch gekennzeichnet, dass man Kaliumpermanganat als Dehydrierungsmittel verwendet.
4. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man ein Alkalimetallsalz einer Verbindung der For mehl 1 herstellt.

The compounds obtainable according to the invention have a weakly acidic character and their salts can therefore be prepared by methods well known for the preparation of salts of strong bases with weak acids. Although the alkali salts can be obtained by evaporating alkaline solutions of the compounds of the present invention, it is preferred to use non-aqueous media. E.g. the corresponding alkali salt is obtained by mixing together an alcoholic solution of a compound obtainable according to the invention with an alcoholic solution which contains a stoichiometric amount of an alkali alkoxide after evaporation of the solvent.
A compound of the general formula II can be dehydrated by known methods by treatment with an appropriate dehydrating agent, preferably potassium permanganate. The dehydrohalogenation can also be carried out by treatment with known dehydrohalogenation agents.
The term cyclopentenyl as used in this description and the appended claims means cyclopentenyl radicals whose double bond is not in conjugation with that of the 1,2,4-benzothiadiazine-1,1-dioxide nucleus, i.e. a 2'-cyclopentenyl or a 3'-cyclopentenyl radical. Apart from the cyclopentenyl radical in position 3, the compounds described here are preferably unsubstituted in the heterocyclic part of the nucleus.
Among the compounds prepared according to the invention, those are preferred which carry hydrogen in positions 5 and 8, particularly 3- (3'-cyclopentenyl) -6,7-dichloro-2H-1, 2,4-benzothiazine-1,1 -dioxide, 3- (3-cyclopentenyl) -6-trifluoro-methyl-7-chloro-2H-1, 2,4-benzothiadiazine-1,1 1-dioxide and 3- (3'-cyclopentenyl) -6-chloro -7-trifluoromethyl 2H-1,2,4-benzothiadiazine-1,1-dioxide.
It is known that a 1,2,4-benzothiadiazine-1,1-dioxide can occur in two tautomeric forms: In one form the double bond is between the atoms in positions 3 and 4, in the other between positions 2 and 3. The present invention encompasses both tautomeric structures.
The compounds obtainable according to the invention can be used for the treatment of high blood pressure.
They are high-melting, white crystalline solids that are practically insoluble in water. However, they are soluble in aqueous alkali and in other polar solvents, such as low aliphatic alcohols.
It is known that certain 1,2,4-benzothiadiazine-1,1-dioxides act on the peripheral vascular system in the sense of a pronounced anti-hypertensive effect. It was expected that such compounds would enjoy wide application in the relief and control of essential hypertension, malignant hypertension, Bürger's disease, Raynaud's disease, and the like. Unfortunately, however, the compounds so far studied also exhibited electrolyte retention and hyperglycemic properties which seriously detracted from their general utility in relieving and controlling hypertension. In contrast to this, the compounds which can be prepared according to the invention show an even stronger antihypertensive effect without simultaneous electrolyte retention and hyperglycemic side effects.
Various pharmacological methods were carried out to determine the hypotensive and hyperglycemic properties and the electrolyte retention of the compounds obtainable according to the invention. Thus, the hypotensive potency of these compounds was first determined by giving groups of hypertensive rats (prepared by implanting deoxycorticosterone acetate pills and a salt-based diet) at widely varied doses. The minimum effective dose for antihypertensive effectiveness has been determined by means of statistical evaluation of the effects of this series of doses. On the basis of this test and other standard evaluation methods, such as the standard in vitro aortic strip test, it has been found that the compounds obtainable according to the invention show greater efficacy values compared to the known compounds.
Similarly, when evaluating suitable pharmacological methods, it was found that the compounds which can be prepared according to the invention show significantly lower electrolyte retention and hyperglycemic properties compared to known 1,2,4-benzothiadiazine-1,1-dioxides. When analyzing the pharmacological test results, it can be seen that the compounds accessible according to the invention represent a significant improvement in the alleviation and control of hypertension.
Quite unexpectedly it has also been found that the antihypertensive effect of the aforementioned 3-cyclopentenyl
1,2,4-benzothiadiazine-1,1-dioxide lasts considerably longer and is stronger than that of the corresponding 3-cyclopentyl-1,2,4 benzothiadiazine-, l-dioxide.
It has been found that 3-cyclopentenyl-1,2,4-benzothiadiazine-1, 1-dioxides which have a nitro or a methyl substituent instead of a 6 or 7 chlorine, bromine or trifluoromethyl substituent give compounds whose pharmacology is not significantly affected.
The invention also includes the production of compounds, such as e.g. 3- (3'-cyclopentenyl) -6-chloro-7 methyl-1,2,4-benzothiadiazine-1, 1-dioxide and 3- (3 'cyclopentenyl) -6-nitro-7-chloro-1,2, 4-benzothiadiazine-1,1-dioxide.
The effective dosage of compounds according to the invention will depend on the severity, stage and details of each case and will be determined by the doctor making the diagnosis and treating. In general, a dose of 0.03 to about 1 mg per kg of body weight per day represents the dosage range for the present class of compounds.
The compounds obtainable according to the invention can be used in the form of pharmaceutical preparations which contain the active ingredient mixed with a pharmaceutical carrier, such as is suitable for enteral or parenteral administration. Such preparations can be in solid form, for example as tablets, capsules or suppositories, or in liquid form, for example as elixirs, emulsions and injection solutions.
For the formulation of pharmaceutical preparations, substances can be added that do not react with the active ingredients, e.g. Water, gelatin, lactose, starch, magnesium stearate, calcium carbonate, talc, vegetable oils, benzyl alcohols, gum, polyalkylene glycols or petrolatum. The active ingredient in question is expediently present in the preparation in a proportion by weight of 0.1 to 50%.

3.36 g of 3- (3'-cyclopentenyl) -6,7-dichloro-3,4-dihydro-2H-1,2,4-benzothiadiazine-1, 1-dioxide are in a solution of 3.6 g Silver nitrate suspended in 1 ml of ethanol and 10 ml of water. A solution of 1.6 g of NaOH in 10 ml of water is added to this suspension with stirring, and the reaction mixture is stirred for a further 16 hours at room temperature. The mixture is then acidified with dilute nitric acid and the solids are filtered off. The solids are extracted with acetone and the extracts evaporated. The desired compound (melting point 344-346 C) is obtained by cooling.

2H-1,2,4-benzothiadiazine-1,1-dioxide
A solution of 6.5 g of 3- (1'-chloro-3'-cyclopentenyl) 6,7-dichloro-2H-3,4-dihydro-1,2,4-benzothiadiazine-1, 1-dioxide and 1, 4 g of potassium cyanide is refluxed for 3 hours. The potassium chloride formed is filtered off and the filtrate is cooled. The desired compound crystallizes out and can be purified by recrystallization from methanol.
