Description
Aufgabe der Erfindung ist ein neues und vorteilhaftes Verfahren zur Herstellung von 3-Hydroxy-4H-pyran-4-on-Deri-vaten. Das Verfahren soll weniger Reaktionsstufen und leichtere Techniken unter milderen Reaktionsbedingungen mit sich bringen, es soll weniger Nebenprodukte erzeugen und von leichter zugänglichen Ausgangsmaterialien ausgehen, sowie weniger Verschmutzung der Luft und des Abwassers, d.h. weniger Umweltbelastung ergeben.
Das erfindungsgemässe Verfahren zur Herstellung von 3-Hydroxy-4H-pyran-4-on-Derivaten der Formel (III) ist im Patentanspruch 1 definiert.
Die Ausgangsmaterialien für das erfindungsgemässe Verfahren, d.h. 3-Oxo-3,6-dihydro-2H-pyran-Derivate der Formel (I) (im folgenden als a,ß-ungesättigte Ketone bezeichnet) sind bekannte Verbindungen und können beispielsweise leicht in guter Ausbeute nach dem folgenden Reaktionsschema erhalten werden:

In den Formeln haben Rt und R2 die im Patentanspruch 1 angegebenen Bedeutungen und R3 ist eine geradkettige oder verzweigte Alkylgruppe mit 1 bis 4 Kohlenstoffatomen.
So kann ein 6-Hydroxy-3-oxo-3,6-dihydro-2H-pyran-Deri-vat durch elektrolytische Oxydation von Furfurylalkohol (4) in Gegenwart von R3OH zu 2,5-Dialkoxy-furfurylalkohol (5) und Behandlung des Alkohols mit einem sauren Katalysator zur Ringerweiterung erhalten werden; und 6-Alkoxy- oder 6-Acyloxy-3-oxo-3,6-dihydro-2H-pyran-Derivate können durch anschliessende Acetalisierung oder Acylierung des 6-Hydroxy-Derivats erhalten werden.
Wie oben gezeigt, können 6-^Iydroxy-3-oxo-3,6-dihydro--2H-pyran-Derivate und 6-Alkoxy- oder 6-Acyloxy-3-oxo-3,6--dihydro-2H-pyran-Derivate als Ausgangsmaterialien für das erfindungsgemässe Verfahren verwendet werden. Die zuletzt genannten 6-Alkoxy- bzw. 6-Acyloxy-3-oxo-3,6-dihydro-2H--pyran-Derivate sind jedoch chemisch stabiler und daher gegenüber den zuerst genannten 6-Hydroxy-Derivaten bevor-:zugt. Obwohl die Acetalisierung und Acylierung zur Herstellung des Ausgangsmaterials nach herkömmlichen Methoden durchgeführt werden können, wurde ein neues, verbessertes Verfahren zur Umwandlung der 6-Hydroxy-Derivate in die 6-Alkoxy-Derivate gefunden; dieses wird weiter unten eingehend beschrieben werden.
Das 6-Alkoxy-Derivat kann durch Acetalisierung des 6--Hydroxv-Derivats erhalten werden. Die Acetalisierung wurde bisher beispielsweise nach der in Tetrahedron, 27,1973-1996 (1971) beschriebenen Methode durchgeführt. Dieses Verfahren erfordert die Anwendung einer starken Lewis-Säure, wie beispielsweise Zinntetrachlorid oder Bortrifluorid als Katalysator und eine niedrige Reaktionstemperatur von 0°C und weniger und ergibt dennoch geringe Ausbeuten. In Anbetracht dieser Nachteile wurde umfangsreiche Forschungsarbeit geleistet, und überraschenderweise wurde ein Verfahren erfunden, nach dem das gewünschte Produkt in hohen Ausbeuten unter milden Reaktionsbedingungen erhalten werden kann. Nach diesem Verfahren wird die Umsetzung in einem Lösungsmittel unter Erhitzen durchgeführt, wobei Ortho-Ameisensäureester oder ein aliphatischer Alkohol als Acetalisierungsmittel und eine schwache Lewis-Säure als Katalysator dienen. Beispiele für Ortho-Ameisensäureester sind Ortho-Ameisensäureme-thylester, Ortho-Ameisensäureäthylester, Ortho-Ameisensäu-repropylester, Ortho-Ameisensäurebutylester usw. Beispiele für aliphatische Alkohole sind Methanol, Äthanol, Isopropa-nol, Butanol usw. Beispiele für schwache Lewis-Säuren, die für die Umsetzung verwendet werden können, sind beispielsweise Calciumchlorid, Ammoniumchlorid, Zinkchlorid, Kaliumchlorid, Magnesiumchlorid, Magnesiumsulfat, Calciumsulfat, Ammoniumnitrat, Natriumnitrat usw. Bevorzugte Lösungsmittel sind u.a. Diäthyläther, Diisopropyläther, und ähnliche aliphatische Äther, Tetrahydrofuran, Dioxan und ähnliche cyclische Äther, Methylchlorid, Chloroform, Kohlenstofftetrachlorid und ähnliche Kohlenwasserstoffhalogenide, Benzol, Toluol, Xylol und ähnliche aromatische Kohlenwasserstoffe

Die Menge des für die Umsetzung eingesetzten Acetali-sierungsmittels ist üblicherweise etwa 0,5 bis 10 Mol, vorzugsweise etwa 0,6 bis 2 Mol, pro Mol Ausgangsmaterial, d.h. 6--Hydroxy-3-oxo-3,6-dihydro-2H-pyran-Derivat. Die Menge des Katalysators beträgt üblicherweise etwa 0,05 bis 1 Mol, 2o vorzugsweise etwa 0,1 bis 0,3 Mol, pro Mol Ausgangsmaterial. Die Reaktionstemperatur, die von dem jeweils verwendeten Lösungsmittel abhängt, liegt üblicherweise zwischen 30 und 150°C, vorzugsweise zwischen etwa 50 und 80°C.
Die Acylierungsreaktion zur Herstellung des 6-Acyloxy-3-25 -oxo-3,6-dihydro-2H-pyran-Derivats aus dem 6-Hydroxy-3--oxo-3,6-dihydro-2H-pyran-Derivat kann nach einer herkömmlichen Methode durchgeführt werden, nämlich durch Umsetzung des Ausgangsmaterials, mit einem Acylierungsmit-tel in Gegenwart eines Katalysators in einem Lösungsmittel. 30 Beispiele für bevorzugte Acylierungsmittel sind Carbonsäureanhydride, wie beispielsweise Essigsäureanhydrid, Propion-säureanhydrid, Buttersäureanhydrid und dgl., sowie Acylhalo-genide, wie beispielsweise Acetylchlorid, Propionylchlorid, Butyrylchlorid und die diesen Chloriden entsprechenden Bro-35 mide. Beispiele für Katalysatoren sind Pyridin, Piperidin, Tri-äthylamin, Phosphorpentoxyd usw. Wertvolle Lösungsmittel sind beispielsweise die oben für die Acetalisierungsreaktion aufgeführten Lösungsmittel. Die Menge an Acylierungsmittel, die für die Umsetzung verwendet werden kann, liegt vorzugs-40 weise im Bereich von etwa 1 bis 1,5 Mol pro Mol Ausgangsmaterial. Die Reaktionstemperatur ist üblicherweise etwa-10 bis +50°C, vorzugsweise etwa 0 bis 30°C.
Beispiele für a, ß-ungesättigte Ketone ( 1 ), die als Ausgangsmaterial für das erfindungsgemässe Verfahren dienen können, 45 sind u.a.:

Nach dem genannten Verfahren wird ein 4,5-Epoxy-3-oxo--tetrahydro-pyran-Derivat (II) (im folgenden als «Epoxyke-ton» bezeichnet) durch Epoxydierung des a,ß-ungesättigten Ketons (I) mit einem Peroxyd hergestellt. Die Verbindung (I), das Ausgangsmaterial, hat eine sehr komplexe Struktur eines cyclischen Äthers, worin das Sauerstoffatom ein Teil des Ace-tals bildet. Obwohl es fast unbekannt war, solche a,ß-ungesättigte Ketone zu epoxydieren, gelang es überraschenderweise leicht, das Keton durch Behandlung mit Peroxyd unter den unten beschriebenen Bedingungen zu epoxydieren. Wertvolle Peroxyde für diese Umsetzung sind beispielsweise ganz allgemein bekannte, wie u.a. Wasserstoffperoxyd, Trifluorperessig-säure, Perbenzoesäure, Monochlorperbenzoesäure, Monoper-phthalsäure, tert.-Butylhydroperoxyd, Di-tert.-butylperoxyd usw. Es wird vorgezogen, die Epoxydierung des erfindungsge-mässen Verfahrens in Gegenwart von mindestens einer alkalischen Substanz durchzuführen, beispielsweise in Gegenwart von Natriumhydroxyd, Kaliumhydroxyd, Natriumcarbonat, Kaliumcarbonat, basisches Ionenaustauschharz oder dgl. Die Epoxydierung kann mit oder ohne Lösungsmittel durchgeführt werden. Beispiele für nützliche Lösungsmittel sind u.a. Wasser, Alkohole, wie beispielsweise Methanol, Äthanol, Iso-propanol, niedrige aliphatische organische Säuren, wie beispielsweise Essigsäure, Propionsäure, aliphatische Äther, wie beispielsweise Diäthyläther, Diisopropyläther, Methylcellosol-ve, Äthylcellosolve, cyclische Äther, wie beispielsweise Tetra-hydrofuran, Dioxan, Ester, wie beispielsweise Äthylacetat, Bu-tylacetat, und Kohlenwasserstoffhalogenide, wie beispielsweise Methylenchlorid, Chloroform, Tetrachlorkohlenstoff usw. Die Menge an Peroxyd, die für die Umsetzung verwendet wird, beträgt üblicherweise 0,5 bis 3 Mol, vorzugsweise etwa 1 bis 2 Mol, pro Mol des a,ß-ungesättigten Ketons. Die Menge an Alkali, die für die Umsetzung verwendet wird, beträgt üblicherweise etwa 0,1 bis 1 Mol, vorzugsweise etwa 0,2 bis 0,5 Mol, pro Mol des a,ß-ungesättigten Ketons. Die Umsetzung wird üblicherweise bei einer Temperatur von etwa -30 bis + 40°C, vorzugsweise etwa-20 bis +20°C, durchgeführt. Nach Beendigung der Umsetzung kann das Reaktionsgemisch üblichen Trenn- und Isolierstufen unterworfen werden. Wenn etwas Peroxyd nicht umgesetzt zurückbleibt, wird vorzugsweise etwas Natriumthiosulfatlösung zugegeben, um das Peroxyd in der Mischung zu inaktivieren und die mögliche Gefahr einer Explosion während der anschliessenden Aufarbeitung zu vermeiden.
Das Reaktionsgemisch wird zweckmässig mit Wasser gewaschen, die erhaltene ölige Schicht wird zweckmässig zur Entfernung des Lösungsmittels destilliert und das erhaltene ölige Produkt kann direkt in die anschliessende Umsetzung eingesetzt werden. Wahlweise kann das ölige Produkt im Vakuum destilliert werden oder der Säulenchromatographie unterworfen werden, um das gereinigte Epoxyketon zu erhalten, das für die anschliessende Umsetzung verwendet werden kann. Es wird vorgezogen, das isolierte gereinigte Epoxyketon für die anschliessende Umsetzung zu verwenden, um die anschliessende Behandlung zu erleichtern.

Das 3-Hydroxy-4H-pyran-4-on-Derivat (3), das nach dem erfindungsgemässen Verfahren erwünschte Produkt, wird 40 leicht durch Erhitzen des Epoxyketons (2) erhalten. Die Umwandlung des Epoxyketons (2) in das 3-Hydroxy-4H-pyran-4--on-Derivat durch Einwirkung von Hitze und vorzugsweise Säure ist ein völlig neues Verhalten, das sich von der üblichen Ringspaltung von Epoxy Verbindungen durch Hydrolyse un-45 terscheidet. Es ist eine neue, überraschende Umsetzung, die hier zum ersten Mal angewendet wird. Die Umsetzung erfolgt ohne Anwesenheit eines Katalysators, kann jedoch vorzugsweise in Gegenwart eines sauren Katalysators, wie beispielsweise Schwefelsäure, Chlorwasserstoffsäure, Bromwasserstoff-50 säure oder ähnlichen anorganischen Säuren, Ameisensäure, Essigsäure, Propionsäure oder ähnlichen organischen Säuren, Methansulfonsäure, Benzolsulfonsäure, Paratoluolsulfonsäure oder ähnlichen Sulfonsäuren, Bortrifluorid, Aluminiumchlorid, Zinkchlorid, Titantetrachlorid, Antimonpentachlorid, 55 Zinntetrachlorid oder ähnlichen Lewis-Säuren oder sauren Ionenaustauschharzen, durchgeführt werden. Bevorzugte saure Katalysatoren sind Schwefelsäure, Benzolsulfonsäure, Paratoluolsulfonsäure und saure lonenaustauschharze. Die für die Umsetzung verwendete Konzentration an Säure selbst kann 60 üblicherweise 10 bis 100 Gew.-%, vorzugsweise etwa 60 bis 90 Gew.-%, betragen, obwohl sie je nach Art der Säure variiert werden kann. Die Konzentration des Katalysators in dem Reaktionssystem kann üblicherweise etwa 0,5 bis 10% (Gew./ Vol.), vorzugsweise etwa 1 bis 5% (Gew./Vol.), betragen. Die 65 Umsetzung geht auch ohne Anwesenheit von Lösungsmittel vor sich, sie wird jedoch vorzugsweise in Gegenwart von Lösungsmittel durchgeführt. Beispiele für Lösungsmittel, die angewendet werden können, sind Wasser, Methanol, Äthanol,

Isopropanol und ähnliche aliphatische Alkohole, Essigsäure, Propionsäure und ähnliche niedrige aliphatische organische Säuren, Diäthyläther, Diisopropyläther, Methylcellosolve, Äthylcellosolve und ähnliche aliphatische Äther, Tetrahydro-furan, Dioxan und ähnliche cyclische Äther, Methylchlorid, Chloroform, Tetrachlorkohlenstoff und ähnliche Kohlenwasserstoffhalogenide. Die Umsetzungstemperatur kann etwa 0 bis 150°C, vorzugsweise etwa 50 bis 100°C, betragen.
Die nach dem erfindungsgemässen Verfahren erhaltene Verbindung der Formel (III) kann leicht abgetrennt und in üblicherweise isoliert werden, beispielsweise, indem das Reaktionsprodukt einer Vakuumdestillation oder der Säulenchromatographie unterworfen wird oder durch Eindampfen des Produkts und Reinigung des Konzentrats durch Sublimation. Beispiele für so erhaltene 3-Hydroxy-4H-pyran-4-on-De-rivate sind u.a.:

Im neuen erfindungsgemässen Verfahren wird ein Ausgangsmaterial eingesetzt, das nicht in der Natur vorkommt, sondern ein 2H-Pyran-3-on-Derivat ist, das leicht aus Furfural hergestellt werden kann. Es liefert das gewünschte Produkt durch eine kurze, zweistufige Umsetzung, die unter milden Bedingungen vor sich geht, keine gefahrlichen Substanzen erfordert und die Möglichkeit der Umweltverschmutzung vermindert. Damit liefert das erfindungsgemässe Verfahren das gewünschte Produkt mit sehr niedrigen Kosten, da es hohe Ausbeuten liefert und die Zahl der erforderlichen Reaktionsstufen vermindert.
Im folgenden werden anhand von Beispielen bevorzugte Ausführungsformen der Erfindung näher erläutert. Angaben in «Prozent» sind Gewichtsangaben.
In 5 Synthesen werden vorerst die Herstellung der 3-Oxo--Derivate der Formel (I) beschrieben:
Synthese 1
12,8 g 2-Methyl-6-hydroxy-3-oxo-3,6-dihydro-2H-pyran und 14,8 g Ortho-Ameisensäureäthylester werden in Gegenwart von 1 g Zinkchlorid in 100 ml Benzol 120 Minuten lang unter Rückflusskühlung zum Sieden erhitzt. Nach der Umsetzung wird das Benzol abdestilliert, der Rückstand wird in 50 ml Wasser gegossen, und die Mischung wird dreimal mit je 50 ml Äther extrahiert. Die ätherische Schicht wird mit einer wässrigen Lösung von Natriumcarbonat gewaschen und dann über Magnesiumsulfat getrocknet. Nach Entfernung des Magnesiumsulfats wird der Äther abdestilliert, und der Rückstand wird bei 30 Torr destilliert, wobei man 12 g 2-Methyl-6-äth-oxy-3-oxo-3,6-dihydro-2H-pyran als gelbe Flüssigkeit bei 82 bis 85°C erhält (Ausbeute 85%).

Synthese 2
14,2 g 2-Äthyl-6-hydroxy-3-oxo-3,6-dihydro-2H-pyran und 50 g Ortho-Ameisensäuremethylester werden in Gegenwart von 10 g Magnesiumsulfat 24 Stunden lang unter Rückflusskühlung zum Sieden erhitzt. Nach der Umsetzung wird das Magnesiumsulfat abfiltriert, und das Filtrat wird im Vakuum von überschüssigem Ortho-Ameisensäuremethylester befreit. Der Rückstand wird in 50 ml Wasser gegossen, die Mischung wird mit Äther extrahiert, und die ätherische Schicht wird mit Natriumcarbonatlösung gewaschen und über Magnesiumsulfat getrocknet. Der Äther wird abdestilliert, und der Rückstand wird bei einem Druck von 12 Torr destilliert, wobei 13,4 g 2-Äthyl-6-methoxy-3-oxo-3,6-dihydro-2H-pyran als gelbe flüssige Fraktion bei 95 bis 90°C in einer Ausbeute von 86% erhalten werden.

Synthese 3
14,2 g 2-Äthyl-6-hydroxy-3-oxo-3,6-dihydro-2H-pyran und 74 g n-Butylalkohol werden in Gegenwart von 10 g Magnesiumsulfat 24 Stunden lang unter Rückflusskühlung zum Sieden erhitzt. Nach der Umsetzung wird das Magnesiumsulfat abfiltriert, und das Filtrat wird unter vermindertem Druck destilliert, um überschüssigen n-Butylalkohol zu entfernen. Der Rückstand wird bei einem Druck von 5 Torr weiter destilliert, und man erhält bei 83 bis 88°C 16,2 g 2-Äthyl-6n-butoxy-3--oxo-3,6-dihydro-2H-pyran als gelbe Flüssigkeit (Ausbeute 82,2%).

Synthese 4
12,8 g 2-Methyl-6-hydroxy-3-oxo-3,6-dihydro-2H-pyran und 25 ml Essigsäureanhydrid werden in Gegenwart von
12.5 ml Pyridin unter Rühren bei Zimmertemperatur 4 Stunden lang miteinander umgesetzt. Nach der Umsetzung wird die Mischung eingedampft, der Rückstand wird in Eiswasser gegossen, und die Mischung wird mit 100 ml Äther extrahiert. Die ätherische Schicht wird mit einer wässrigen Lösung von Natriumcarbonat gewaschen und dann über Magnesiumsulfat getrocknet. Das Magnesiumsulfat wird abfiltriert, die ätherische Schicht wird eingedampft, und der Rückstand wird im Vakuum bei 4 Torr destilliert, wobei man bei 80 bis 83°C
12.6 g 2-Methyl-6-acetoxy-3-oxo-3,6-dihydro-2H-pyran als farblose Flüssigkeit in einer Ausbeute von 80% erhält.

14,2 g 2-Äthyl-6-hydroxy-3-oxo-3,6-dihydro-2H-pyran und 9 g Pyridin werden in 100 ml Äther gelöst, zu der Lösung werden unter Rühren und Kühlen der Lösung auf eine Temperatur von etwa 5°C langsam und tropfenweise 12 g Isovaleryl-chlorid gegeben. Die Mischung wird dann 60 Minuten lang bei Zimmertemperatur reagieren gelassen. Die erhaltene Reaktionsmischung wird filtriert, um Pyridinhydrochlorid zu entfernen. Die Mutterlösung wird eingedampft, der Rückstand wird in Eiswasser gegossen, und die Mischung wird mit Natriumcarbonat neutralisiert und dann mit 100 ml Äther extrahiert. Die ätherische Schicht wird über Magnesiumsulfat getrocknet und dann eingedampft. Der Rückstand wird durch Säulenchromatographie abgetrennt, wobei ein Gemisch von Benzol und Äthylacetat (Volumenverhältnis 4 : I) als Elu-tionsflüssigkeit verwendet wird. Eindampfen des Lösungsmittels ergibt 18,6 g Rückstand, was einer Ausbeute von 82,4% entspricht.

In den Beispielen 1 bis 5 wird die Epoxydierung beschrieben:
Beispiel 1
12,8 g 2-Methyl-6-äthoxy-3-oxo-3,6-dihydro-2H-pyran, 13 g 30%ige wässrige Lösung von Wasserstoffperoxid und 100 ml Methanol werden unter Rühren auf eine Temperatur von etwa 10°C abgekühlt. Über einen Zeitraum von 30 Minuten verteilt werden tropfenweise 10 ml l%ige wässrige Lösung von Natriumhydroxyd zugegeben. Die Mischung wird dann 1 Stunde lang ausreagieren gelassen. Nach der Umsetzung wird überschüssiges Wasserstoffperoxyd durch Zugabe von Natriumthiosulfat zerstört. Die Reaktionsmischung wird dann bei vermindertem Druck eingedampft, der Rückstand wird dreimal mit je 100 ml Äther extrahiert, und die ätherische Schicht wird eingedampft. Der Rückstand wird an einer Sili-kagelsäule chromatographiert, wobei ein Gemisch von Benzol und Äthylacetat (V olumenVerhältnis 1: 3) als Elutionsflüssig-keit verwendet wird. Destillation des Lösungsmittels bei vermindertem Druck ergibt 11,6 g der entsprechenden Verbindung der Formel (II) als farblosen flüssigen Rückstand in einer Ausbeute von 88,7%.

15,6 g 2-Äthyl-6-methoxy-3-oxo-3,6-dihydro-2H-pyran, 15 g 30%iger wässriger Wasserstoffperoxydlösung und 100 ml Methanol werden unter Rühren auf eine Temperatur von etwa 10°C abgekühlt. Dann wird das gleiche Verfahren wie in Beispiel 1 angewendet, wobei ein Konzentrat erhalten wird, das bei einem Druck von 3 Torr destilliert wird, wobei bei 83 bis 88°C 14,8 g 6-Methoxy-4,5-epoxy-3-oxo-2-äthyl-tetrahydropy-ran der Formel (II) als farblose Flüssigkeit in einer Ausbeute von 86,3% erhalten werden.

24,4 g 2-Octyl-6-äthoxy-3-oxo-3,6-dihydro-2H-pyran, 36 g einer 25%igen Lösung von Peressigsäure in Essigsäure und
100 ml Methanol werden unter Rühren auf eine Temperatur von 5°C abgekühlt. Über einen Zeitraum von 30 Minuten verteilt werden tropfenweise 15 ml l%ige wässrige Natriumhydroxydlösung zu der Lösung gegeben. Die Mischung wird s dann 2 Stunden lang ausreagieren gelassen. Unter Anwendung des gleichen Verfahrens wie in Beispiel I wird ein Konzentrat erhalten, das an einer Silikagelsäule chromatographiert wird, wobei ein Gemisch von Benzol und Äthylacetat (Volumenver-hältnis 1 : 3) als Elutionsflüssigkeit verwendet wird. DestillaT io tion bei vermindertem Druck ergibt 20,1 g (77,4% Ausbeute) der entsprechenden Verbindung der Formel (II) als blassgelbe Flüssigkeit.

Beispiel 4
26,8 g 2-n-Decyl-6-methoxy-3-oxo-3,6-dihydro-2H-pyran, 17,2 g m-Chlorperbenzoesäure und 100 ml Methanol werden unter Rühren auf eine Temperatur von etwa 5°C abgekühlt. Über einen Zeitraum von 30 Minuten verteilt werden tropfenweise 5 ml 1 n wässrige Natronlauge zugesetzt. Man lässt die Mischung dann 2 Stunden lang ausreagieren. Unter Anwendung des gleichen Verfahrens wie in Beispiel 1 wird ein Konzentrat erhalten, das an einer Silikagelsäule chromatographiert wird, wobei ein Gemisch von Benzol und Äthylacetat (Volumenverhältnis 1 : 3) als Elutionsflüssigkeit verwendet wird. Destillation bei vermindertem Druck ergibt 21,7 g (76,7% Ausbeute) der entsprechenden Verbindung der Formel (II) in Form einer blassgelben Flüssigkeit.

Beispiel 5
19,8 g 2-Methyl-6-isobutyryloxy-3-oxo-3,6-dihydro-2H-py-ran, 13 g 30%ige wässrige Wasserstoffperoxydlösung und 40 100 ml Methanol werden unter Rühren auf etwa 10°C abgekühlt. Über einen Zeitraum von 30 Minuten verteilt werden tropfenweise 10 ml l%ige Natronlauge zugegeben. Man lässt die Mischung dann 1 Stunde lang ausreagieren. Unter Anwendung des gleichen Verfahrens wie in Beispiel 1 wird ein Kon-45 zentrat erhalten, das der Silikagelsäulenchromatographie unterworfen wird, wobei eine Benzoläthylacetatmischung verwendet wird. Abdestillieren des Lösungsmittels bei vermindertem Druck ergibt 15,9 g (74,5%) der entsprechenden Verbindung der Formel (II) in Form eines farblosen, flüssigen so Rückstands.

6o 15,8 g 6-Äthoxy-4,5-epoxy-3-oxo-2-methyltetrahydropyran werden in 500 ml Dioxan gelöst und 114 g 80%ige wässrige Schwefelsäure werden unter Rühren tropfenweise zugesetzt. Man erhitzt die Mischung eine Stunde lang unter Rückflusskühlung zum Sieden. Die andere Reaktionsmischung wird auf 65 Zimmertemperatur abgekühlt und mit Natriumcarbonat neutralisiert. Das ausgefallene Natriumsulfat wird abfiltriert, und das Filtrat wird bei vermindertem Druck eingedampft, wobei braune Kristalle erhalten werden. Die Kristalle werden durch

Sublimation gereinigt, und man erhält 9,6 g (76,4% Ausbeute) 2-Methyl-3-oxo-4H-pyran-4-on in Form von farblosen Kristallen.

17,2 g 6-Methoxy-4,5-epoxy-3-oxo-2-äthyltetrahydropyran werden in 500 ml Dioxan gelöst, und unter Rühren werden tropfenweise 114 g 80%ige Schwefelsäure zugesetzt. Man erhitzt die Mischung 1 Stunde lang unter Rückflusskühlung zum Sieden. Nach dem gleichen Verfahren wie in Beispiel 6 werden dann 10,5 g (Ausbeute 75,6%) 2-Äthyl-3-hydroxy-4H-py-ran-4-on in Form von farblosen Kristallen erhalten. Elementaranalyse :

Die Mischung wird dann 1 Stunde lang unter Rückflusskühlung zum Sieden erhitzt. Unter Anwendung des gleichen Verfahrens wie in Beispiel 6 werden dann 10,8 g (62,5% Ausbeute) 2-n-Butyl--3-hydroxy-4H-pyran-4-on in Form von farblosen Kristallen erhalten.

Die Mischung wird dann 1 Stunde lang unter Rückflusskühlung zum Sieden erhitzt. Unter Anwendung des gleichen Verfahrens wie in Beispiel 6 werden dann 15,3 g (78,4% Ausbeute) 2-n-Hexyl--3-hydroxy-4H-pyran-4-on in Form von farblosen Kristallen 5 erhalten.

, 28,5 g 6-Äthoxy-4,5-epoxy-3-oxo-2-n- decyltetrahydropyran werden in 500 ml Dioxan gelöst, und unter Rühren werden tropfenweise 114 g 80%ige Schwefelsäure zugegeben. Die Mischung wird dann 1 Stunde lang unter Rückflusskühlung zum Sieden erhitzt. Unter Anwendung des gleichen Verfahrens wie in Beispiel 6 werden dann 20 g (79,5% Ausbeute) 2-n-Decyl-3--hydroxy-4H-pyran-4-on in Form von farblosen Kristallen erhalten.

Beispiel II
15,8 g 6-Äthoxy-4,5-epoxy-3-oxo-2-methyltetrahydropyran werden zu 100 ml Wasser gegeben, und die Mischung wird 24 Stunden lang unter Rückflusskühlung zum Sieden erhitzt. Die erhaltene Reaktionsmischung wird dann mit Wasserdampf destilliert, wobei das gewünschte Produkt als Destillat erhalten wird. Das Destillat wird mit Dichlormethan extrahiert, und die Dichlormethanschicht wird über Magnesiumsulfat getrocknet und dann unter vermindertem Druck eingedampft, wobei 1,2 g (9,5% Ausbeute) farblose Kristalle von 2-Methyl-3-hy-droxy-4H-pyran-4-on erhalten werden.

Verfahren gemäss Patentanspruch 1, dadurch gekennzeichnet, dass R2 eine geradkettige oder verzweigte Alkylgruppe mit 1 bis 6 Kohlenstoffatomen oder eine geradkettige oder verzweigte Acylgruppe mit 1 bis 6 Kohlenstoffatomen bedeutet.3. Verfahren gemäss Patentanspruch 1, dadurch gekennzeichnet, dass die Epoxydierung in Gegenwart einer alkalischen Substanz, vorzugsweise in Gegenwart von Natriumhydroxyd, Kaliumhydroxyd, Natriumcarbonat, Kaliumcarbonat oder einem basischen Ionenaustauschharz, insbesondere in einer Menge von 0,1 bis 1 Mol pro Mol des 3-Oxo-3,6-dihy-dro-2H-pyran-Derivats durchgeführt wird.4. Verfahren gemäss einem der Patentansprüche 1 bis 3, dadurch gekennzeichnet, dass die Epoxydierung bei einer Temperatur im Bereich von -30 bis +40°C, vorzugsweise im Bereich von-20 bis +20°C, durchgeführt wird.5. Verfahren gemäss Patentanspruch 1, dadurch gekennzeichnet, dass das 4,5-Epoxy-3-oxo-tetrahydropyran-Derivat in Gegenwart eines sauren Katalysators, vorzugsweise Schwefelsäure, Benzolsulfonsäure, Paratoluolsulfonsäure oder einem sauren Ionenaustauschharz erhitzt wird.6. Verfahren gemäss Patentanspruch 1 oder 5, dadurch gekennzeichnet, dass das 4,5-Epoxy-3-oxo-tetrahydropyran-Derivat auf eine Temperatur im Bereich von 0 bis 150°C, vorzugsweise 60 bis 100°C, erhitzt wird.7. Verfahren gemäss Patentanspruch 1, dadurch gekennzeichnet, dass das 3-Oxo-3,6-dihydro-2H-pyran-Derivat in Gegenwart von 0,1 bis 1 Mol alkalischer Substanz pro Mol des Derivats epoxydiert wird und dass das 4,5-Epoxy-3-oxo-tetra-hydropyran-Derivat in Gegenwart eines sauren Katalysators erhitzt wird.8. Nach dem Verfahren gemäss Patentanspruch 1 hergestellte 3-Hydroxy-4H-pyran-4-on-Derivate.2PATENTANSPRÜCHE 1. Verfahren zur Herstellung von 3-Hydroxy-4H-pyran-4--on-Derivaten der Formel (III)0worin Rj Wasserstoff oder eine Alkylgruppe mit I bis 10 Kohlenstoffatomen bedeutet, dadurch gekennzeichnet, dass ein 3-0x0-3,6-dihydro-2H-pyran-Derivat der Formel (I)(I)R20 Rworin Rj die oben angegebene Bedeutung hat und R2 Wasserstoff, eine Alkylgruppe mit 1 bis 6 Kohlenstoffatomen oder eine Acylgruppe mit 1 bis 6 Kohlenstoffatomen bedeutet, mit einem Peroxyd epoxydiert wird und dass das so erhaltene 4,5--Epoxy-3-oxotetrahydropyrin-Derivat der Formel (II)R2o ^ Rx worin Rj und R2 die oben angegebenen Bedeutungen haben, erhitzt wird, wodurch die Verbindung der Formel (III) erhalten wird.
- 3-Hydroxy-4H-pyran-4-on-Derivate sind wertvolle Aromastoffe für Nahrungsmittel. Beispielsweise sind 2-Methyl-3--hydroxy-4H-pyran-4-on und 2-Äthyl-3-hydroxy-4H-pyran-4--on gut bekannte Aromastoffe. Daher sind die Derivate wichtig als Zusatzstoffe zur Verbesserung des Geschmacks und des Aromas von Nahrungsmitteln, und es sind zahlreiche Verfahren zur Herstellung dieser Derivate veröffentlicht, beispielsweise in den japanischen Patentschriften Nr. 110/67,6813/70, 17237/71 und 14105/72, in J. Am. Chem. Soc. 68,2557-61 (1946), 69,2908-9 (1947), 70,2321-5 (1948) und 73,5912-13 (1951), usw. Die vorgeschlagenen Verfahren sind jedoch nicht wirtschaftlich, da sie von Ausgangsstoffen ausgehen, die nicht leicht erhältlich sind und umständliche Verfahren betreffen, die niedrige Ausbeuten ergeben. Diese Verfahren nehmen ihre Zuflucht zur Methode der Behandlung von 4H-Pyran-4-on--Derivaten, die als Ausgangsmaterial bzw. als Zwischenprodukte dienen, sie umfassen eine grosse Anzahl von Reaktionsstufen und liefern grosse Mengen an Nebenprodukten, sie beruhen im wesentlichen auf dem konventionellen Konzept der Synthese. Ferner werden diese herkömmlichen Methoden beispielsweise bei sehr hohen Temperaturen im Bereich von 200 bis 600°C ausgeführt und bringen Probleme mit sich, wie beispielsweise die Vernichtung von Schwermetallen, die als Katalysatoren bei der Umsetzung verwendet wurden, sowie die Beseitigung von Fermentationsrückständen, was erhebliche Probleme für die Umweltbelastung mit sich bringt.
