Description
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von 9-Aminoalkyl-9,10-dihydro-9,10-metha-noanthracenderivaten und von nichttoxischen, pharmazeutisch zulässigen Salzen davon, welche sich zu pharmazeutischen Präparaten verarbeiten lassen.
Das 9,10-Dihydro-9,10-methanoanthracen selbst ist seit 1920 bekannt, wobei verschiedene chemische Studien mit 9,10-Dihydro-9,10-methanoanthracenderivaten angestellt worden sind. Es ist aber kein Bericht bezüglich der Synthese von 9-Aminoalkyl-9,10-dihydro-9,10-methanoanthracenderi-vaten oder bezüglich einer pharmakologischen Studie mit 9,10-Dihydro-9,10-methanoanthracenderivaten veröffentlicht worden.
Ferner waren durch das US-Patent Nr. 3 399 201 von p. Schmidt et al. Äthanoanthracenderivate der Formel:
a bekannt. Diese Verbindungen stellen Homologe der erfin-dungsgemäss erhältlichen Methanoanthracenderivate dar, letztere erweisen sich aber in pharmakologischer Hinsicht als weit überlegen sowohl durch eine stärkere Antitetrabenazinwir-kung als auch durch eine geringere akute Toxizität.

Es wurde nun festgestellt, dass die neuen 9-Aminoalkyl-9,10-dihydro-9,10-methanoanthracenderivate der folgenden Formel (I) und deren nichttoxische, pharmazeutisch zulässige Salze sich durch verschiedene pharmakologische Eigenschaften auszeichnen. Diese Verbindungen entsprechen der folgenden Formel:
a worin A einen Alkylenrest mit 1 bis 4 Kohlenstoffatomen oder einen Alkenylenrest mit 3 oder 4 Kohlenstoffatomen und Ri und R2 jeweils Wasserstoff atome, Alkylreste mit 1 bis 4 Kohlenstoffatomen, Alkenylreste mit 3 oder 4 Kohlenstoffatomen, Alkinylreste mit 3 oder 4 Kohlenstoffatomen, Cycloalkylalkylreste, deren Cycloalkylkomponente 3 bis 6 Kohlenstoffatome und deren Alkylkomponente 1 bis 3 Kohlenstoffatome aufweisen, Aralkylreste, deren Alkylkomponente 1 bis 3 Kohlenstoffatome aufweist, oder Polyhalogenalkyl-reste, deren Alkylkomponente 2 bis 4 Kohlenstoffatome aufweist, bedeuten oder aber Rt und R2, wenn sie mit dem benachbarten Stickstoffatom zusammengenommen werden, einen 5- bis 7gliedrigen, stickstoffhaltigen, heterocyclischen Ring bilden, welcher ein weiteres Heteroatom enthalten kann.
Bei den obigen Bedeutungen bedeutet der Ausdruck «Alkylenrest mit 1 bis 4 Kohlenstoffatomen» eine geradkettige oder verzweigte Alkylengruppe mit 1 bis 4 Kohlenstoffatomen, z. B. Methylen, Äthylen, Propylen, Butylen, 1-Methyläthylen, 1-Methylpropylen oder 2-Methylpropylen. Die Bezeichnung «Alkenylenreste mit 3 oder 4 Kohlenstoffatomen» umfassen insbesondere 1-Propenylen, 1-Butenylen, 2-Butenylen, 1-Me-thyl-l-propenylen und 2-Methyl-l-propenylen, wobei die Numerierung vom Kohlenstoffatom ausgeht, welches am 9,10-Di-hydro-9,10-methanoanthracenskelett haftet. Die Bezeichnung «Alkylrest mit 1 bis 4 Kohlenstoffatomen» bedeutet geradkettige oder verzweigte Alkylreste mit 1 bis 4 Kohlenstoffatomen, wie Methyl, Äthyl, n-Propyl, Isopropyl, n-Butyl, sek.-Butyl oder Isobutyl. Die Bezeichnung «Alkenylreste mit 3 oder 4 Kohlenstoffatomen» bedeutet eine geradkettige oder verzweigte Alkenylgruppe mit 3 oder 4 Kohlenstoffatomen, wie Propenyl oder Butenyl. Die Bezeichnung «Alkinyl mit 3 oder 4 Kohlenstoffatomen» bedeutet eine geradkettige oder verzweigte Alkinylgruppe mit 3 oder 4 Kohlenstoffatomen, wie Propargyl. Die Bezeichnung «Cycloalkylalkylreste» bedeutet eine geradkettige oder verzweigte Alkylgruppe mit 1 bis 3 Kohlenstoffatomen, welche eine Cycloalkylgruppe mit 3 bis 6 Kohlenstoffatomen aufweist, wie Cyclopropylmethyl oder Cyclobutylmethyl. Die Bezeichnung «Aralkyl» bedeutet eine geradkettige oder verzweigte Alkylgruppe mit 1 bis 3 Kohlenstoffatomen, z. B. Methyl, Äthyl oder Propyl, wobei diese Alkylgruppe eine Arylgruppe, z. B. die Phenylgruppe, trägt. Die Bezeichnung «Polyhalogenalkyl» bedeutet eine verzweigte oder geradkettige Alkylgruppe mit 2 bis 4 Kohlenstoffatomen, welche zwei oder mehr Halogenatome enthält, wie Trifluoräthyl, Trichloräthyl oder Trifluorpropyl. Als 5-bis 7gliedriger, stickstoffhaltiger, heterocyclischer Ring seien die Pyrrolidino-, Piperidino-, Morpholino- und Thiomorpho-linoreste genannt.

können organische oder anorganische Säureadditionssalze davon sein, wie z. B. Hydrochloride, Hydrobromide, Acetate, Oxalate, Citrate, Tartrate, Succinate, Fumarate und Lactate.
Die 9-Aminoalkyl-9,10-dihydro-9,1O-methanoanthracen-derivate (nachstehend als 9-Aminoalkylmethanoanthracen-derivate bezeichnet) der Formel (I) zeichnen sich durch die Anwesenheit einer Aminoalkylseitenkette in der 9-Stellung des 9,10-Dihydro-9,10-methanoanthracenskeletts aus.
Wenngleich viele tricyclische Dibenzoverbindungen bekanntgeworden sind und auch einige davon klinisch als Arzneimittel, insbesondere als psychotrope Mittel, Verwendung finden, ist doch festzuhalten, dass bisher keine tricyclische Dibenzoverbindungen, welche einen 9,10-Dihydro-9,10-me-thanoanthracenring als tricyclisches Dibenzoskelett aufweisen, für solche Zwecke Verwendung gefunden haben. Die Schaffung von 9-Aminoalkylmethanoanthracenderivaten der Formel (I) konnte deshalb mit Erfolg nun geschehen, weil es gelang, das Schlüsselzwischenprodukt, nämlich 9-Formyl-9,10-dihydro-9,10-methanoanthracen der Formel:
mit Erfolg zu synthetisieren.
Die 9-Aminoalkyl-methanoanthracenderivate der Formel (I) sind neue Verbindungen und zeichnen sich durch eine grosse Vielfalt von wertvollen pharmakologischen Eigenschaften, insbesondere bezüglich des zentralen und autonomen Nervensystems, aus. Insbesondere besitzen die 9-Aminoalkyl-methanoanthracenderivate der Formel (I), worin A den Methylenrest darstellt, welcher durch einen Alkylrest mit 1 bis 3 Kohlenstoffatomen substituiert sein kann, eine potenzierende Wirkung der Hexobarbitalanästhesie, eine Hypothermie- und Ptosiswirkung sowie eine muskelrelaxierende Wirkung und auch eine Antitetrabenazinwirkung. Diese neuen Verbindungen eignen sich daher als anxiolytische und antidepressive Arzneimittel und ebenfalls als Major Tranquilizers.
Die 9-Aminoalkylmethanoanthracenderivate der Formel (I), worin A den Äthylenrest darstellt, welcher als Sub-stituenten mindestens einen Alkylrest mit 1 oder 2 Kohlenstoffatomen tragen kann, besitzen starke antihistaminische und anticholinergische Wirkungen und Antiserotoninwirkungen. Sie besitzen ferner eine Antitetrabenzinwirkung. Daher eignen sie sich als Antihistaminika und als Antiallergika.
Die 9-Aminoalkylmethanoanthracenderivate der Formel (I), worin A einen Alkylenrest mit 3 oder 4 Kohlenstoffatomen oder Alkenylenrest mit 3 oder 4 Kohlenstoffatomen bedeutet, besitzen eine starke Antitetrabenazinwirkung. Sie entfalten auch eine Noradrenalin-potenzierende Wirkung und Antireserpin-, Antihistamin-, Antiacetylcholin- und Antisero-toninwirkung. Zudem besitzen diese Verbindungen eine sehr geringe akute Toxizität und akute Kardiotoxizität. Die neuen Verbindungen eignen sich daher als antidepressive Mittel und als Antihistaminika.
Generell betrachtet, besitzen die 9-Aminoalkylmethano-anthracenderivate der Formel (I) durchwegs Antitetra-benazin-, Antiacetylcholin-, Antihistamin- und Antiserotoninwirkungen und auch sedative Wirkungen. Als Mittel gegen Angstgefühle (Anxiolytica) werden jene Verbindungen der Formel (1) bevorzugt, worin Rj einen Alkylrest mit 1 oder 2 Kohlenstoffatomen, R2 das Wasserstoffatom oder einen Alkylrest mit 1 oder 2 Kohlenstoffatomen und A den Methylenrest bedeuten. Als Antihistaminika oder Antiallergika werden jene Verbindungen der Formel (I) bevorzugt, worin R! einen Alkylrest mit 1 oder 2 Kohlenstoffatomen, R2 das Wasserstoffatom oder einen Alkylrest mit 1 oder 2 Kohlenstoffatomen und A die Äthylengruppe bedeuten. Bevorzugte antidepressive Mittel sind Verbindungen der Formel (I), worin A den Propylen- oder Propenylenrest bedeutet. Besonders bevorzugt werden jene Verbindungen der Formel (I), worin Ri und R2, welche gleich oder verschieden sein können, Wasserstoffatome oder Alkylreste mit 1 oder 2 Kohlenstoffatomen und A den Propylen- oder Propenylenrest bedeuten. Die besonders bevorzugten Verbindungen sind jene, worin A den Propylenrest, Ri das Wasserstoffatom oder den Methylrest und R2 den Methylrest bedeuten.
Die 9-Aminoalkylmethanoanthracenderivate der Formel (I) und deren nichttoxische, pharmazeutisch zulässige iSalze können oral oder parenteral in Dosierungen von normalerweise 5 bis 500 mg/Körpergewicht, vorzugsweise 25 bis 500 mg/Körpergewicht (ungefähr 60 kg Körpergewicht) pro Tag in Form eines üblichen, pharmazeutischen Präparates verabreicht werden.
So kann man diese Verbindungen beispielsweise in Form von üblichen festen, pharmazeutischen Präparaten, wie Pulvern, Granulaten, Tabletten oder Kapseln, oder in Form von üblichen flüssigen pharmazeutischen Präparaten, z. B. Suspensionen, Emulsionen oder Lösungen, verabreichen. Solche Präparate können dadurch erhalten werden, dass man ein 9-Ami-noalkylmethanoanthracenderivat der Formel (I) oder ein nichttoxisches, pharmazeutisch zulässiges Salz davon entweder allein verwendet oder in geeignete Hilfsmitteln, wie Stärke, Lactose oder Talk, in bekannter Weise einarbeitet.
Die 9-Aminoalkylmethanoanthracenderivate der Formel (I) können aus 9-Formyl-9,10-dihydro-9,10-methano-anthracen der Formel (II), in der Praxis durch Vermittlung geeigneter Derivate, erhalten werden.

durch Reduktion erhalten.
Diese Reduktion kann man mit Vorteil mit einem Reduktionsmittel, wie z. B. einem Alkalimetall in einem alkoholischen Lösungsmittel, einem Metallhydrid oder dergleichen, er- s folgen. Man kann sich auch einer elektrolytischen Reduktion bedienen.
Vorzugsweise wird man ein Metallhydrid, z. B. Lithiumaluminiumhydrid, Natriumaluminiumdiäthyldihydrid oder Na-trium-bis-(2-methoxyäthoxy)-aluminiumhydrid, in einem iner- io ten organischen Lösungsmittel, z. B. in einem Äther, wie Di-äthyläther, Diisopropyläther, Tetrahydrofuran, Dioxan oder Äthylenglycoldimethyläther, in einem aliphatischen Kohlenwasserstoff, z. B. Heptan, Hexan oder Cyclohexan, in einem aromatischen Kohlenwasserstoff, z. B. Benzol oder Toluol, 15 oder in einem Gemisch davon verwenden. Die für diese Behandlung erforderliche Temperatur schwankt zwischen Eiskühlung und Rückflusstemperatur des Reduktionssystems.
Natriumborhydrid ist ein anderes Beispiel von in der Praxis als Reduktionsmittel verwendbaren Metallhydriden und 20 insbesondere dann, wenn man in Gegenwart eines Salzes, z. B. Aluminiumchlorid, oder durch Aktivierung der Carboxamid-gruppe in einer Verbindung der Formel (III) mit Triäthyloxo-niumfluoroborat oder dergleichen arbeitet. Diboran ist ein weiteres Beispiel eines Metallhydrids, das als Reduktionsmit- 25 tel wirksam ist.

Diese Reduktion kann in der gleichen Weise, wie sie im Zusammenhang mit der Reduktion einer Verbindung der Formel (III) weiter oben beschrieben worden ist, oder durch ka-talytische Hydrierung erfolgen.
Die 9-AminoalkyImethanoanthracenverbindungen der Formel (I) lassen sich aus dem Reaktionsgemisch in bekannter Weise abtrennen und reinigen. Die 9-Aminoalkylmethano-anthracenverbindungen können in an sich bekannter Weise in die entsprechenden Salze übergeführt werden, wobei aber auch die erwähnten Salze in an sich bekannter Weise wiederum in die ursprüngliche freie Base übergeführt werden können.
Die als Schlüsselfigur zu bezeichnende Zwischenverbindung, d. h. 9-Formyl-9,10-dihydro-9,10-methanoanthracen der Formel (II), kann aus 9-Amino-12-hydroxy-9,10-dihydro-9,10-äthanoanthracen der Formel:

Diese Umlagerung von Aminen und a-Aminoalkoholderi-vaten durch Behandeln mit salpetriger Säure ist als Demjanov-Umlagerung und Tiffeneu-Demjanov-Umlagerung [Organic Reactions, Bd. 11, S. 157, John Wiley & Sons, Inc.] bekannt. Diese Umlagerungsumsetzungen wurden in den meisten Fällen, über die berichtet wurde, zur Ringerweiterung angewandt, während nur wenige Fälle einer Anwendung der Umlagerungs-reaktion zur Ringverengung bekanntgeworden sind. Die Umlagerung eines 9,10-Äthanoanthracenderivates in 9-Formyl-9,10-dihydro-9,10-methanoanthracen wurde bisher nicht ge-offenbart, so dass es sich um ein neues Verfahren für die Herstellung von 9-Formyl-9,10-dihydro-9,10-methanoanthracen handelt.
Die Umlagerung von 9-Amino-12-hydroxy-9,10-dihydro-9,10-äthanoanthracen in 9-Formyl-9,10-dihydro-9,10-metha-noanthracen kann durch Behandlung mit salpetriger Säure durchgeführt werden. Man behandelt daher 9-Amino-12-hy-droxy-9,10-dihydro-9,10-äthanoanthracen mit salpetriger Säure oder einem Metallnitrit, z. B. Natriumnitrit, Kaliumnitrit, in einem sauren Medium, wie Essigsäure, Ameisensäure, Salzsäure, Bromwasserstoffsäure, Schwefelsäure oder Phosphorsäure, oder einer Lösung eines Gemisches solcher Säuren. Man kann auch ein inertes Lösungsmittel, z. B. Wasser, Methanol, Äthanol, Aceton, Benzol, Toluol, Chloroform, Dichloräthan, Dichlormethan, Diäthyläther, Äthylenglycol-dimethyläther, Tetrahydrofuran, Äthylacetat, Dimethylsulf-oxyd oder Dimethylformamid oder ein Gemisch davon, in diesem Reaktionssystem verwenden. Die Temperatur für die Behandlung in diesem Falle schwankt zwischen Eiskühlung und Rückflusstemperatur des Reaktionssystems.
Das so erhaltene 9-Formyl-9,10-dihydro-9,10-methano-anthracen der Formel (II) kann aus dem Reaktionsgemisch in bekannter Weise abgetrennt und gereinigt werden.
Die Verbindung [A], d. h. das 9-Amino-12-hydroxy-
9,10-dihydro-9,10-äthanoanthracen, kann aus einer Verbindung der folgenden Formel:

worin R das Wasserstoffatom oder eine die Hydroxylgruppe 15 schützende Gruppe, wie die Acetylgruppe, Benzoylgruppe oder Tetrahydopyranylgruppe, bedeutet, durch Umlagerung, z. B. gemäss Curtius-Reaktion oder Hoffman-Umlagerung, und Hydrolyse erhalten werden. Die Umlagerung kann beispielsweise nach üblichen Methoden gemäss Curtius-Reaktion 2o [Organic Reactions, Bd. 3, S. 337, John Wiley & Sons, Inc.] erfolgen, während die Hydrolyse unter üblichen Hydrolysebedingungen, wie sie für Urethan- und Isocyanatderivate bekannt sind, durchgeführt werden kann.
Die Zwischenprodukte für die Synthese der 9-Aminoalkyl-25 methanoanthracenverbindungen der Formel (I) können aus 9-Formyl-9,10-dihydro-9,10-methanoanthracen der Formel (II) unter Anwendung üblicher Reaktionsbedingungen, wie Oxydation, Reduktion, Hydrolyse, Reaktion zur Verlängerung der Kohlenstoffkette (Substitution, Wittigreaktion, 30 Reformatskyreaktion, Grignardreaktion) usw. hergestellt werden.
Die für die Synthese von 9-Aminomethyl-9,10-dihydro-9,10-methanoanthracenderivaten verwendeten Ausgangsmaterialien können beispielsweise wie folgt hergestellt werden:

(2 ) 9-Hydroxymethyl-9,10-dihydro-9,1O-methanoanthra-cen wird aus 9-Formyl-9,10-dihydro-9,10-methanoanthracen hergestellt, und zwar durch Behandlung mit einem Reduktionsmittel, wie z. B. Natriumborhydrid oder Lithiumaluminiumhydrid, in einem inerten Lösungsmittel;
(3) 9-Tosyloxymethyl-9,10-dihydro-9,10-methanoanthra-cen wird aus 9-Hydroxymethyl-9,10-dihydro-9,10-methano-anthracen hergestellt, und zwar durch Behandlung mit p-To-luolsulfonylchlorid in Gegenwart einer Base und in einem inerten Lösungsmittel;
(4)9,10-Dihydro-9,10-methanoanthracen-9-carbonsäure wird durch Umsetzung mit Thionylchlorid in Gegenwart oder in Abwesenheit eines inerten Lösungsmittels in das entsprechende Säurechlorid übergeführt, worauf dieses Säurechlorid durch Umsetzung mit Ammoniak in an sich bekannter Weise in das 9,10-Dihydro-9,10-methanoanthracen-9-carboxamid 5 übergeführt wird;
(5) die Dehydratisierung von 9,10-Dihydro-9,10-metha-noanthracen-9-carboxamid in 9,10-Dihydro-9,10-methano-anthracen-9-carbonitril erfolgt unter Verwendung von Phos-phoroxychlorid in Gegenwart oder in Abwesenheit eines iner-10 ten Lösungsmittels.
Die Ausgangsmaterialien für die Synthese von 9-/3-Amino-äthyl-9,10-dihydro-9,10-methanoanthracenderivaten kann beispielsweise aus 9-Formyl-9,10-dihydro-9,10-methanoan-thracen der Formel (II) oder einem Derivat davon in folgen-15 der Weise durchgeführt werden:

(10) Der [9,10-Dihydro-9,10-methano-9-anthryl]-acet-aldehyd wird aus 9-Formyl-9,10-dihydro-9,10-methano-anthracen unter Anwendung des Verfahrens nach Wittig mittels Methoxymethyltriphenylphosphoniumchlorid und durch saure Hydrolyse erhalten.
(11) [9,10-Dihydro-9,10-methano-9-anthryl]-acetonitril kann aus 9-Tosyloxymethyl-9,10-dihydro-9,10-methano-anthracen durch Umsetzung mit einem Metallcyanid in einem inerten Lösungsmittel erhalten werden.

(17) und (18) die /J-[9,10-Dihydro-9,10-methano-9-an-thryl]-propionsäure wird in der oben beschriebenen Weise in /?-[9,10-Dihydro-9,10-methanoanthryl]-propionitril übergeführt;
(19) ß-[9,1 O-Dihydro-9,10-methano-9-anthryl]-acryl-säurealdehyd wird gemäss Wittig-Reaktion mittels Formyl-methylentriphenylphosphoran aus 9-Formyl-9,10-dihydro-9,10-methanoanthracen erhalten.
Die Derivate dery-[9,10-Dihydro-9,10-methano-9-an-thrylj-buttersäure können in der gleichen Weise, wie dies oben beschrieben worden ist, erhalten werden.
Die Verbindungen der Formel (III) können aus den entsprechenden Carbonsäurederivaten in an sich bekannter Weise mit den entsprechenden Aminverbindungen hergestellt werden. Andere Zwischenprodukte, z. B. Verbindungen der Formel (VII), können aus einer Verbindung der Formel (Id)
durch Umsetzung mit einer Verbindung der folgenden Formel:
R'4COY oder R'4C0-0-C0R'4
worin R'4 die obige Bedeutung hat und Y ein Halogenatom, z. B. das Chlor- oder Bromatom, darstellt, unter üblichen Bedingungen für die Acylierung einer Aminverbindung erhalten werden.
Die folgenden Beispiele erläutern die vorliegende Erfindung.
Herstellung der Zwischen- und Ausgangsprodukte
(a) Zu einer Lösung von 3,0 g 9-Amino-12-hydroxy-
9,10-dihydro-9,10-äthanoanthracen in 240 ml Essigsäure gibt man eine Lösung von 6,7 g Natriumnitrit in 120 ml Wasser bei 2 bis 5° C hinzu, worauf man das Gemisch während 1 Stunde bei der gleichen Temperatur und hierauf während 5 Stunden bei 95 bis 105° C rührt. Das Reaktionsgemisch wird hierauf mit Wasser verdünnt und mit Benzol extrahiert. Die Benzolschicht wird mit Wasser gewaschen, über Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man 2,8 g rohe Kristalle von 9-Formyl-9,10-dihydro-9,10-methanoanthracen erhält, welche man umkristallisiert. Auf diese Weise gelangt man zu farblosen Kristallen in einer Menge von 2,45 g vom Schmelzpunkt 99 bis 100° C. Durch weitere Reinigung durch Umkristallisieren erhält man analytisch reines 9-Formyl-9,10-dihydro-9,10-methanoanthracen vom Schmelzpunkt 102,5 °C.
(b) Zu einer Lösung von 50 mg 9-Amino-12-hydroxy-9,10-dihydro-9,10-äthanoanthracen in 2 ml konz. Salzsäure und 2 ml Wasser gibt man eine Lösung von 112 mg Natriumnitrit in 1,0 ml Wasser bei 0° C hinzu. Das so erhaltene Gemisch wird 1 Stunde bei 0° C und dann während 5 Stunden bei Zimmertemperatur gerührt. Das Reaktionsgemisch wird schliesslich mit Wasser verdünnt und mit Benzol extrahiert. Dann wird die Benzolschicht mit Wasser gewaschen, über Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man 35 mg rohe Kristalle von 9-Formyl-9,10-dihydro-9,10-methanoanthracen erhält.
(c) Eine Lösung von 1,0 g 12-Acetoxy-9,10-dihydro-9,10-äthanoanthracen-9-carbonsäure in 10 ml Benzol und 4,0 ml Thionylchlorid wird während 4 Stunden unter Rück-fluss zum Sieden erhitzt. Durch Verdampfen des überschüssigen Thionylchlorids und des Benzols erhält man 12-Acetoxy-9,10-dihydro-9,10-äthanoanthracen-9-carbonsäurechlorid. Das Säurechlorid wird dann in 25,0 ml trockenem Aceton gelöst und diese Lösung unter Kühlen mit Eis mit einer Lösung von 0,63 g Natriumazid in 1,3 ml Wasser versetzt. Das erhaltene Gemisch wird unter Eiskühlung während 2 Stunden gerührt. Dann wird das Reaktionsgemisch mit Wasser verdünnt und mit Benzol extrahiert. Der Benzolextrakt wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet, während 2 Stunden unter Rückfluss zum Sieden erhitzt und zur Trockne eingedampft, wobei man 9-Isocyanato-12-acet-oxy-9,10-dihydro-9,10-äthanoanthracen erhält.
Die Isocyanatverbindung wird in 12,0 ml Äthanol und in 12,0 ml einer 20%igen wässrigen Natriumhydroxydlösung gelöst und die so erhaltene Lösung während 6 Stunden unter Rückfluss zum Sieden erhitzt. Nach dem Verdampfen des Äthanols wird das Reaktionsgemisch mit Wasser verdünnt und mit Äthylacetat extrahiert. Der Äthylacetatextrakt wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man 0,72 g Kristalle von 9-Amino-12-hydroxy-9,10-dihydro-9,10-äthanoanthracen erhält. Diese Kristalle schmelzen bei 181 bis 181,5° C. Durch Umkristallisieren aus Benzol erhält man analytisch reine Kristalle von 9-Amino-12-hydroxy-9,10-dihydro-9,10-äthanoanthracen vom Schmelzpunkt 183,5° C.
(d) Ein Gemisch von 110 mg 9-Formyl-9,10-dihydro-9,10-methanoanthracen und 186 mg/3-Carboxyäthyltriphe-nylphosphoniumchlorid in 2 ml Dimethylsulfoxyd und 2 ml Tetrahydrofuran wird mit einer 65,4%igen Natriumhydriddispersion in 37 mg Mineralöl bei 0° C in einer Stickstoffatmosphäre versetzt. Das Reaktionsgemisch wird während
6 Stunden bei 0° C gerührt, dann mit Wasser verdünnt, mit Salzsäure angesäuert und anschliessend mit Benzol extrahiert. Der Benzolextrakt wird mit wässriger 2n Natriumhydroxydlösung geschüttelt. Die basische Schicht wird mit Salzsäure angesäuert und mit Äthylacetat extrahiert. Der Äthylacetatextrakt wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man y-(9,1 O-Dihydro-9,10-methano-9-anthryl)-/3 -buten-säure vom Schmelzpunkt 166 bis 167° C erhält.
(e) 2,65 g Triäthylphosphonoacetat in Benzol wird mit einer 50%igen Natriumhydriddispersion in 0,66 g Mineralöl behandelt und dann bei Zimmertemperatur in einer Stickstoffatmosphäre mit einer Lösung von 2,0 g 9-Formyl-9,10-di-hydro-9,10-methanoanthracen in 20,0 ml Benzol versetzt. Das Reaktionsgemisch wird hierauf während 5 Stunden bei Zimmertemperatur und dann anschliessend während 1 Stunde bei 70° C gerührt, mit Wasser verdünnt und mit Äthylacetat extrahiert. Der Äthylacetatextrakt wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man den/3-(9,10-Dihydro-9,10-methano-9-anthryl)-acrylsäureäthylester erhält. Eine Lösung dieses Äthylesters in 53 ml Methanol und 12 ml einer 10%igen wässrigen Natriumhydroxydlösung wird während 4 Stunden unter Rückfluss zum Sieden erhitzt. Dann wird das Reaktionsge5

misch mit Wasser verdünnt, mit Salzsäure angesäuert und mit Äthylacetat extrahiert. Der Äthylacetatextrakt wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man/3-(9,10-Dihydro-9,10-methano-9-anthryl)-acrylsäure erhält. Schmelzpunkt 219,5 bis 222° C.
(f) Ein Gemisch von 612 mg/ö-(9,10-Dihydro-9,10-me-thano-9-anthryl)-acrylsäure und 120 mg 5%igem Palladiumauf-Kohle in Äthanol wird in einer Wasserstoffatmosphäre während 2 Stunden bei Zimmertemperatur gerührt. Der Katalysator wird durch Filtrieren entfernt und die Lösung zur Trockne eingedampft, wobei man /3-(9,l O-Dihydro-9,10-me-thano-9-anthryI)-propionsäure vom Schmelzpunkt 185 bis
189° C erhält.
(g) Eine Lösung von ß-(9,1 O-Dihydro-9,10-methano-9-anthryl)-propionsäure und Thionylchlorid in Benzol wird während 4 Stunden unter Rückfluss zum Sieden erhitzt. Durch Verdampfen des überschüssigen Thionylchlorids und des Benzols erhält man ß-(9,1 O-Dihydro-9,10-methano-9-anthryl)-propionsäurechlorid, das man in trockenem Tetrahydrofuran löst. Diese Lösung wird einer 30%igen wässrigen Monomethyl-aminlösung bei 0 bis 5° C hinzugegeben. Dann wird das Reaktionsgemisch bei 0 bis 15° C gerührt, mit Wasser verdünnt und mit Äthylacetat extrahiert. Die Äthylacetatschicht wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man
/5-(9,l O-Dihydro-9,10-methano-9-anthryl)-propionsäure-monomethylamid vom Schmelzpunkt 200 bis 201° C erhält.
(h) Eine Lösung von 0,7 g /3-(9,1 O-Dihydro-9,10-me-thano-9-anthryl)-propionsäure und Thionylchlorid in Benzol wird während 2 Stunden unter Rückfluss zum Sieden erhitzt und dann zur Trockne eingedampft, wobei man das entsprechende Säurechlorid erhält, das man in 3,7 ml Aceton löst.
Eine Lösung von 0,52 g Natriumazid in Wasser wird unter Eiskühlung der Acetonlösung hinzugegeben. Dann wird das Gemisch während 2 Stunden bei 0°C gerührt. Das Reaktionsgemisch wird anschliessend mit Wasser verdünnt und mit Benzol extrahiert. Die Benzolschicht wird mit Wasser gewaschen, über Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man das entsprechende Säureazid erhält. Eine Lösung dieses Säureazids in 7,5 ml Äthanol wird während 10 Stunden unter Rückfluss zum Sieden erhitzt und zur Trockne eingedampft, wobei man 9-/3-Äthoxycarbonylaminoäthyl-9,10-dihydro-9,10-methanoanthracen vom Schmelzpunkt 122 bis 123° C erhält.
(i) Eine Lösung von 235 mg 9-Aminomethyl-9,10-di-hydro-9,10-methanoanthracen und 217 mg Essigsäureanhydrid in 5,0 ml Äthanol wird während 3 Stunden unter Rückfluss zum Sieden erhitzt. Das Reaktionsgemisch wird hierauf mit Wasser verdünnt und mit Äthylacetat extrahiert. Der Äthylacetatextrakt wird mit Wasser, dann mit wässriger Na-triumbicarbonatlösung und erneut mit Wasser gewaschen,
über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man 9-Acetylaminomethyl-9,10-dihydro-9,10-methanoanthracen vom Schmelzpunkt 184 bis 185,5° C erhält.
(j) Eine Lösung von 30 mg 9,1 O-Dihydro-9,10-methano-anthracen-9-carboxamid und 0,15 ml Thionylchlorid in 1 ml Toluol wird während 18 Stunden unter Rückfluss zum Sieden erhitzt. Durch Verdampfen des Toluols und des überschüssigen Thionylchlorids erhält man 9,10-Dihydro-9,10-methano-anthracen-9-carbonitril vom Schmelzpunkt 120 bis 123° C.
(k) Ein Gemisch von 188 g 9-Tosyloxymethyl-9,10-di-hydro-9,10-methanoanthracen und 40 mg Kaliumcyanat in 2 ml Dimethylformamid wird während 7 Stunden auf 150° C erhitzt. Das Reaktionsgemisch wird dann mit Wasser verdünnt und mit Benzol extrahiert. Der Benzolextrakt wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man rohe Kristalle von (9,10-Dihydro-9,10-methano-9-anthryl)-acetonitril erhält. Dieses Material wird aus Isopropanol umkristallisiert, wobei man zu reinen Kristallen vom Schmelzpunkt 130 bis 131 °C gelangt.
Beispiel 1
Ein Gemisch von 1,0 g ß-(9,1 O-Dihydro-9,10-methano-9-anthryl)-propionsäure-monomethylamid und 0,5 g Lithiumaluminiumhydrid wird während 2 Stunden bei 50° C gerührt. Das überschüssige Lithiumaluminiumhydrid wird durch Zugabe von Wasser zersetzt. Dann wird das Reaktionsgemisch mit Äthylacetat verdünnt, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man 9-/-MethyIaminopropyl-9,10-dihydro-9,10-methano-anthracen erhält, das man in das Hydrochlorid vom Schmelzpunkt 247 bis 249° C überführt. Durch Umkristallisieren aus Isopropylalkohol erhält man farblose Kristalle vom Schmelzpunkt 259 bis 260° C.
Beispiel 2
Ein Gemisch von 70 mg 9-y-Acetylaminopropyl-9,10-di-hydro-9,10-methanoanthracen und 35 mg Lithiumaluminiumhydrid in 2 ml Dioxan wird während 9 Stunden bei 40 bis 50° C gerührt. Das überschüssige Lithiumaluminiumhydrid wird durch Zugabe von Wasser zersetzt. Dann wird das Reaktionsgemisch mit Äthylacetat verdünnt, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man 9-y-Äthylaminopropyl-9,10-dihydro-9,10-methanoan-thracen erhält, das man in das Chlorhydrat vom Schmelzpunkt 182 bis 186° C überführt.
Beispiel 3
Ein Gemisch von 100 mg 9-y-N-Acetyl-N-allylaminopro-pyl-9,10-dihydro-9,10-methanoanthracen in Äthanol und 25 %iger wässriger Natriumhydroxydlösung wird während 10 Stunden unter Rückfluss zum Sieden erhitzt. Dann wird das Reaktionsgemisch mit Wasser verdünnt und mit Äthylacetat extrahiert. Die Äthylacetatschicht wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man 9-y-Allylaminopropyl-9,10-dihydro-9,10-methanoanthracen erhält, das man in das Chlorhydrat vom Schmelzpunkt 227 bis 228° C überführt.
Beispiel 4
Ein Gemisch von 250 mg /3-(9,1 O-Dihydro-9,10-methano-9-anthryl)-propionitril und 100 mg Lithiumaluminiumhydrid in 12 ml Dioxan wird während 5 Stunden bei 60° C gerührt. Das überschüssige Lithiumaluminiumhydrid wird hierauf durch Zugabe von Wasser versetzt. Das Reaktionsgemisch wird anschliessend mit Äthylacetat verdünnt, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man 9-y-Aminopropyl-9,10-dihydro-9,10-methano-anthracen erhält, das man in das Hydrochlorid vom Schmelzpunkt 275 °C (unter Zersetzung) überführt.
Die folgenden Verbindungen lassen sich in ähnlicher Weise herstellen:

- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass die Reduktion elektrolytisch erfolgt oder dass man hierfür ein Reduktionsmittel verwendet.2PATENTANSPRÜCHE 1. Verfahren zur Herstellung von Verbindungen der folgenden Formel:
- *3 CH2"r4worin A einen Alkylenrest mit 1 bis 4 Kohlenstoffatomen oder einen Alkenylenrest mit 3 oder 4 Kohlenstoffatomen, R3 das Wasserstoffatom, einen Alkylrest mit 1 bis 4 Kohlenstoffatomen, einen Alkenylrest mit 3 oder 4 Kohlenstoffatomen, einen Cycloalkylalkylrest, dessen Cycloalkylkomponente 3 bis 6 Kohlenstoffatome und dessen Alkylkomponente 1 bis 3 Kohlenstoffatome aufweisen, einen Aralkylrest, dessen Alkylkomponente 1 bis 3 Kohlenstoffatome aufweist, oder einen Polyhalogenalkylrest mit 2 bis 4 Kohlenstoffatomen und R4 das Wasserstoffatom, einen Alkylrest mit 1 bis 3 Kohlenstoffatomen, einen Alkenylrest mit 2 oder 3 Kohlenstoffatomen, einen Arylrest oder einen Cycloalkylrest mit 3 bis 6 Kohlenstoffatomen bedeuten, sowie von nichttoxischen, pharmazeutisch zulässigen Salzen davon, dadurch gekennzeichnet, dass man eine Verbindung der folgenden Formel:3540worin A und R3 die obigen Bedeutungen haben und R/ das Wasserstoffatom, einen Alkylrest mit 1 bis 3 Kohlenstoffatomen, einen Alkenylrest mit 2 oder 3 Kohlenstoffatomen, einen Arylrest, einen Cycloalkylrest mit 3 bis 6 Kohlenstoffatomen oder einen Alkoxyrest mit 1 bis 4 Kohlenstoffatomen bedeutet, reduziert.3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man als Reduktionsmittel ein Alkalimetall in einem alkoholischen Lösungsmittel oder ein Metallhydrid verwendet.
- 4. Verfahren nach Anspruch 2, dadurch gekennzeichnet, dass man die Reduktion in einem inerten organischen Lösungsmittel durchführt.
- 5. Verfahren nach Anspruch 2, dadurch gekennzeichnet, dass man die Reduktion bei einer Temperatur zwischen Eiskühlung und Rückflusstemperatur durchführt.5055
- 6. Verfahren zur Herstellung von Verbindungen der folgenden Formel:
- 7. Verfahren nach Anspruch 6, dadurch gekennzeichnet, dass man die Reduktion elektrolytisch oder unter Verwendung eines Reduktionsmittels durchführt.
- 8. Verfahren nach Anspruch 7, dadurch gekennzeichnet, dass man als Reduktionsmittel ein Alkalimetall in einem alko-60 holischen Lösungsmittel oder ein Metallhydrid verwendet.
- 9. Verfahren nach Anspruch 6, dadurch gekennzeichnet, dass man die Reduktion in einem inerten Lösungsmittel durchführt.
- 10. Verfahren nach Anspruch 6, dadurch gekennzeichnet, 65 dass man die Reduktion bei einer Temperatur zwischen Eiskühlung und Rückflusstemperatur durchführt.1015worin A' eine Direktbindung, einen Alkylenrest mit 1 bis 3 Kohlenstoffatomen oder einen Alkenylenrest mit 2 oder 3 Kohlenstoffatomen und Ra und R2 jeweils Wasserstoffatome, Alkylreste mit 1 bis 4 Kohlenstoffatomen, Alkenylreste mit 3 oder 4 Kohlenstoffatomen, Alkinylreste mit 3 oder 4 Kohlenstoffatomen, Cycloalkylalkylreste, deren Cyclo-alkylkomponente 3 bis 6 Kohlenstoffatome und deren Alkyl-komponente 1 bis 3 Kohlenstoffatome aufweisen, Aralkyl-reste, deren Alkylkomponente 1 bis 3 Kohlenstoffatome aufweist, oder Polyhalogenalkylreste, deren Alkylkomponente 2 bis 4 Kohlenstoffatome aufweist, bedeuten oder aber Rx und R2, wenn sie mit dem benachbarten Stickstoffatom zusammengenommen werden, einen 5- bis 7gliedrigen, stickstoffhaltigen, heterocyclischen Ring bilden, welcher ein weiteres He-teroatom enthalten kann, sowie von nichttoxischen, pharmazeutisch zulässigen Salzen davon, dadurch gekennzeichnet,dass man eine Verbindung der Formel:202530worin A', R, und R2 die obigen Bedeutungen haben, reduziert.
- 11. Verfahren zur Herstellung von Verbindungen der folgenden Formel:A'If2' nh2worin A' eine Direktbindung, einen Alkylenrest mit 1 bis 3 Kohlenstoffatomen oder einen Alkenylenrest mit 2 oder 3 Kohlenstoffatomen bedeutet, sowie von pharmazeutisch zulässigen Salzen davon, dadurch gekennzeichnet, dass man eine Verbindung der folgenden Formel:A»worin A' die obige Bedeutung hat und R7 eine Nitrilgruppe (-C=N) oder eine Carbaldehydoximgruppe (-CH=NOH) bedeutet, einer Reduktion unterwirft.
